Hologenomic insights into mammalian adaptations to myrmecophagy

Highly specialized myrmecophagy(ant-and termite-eating) has independently evolved multiple times in species of various mammalian orders and represents a textbook example of phenotypic evolutionary convergence. We explored the mechanisms involved in this unique dietary adaptation and convergence through multi-omic analyses, including analyses of host genomes and transcriptomes, as well as gut metagenomes, in combination with validating assays of key enzymes’ activities, in the species of three mammalian orders(anteaters, echidnas and pangolins of the orders Xenarthra, Monotremata and Pholidota, respectively) and their relatives. We demonstrate the complex and diverse interactions between hosts and their symbiotic microbiota that have provided adaptive solutions for nutritional and detoxification challenges associated with high levels of protein and lipid metabolisms, trehalose degradation, and toxic substance detoxification. Interestingly, we also reveal their spatially complementary cooperation involved in degradation of ants’ and termites’ chitin exoskeletons. This study contributes new insights into the dietary evolution of mammals and the mechanisms involved in the coordination of physiological functions by animal hosts and their gut commensals.

Landslide Hazard and Community-based Risk Reduction Effort in Karanganyar and the Surrounding Area,Central Java,Indonesia

Karanganyar and the surrounding area are situated in a dynamic volcanic arc region, where landslide frequently occurs during the rainy season. The rain-induced landslide disasters have been resulting in 65 fatalities and a substantial socioeconomical loss in last December 2007. Again, in early February 2009, 6 more people died, hundreds of people temporary evacuated and tens of houses damaged due to the rain-induced landslide. Accordingly, inter-disciplinary approach for geological, geotechnical and social investigations were undertaken with the goal for improving community resilience in the landslide vulnerable villages. Landslide hazard mapping and communitybased landslide mitigation were conducted to reduce the risk of landslides. The hazard mapping was carried out based on the susceptibility assessment with respect to the conditions of slope inclination, types and engineering properties of lithology/soil as well as the types of landuse. All of those parameters were analyzed by applying weighing and scoring system which were calculated by semi qualitative approach (Analytical Hierarchical Process). It was found that the weathered andesitic-steep slope (steeper than 30o) was identified as the highest susceptible slope for rapid landslide, whilst the gentle colluvial slope with inter-stratification of tuffaceous clay-silt was found to be the susceptible slope for creeping. Finally, a programme for landslide risk reduction and control were developed with special emphasize on community-based landslide mitigation and early warning system. It should be highlighted that the social approach needs to be properly addressed in order to guarantee the effectiveness of landslide risk reduction.

Aggrecan is the major proteoglycan in the articular cartilage. This molecule is important in the proper functioning of articular cartilage because it provides a hydrated gel structure (via its interaction with hyaluronan and link protein) that endows the cartilage with load-bearing properties. It is also crucial in chondroskeletal morphogenesis during development. Aggrecan is a multimodular molecule expressed by chondrocytes. Its core protein is composed of three globular domains (G1, G2, and G3) and a large extended region (CS) between G2 and G3 for glycosaminoglycan chain attachment. G1 comprises the amino terminus of the core protein. This domain has the same structural motif as link protein. Functionally, the G1 domain interacts with hyaluronan acid and link protein, forming stable ternary complexes in the extracellular matrix. G2 is homologous to the tandem repeats of G1 and of link protein and is involved in product processing. G3 makes up the carboxyl terminus of the core protein. It enhances glycosaminoglycan modification and product secretion. Aggrecan plays an important role in mediating chondrocyte-chondrocyte and chondrocyte-matrix interactions through its ability to bind hyaluronan.

China’s Three Level Game in the Middle East

How has China responded to regional tensions and conflicts in the Middle East?Broadly,it has done so in the form of a threelevel game,at the global level(involving powerful outside parties like the US,Russia and the Europeans),at the regional level(and specifically the main regional powers like Saudi Arabia,Iran,Israel and Turkey)and at the sub-regional level where smaller and weaker states are *** existence of this multipolar and competitive environment in which no one state or group of states is sufficiently capable to overcome another means that China’s relations with the region involve a range of sometimes contrary responses,including cooperation with these states while avoiding being caught up in their *** strategy may be summarised as‘strategic ambiguity’or‘hedging’and while it has served China well,it may be reaching the endpoint of its current utility.

What has been China’s approach to conflict management in the Middle East?The article begins with a review of Chinese responses to conflict,its wariness at intervention and the emergence of a new model of third-party intervention labelled‘quasimediation’.The article then examines three cases of conflict in the region since 2011 and China’s engagement with them:in Libya,Syria and *** quasi-mediation is not the principal form of Chinese behavior in each case,several common themes are *** include Chinese wariness at the initial protests;rhetorical appeals for political dialogue between government and opposition and national unity;deference to powerful external actors with interests in the conflict-affected state;and a willingness to pursue commercial opportunities,which advantage some actors and marginalize *** article therefore suggests that China is not currently making use of quasi-mediation in the Middle East,but that cross-regional comparative studies may reveal if it is occurring elsewhere.

Recent studies have shown that global translational reprogramming is an early activation event in pattern-triggered immunity,when plants recognize microbe-associated molecular ***,it is not fully known whether translational regulation also occurs in subsequent immune responses,such as effector-triggered immunity(ETI).In this study,we performed genome-wide ribosome profiling in Arabidopsis upon RPS2-mediated ETI activation and discovered that specific groups of genes were translationally regulated,mostly in coordination with *** genes encode enzymes involved in aromatic amino acid,phenylpropanoid,camalexin,and sphingolipid *** functional significance of these components in ETI was confirmed by genetic and biochemical *** findings provide new insights into diverse translational regulation of plant immune responses and demonstrate that translational coordination of metabolic gene expression is an important strategy for ETI.

（1,3;1,4）-β-D-Glucans in Cell Walls of the Poaceae, Lower Plants, and Fungi： A Tale of Two Linkages

（1,3;1,4）-β-D-Glucans consist of unbranched and unsubstituted chains of （1,3）- and （1,4）-β-glucosyl residues, in which the ratio of （1,4）-β-D-glucosyl residues to （1,3）-β-D-glucosyl residues appears to influence not only the physicochemical properties of the polysaccharide and therefore its functional properties in cell walls, but also its adoption by different plant species during evolution. The （1,3; 1,4）-β-D-glucans are widely distributed as non-cellulosic matrix phase polysaccharides in cell walls of the Poaceae, which evolved relatively recently and consist of the grasses and commercially important cereal species, but they are less commonly found in lower vascular plants, such as the horsetails, in algae and in fungi. The （1,3;1,4）-β-D-glucans have often been considered to be components mainly of primary cell walls, but recent observations indicate that they can also be located in secondary walls of certain tissues. Enzymes involved in the depolymerisation of （1,3;1,4）-β-D-glucans have been well characterized. In contrast, initial difficulties in purifying the enzymes responsible for （1,3;1,4）-β-D-glucan biosynthesis slowed progress in the identification of the genes that encode （1,3;1,4）-β-D-glucan synthases, but emerging comparative genomics and associated techniques have allowed at least some of the genes that contribute to （1,3;1,4）-β-D-glucan synthesis in the Poaceae to be identified. Whether similar genes and enzymes also mediate （1,3;1,4）-β-D-glucan biosynthesis in lower plants and fungi is not yet known. Here, we compare the different fine structures of （1,3;1,4）-β-D-glucans across the plant kingdom, present current information on the genes that have been implicated recently in their biosynthesis, and consider aspects of the cell biology of （1,3;1,4）-β-D-glucan biosynthesis in the Poaceae.
