The effect of monosialylganglioside mix modifying the PEGylated liposomal epirubicin on the accelerated blood clearance phenomenon

PEGylated liposomes are potential candidates to improve the pharmacokinetic characteristics of encapsulated drugs, to extend their circulation half-life and facilitate their passive accumulation at tumour sites. However, PEG-modified liposomes can induce accelerated blood clearance(ABC) upon repeated administration, and the extent of ABC phenomenon on the cytotoxic drugs-containing PEGylated liposomes is related to the dose of the cytotoxic *** this study, EPI served as a model cytotoxic drug, a hydrophilic surfactant molecule,monosialylganglioside(GM1) was chosen and modified on the liposomes together with *** was shown that upon mixed modification, when GM1 contents reached 10% or 15% mol,the ABC phenomenon of the PEGylated liposomal EPI significantly reduced. We also found that GM1 played an important role in abrogating the ABC phenomenon in both the induction phase and the effectuation phase. The results suggested that GM1 incorporation unfortunately did not avoid occurrence of ABC phenomenon completely, but GM1 modification on PEGylated liposomes may provide a significant improvement in clinical practice of PEGylated liposomes. Further study must be necessary.

Deep neural networks(DNNs)have recently shown great potential in solving partial differential equations(PDEs).The success of neural network-based surrogate models is attributed to their ability to learn a rich set of solution-related ***,learning DNNs usually involves tedious training iterations to converge and requires a very large number of training data,which hinders the application of these models to complex physical *** address this problem,we propose to apply the transfer learning approach to DNN-based PDE solving *** our work,we create pairs of transfer experiments on Helmholtz and Navier-Stokes equations by constructing subtasks with different source terms and Reynolds *** also conduct a series of experiments to investigate the degree of generality of the features between different *** results demonstrate that despite differences in underlying PDE systems,the transfer methodology can lead to a significant improvement in the accuracy of the predicted solutions and achieve a maximum performance boost of 97.3%on widely used surrogate models.

Moringa oleifera(M. oleifera) have laxative effects， but their active compositions and mechanisms are not very clear thus far. To this end， we systematically explored the active components and mechanism of M. oleifera leaves in relieving constipation by using the slow transit constipation (STC) mouse model and network pharmacology. The results of animal experiments showed that M. oleifera aqueous extract (MOA) had good laxative activity， and its 70% alcohol soluble part (ASP) also showed significant laxative activity(P < 0.01). Network pharmacological prediction results suggested that L-phenylalanine (Phe) was the key compound of ASP， and it might relieve constipation through tachykinin receptor 1 (TACR1) and three kinds of adrenergic receptors， including alpha-1a (ADRA1A)， alpha-2a (ADRA2A)， and alpha-2b (ADRA2B). Further animal experiment results showed that Phe significantly promoted gastrointestinal motility. Phe may relieve STC by enhancing the release of substance P (SP) and upregulating the mRNA expression of TACR1 in the ileum. Importantly， Phe may also promote intestinal movement by downregulating ADRA2A and ADRA2B and upregulating calmodulin and the mRNA and protein expression of myosin light chain 9 (MYL9) in the ileum， thereby activating the GPCR-MLC signaling pathway. These results lay a foundation for the application of M. oleifera and Phe in constipation.

CRISPR （clustered regularly interspaced short palindromic repeats）/Cas9 system, which is a newly developed technology for targeted genome modification, has been successfully used in a number of species. In this study, we applied this technology to carry out targeted genome modification in maize. A marker gene Zmzb7 was chosen for targeting. The sgRNA-Cas9 construct was transformed into maize protoplasts, and indel （insertion and deletion） mutations could be detected. A mutant seedling with an expected albino phenotype was obtained from screening 120 seedlings generated from 10 callus events. Mutation efficiency in maize heterochromatic regions was also investigated. Twelve sites with different expression levels in maize centromeres or pericentromere regions were selected. The sgRNA- Cas9 constructs were transformed into protoplasts followed by sequencmg the transformed protoplast genomic DNA. The results show that the genes in heterochromatic regions could be targeted by the CRISPR/Cas9 system efficiently, no matter whether they are expressed or not. Meanwhile, off-target mutations were not found in the similar sites having no PAM （protospacer adjacent motif） or having more than two mismatches. Together. our results show that the CRISPR/Cas9 system is a robust and efficient tool for genome modification in both euchromatic and heterochromatic regions in maize.

针对当前多模态模型不能充分挖掘图像中非显著区域的空间关系和上下文间的语义关系,导致多模态关系推理效果不佳的问题,提出了一个基于跨模态多维关系增强的多模态模型(multi-dimensional relationship enhancement model,MRE),用于提取潜层结构下图像各要素之间的空间关系信息,并推理出视觉—语言间的语义相关性。设计了特征多样性模块用于挖掘图像中与显著区域相关的次显著区域特征,从而增强图像空间关系特征表示。同时设计了上下文引导注意模块来引导模型学习语言上下文在图像中的关系,实现跨模态关系对齐。在MSCOCO数据集上的实验表明所提模型获得了更好的性能,其中BLEU-4和CIDEr分数分别提升了0.5%和1.3%。将这种方法应用到视觉问答任务中,在VQA 2.0数据集上性能得到了0.62%的提升,证明了该方法在多模态任务方面的广泛适用性。

Objective To identify the prominent molecular signaling in acupoints and explore their roles in initiating the analgesia effect of manual acupuncture(MA).Method A three-step study was conducted,the experiment 1 was a genome-wide analysis of the tissue at acupoint Zusanli(ST 36),including 12 Wistar rats which were divided into control,control+MA1,and control+MA7 *** the experiment 2,the paw withdrawal latency(PWL),immunohistochemistry and Western blot analysis of phospho-nuclear factor kappa B(NFκB)p65(p-p65),phospho-NFκB p50(p-p50)at ST 36 were performed on rats of saline,saline+MA,and complete Freund’s adjuvant(CFA)+MA groups(n=6).In experiment 3,24 rats were divided into saline+DMSO,CFA+DMSO,CFA+DMSO+MA,and CFA+BAY 11–7082+MA groups,the PWL and immunofluorescence assay of NFκB p65 at ST 36 was ***(1)The gene:inhibitor of NFκB(Nfkbia),interleukin-1β(Il1b),interleukin-6(Il6),chemokine c-x-c motif ligand 1(Cxcl1),monocyte chemoattractant protein-1(MCP-1/Ccl2)expressions in the control+MA7 group were significantly increased(P<0.05 or P<0.01),and the expression of NFκB p65(Rela),NFκB p50(Nfkb1)were increased in the control+MA7 group(P<0.05).(2)CFA+MA groups showed increased PWL from day 1 to 7(P<0.01 ***),and the Western blot results were consistent with immunohistochemistry,the expression of NFκB p-p65 and NFκB p-p50 were significantly increased in the MA-related groups compared with control and CFA groups(P<0.05).(3)Compared with the CFA+DMSO+MA group,the PWL of the CFA+BAY 11–7082+MA group decreased significantly and continued until day 5 and 7(P<0.05 and P<0.01,respectively),and the NFκB p65 expression of CFA+BAY 11–7082+MA was significantly reduced compared with CFA+DMSO+MA(P<0.01).Conclusion Local NFκB signaling cascade in acupoint caused by MA is an important step in initiating the analgesic effect,which would provide new evidence for the initiation of MA-effect and improve the understanding of the scientific basis of acupuncture analgesia.

Neurobiological disruptions induced in brains of the rats fed with mercury contami- nated rice collected from experimental fields in Guizhou Province, China

Guizhou Province, located in the southwestern China, is an important mercury (Hg) production center. High levels of Hg were found in local environmental and biomass samples. The neurobiological disruptions induced in brains of rats fed with Hg polluted rice collected from two experimental plots in Guizhou Province were studied. The plots are close to the Hg mine of Wanshang area and a chemical plant (where Hg is used as a catalyst for the pro-duction of acetaldehyde) of Qingzhen area. Rats were fed for 30 d before toxicological end points were tested. The results showed that the accumulations of Hg in rat’s brain of Qingzhen chemical plant group and Wanshang mine group were significantly higher than those of Shanghai group (con-trol group) (P<0.01). The neural transmitters, including ace-tylcholine (Ach), acetylcholine enzyme (AchE), nitric oxide and nitricoxide synthase (NOS), in rat’s brain were signifi-cantly affected. Oxidative stress was observed in the expo-sure groups. The expression levels of the immediately early gene (IEG), c-fos and c-FOS proteins were induced by the Hg polluted rice. It was noted that the IEG (c-fos) participated in the neurotoxic process induced by the Hg polluted rice. IEG response to Hg might be dependent on interactions between neural transmitter and oxidative damage. c-fos could be used as an effective index of detecting and assessing the neurotox-icity of Hg. The results suggested that it should be alert to mental health problem in human beings eating Hg-polluted food. More efforts are necessary to protect the local ecosys-tem and human health in the Hg polluted areas such as Wangshan and Qingzhen in Guizhou Province of China.

Dion-Jacobson phase two-dimensional(DJ 2D)perovskites,recently attracting considerable interests,exhibit excellent environmental stability and structural tunability,but their solar cells still offer unsatisfactory power conversion efficiencies(PCEs).Herein,we develop DJ 2D perovskites employing formamidinium(FA+)as a ternary cation in the perovskite cages((PDA)(FA)x(MA)3-xPb4 I13,χ=0,0.15,0.3 and 0.6,PDA=1,3-propanediammonium)for highly efficient and stable perovskite solar cells(PSCs).We found that the DJ 2D perovskite with a 10%FA+fraction presents improved crystallinity,preferred vertical orientation,and longer charge carrier lifetime compared to that without FA+*** a result,the FAdoped DJ 2D PSCs exhibit a champion PCE of 14.74%with superior device *** unencapsulated devices sustain over 92%of its initial PCE after storage at a constant relative humidity(RH)of 65%for 6000 h,90%by heat at 85℃in air for 800 h,and 94%under 1-sun illumination for 5000 *** findings demonstrate that the incorporation of FA cation into the DJ 2D perovskite is a promising strategy to develop highly efficient and stable DJ 2D PSCs.
