Molecular regulation of calcium-sensing receptor(CasR)-mediated signaling

Calcium-sensing receptor(CasR),a family C G-protein-coupled receptor,plays a crucial role in regulating calcium homeostasis by sensing small concentration changes of extracellular Ca^(2+),Mg^(2+),amino acids(e.g.,L-Trp and L-Phe),small peptides,anions(e.g.,HCO_(3)^(-)and PO_(4)^(3-)),and ***-mediated intracellular Ca^(2+)signaling regulates a diverse set of cellular processes including gene transcription,cell proliferation,differentiation,apoptosis,muscle contraction,and neuronal *** of CasR with mutations results in diseases such as autosomal dominant hypocalcemia,familial hypocalciuric hypercalcemia,and neonatal severe *** also influences calciotropic disorders,such as osteoporosis,and noncalciotropic disorders,such as cancer,Alzheimer's disease,and pulmonary arterial *** study first reviews recent advances in biochemical and structural determination of the framework of CasR and its interaction sites with natural ligands,as well as exogenous positive allosteric modulators and negative allosteric *** establishment of the first CasR protein-protein interactome network revealed 94 novel players involved in protein processing in endoplasmic reticulum,trafficking,cell surface expression,endocytosis,degradation,and signaling *** roles of these proteins in Ca^(2+)-dependent cellular physiological processes and in CasR-dependent cellular signaling provide new insights into the molecular basis of diseases caused by CasR mutations and dysregulated CasR activity caused by its protein interactors and facilitate the design of therapeutic agents that target CasR and other family C G-protein-coupled receptors.

OBjECTIVE:To investigate the impact of Yemazhui(Herba Eupatorii Lindleyani,HEL)against lipopolysaccharide(LPs)-induced acute lung injury(ALI)and explore its underlying mechanism in ***:The chemical constituents of HEL were analyzed by ultra-high performance liquid chromatographyquadrupole time-of-flight mass spectrometry ***,HEL was found to suppress LPs-induced ALI in ***-week-old male sprague-Dawley rats were randomly divided into 6 groups:control,LPs,Dexamethasone(Dex),HEL low dose 6 g/kg(HEL-L),HEL medium dose 18 g/kg(HEL-M)and HEL high dose 54 g/kg(HEL-H)*** model rats were intratracheally injected with 3 mg/kg LPs to establish an ALI *** counts,lung wet/dry weight ratio,as well as myeloperoxidase(MPO)activity were determined followed by the detection with hematoxylin and eosin staining,enzyme linked immunosorbent assay,quantitative real time polymerase chain reaction,western blotting,immunohistochemistry,and ***,to explore the effect of HEL on ALI-mediated intestinal flora,we performed 16s rRNA sequencing analysis of intestinal ***:HEL attenuated LPs-induced inflammation in lung tissue and intestinal flora *** study indicated that HEL suppressed the lung coefficient and wet/dry weight ratio of LPs-induced ALI in rats,inhibited leukocytes exudation and MPO activity,and improved the pathological injury of lung *** addition,HEL reduced the expression of tumor necrosis factoralpha,interleukin-1beta(IL-1β)and interleukin-6(IL-6)in bronchoalveolar lavage fluid and serum,and inhibited nuclear displacement of nuclear factor kappa-B p65(NF-κBp65).And 18 g/kg HEL also reduced the expression levels of toll-like receptor 4(TLR4),myeloid differentiation factor 88,NF-κBp65,phosphorylated inhibitor kappa B alpha(phospho-IκBα),nod-like receptor family pyrin domain-containing 3 protein(NLRP3),IL-1β,and interleukin-18(IL-18)in lung tissue,and regulated intestinal flora disturban

Crystalline-amorphous M@MN_x(M=Co,Fe,Ni)encapsulated in nitrogen-doped carbon for enhanced efficient and durable hydrogen evolution reaction

Developing earth-abundant electrocatalysts for hydrogen evolution reaction(HER)is important for the sustainable energy ***,efficient and stable heterocatalysts consisting of crystalline-amorphous M@MN_(x)(M=Co,Fe,Ni)encapsulated in N-doped carbon layers supported with N-doped graphene sheets(denoted as M@MN_(x)@NC-NG)are synthesized by facile hydrothermal reaction and nitridation *** the nitriding process,metal ions in M(tzbc)_2(H_(2)O)_4(tzbc=4-(1H-1,2,4-triazol-1-yl)benzoic acid)complexes are reduced to crystalline M cores,accompanied by the formation of amorphous MN_(x)shells;the tzbc ligands are insitu carbonized to form outermost N-doped carbon(NC)layers that connect with inner MN_(x)via M-N-C motifs inherited from the complex precursors and inhibit the transition of MN_(x)from amorphous to crystalline *** Co@CoN_(x)@NC-NG catalyst exhibits excellent HER activity with small overpotentials of 45 and 64 mV at a cathode current density of 10 mA·cm^(-2)and low Tafel slopes of 40 and 85 mV·dec^(-1)in 0.5 mol·L^(-1)H_(2)sO_4 and1.0 mol·L^(-1)KOH electrolytes,*** Co@CoN_(x)@NC-NG retains 97%of the initial overpotential after 100,000 s in both acidic and alkaline *** outstanding HER performance originates from the crystalline-amorphous Co@CoN_(x)that redistributes electrons around the heterointerfaces,facilitating the conversion process of H^(+)/H_(2)O to hydrogen and thereby promoting HER *** outermost NC layers serve as the armor of Co@CoN_(x),and graphene nanosheets act as carriers of egg-like Co@CoN_(x)@NC and conduction paths for electron shuttles,ensuring stable and continuous electrocatalytic hydrogen production.

The role of exosomes in adult neurogenesis:implications for neurodegenerative diseases

Exosomes are cup-shaped extracellular vesicles with a lipid bilayer that is approximately 30 to 200 nm in *** are widely distributed in a range of body fluids,including urine,blood,milk,and *** exert biological function by transporting factors between different cells and by regulating biological pathways in recipient *** an important form of intercellular communication,exosomes are increasingly being investigated due to their ability to transfer bioactive molecules such as lipids,proteins,mRNAs,and microRNAs between cells,and because they can regulate physiological and pathological processes in the central nervous *** neurogenesis is a multistage process by which new neurons are generated and migrate to be integrated into existing neuronal *** the adult brain,neurogenesis is mainly localized in two specialized niches:the subventricular zone adjacent to the lateral ventricles and the subgranular zone of the dentate *** increasing body of evidence indicates that adult neurogenesis is tightly controlled by environmental conditions with the *** recent studies,exosomes released from different sources of cells were shown to play an active role in regulating neurogenesis both in vitro and in vivo,thereby participating in the progression of neurodegenerative disorders in patients and in various disease ***,we provide a state-of-the-art synopsis of existing research that aimed to identify the diverse components of exosome cargoes and elucidate the therapeutic potential of exosomal contents in the regulation of neurogenesis in several neurodegenerative *** emphasize that exosomal cargoes could serve as a potential biomarker to monitor functional neurogenesis in *** addition,exosomes can also be considered as a novel therapeutic approach to treat various neurodegenerative disorders by improving endogenous neurogenesis to mitigate neuronal loss in the central nervous system.

Bioinspired supramolecular macrocycle hybrid membranes with enhanced proton conductivity

Enhancing the proton conductivity of proton exchange membranes(PEMs)is essential to expand the applications of proton exchange membrane fuel cells(PEMFCs).Inspired by the proton conduction mechanism of bacteriorhodopsin,cucurbit[n]urils(CB[n],where n is the number of glycoluril units,n=6,7,or 8)are introduced into sulfonated poly(ether ether ketone)(sPEEK)matrix to fabricate hybrid PEMs,employing a nature-inspired chemical engineering(NICE)*** carbonyl groups of CB[n]act as proton-conducting sites,while the host–guest interaction between CB[n]and water molecules offers extra protonconducting ***,the molecular size of CB[n]aids in their dispersion within the sPEEK matrix,effectively bridging the unconnected proton-conducting sulfonic group domains within the sPEEK ***,all hybrid membranes exhibit significantly enhanced proton ***,the sPEEK membrane incorporating 1 wt.%CB[8](CB[8]/sPEEK-1%)demonstrates the highest proton conductivity of 198.0 ms·cm^(−1) at 60°C and 100%relative humidity(RH),which is 228%greater than that of the pure sPEEK membrane under the same ***,hybrid membranes exhibit superior fuel cell *** CB[8]/sPEEK-1%membrane achieves a maximum power density of 214 mW·cm^(−2),representing a 140%improvement over the pure sPEEK membrane(89 mW·cm^(−2))at 50°C and 100%*** findings serve as a foundation for constructing continuous proton-conducting pathways within membranes by utilizing supramolecular macrocycles as fuel cell electrolytes and in other applications.

Energy Absorption by 3D-Printed Mesh structures with a Negative Poisson’s Ratio

A negative Poisson's ratio(NPR)structure represents optimal impact-resistance with applications in various fields,including the crash box in vehicles,which absorbs impact kinetic *** crash box is designed to deform in response to impact,increasing local structural density,which enhances impact resistance *** studies have only focused on the NPR effect in the plane dimension at low-speed *** studies have considered high-speed impact loads on three-dimensional NPR *** have developed two types of Alsi10Mg alloy energy-absorbing structures with NPR using three-dimensional printing technology,and have compared our systems with a conventional hexagonal mesh *** testing involved split-Hopkinson pressure bar measurements,which showed good agreement with dynamic numerical *** subjected to an impact load,the NPR structure exhibited better impact resistance and energy absorption compared with the positive Poisson's ratio *** proposed dual-layer hexagonal structure ensures an NPR effect while exhibiting higher strength and improved stability relative to the conventional concave hexagon structure.

s-scheme heterojunction construction of Fe/BiOCl/BiVO_(4) for enhanced photocatalytic degradation of ciprofloxacin

With the growing problem of water pollution caused by antibiotics,the development of photocatalysts with high photogenerated carrier separation efficiency is crucial.A high-efficiency microsphere Fe/BiOCl/BiVO_(4)with sscheme heterojunction was synthesized by solvothermal method and its ciprofloxacin(CIP)degradation performance were investigated under visible ***,FT-IR,sEM,EDs,HRTEM and XPs results show that the photocatalytic have good crystallization,morphology and the formed a *** photocatalytic performance of Fe/BiOCl/BiVO_(4)for CIP was superior to pure BiOCl and BiVO_(4)due to the microsphere and formed heterostructure between BiOCl and BiVO_(4).The influencing factors of CIP degradation by Fe/BiOCl/BiVO_(4)were investigated,and the results showed that Fe/BiOCl/BiVO_(4)had high degradation efficiency not only at pH 5-9,but also in the presence of inorganic Cl^(-),NO_(3)^(-)and metal *** the optimal conditions,the degradation rate of CIP was up to 100%in 75 *** addition to CIP,the Fe/BiOCl/BiVO_(4)photocatalysts degraded other organic pollutants,such as tetracycline,oxytetracycline,chlortetracycline,ofloxacin,levofloxacin,and rhodamine B,by more than 92%.The main active species were photogenerated holes(h^(+))and superoxide radicals(·O_(2)^(-)).In addition,possible intermediates and toxicity of intermediates were analyzed and five potential pathways for CIP degradation were proposed.
