Aim:To explore whether overexpression of the small heat shock protein HSP2. inrat cardiomyocytes protects against simulated ischemia/reperfusion(SI/R)***:Recombinant adenovirus expressing HSP2. was used to infect rat H9c2.ardiomyocytes at high efficiency,as assessed by green fluorescent protein.H9c2.ells were subjected to SI/R stress;survival was estimated through assessment oflactate dehydrogenase and cell apoptosis through caspase-3.***:Overexpression of HSP2. decreased lactate dehydrogenase release by 2..5% andcaspase-3.activity by 58.8%.Pretreatment with the protein kinase C inhibitor Ro-3.-82.0(0.1 μmol/L)for 3. min before SI/R canceled the protective effect of *** selective mitochondrial K+awP channel inhibitor 5-hydroxydecanoate(100 μmol/L)had a similar ***,the non-selective K~+ATPchannel inhibitorglibenclamide(100 μmol/L)had no significant ***:These dataindicate that the protective effect of HSP2. in vitro is primarily due to reducednecrotic and apoptotic death of cardiomyocytes,possibly via the protein kinaseC/mitochondrial K~+ATP pathway.

Aim: To explore whether overexpression of HSP2. in the myocardium could protect against ischemia/reperfusion injury in rats. Methods: Rat hearts were injected with vector, recombinant adenovirus encoding green fluorescent protein(***) or recombinant adenovirus encoding wild-type HSP2. (***2.) in the left ventricle. Four days later, hearts were removed and expression of HSP2.was measured in the left ventricle. Subsets of animals in the vector-, ***-, ***2.-treated groups were subjected to 2.-rain ischemia and 12.-rainreperfusion. Myocardial injury was evaluated by infarct size and level of serumcardiac troponin T and creatine phosphokinase. Apoptosis of cardiomyocyteswas determined by TUNEL staining. Cardiac function was evaluated by hemodyamic indexes. Results: Infarct size and serum cardiac troponin T and creatinephosphokinase levels were significantly reduced in ***2.-treated hearts compared with vector-and ***-treated hearts. The ratio of TUNEL-positivecardiomyocytes to total number of cardiomyocytes in the ***2. group wassignificantly reduced as compared with the vector and *** groups. Left ventricular end systolic pressure, and maximal rate of pressure increase (+dp/dtmax)and decrease (-dp/dtmin) values were increased significantly, while left ventricularend diastolic pressure was decreased significantly in ***2.-treated heartscompared with vector- and ***-treated hearts. Conclusion: These data indicate that the cardioprotective effects of HSP2. may contribute to the reduction ofmyocardial necrosis and apoptosis in ischemia/reperfusion injury in rats.

The Circular Electron Positron Collider(CEPC)is a large scientific project initiated and hosted by china,fostered through extensive collaboration with international *** complex comprises four accelerators:a 3. GeV Linac,a 1.1 GeV Damping Ring,a Booster capable of achieving energies up to 180 GeV,and a Collider operating at varying energy modes(Z,W,H,and tt).The Linac and Damping Ring are situated on the surface,while the subterranean Booster and Collider are housed in a 100 km circumference underground tunnel,strategically accommodating future expansion with provisions for a potential Super Proton Proton Collider(SPPC).The CEPC primarily serves as a Higgs *** its baseline design with synchrotron radiation(SR)power of 3. Using e^(+)e^(−)annihilation data corresponding to an integrated luminosity of 2.93.fb^(−1)taken at the center-of-mass energy√s=3.773.GeV with the BESIII detector,a joint amplitude analysis is performed on the decays D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η).The fit fractions of individual components are obtained,and large interferences among the dominant components of the decays D^(0)→a_(1)(12.0)π,D^(0)→π(13.0)π,D^(0)→ρ(770)ρ(770),and D^(0)→2.ππ)_(S)are observed in both *** the obtained amplitude model,the CP-even fractions of D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η)are determined to be(75.2.1.1_(stat).±1.5_(syst.))%and(68.9±1.5_(stat).±2.4_(syst.))%,*** branching fractions of D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η)are measured to be(0.688±0.010_(stat.)±0.010_(syst.))%and(0.951±0.02._(stat.)±0.02._(syst.))%,*** amplitude analysis provides an important model for the binning strategy in measuring the strong phase parameters of D^(0)→4πwhen used to determine the CKM angleγ(ϕ_(3.)via the B^(−)→DK^(−)decay.

Despite a growing neuroimaging literature on the pathophysiology of major depressive disorder(MDD),repro-ducible findings are lacking,probably reflecting mostly small sample sizes and heterogeneity in analytic *** address these issues,the Depression Imaging REsearch ConsorTium(DIRECT)was *** REST-meta-MDD project,pooling 2.2. functional brain images processed with a standardized pipeline across all participating sites,has been the first effort from *** this review,we present an overview of the moti-vations,rationale,and principal findings of the studies so far from the REST-meta-MDD *** from the first round of analyses of the pooled repository have included alterations in functional connectivity within the default mode network,in whole-brain topological properties,in dynamic features,and in functional *** well-powered exploratory observations have also provided the basis for future longitudinal hypothesis-driven *** these fruitful explorations,DIRECT has proceeded to its second stage of data sharing that seeks to examine ethnicity in brain alterations in MDD by extending the exclusive Chinese original sample to other ethnic groups through international collaborations.A state-of-the-art,surface-based preprocessing pipeline has also been introduced to improve *** images from patients with bipolar disorder and schizophrenia will be included to identify shared and unique abnormalities across diag-nosis *** addition,large-scale longitudinal studies targeting brain network alterations following antidepressant treatment,aggregation of diffusion tensor images,and the development of functional magnetic resonance imaging-guided neuromodulation approaches are *** these endeavours,we hope to accelerate the translation of functional neuroimaging findings to clinical use,such as evaluating longitudinal effects of antidepressant medications and developing individualized

遗传性耳聋是一种常见的遗传性疾病,新生儿的发病率为1‰~3.。患者终身携带致病突变,导致不可逆性耳聋,至今尚无临床可用的治疗药物。在部分遗传性耳聋动物模型中,基因治疗已被证实为潜在有效的治疗方法。国内外已开展了多项遗传性耳聋基因治疗的临床试验。其中,由东南大学附属中大医院牵头,联合国内多家医院成功开展的遗传性耳聋基因治疗临床试验,取得了显著的疗效,并积累了丰富的经验。为规范遗传性耳聋基因治疗并提供临床研究指导,全国多家医院的耳科专家讨论制定了遗传性耳聋基因治疗临床指南,内容涵盖了伦理审查、遗传性耳聋的诊断和入组标准、临床前准备、药物、疗效与安全性评估标准、术后言语康复等。本指南将为耳聋基因治疗的临床诊疗提供规范的理论和技术指导。
