Rheumatoid arthritis(RA)is a chronic autoimmune disorder marked by persistent synovial inflammation and joint degradation,posing challenges in the development of effective ***,an alkaloid found in lotus leaf,has shown promising anti-inflammatory and anti-tumor effects,yet its efficacy in RA treatment remains *** study investigated the antiproliferative effects of nuciferine on the MH7A cell line,a human RA-derived fibroblast-like synoviocyte,revealing its ability to inhibit cell proliferation,promote apoptosis,induce apoptosis,and cause G1/S phase ***,nuciferine significantly reduced the migration and invasion capabilities of MH7A *** therapeutic potential of nuciferine was further evaluated in a collagen-induced arthritis(CIA)rat model,where it markedly alleviated joint swelling,synovial hyperplasia,cartilage injury,and inflammatory *** also improved collagen-induced bone erosion,decreased pro-inflammatory cytokines and serum immunoglobulins(IgG,IgG1,IgG2a),and restored the balance between T helper(Th)17 and regulatory T cells in the spleen of CIA *** results indicate that nuciferine may offer therapeutic advantages for RA by decreasing the proliferation and invasiveness of FLS cells and correcting the Th17/Treg cell imbalance in CIA rats.

High-temperature stress threatens the growth and yield of crops. Basic helix-loop-helix(bHLH) transcription factors(TFs) have been shown to play important roles in regulating high-temperature resistance in plants. However, the bHLH TFs responsible for high-temperature tolerance in cucumbers have not been identified. We used transcriptome profiling to screen the high temperature-responsive candidate bHLH TFs in cucumber. Here, we found that the expression of 75 CsbHLH genes was altered under high-temperature stress. The expression of the CsSPT gene was induced by high temperatures in TT(Thermotolerant) cucumber plants. However, the Csspt mutant plants obtained by the CRISPR-Cas9 system showed severe thermosensitive symptoms, including wilted leaves with brown margins and reduced root density and cell *** Csspt mutant plants also exhibited elevated H_(2)O_(2) levels and down-regulated photosystem-related genes under normal ***, there were high relative electrolytic leakage(REC), malondialdehyde(MDA), glutathione(GSH), and superoxide radical(O_(2)^(·-)) levels in the Csspt mutant plants, with decreased Proline content after the high-temperature treatment. Transcriptome analysis showed that the photosystem and chloroplast activities in Csspt mutant plants were extremely disrupted by the high-temperature stress compared with wildtype(WT) plants. Moreover, the plant hormone signal transduction, as well as MAPK and calcium signaling pathways were activated in Csspt mutant plants under high-temperature stress. The HSF and HSP family genes shared the same upregulated expression patterns in Csspt and WT plants under high-temperature conditions. However, most bHLH, NAC, and bZIP family genes were significantly down-regulated by heat in Csspt mutant plants. BACKGROUND Gastric cystica profunda(GCP)represents a rare condition characterized by cystic dilation of gastric glands within the mucosal and/or submucosal *** is often linked to,or may progress into,early gastric cancer(EGC).AIM To provide a comprehensive evaluation of the endoscopic features of GCP while assessing the efficacy of endoscopic treatment,thereby offering guidance for diagnosis and *** This retrospective study involved 104 patients with GCP who underwent endoscopic *** demographic and clinical data,regular patient followups were conducted to assess local *** Among the 104 patients diagnosed with GCP who underwent endoscopic resection,12.5%had a history of previous gastric *** primary site predominantly affected was the cardia(38.5%,n=40).GCP commonly exhibited intraluminal growth(99%),regular presentation(74.0%),and ulcerative mucosa(61.5%).The leading endoscopic feature was the mucosal lesion type(59.6%,n=62).The average maximum diameter was 20.9±15.3 mm,with mucosal involvement in 60.6%(n=63).Procedures lasted 73.9±57.5 min,achieving complete resection in 91.3%(n=95).Recurrence(4.8%)was managed via either surgical intervention(n=1)or through endoscopic resection(n=4).Final pathology confirmed that 59.6%of GCP cases were associated with *** analysis indicated that elderly males were more susceptible to GCP associated with ***,multivariate analysis identified lesion morphology and endoscopic features as significant risk *** analysis demonstrated no statistically significant difference in recurrence between GCP with and without EGC(P=0.72).CONCLUSION The findings suggested that endoscopic resection might serve as an effective and minimally invasive treatment for GCP with or without EGC.

Bacterial pathogens destroy the structural integrity of functional organs in fish,leading to severe challenges in the aquaculture *** D_(3)(VD_(3))prevents bacterial infections and strengthens immune system function via vitamin D receptor(VDR).However,the correlation between VD_(3)/VDR and the structural integrity of functional organs remains *** study aimed to investigate the influence of VD_(3)supplementation on histological characteristics,apoptosis,and tight junction characteristics in fish intestine during pathogen infection.A total of 540 healthy grass carp(257.24±0.63 g)were fed different levels of VD_(3)(15.2,364.3,782.5,1,167.9,1,573.8,and 1,980.1 IU/kg)for 70 ***,fish were challenged with Aeromonas hydrophila,a pathogen that causes intestinal *** present study demonstrated that optimal supplementation with VD_(3)(1)alleviated intestinal structural damage,and inhibited oxidative damage by reducing levels of oxidative stress biomarkers;(2)attenuated excessive apoptosisrelated death receptor and mitochondrial pathway processes in relation to p38 mitogen-activated protein kinase signaling(P<0.05);(3)enhanced tight junction protein expression by inhibiting myosin light chain kinase signaling(P<0.05);and(4)elevated VDR isoform expression in fish intestine(P<0.05).Overall,the results demonstrated that VD_(3)alleviates oxidative injury,apoptosis,and the destruction of tight junction protein under pathogenic infection,thereby strengthening pathogen defenses in the *** finding supports the rationale for VD_(3)intervention as an essential practice in sustainable aquaculture.

Efforts are underway to establish a permanent lunar base on the Moon. In situ lunar regolith is anticipated to be useful as a building material after sintering. However, sintering lunar regolith into a large-scale structure presents challenges. Therefore, the key to lunar construction lies in assembling multiple small-sized sintered modules into a stable, large-sized structure. This study explored the feasibility of welding the sintered HUST-1 lunar regolith simulant(HLRS) using a laser device and conducted experiments using lasers of varying power. The microstructure, mineral composition, element distribution, and shear strength of the welded joint were investigated. A few low-melting minerals were fused and vaporized during welding, leading to the generation of thermal decomposition gas. Furthermore, the welded joint exhibited numerous micro-cracks, pores, and bubbles,resulting in reduced weld shear strength. Finally, the influence of laser power on weld shear strength was investigated, revealing that the highest shear strength(15.69 N/cm) was achieved at a laser power of 1000 W. This study demonstrates the feasibility of laser welding of sintered HLRS for the first time, with potential applications in lunar base construction.

Unusual photocarrier and coherent phonon dynamics behaviors of layered PdSeunveiled by ultrafast spectroscopy of the edge surface

Layered materials exhibit different electronic and phonon properties along in-plane and out-of-plane directions; existing studies focus on their in-plane behaviors, and the influence of such anisotropies on the dynamics of photocarriers and phonons is unknown. Here, we fabricate layered PdSe2crystals with flat edge surfaces and compare the time-resolved ultrafast spectroscopies on their basal and edge surfaces. Pronounced differences in the transient reflection spectroscopies reveal the inconsistent photocarrier and phonon dynamics behaviors on the two surfaces: the slow hot carrier relaxation process is accelerated and the thermoelasticity-induced longitudinal coherent acoustic phonon oscillation completely vanishes on the edge surface, as compared with the basal surface. Theoretical analysis reveals that the inconsistent hot carrier dynamics originate from the anisotropic properties of low-energy phonons in PdSe2, and the absence of phonon oscillation on the edge surface results from the wavevector-limited sensitivity of acoustic B1umode. Moreover, polarization-dependent spectroscopies indicate the diverse optical anisotropies beyond the in-plane of PdSe2. This work provides a new method to explore unique physical properties and modulate the optical anisotropy of layered materials.
