Chronic administration of triclosan leads to liver fibrosis through hepcidin-ferroportin axis-mediated iron overload

Triclosan(TCS)has been manufactured as an antibacterial compound for half a century.Currently,it is widely used in various personal care products;however,its potential adverse effects raise a lot of attention.Here,we create a long-term oral administration mouse model and identify the corresponding hepatotoxicity of TCS.We discover that daily intragastric administration of 10 mg/kg TCS to mice for 12 weeks results in severe hepatic fibrosis.Further study displays that hepatic iron increased 18%,23%and 29%upon oral TCS treatment for4,8 and 12 weeks,respectively.Accompanied by hepatic iron variation,splenic and duodenal iron are increased,which indicates systemic iron disorder.Not only excessive iron accumulated in the liver,abnormal hepatic malondialdehyde,prostaglandin synthase 2 and glutathione peroxidase 4 are pointed to ferroptosis.Additional study uncovers that hepcidin expression increases 7%,10%,4%in serum and 2.4-,4.8-,and 2.3-fold on transcriptional levels upon TCS exposure for 4,8 and 12 weeks,individually.Taken together,the mice in the TCS-treated group show disordered systemic iron homeostasis via the upregulated hepatic hepcidin-ferroportin axis.Meanwhile,both hepatic iron overload(systemic level)and hepatocyte ferroptosis(cellular level)are accused of TCS-induced liver fibrosis.Ferriprox■,an iron scavenger,significantly ameliorates TCS-induced liver fibrosis.In summary,this study confirms the impact of TCS on liver fibrosis;a critical signal pathway is also displayed.The significance of the current study is to prompt us to reevaluate the“pros and cons”of TCS applications.

Recently, the physics-of-failure(PoF) method has been more and more popular in engineering to understand the failure mechanisms(FMs) of products.However, due to the lack of system modeling methods and problem-solving algorithms,the information of FMs cannot be used to evaluate system reliability.This paper presents a system reliability evaluation method with failure mechanism tree(FMT) considering physical dependency(PDEP) such as competition, trigger, acceleration, inhibition, damage accumulation, and parameter combination.And the binary decision diagram(BDD) analytical algorithm is developed to establish a system reliability model.The operation rules of ite operators for generating BDD are discussed.The flow chart of system reliability evaluation method based on FMT and BDD is proposed.The proposed method is applied in the case of an electronic controller drive unit.Results show that the method is effective to evaluate system reliability from the perspective of FM.

We conceive a model of nanosystems where Co is uniformly,randomly distributed into a CoFe_(2)O_(4)(CFO)matrix,and study the influences of composition and CFO antiferromagnetic exchange coupling(JCFO)on zero-field-cooled/field-cooled magnetization behavior and the dependencies of exchange bias field(H_(E))and coercivity(H_(C))on Co/CFO interfacial coupling(J_(Co/CFO)),composition and cooling field(H_(FC)),based on a modified Monte Carlo method.With decreasing temperature,the zero-field-cooled and field-cooled magnetization curves are overlapped and increasing initially,and then splitting at a critical temperature(T_(P)),below which the field-cooled magnetization continues increasing while the zero-field-cooled magnetization exhibits a peak and drops,indicating a magnetically frozen state.A larger J_(CFO) induces an elevated T_(P) and a reduced magnetization,while the opposite results are obtained by a higher Co occupation fraction(P_(Co)).On the other hand,H_(E) is nonmonotonic with J_(Co/CFO )and the peak decreases and shifts to a lower J_(Co/CFO)value for a larger P_(Co).H_(C) increases monotonically with increasing J_(Co/CFO).Moreover,H_(E) is inversely proportional to P_(Co) when PCo varies from 0.11 to 0.30,indicating the exchange bias is still an interfacial effect.Finally,for a nonzero J_(CFO),H_(E) may be suppressed and accompanied by the recovery of H_(C) with increasing H_(FC).Otherwise,H_(C) for zero J_(CFO) becomes high and insensitive to H_(FC).It is ascribed to the uniform contact between Co and CFO,leading to the Co magnetization reversal and the CFO magnetization precisely controlled by H_(FC).This work opens a new avenue to control the exchange bias in nanosystems through interfacial randomization.

Based on the high positioning accuracy,low cost and low-power consumption,the ultra-wide-band(UWB)is an ideal solution for indoor unmanned aerial vehicle(UAV)localization and navigation.However,the UWB signals are easy to be blocked or reflected by obstacles such as walls and furniture.A resilient tightly-coupled inertial navigation system(INS)/UWB integration is proposed and implemented for indoor UAV navigation in this paper.A factor graph optimization(FGO)method enhanced by resilient stochastic model is established to cope with the indoor challenging scenarios.To deal with the impact of UWB non-line-of-sight(NLOS)signals and noise uncertainty,the conventional neural net-works(CNNs)are introduced into the stochastic modelling to improve the resilience and reliability of the integration.Based on the status that the UWB features are limited,a‘two-phase'CNNs structure was designed and implemented:one for signal classification and the other one for measurement noise prediction.The proposed resilient FGO method is tested on flighting UAV platform under actual indoor challenging scenario.Compared to classical FGO method,the overall positioning errors can be decreased from about 0.60 m to centimeter-level under signal block and reflection scenarios.The superiority of resilient FGO which effectively verified in constrained environment is pretty important for positioning accuracy and integrity for indoor navigation task.

The Rayleigh-Taylor mixing with a premixed layer is investigated in the present study.ABHR-II turbulence model is employed to simulate the Rayleigh-Taylor mixing with different premixed layers.The implementation of the BHR-II model is described in detail and validated in canonical problems.Afterwards,the classic Rayleigh-Taylor instability(RTI)with a premixed layer is studied.Effects of density ratio,layer thickness and density profiles on the late time behaviour of classic RTI are analysed,and the results are also compared with ILES data.It turns out that the premixed layer delays the transition time of turbulent mixing.The late time behaviours of different premixed cases show a similar trend with the similarity of their distributions of turbulent kinetic energy.It is also shown that the premixed layer has little effect on the tilted angle in the canonical tilted rig case while the temporal evolution of the turbulent mixing at the early beginning is distinctly influenced by the premixed layer.

We investigate the relative importance of spring sea surface temperatures(SSTs)in different Indian Ocean(IO)domains,especially the northern and southern IO,for the development and intensity of the Asian summer monsoon.By performing unsupervised neural network analysis,the self-organizing map,we extract distinct patterns of springtime IO SST.The results show that the uniform warming(cooling)of the southern IO plays a crucial role in the warming(cooling)of both the basin-wide IO and tropical IO.The southern IO thus well represents the associations of basinwide IO and tropical IO with the Asian summer monsoon,and is instrumental in the relationship between the IO and summer monsoon.A warming in the southern IO is closely related to the weakening of large-scale meridional monsoon circulation in May and summer(June–August),including suppression of the South Asian monsoon development in May and the East Asian monsoon in summer.On the other hand,a warming in the northern IO appears to be associated with an earlier South Asian monsoon onset and a stronger East Asian monsoon.In summer,the connection of the springtime IO SST with the South Asian monsoon weakens,but that with the East Asian monsoon strengthens.Finally,a robust negative correlation is found between the warming of various IO domains and the development and intensity of the Southeast Asian monsoon.

A radiant floor cooling system(RFCS)is a high-comfort and low energy consumption system suitable for residential buildings.Radiant floor systems usually work with fresh air,and their operating performance is affected by climatic conditions.Indoor and outdoor environmental disturbances and the system’s control strategy affect the indoor thermal comfort and energy efficiency of the system.Firstly,a multi-story residential building model was established in this study.Transient system simulation program was used to study the operation dynamics of three control strategies of the RFCS based on the calibrated model.Then,the performance of the control strategies in five climate zones in China were compared using multi-criteria decision-making in combination.The results show that control strategy has a negligible effect on condensation risk,but the thermal comfort and economic performance differ for different control strategies.The adaptability of different control strategies varies in different climate zones based on the consideration of multiple factors.The performance of the direct-ground cooling source system is better in Hot summer and warm winter zone.The variable air volume control strategy scores higher in Serve cold and Temperate zones,and the hours exceeding thermal comfort account for less than 3%of the total simulation period.Therefore,it is suggested to choose the RFCS control strategy for residential buildings according to the climate zone characteristics,to increase the energy savings.Our results provide a reliable reference for implementing RFCSs in residential buildings.

The soil particles are often regarded as completely hydrophilic solids and it is assumed that the contact angle between soil particles and pore water is constant in geotechnical engineering. However, the contact angle hysteresis exists on nonideal solid surfaces. The results of evaporation tests of liquid drops on silt surfaces with different initial contact angles indicate that the contact angle decreases continuously during the evaporation process, but the wetting diameter remains constant. The soil-water characteristic curve (SWCC) experiment is carried out based on these test results. The ideal spherical particle model is established to study the soil-water characteristic curve under the constant contact angle (non-evaporation) and continuous changes in contact angle (evaporation), respectively. When the initial contact angle of soil is small, the variation of SWCC is small under both constant and continuous variation of contact angles. Thus it is reasonable to ignore the impact of contact angle on soil-water characteristic curve when the initial contact angle of soil is small. However, the SWCC is greatly affected by both constant and continuous variation of contact angles when the initial contact angle of soil is large. Under the same moisture content, matric suction measured in samples with continuous changes in contact angle is obviously higher than that of soil samples with a constant contact angle. The matric suction error caused by changes in contact angle can reach 56.1%. Many tests related to unsaturated soil mechanics involve soil interface problems in geotechnical engineering. The principle of interface chemistry in the soil should be fully considered in the design of the test instruments that are more reasonable and the test result can be much closer to the real situation. © 2020, Science Press. ●Characterization of mollisol soil DOM by untargeted metabolomics is possible.●The polarity of the extractants determines the polarity of the extracted DOM.●Land use patterns affect the biological functions and co-network interaction of DOM.Mollisol soil is a major contributor to food production.Clarification of the molecular characteristics of dissolved organic matter(DOM)will contribute to the overall understanding and management of mollisol soil.However,the complexity of DOM poses a challenge to understanding its molecular characteristics.In this study,we investigated the molecular characteristics of DOM(<1000 Da)in mollisol soils with different soil use patterns(forestland and dryland)based on untargeted metabolomics.Here,we confirmed the feasibility of untargeted metabolomics for the molecular characterization of DOM in mollisol soils.DOM in forestland is mainly derived from plant metabolites,and DOM can perform more biological functions.However,DOM in dryland has complex composition and has powerful co-occurrence network interactions due to human activities.Water has better extraction efficiency for polar DOM,while organic reagents can efficiently extract lipid-like DOM,but the polarity of the extractant has less influence on the DOM than the soil physicochemical properties.Meanwhile,14-dihydroxyzeatin screened based on metabolomics can be used as a potential indicator for corn land.Therefore,untargeted metabolomics can be an effective method to characterize the DOM molecules of mollisol soil,which provides new insights for management of mollisol soil and sustainable agricultural development.
