Objective Renal fibrosis is the ultimate pathway of various forms of acute and chronic kidney ***,the knockout of transient receptor potential channel 6(TRPC6)has shown promise in alleviating renal ***,the regulatory impact of TRPC6 on renal fibrosis remains *** In vivo,TRPC6 knockout(TRPC6−/−)mice and age-matched 129 SvEv(WT)mice underwent unilateral renal ischemia-reperfusion(uIR)injury surgery on the left renal pedicle or sham *** and serum were collected on days 7,14,21,and 28 after *** vitro,primary tubular epithelial cells(PTECs)were isolated from TRPC6−/−and WT mice,followed by treatment with transforming growth factorβ1(TGFβ1)for 72 *** anti-fibrotic effect of TRPC6−/−and the underlying mechanisms were assessed through hematoxylin-eosin staining,Masson staining,immunostaining,qRT-PCR,and Western *** Increased TRPC6 expression was observed in uIR mice and PTECs treated with TGFβ***6−/−alleviated renal fibrosis by reducing the expression of fibrotic markers(Col-1,α-SMA,and vimentin),as well as decreasing the apoptosis and inflammation of PTECs during fibrotic progression both in vivo and in ***,we found that the phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)/glycogen synthase kinase 3 beta(GSK3β)signaling pathway,a pivotal player in renal fibrosis,was down-regulated following TRPC6 *** These results suggest that the ablation of TRPC6 may mitigate renal fibrosis by inhibiting the apoptosis and inflammation of PTECs through down-regulation of the PI3K/AKT/GSK3β*** TRPC6 could be a novel therapeutic strategy for preventing chronic kidney disease.

Background and objective： It has been shown that macrophages play an important role in the development of severe acute pancreatitis （SAP）, and eventually lead to multiple organ failure （MOF）. Clodronate-liposome selectively depleted macrophages. This study was to investigate the role of renal macrophage infiltration in acute renal injury in rats with SAP and to evaluate the potential of superparamagnetic iron oxide （SPIO）-enhanced magnetic resonance imaging （MRI） for diagnosis. Methods： Superparamagnetic Fe3O4 nanoparticles were prepared by chemical coprecipitation. SPIO-liposomes and SPIO-clodronate-liposomes were prepared by the thin film method. SAP models were prepared by injection of sodium taurocholate into the subcapsular space of rat pancreas. Sprague-Dawley rats were randomly divided into a control group, SAP plus SPIO-liposome （P） group, and SAP plus SPIO-clodronatecontaining liposome （T） group. Kidney injury was evaluated by T2-weighted MRI scan. The levels of serum amylase （SAM）, blood urea nitrogen （BUN）, and serum creatinine （SCr） were measured by an automated enzymatic method. Serum tumor necrosis factor-α （TNF-α） was measured by enzyme-linked immunosorbent assay （ELISA）. Pathological changes in the pancreas and kidney were observed using hematoxylin and eosin （H＆E） staining, while cell apoptosis was detected with terminal deoxynucleotidyl transferase dUTP nick end labeling （TUNEL） staining. In addition, the macrophage markers （CD68） of the renal tissue were detected with immunohistochemistry. Results： The pathological changes in the pancreas and kidneys of rats in the T group were milder than those in the P group. The MRI signal intensity of the kidneys in the P and T groups was significantly lower than that in the control group. There were significant changes in the two experimental groups （P〈0.01）. BACKGROUND Grasping the underlying mechanisms of Alzheimer's disease(AD)is still a work in progress,and existing diagnostic techniques encounter various ***,the discovery of dependable biomarkers is essential for early detection,tracking the disease's advancement,and steering treatment *** To explore the diagnostic potential of serum CXCL12,sCD22,Lp-PLA2,and their ratios in AD,aiming to enhance early detection and inform targeted treatment *** The study was conducted in Dongying people's Hospital from January 2021 to December *** included 60 AD patients(AD group)and 60 healthy people(control group).Using a prospective case-control design,the levels of CXCL12,sCD22 and Lp-PLA2 and their ratios were detected by enzyme-linked immunosorbent assay kit in the diagnosis of *** differences between the two groups were analyzed by statistical methods,and the corresponding ratio was constructed to improve the specificity and sensitivity of *** Serum CXCL12 levels were higher in the AD group(47.2±8.5 ng/mL)than the control group(32.8±5.7 ng/mL,P<0.001),while sCD22 levels were lower(14.3±2.1 ng/mL vs 18.9±3.4 ng/mL,P<0.01).Lp-PLA2 levels were also higher in the AD group(112.5±20.6 ng/mL vs 89.7±15.2 ng/mL,P<0.05).Significant differences were noted in CXCL12/sCD22(3.3 vs 1.7,P<0.001)and Lp-PLA-2/sCD22 ratios(8.0 vs 5.2,P<0.05)between the *** operating characteristic analysis confirmed high sensitivity and specificity of these markers and their ratios in distinguishing AD,with area under the curves ranging from CONCLUSION Serum CXCL12 and Lp-PLA2 levels were significantly increased,while sCD22 were significantly decreased,as well as increases in the ratios of CXCL12/sCD22 and Lp-PLA2/sCD22,are closely related to the onset of *** biomarkers and their ratios can be used as potential diagnostic indicators for AD,providing an important clinical reference for early intervention and treatment.

Photoinduced ligand-to-metal charge transfer(LMCT) has emerged as an effective strategy for synthesizing organic molecules in a sustainable manner. However, the majority of existing reports on selective C(sp^(3))–H bond functionalization via photoinduced LMCT focus on the use of late transition metals or rare-earth metals for radical additions or cross-couplings. In contrast, the utilization of photoinduced LMCT with 3d early transition metals poses a significant challenge. Herein, we describe an unprecedented approach to allylic C(sp^(3))–H addition to aldehydes, employing chromium(Cr) complexes as catalysts through visible-light-induced LMCT. By investigating the reaction pathway through various mechanistic studies, including radical trapping, kinetic isotope effect(KIE) analysis, and transient absorption spectroscopy, valuable insights have been gained. The proposed mechanism suggests the intermediacy of bromine radicals through homolysis of the Cr–Br bond. Notably, this protocol expands our understanding of the photochemical properties of earth-abundant Cr complexes.

Effects of formal credit on pastoral household expense: Evidence from the Qinghai-Xizang Plateau of China

Formal credit is critical in agricultural production,allowing more expenditure and productive input,thereby improving farmers'*** pastoral China,formal financial institutions are gradually ***,a limited understanding remains of how formal credit affects herders'household *** on a survey of 544 herders from the Qinghai-Xizang Plateau of China,this study adopted the propensity score matching approach to identify the effect of formal credit on herders'total household expenses,daily expenses,and productive *** results found that average age,grassland mortgage,and other variables significantly affected herders'participation in formal *** credit could significantly improve household expenses,especially productive expenses.A heterogeneity analysis showed that formal credit had a greater impact on the household total expense for those at higher levels of wealth;however,it significantly affected the productive expense of herders at lower wealth ***,the mediating effect indicated that formal credit could affect herders'household income,thus influencing their household ***,this study suggests that policies should improve herders'accessibility to formal credit.

Accurate prediction of Shock-Wave/Boundary Layer Interaction(SWBLI)flows has been a persistent challenge for linear eddy viscosity models.A major limitation lies in the isotropic representation of the Reynolds stress,as assumed under the Boussinesq *** studies have shown promise in improving the prediction capability for incompressible separation flows by perturbing the Reynolds-stress anisotropy ***,it remains uncertain whether this approach is effective for SWBLI flows,which involve compressibility and *** address this issue,this study systematically quantifies the structural uncertainty of the anisotropy for oblique SWBLI *** eigenspace perturbation method is applied to perturb the anisotropy tensor predicted by the Menter Shear–Stress Transport(SST)model and reveal the impacts of anisotropy on the prediction of quantities of interest,such as separation and reattachment positions,wall static pressure,skin friction,and heat *** results demonstrate the potential and reveal the challenges of eigenspace perturbation in improving the SST ***,a detailed analysis of turbulent characteristics is performed to identify the source of *** findings indicate that eigenspace perturbation primarily affects turbulent shear stress,while the prediction error of the SST model is more related to turbulent kinetic energy.

The one-bit compressed sensing problem is of fundamental importance in many areas,such as wireless communication,statistics,and so ***,the optimization of one-bit problem coustrained on the unit sphere lacks an algorithm with rigorous mathematical proof of convergence and *** this paper,an iteration algorithm is established based on difference-of-convex algorithm for the one-bit compressed sensing problem constrained on the unit sphere,with iterating formula■,where C is the convex cone generated by the one-bit measurements andη_(1)>η_(2)>1/*** new algorithm is proved to converge as long as the initial point is on the unit sphere and accords with the measurements,and the convergence to the global minimum point of the l_(1)norm is discussed.

Isotope shifts among different isotopes can be effectively addressed using narrow-linewidth lasers,facilitating laser isotope separation and achieving significant enrichment at a single *** separation of potassium isotopes,employing optical pumping and magnetic deflection,has proven to be *** further improve the enrichment of ^(40)K,we introduce 2D transverse cooling to minimize the divergence *** this modification,we demonstrate enrichment of ^(40)K,elevating it from 0.012%to 12%–20%.This represents an enrichment increase by three orders of magnitude,surpassing our previous result by one *** method is particularly well-suited for isotope enrichment of elements with extremely low abundance.

The conventional multilevel inverters(MLIs)have the disadvantages of numerous devices,incapacity of boost,unbalance for capacitor’s voltage,high complexity for control,and *** by this issue,a seven-level boost inverter(7LBI)based on a switched capacitor is presented for singlephase applications in this *** proposed 7LBI using only seven transistors can achieve seven output levels,1.5 voltage gain,and natural balance of capacitors’voltages without sensors or other auxiliary methods,which illustrates its suitability for the applications of renewable energy *** configuration of topology and operating principles are illustrated in *** natural balance of capacitors and capacitance calculations are deduced as ***,the comparative study is conducted for different types of *** results illustrate the merits of the proposed 7LBI with respect to reduced devices,lower voltage stress,and less power ***,a simulation for the proposed 7LBI with PWM modulation is realized based on the theoretical analysis;an experimental prototype is also implemented,verifying multilevel output,boost ability,natural balance for switched capacitors,and performance of transient response.
