Gentiopicroside Alleviates Atherosclerosis by Suppressing Reactive Oxygen Species-Dependent NLRP3 Inflammasome Activation in Vascular Endothelial Cells via SIRT1/Nrf2 Pathway

Objective:To evaluate the protective effects of gentiopicroside(GPS)against reactive oxygen species(ROS)-induced NOD-like receptor family,pyrin domain containing 3(NLRP3)inflammasome activation in endothelial cells,aiming to reduce ***:Eight-week-old male ApoE-deficient mice were randomly divided into 2 groups(n=10 per group):the vehicle group and the GPS treatment *** groups were fed a high-fat diet for 16 ***(40 mg/kg per day)was administered by oral gavage to the GPS group,while the vehicle group received an equivalent volume of the vehicle *** the end of the treatment,blood and aortic tissues were collected for assessments of atherosclerosis,lipid profiles,oxidative stress,and molecular expressions related to NLRP3 inflammasome activation,ROS production,and ***,in vitro experiments on human aortic endothelial cells treated with oxidized low-density lipoprotein(ox-LDL)were conducted to evaluate the effects of GPS on NLRP3 inflammasome activation,pyroptosis,apoptosis,and ROS production,specifically examining the role of the sirtuin 1(SIRT1)/nuclear factor erythroid 2-related factor 2(Nrf2)***1 and Nrf2 inhibitors were used to confirm the pathway's ***:GPS treatment significantly reduced atherosclerotic lesions in the en face aorta(Plike protein containing a CARD,cleaved-caspase-1,and cleavedgasdermin D expressions(all P<0.01).GPS also decreased ROS production,apoptosis,and pyroptosis,with the beneficial effects being significantly reversed by SIRT1 or Nrf2 ***:GPS exe

Genomic selection for meat quality traits based on VIS/NIR spectral information

The principle of genomic selection(GS) entails estimating breeding values(BVs) by summing all the SNP polygenic effects. The visible/near-infrared spectroscopy(VIS/NIRS) wavelength and abundance values can directly reflect the concentrations of chemical substances, and the measurement of meat traits by VIS/NIRS is similar to the processing of genomic selection data by summing all ‘polygenic effects' associated with spectral feature peaks. Therefore, it is meaningful to investigate the incorporation of VIS/NIRS information into GS models to establish an efficient and low-cost breeding model. In this study, we measured 6 meat quality traits in 359Duroc×Landrace×Yorkshire pigs from Guangxi Zhuang Autonomous Region, China, and genotyped them with high-density SNP chips. According to the completeness of the information for the target population, we proposed 4breeding strategies applied to different scenarios: Ⅰ, only spectral and genotypic data exist for the target population;Ⅱ, only spectral data exist for the target population;Ⅲ, only spectral and genotypic data but with different prediction processes exist for the target population;and Ⅳ, only spectral and phenotypic data exist for the target *** 4 scenarios were used to evaluate the genomic estimated breeding value(GEBV) accuracy by increasing the VIS/NIR spectral information. In the results of the 5-fold cross-validation, the genetic algorithm showed remarkable potential for preselection of feature wavelengths. The breeding efficiency of Strategies Ⅱ, Ⅲ, and Ⅳ was superior to that of traditional GS for most traits, and the GEBV prediction accuracy was improved by 32.2, 40.8 and 15.5%, respectively on average. Among them, the prediction accuracy of Strategy Ⅱ for fat(%) even improved by 50.7% compared to traditional GS. Improving atmospheric pressure vertical correction model using Gaussian function

The Zenith Hydrostatic Delay(ZHD) is essential for high-precision Global Navigation Satellite System(GNSS) and Very Long Baseline Interferometry(VLBI) data processing. Accurate estimation of ZHD relies on in situ atmospheric pressure, which is primarily variable in the vertical direction. Current atmospheric pressure is either site-specific or has limited spatial coverage, necessitating vertical corrections for broader applicability. This study introduces a model that uses a Gaussian function for the vertical correction of atmospheric pressure when in situ meteorological observations are unavailable. Validation with the fifth-generation European Centre for Medium-Range Weather Forecasts reanalysis(ERA5) reveals an average Bias and RMS for the new model of 0.31 h Pa and 2.96 h Pa,respectively. This corresponds to improvements of 37.5% and 80.3% in terms of RMS compared to two commonly used models(T0and Tvmodels) that require in situ meteorological observations,respectively. Additional validation with radiosonde data shows an average Bias and RMS of 1.85 h Pa and 4.87 h Pa, corresponding to the improvement of 42.8% and 71.1% in RMS compared with T0and Tv models, respectively. These accuracies are sufficient for calculating ZHD to an accuracy of 1 mm by performing atmospheric pressure vertical correction. The new model can correct atmospheric pressure from meteorological stations or numerical weather forecasts to different heights of the troposphere.

Acupuncture Combined with Periocular Injection for Treatment of Hydroxychloroquine Retinopathy with Cystoid Macular Edema:A Case Report

Hydroxychloroquine(HCQ)is widely used in the treatment of autoimmune disorders such as systemic lupus erythematosus(SLE)and rheumatoid arthritis,and in recent years it has also been used in diseases such as cancer,neurodegenerative diseases,metabolic diseases,and microbial infections.(1)Due to the complex pharmacokinetic characteristics of HCQ,overdose or long-term use of the drug will lead to accumulation in the tissue of organism,and severe HCQ-related retinal toxicity may occur.A cohort study in America showes that after 10 and 15 years of HCQ use,the incidence of HCQ retinopathy will be 2.5%and 8.6%,respectively.(2)The clinical manifestations of HCQ retinopathy are various,mainly photoreceptor and retinal pigment epithelium(RPE)defects,some combined with cystoid macular edema(CME),and ireversible central vision loss.
