In vitro Cytotoxicity of TCDD on SPC-A1 Cells

Objective The toxicology of TCDD （2,3,7,8-tetrachlorodibenzo-p-dioxin） has been studied mainly with regard to the carcinogenicity of its metabolites, but its phototoxicity is not well understood. Although some studies have indicated the lethal phototoxicity of TCDD, this study was designed to investigate its effect on SPC-Al cells. Methods SPC-Al cells were cultured in 1640 medium and treated with 10 nmol/L, 0.1 μmol/L, 1 μmol/L TCDD for either 24 h or 96 h at each concentration. SPC-Al cells were co-cultured with TCDD at different concentrations. Then the cell morphology, DNA fragment electrophoresis, and cell cycle were analyzed by flow cytometry, and enzyme assays were used to observe the effect of TCDD on the morphology, growth rate, and enxyme change of SPC-A1 cells. Results With the increasing concentrations of TCDD and prolongation of culture time, the morphology of SPC-Alcells was changed from round shape to spindle, and the ability of SPC-Al cells to adhere to wall was decreased. With debris emitted around the cells, the morphologic changes included reduction in cell volume. Nuclear ehromatin condensation and PI were observed. With the increasing concentrations of TCDD, DNA ladder occurred. After treatment with TCDD, extraction of cancer cells exhibited typical DNA fragmentation, and flow eytometry analysis showed apoptosis in a dose-dependent manner. As the concentration of TCDD rose from 10 nmol/L to 1 μmol/L, the ratio of apoptotic cells increased from 10.76% to 21.82%. Conclusions TCDD has in vitro cytotoxicity on SPC-Al cells, and the cytotoxicity is positively related to its concentration and culture time. TCDD may inhibit the growth and proliferation of SPC-A lcells through the pathway of apoptosis introduction.

Experimental Study on Denitrification Using Coated Electrode of Immobilized Denitrifying Bacteria

Objective To develop a coated electrode of immobilized denitrificants and to evaluate the performance of a bioelectrochemical reactor to enhance and control denitrification, Methods Denitrifying bacteria were developed by batch incubation and immobilized with polyvinyl alcohol （PVA） on the surface of activated carbon fiber （ACF） to make a coated electrode. Then the coated electrode （cathode） and graphite electrode （anode） were transferred to the reactor to reduce nitrate. Results After acclimated to the mixtrophic and autotrophic denitrification stages, the denitrifying bacteria could use hydrogen as an electron donor to reduce nitrate, When the initial nitrate concentration was 30.2 mg NO3-N/L, the denitrification efficiency was 57.3% at an applied electric current of 15 mA and a hydraulic retention time （HRT） of 12 hours. Correspondingly, the current density was 0.083 mA / cm^2. The nitrate removal rate of the reactor was 34,4 g NO3-N / m^3,d, and the surface area loading was 1.34 g NO3-N / m^2.d. Conclusion The coated electrode may keep high quantity of blomass, thus achieving a high denitrification rate. Denitrification efficiencies are related to HRT, current density, oxidation reduction potential （ORP）, dissolved oxygen （DO）, pH value, and temperature,

Removal of Nitrogen, Phosphorus, and Organic Pollutants From Water Using Seeding Type Immobilized Microorganisms

Objective To study the possibility of removing nitrogen, phosphorus, and organic pollutants using seeding type immobilized microorganisms. Methods Lakes P and M in Wuhan were chosen as the objects to study the removal of nitrogen, phosphorus, and organic pollutants with the seeding type immobilized microorganisms. Correlations between the quantity of heterotrophic bacteria and the total nitrogen （TN）, total phosphorus （TP）, and total organic carbon （TOC） in the two lakes were studied. The dominant bacteria were detected, inoculated to the sludge and acclimated by increasing nitrogen, phosphorus and decreasing carbon source in an intermittent, time-controlled and fixed-quantity way. The bacteria were then used to prepare the seeding type immobilized microorganisms, selecting diatomite as the adsorbent cartier. The ability and influence factors of removing nitrogen, phosphorus, and organic pollutant from water samples by the seeding type immobilized microorganisms were studied. Results The coefficients of the heterotrophic bacterial quantity correlated with TOC, TP, and TN were 0.9143, 0.8229, 0.7954 in Lake P and 0.9168, 0.7187, 0.6022 in Lake M. Ten strains of dominant heterotrophic bacteria belonging to Pseudomonas, Coccus, Aeromonas, Bacillus, and Enterobateriaceae, separately, were isolated. The appropriate conditions for the seeding type immobilized microorgansims in purifying the water sample were exposure time=24 h, pH=7.0-8.0, and quantity of the immobilized microorganisms=0.75-1g/50 mL. The removal rates of TOC, TP, and TN under the above conditions were 80.2%, 81.6%, and 86.8%, respectively. Conclusion The amount of heterotrophic bacteria in the two lakes was correlated with TOC, TP, and TN. These bacteria could be acclimatized and prepared for the immobilized microorganisms which could effectively remove nitrogen, phosphorus, and mixed organic pollutants in the water sample.

Fish species diversity in the Huanghe Estuary

This paper lists 50 fish specs in the Huanghe Estuary, which belong to 9 orders, 22 families and 37 genera. The paper describes the seasonal distributions of these fish species and the succession of the important fish species during the past 50 a. The authors discuss the main factors that have resulted in the changes in fish species diversity and the fishery resources. These include the overexploitation of the fishery and the sharp reduction in the fresh water discharges from the Huanghe River.

The ability of plants to cope with frost events relies on the physiological and molecular triggered by cold *** oak,Quecus wutaishanica Mayr. and Q. acutissima Carruth.,are economically and ecologically important trees species in the family Fagaceae with a wide distribution in *** two species are also widely planted because of their outstanding performance in breeding Antheraea ***,the acclimation process varies between Q. wutaishanica and Q. *** identify metabolites responsible for these two oak trees,two acclimation regimes were imposed to branches and leaves submitted to biochemical and metabolic ***,seedling were used for simulated frosts in order to test the acclimation status effect on frost tolerance.Q. wutaishanica showed higher frost tolerance than Q. acutissima under control and low temperature,possibly due to its higher accumulation of soluble proteins and soluble sugars,also lower malondialdehyde content and higher peroxidase ***,metabolic profiles differed between the two species.Q. wutaishanica accumulated more flavonoids compounds than Q. acutissima at NT,while under low temperature stress,Q. acutissima accumulated more flavonoids compounds than Q. *** can be inferred that flavonoids play a key role in these two oak species under low temperature stress.

Based on additive manufacturing of three-dimensional structures,PEDOT realizes the implementation in the efficient production of flexible bioelectronics, the construction of intricate electrode-neural implantations, and the development of smart brain-computer-interfaces [2].

Edible insects are considered rich in protein and a variety of micronutrients and are therefore regarded as one of the potential contributors to food *** Chinese oak silkworm,Antheraea pernyi（Guérin-Méneville 1855）,has been collecting from oak trees in the wild for at least 2000 years,and Chinese farmers developed the artificial rearing of Chinese oak silkworm for silk production before the 17th ***,about 5×104 t of *** including the fifth-instar larvae,pupae and moths are consuming as delicious food in China per year.A total of 18 amino acids,16 fatty acids,13 minerals,five vitamins and seven human hormones in the human diet have been detected in larvae,pupae and adults of *** this review,we summarised the complete nutrient content of four developmental stages of this economically important *** available data suggest that Chinese oak silkworm is a nutrition-rich and healthy and sustainable food item of the *** review could provide podium to food and feed industry for framing a strategy for molecular nutritional component intake into the diets of humans and ***,this paper would contribute to advance exploring for developmental and metabolic biology of Chinese oak silkworm.

Eukaryotic initiation factor 4 A（eIF-4 A） is an essential component for protein translation in *** eIF-4 A gene（ApeIF-4 A） was isolated and characterized from Antheraea *** obtained cDNA sequence was 1435 bp long with an open reading frame（ORF） of 1266 bp encoding 421 amino *** predicted amino acid sequence shared several conserved features as found in known eIF-4 As,and revealed 74 and 78% identities with eIF-4 As of Homo sapiens and Drosophila melanogaster,***-PCR analysis showed that ApeIF-4 A was transcribed at four developmental stages and in all tissues tested,suggesting that it plays an important role in development of *** alignment suggested that eIF-4 As are highly conserved throughout evolution of eukaryote *** trees based on the amino acid and nucleotide sequences of eIF-4 A demonstrated a similar topology with the classical systematics,suggesting that it has the potential value in phylogenetic inference of eukaryotes.

Qinghuang 1 is the first modern inbred strain of Chinese oak silkworm Antheraea pernyi systematically selected from local ***,the complete mitochondrial genome of this strain was sequenced and compared to other known *** mitochondrial *** mitogenome of Qinghuang 1 is 15,573 bp in length and harbors a typical gene composition and organization for completely sequenced silkworm *** nucleotide composition of the genome is highly A+T biased,accounting for 80.22%,with a slightly negative AT skewness(-*** protein-coding genes（PCGs） are initiated by ATN codons,except for cytochrome oxidase subunits I（COI）,which is tentatively designated by the CGA *** of 13 PCGs harbor an incomplete termination codons,T or *** t RNAs have a typical clover-leaf structure of mitochondrial t RNA,except for t RNASer（AGN）,the DHU arm of which could not form a stable stem-loop *** PCGs evolved under strong purifying selection with highest evolutionary rates observed for ND5 while lowest for COI *** phylogenetic analyses confirmed the basic position of Qinghuang 1 among the modern Chinese oak silkworm *** complete annotated mitochondrial genome is available in GenBank（MN064713）.
