The Citrus tristeza virus (CTV) uses 3 silencing suppressor genes, p20, p23 and p25, to resist the attacks from its Citrus hosts. Inactivating these genes is therefore obviously a potential defensive option in addition to the current control strat-egies including aphid management and the use of mild strain cross protection. In this study, we cloned partial DNA frag-ments from the three genes, and used them to construct vectors for expressing hairpin RNAs (hpRNAs). To facilitate the formation of hpRNAs, the constructs were introduced in a loop structure. Fol owing transformation of sour orange (Citrus aurantium) with these constructs, 8 p20 hpRNA (hp20) and 1 p25 hpRNA (hp25) expressing lines were obtained. The 7 hp20 transgenic lines were further characterized. Their reactions to CTV were tested fol owing inoculation with CT14A and/or TR-L514, both of which are severe strains. Results showed that 3 lines (hp20-5, hp20-6 and hp20-8) were completely resistant to TR-L514 under greenhouse conditions for no detectable viral load was found in their leaves by PCR. However, they exhibited only partial suppression of TR-L514 under screen house conditions since the virus was detected in their leaves, though 2 months later compared to non-transgenic controls. Further tests showed that hp20-5 was tolerant also to CT14A under screen house conditions. The growth of hp20-5 was much better than others including the controls that were concurrently chal enged with CT14A. These results showed that expressing p20 hpRNA was sufifcient to confer sour orange with CTV resistance/tolerance.

Clay mineral proxies have been widely used to reconstruct changes in provenance and the ***,high-altitude clay mineral indications and long-term and orbital variations have not been fully ***,we present an800-ka clay-mineral record from a lacustrine sediment core(ZB13-C2)from the Zoige Basin on the eastern Tibetan Plateau(TP).X-ray diffraction and scanning electron microscopy were used to identify the clay mineral assemblages and their micromorphological characteristics of core *** results revealed that the clay minerals in core ZB13-C2 are mainly illite,with small amounts of chlorite,kaolinite,and smectite(including irregular mixed-layers of illite-smectite(I/S)and chloritesmectite(C/S)).The clay mineral assemblages and proxies exhibit three stages of change,which are mainly controlled by the Zoige watershed range under different climate *** watershed was limited to the southeastern region with altitudes ranging from~3500 to~4200 meters above sea level(m a.s.l.),and glacial-interglacial hydraulic erosion controlled the input of bedrock debris during 800-621 ka *** expanded to higher altitudes(~4200 to~4700 m a.s.l.)due to stronger seasonal freezethaw weathering without significant glacial-interglacial variations under a cooling and drying climate during 621-100 ka ***,it expanded to its highest altitudes of~4700 to~5200 m *** of the watershed after 100 ka BP,and hydraulic erosion increased obviously under a warm climate of the interstadial MIS 3 on the *** watershed underwent two major expansions under different climatic conditions at~621 ka and 100 ka BP,with the wet/dry alternation became strong at~621 ka *** results reveal that long-term temperature changes and the wet/dry alternation at high altitudes controlled the watershed range and degree of the physical erosion,and that the indications of clay minerals should be carefully clarified before they are used as climate indicators,especi

Next generation power system needs dielectrics with increased dielectric energy ***,the low energy density of dielectrics limits their ***,an asymmetric trilayered nanocomposite,with a transition layer(TL),an insulation layer(IL),and a polarization layer(PL),is designed based on poly(vinylidene fluoride)-polymethyl methacrylate(PVDF-PMMA)matrix using KNbO_(3)(KN)and TiO_(2)(TO)as the *** morphology and defect control of the two-dimensional nano KN and nano TO fillers are realized via a hydrothermal method to increase the composite breakdown strength(E_(b))and the composite energy density(U_(e)).The asymmetric trilayered structure leads to a gradient electric field distribution,and the KN and TO nanosheets block charges transfer along z *** a result,the development path of the electrical trees is greatly curved,and E_(b) is effectively *** the Ue value of the nanocomposites reaches 17.79 J·cm^(-3) at 523 MV·m^(-1).On the basis,the composite Ue is further improved by defect control in TO *** nanocomposite KN/TO/PVDF-PMMA containing TO with less oxygen vacancy concentration(calcined at oxygen atmosphere)acquires a high Ue of 21.61 J·cm^(-3) at 548 MV·m^(-1).This study provides an idea for improving the energy storage performance by combining the design of the composite dielectric structure and the control of nanofillers’defect and morphology.

As the most economically developed metropolitan area in China’s yangtze River Delta,the rapid changing land use patterns of Suzhou-Wuxi-Changzhou(Su-Xi-Chang) metropolitan area have profound impacts on the ecosystem service value(ESV).Based on the patterns of land use change and the ESV change in Su-Xi-Chang metropolitan area from 2000 to 2020,we set up four scenarios:natural development scenario,urban development scenario,arable land protection scenario and ecological protection scenario,and simulated the impact of land use changes on the ESV in these *** results showed that:1) the area of built-up land in the Su-XiChang metropolitan area increased significantly from 2000 to 2020,and the area of other types of land *** land underwent the highest transfer-out area,and was primarily converted into built-up *** total ESV of Su-Xi-Chang metropolitan area increased initially then declined from 2000–2020,and the value of almost all individual ecosystem services decreased.2) Population density,GDP per area,night lighting intensity,and road network density can negatively impact the ESV.3) The total ESV loss under the natural development and urban development scenarios was high,and the expansion of the built-up land and the drastic shrinkage of the arable land contributed to the ESV decline under both *** total ESV under arable land protection and ecological protection scenarios increases,and therefore these scenarios are suitable for future land use optimization in *** study could provide a certain reference for land use planning and allocation,and offer guidance for the rational allocation of land resources.

Preparing antioxidant coatings to address the inherent oxidation sensitivity of carbon fiber-reinforced carbon aerogel(C/CA)composites is a feasible way to promote their application in oxidizing environments as thermal insulation ***,preparing the coatings with excellent oxidation and ablation resistance while avoiding evident damage to the C/CA substrate still remains a ***,a SiC@SiO_(2)nanowire-toughened ZrB2–SiC/SiC bilayer coating with a large thickness of 500μm was prepared on C/CA using a one-step low-temperature reaction sintering method,which simultaneously formed a sintered outer layer with even-distributed nanowires and a siliconized gradient inner *** courtesy of the synergic thermal response of the layers and the crack deflection induced by the nanowires,the resulting coating has moderate residual compressive stress of 0.08–1.22 GPa in the interface,high interfacial bonding strength of 6.02 MPa,and good fracture toughness of 4.36 MPa·m^(1/2).Benefited from the optimum components and improved structure,the coating shows excellent cyclic ablation resistance with linear ablation rates of 0.1μm/s at 1650℃for 1500 s(300 s×5 cycles)and 0.4μm/s at 1850℃for 900 s(300 s×3 cycles).The one-step preparation strategy contributes to little damage to the substrate,thus showing the well-preserved mechanical and thermal insulation properties.

Yarn-based flexible strain sensors with advantages in wearability and integrability have attracted wide ***,it is still a big challenge to achieve yarn-based strain sensors with a wide linear strain range,low hysteresis,and durability synchronously that can be used for full range detection of human body ***,a new structure,double-threaded conductive yarn with rhythmic strain distribu-tion,is reported to markedly widen the linear strain range of microcrack-based stretchable strain sensors.A new method of winding and thermally adhering hot-melt filaments on the surface of the elastic fiber is used to achieve double-threaded yarn(DTY)with rhythmic strain *** proposed strategy,the integration of heterogeneous materials,is reported to significantly reduce the mechanical hysteresis of composite *** strain distribution of the DTY during stretching causes multi-level micro-cracks in different regions of the carbon nanotube(CNT)conductive layer deposited on the surface of the ***,the sensing performance of DTY-based strain sensor can be adjusted by designing the structural *** final prepared flexible strain sensor has the advantages of a wide linear strain range(100%),great sensitivity(GF=12.43),low hysteresis,rapid response(158 ms),high repeatability(>2000 cycles at 50%strain),and hydrophobicity,*** sensor can monitor human motion repeatedly and stably well,and shows great advantages in flexible wearable devices.

Mosquitoes are of great medical significance as vectors of many deadly *** have been widely used in phylogenetic studies,but mitogenome knowledge within the family Culicidae is limited,and Culicidae phylogeny is far from *** this study,we surveyed the mitogenomes of 149 Culicidae species,including 7 newly sequenced *** analysis of 149 mosquito mitogenomes shows gene composition and order to be identical to that of an ancestral insect,and the AT bias,length variation,and codon usage are all consistent with that of other reported Dipteran *** analyses based on the DNA sequences of the 13 protein-coding genes from the 149 species robustly support the monophyly of the subfamily Anophelinae and the tribes Aedini,Culicini,Mansonini,Sabethini,and *** resolve ambiguous relationships between clades within the subfamily Culicinae,we performed topological tests and show that Aedini is a sister to Culicini and that Uranotaenini is a sister to(Mansonini)+(Toxorhynchitini+Sabethini).In addition,we estimated divergence times using a Bayesian relaxation clock based on the sequence data and 3 fossil calibration *** results show mosquitoes diverged during the Early Jurassic with massive Culicinae radiations during the Cretaceous,coincident with the emergence of angiosperms and the burst of mammals and ***,this study,which uses the largest number of Culicidae mitogenomes sequenced to date,comprehensively reveals the mitogenome characteristics and mitogenome-based phylogeny and divergence times of Culicidae,providing information for further studies on the mitogenome,phylogeny,evolution,and taxonomic revision of Culicidae.
