MicroRNAs are a large new class of tiny regulatory RNAs found in nematodes, plants, insects and mammals. They have diverse expression patterns and might regulate various developmental and physiological processes by targeting mRNAs for cleavage , translational repression or un-stabilizing the target mRNA. Although several hundred microRNA genes have been identified, little is known about the regulation and pattern of their expression. Relative expression levels of micro RNAs in different tissues can provide clues about their physiological functions. The short length and uniqueness of each microRNA render many conventional tools ineffective.

As the most common internal modification of mRNA,Ne-methyladenosine(m^(6)A)and its regulators modulate gene expression and play critical roles in various biological and patholog-ical processes including *** was reported previously that m^(6)A methyltransferase(writer),methyltransferase-like 3(METTL3)adds m^(6)A in primary microRNAs(pri-miRNAs)and fa-cilitates its processing into precursor miRNAs(pre-miRNAs).However,it is unknown whether m^(6)A modification also plays a role in the maturation process of pre-miRNAs and(if so)whether such a function contributes to ***,we found that YTHDF2 is aberrantly overexpressed in acute myeloid leukemia(AML)patients,especially in relapsed patients,and plays an onco-genic role in ***,YTHDF2 promotes expression of miR-126-3p(also known as miR-126,as it is the main product of precursor miR-126(pre-miR-126)),a miRNA that was reported as an oncomiRNA in AML,through facilitating the processing of pre-miR-126 into mature ***,YTHDF2 recognizes m^(6)A modification in pre-miR-126 and recruits AGO2,a regulator of pre-miRNA processing,to promote the maturation of ***2 posi-tively and negatively correlates with miR-126 and miR-126's downstream target genes,respec-tively,in AML patients,and forced expression of miR-126 could largely rescue YTHDF2/Ythdf2 depletion-mediated suppression on AML cell growth/proliferation and leukemogenesis,indi-cating that miR-126 is a functionally important target of YTHDF2 in ***,our studies not only reveal a previously unappreciated YTHDF2/miR-126 axis in AML and highlight the ther-apeutic potential of targeting this axis for AML treatment,but also suggest that m^(6)A plays a role in pre-miRNA processing that contributes to tumorigenesis.

Dear Editor： Gao et al. published data in Nature Biotechnology （Nat Biotechnol. 2016 May 2） showing that DNA-guided genome editing using the Natronobacterium gregoryi Argonaute （NgAgo） protein targeted 47 mammalian genomic loci with a 100% success rate and an efficiency of 21.3%-41.3% at various targets. This report led us to test NgAgo＇s utility in various cells and organisms such as mouse and zebrafish for gene editing.

Exploring the cross-talk between the immune system and advanced biomaterials to treat sARs-CoV-2 infection is a promising ***,we show that ACE2-overexpressing A549 cell-derived microparticles(AO-MPs)are a potential therapeutic agent against sARs-CoV-2 *** administered AO-MPs dexterously navigate the anatomical and biological features of the lungs to enter the alveoli and are taken up by alveolar macrophages(AMs).Then,AO-MPs increase the endosomal pH but decrease the lysosomal pH in AMs,thus escorting bound sARs-CoV-2 from phago-endosomes to lysosomes for *** pH regulation is attributable to oxidized cholesterol,which is enriched in AO-MPs and translocated to endosomal membranes,thus interfering with proton pumps and impairing endosomal *** addition to promoting viral degradation,AO-MPs also inhibit the proinflammatory phenotype of AMs,leading to increased treatment efficacy in a sARs-CoV-2-infected mouse model without side *** findings highlight the potential use of AO-MPs to treat sARs-CoV-2-infected patients and showcase the feasibility of MP therapies for combatting emerging respiratory viruses in the future.

Acyl-CoA:cholesterol acyltransferase(ACAT) is an intracellular exclusive enzyme which utilizes long-chain fatty acyl-CoA and free cholesterol as substrates to form cholesteryl esters. It is a key enzyme involved in maintaining cellular cholesterol homeostasis, and plays important roles in some serious human diseases including atherosclerosis, Alzheimer’s disease(AD) as well as cancer. In mammals, two ACAT genes encoding ACAT1 and ACAT2 have been identified. In this work, we report the impact of human ACAT1 on the dynamic free cholesterol in plasma membrane(PM-FC) of neural cell line sK-N-sH. First, the Filipin staining and PM-FC determining assays indicate that the increase of the PM-FC content of neural cell line sK-N-sH is in dose and time dependent manners by the ACAT1 inhibition, while the western blot analyses display that the α-processing of amyloid precursor protein(APP) mainly on the plasma membrane can be inversely decreased. More importantly, we have observed that the decreased α-processing of APP by the ACAT1 inhibition can be conversely enhanced when depleting the increased PM-FC directly by methyl-β-cyclodextrin, meaning that the ACAT1 inhibition can impact the dynamic PM-FC which leads to the change of APP α-processing on the plasma membrane of neural cell line sK-NsH. Then, under the different levels of cellular cholesterols by inhibiting the absorption, efflux, synthesis of cholesterolsand/or increasing cholesterols by adding LDL or mevalonate in culture media, the results show that the ACAT1 inhibition can evidently impact the dynamic PM-FC. Finally, further evidences confirm that the α-processing of APP affected by the dynamic PM-FC through the ACAT1 activity change can be mimicked by the RNAi knockdown and overexpression of ACAT1 in neural cell line sK-N-sH. These data suggest that ACAT1 impacting the dynamic PM-FC may play important role in neural cells.
