Input associativity underlies fear memory renewal

Synaptic associativity, a feature of Hebbian plasticity wherein coactivation of two inputs onto the same neuron produces synergistic actions on postsynaptic activity, is a primary cellular correlate of associative learning. However, whether and how synaptic associativity are implemented into context-dependent relapse of extinguished memory(i.e. fear renewal) is unknown. Here, using an auditory fear conditioning paradigm in mice, we show that fear renewal is determined by the associativity between convergent inputs from the auditory cortex(ACx) and ventral hippocampus(v HPC) onto the lateral amygdala(LA) that reactivate ensembles engaged during learning. Fear renewal enhances synaptic strengths of both ACx to LA and the previously unknown v HPC to LA monosynaptic inputs. While inactivating either of the afferents abolishes fear renewal, optogenetic activation of their input associativity in the LA recapitulates fear renewal. Thus, input associativity underlies fear memory renewal.

Carbon efficiency evaluation method for urban energy system with multiple energy complementary

Urban energy systems(UESs)play a pivotal role in the consumption of clean energy and the promotion of energy cascade utilization.In the context of the construction and operation strategy of UESs with multiple complementary energy resources,a comprehensive assessment of the energy efficiency is of paramount importance.First,a multi-dimensional evaluation system with four primary indexes of energy utilization,environmental protection,system operation,and economic efficiency and 21 secondary indexes is constructed to comprehensively portray the UES.Considering that the evaluation system may contain a large number of indexes and that there is overlapping information among them,an energy efficiency evaluation method based on data processing,dimensionality reduction,integration of combined weights,and gray correlation analysis is proposed.This method can effectively reduce the number of calculations and improve the accuracy of energy efficiency assessments.Third,a demonstration project for a UES in China is presented.The energy efficiency of each scenario is assessed using six operational scenarios.The results show that Scenario 5,in which parks operate independently and investors build shared energy-storage equipment,has the best results and is best suited for green and low-carbon development.The results of the comparative assessment methods show that the proposed method provides a good energy efficiency assessment.This study provides a reference for the optimal planning,construction,and operation of UESs with multiple energy sources.

Preweaning period is a critical window for rumen microbial regulation of average daily gain in Holstein heifer calves

Background Rumen bacterial groups can affect growth performance,such as average daily gain(ADG),feed intake,and efficiency.The study aimed to investigate the inter-relationship of rumen bacterial composition,rumen fermentation indicators,serum indicators,and growth performance of Holstein heifer calves with different ADG.Twelve calves were chosen from a trail with 60 calves and divided into higher ADG(HADG,high pre-and post-weaning ADG,n=6)and lower ADG(LADG,low pre-and post-weaning ADG,n=6)groups to investigate differences in bacterial composition and functions and host phenotype.Results During the preweaning period,the relative abundances of propionate producers,including g_norank_f_Butyricicoccaceae,g_Pyramidobacter,and g_norank_f_norank_o_Clostridia_vadin BB60_group,were higher in HADG calves(LDA>2,P<0.05).Enrichment of these bacteria resulted in increased levels of propionate,a gluconeogenic precursor,in preweaning HADG calves(adjusted P<0.05),which consequently raised serum glucose concentrations(adjusted P<0.05).In contrast,the relative abundances of rumen bacteria in post-weaning HADG calves did not exert this effect.Moreover,no significant differences were observed in rumen fermentation parameters and serum indices between the two groups.Conclusions The findings of this study revealed that the preweaning period is the window of opportunity for rumen bacteria to regulate the ADG of calves.

Protective action of aspirin and its against endothelial Nlrp3 inflammasome activation in response to LPS stimuli

OBJECTIVE Recent studies have demonstrated that the Nlrp3 inflammasome serve as a central role in the pathogenesis of cardiovascular diseases and endothelial dysfunction occurs in association with several cardiovascular risk factors.Given the demonstrated anti-inflammatory effects of aspirin,the present study was designed to test whether aspirin diminish NLRP3 inflammasome activation and prevent endothelium injury and associated coronary artery damage during LPS.METHODS Mouse carotid arterial endothelial cells(CAECs)were cultured and treated with 0.1-3 mmol·L^(-1) of aspirin in response to LPS(2μg·mL^(-1))stimuli.After 24 h,the Nlrp3 inflammasome complexes consist of varied proteins were analyzed by WB.NO and T-AOC in the supernatant was detected by ELISA.Intracellular reactive oxygen species(ROS)generation for 24 h was observed by DCF fluorescence.The mice were treated with aspirin(12.5 mg·kg^(-1) per day,62.5 mg·kg^(-1) per day,125 mg·kg^(-1) per day)and dexamethasone(0.0182 mg·kg^(-1)per day)for 7 d.The level of IL^(-1)β,IL^(-1)8 protein was detected by ELISA.RESULTS Immunofluorescence results showed the colocalization of Nlrp3 with ASC or caspase 1decrease in a concentration-dependent manner.Meanwhile,the expression of Nlrp3 and caspase 1protein was decreased with the concentration of aspirin,but no changes the expression of ASC protein.Nlrp3 protein levels in CAECs were 0.33-0.8-fold and cle-caspase 1 protein levels in CAECs were 0.48-1-fold compared to those in LPS stimulation when treated with 0.1-3 mmol·L^(-1) aspirin for 24 h(P<0.01).Aspirin significantly antagonized the effect of LPS on NO(1.22-1.91-fold that of LPS stimulation,P<0.01)and T-AOC expression(1.02-1.90-fold that of LPS stimulation,P<0.01).As the different concentration of aspirin treated,the generation of ROS was 0.51-1.10-fold that of LPS stimulation(P<0.01).In vivo data shown the level of IL^(-1)β,IL^(-1)8 protein from serum are in concordance with the level of Nlrp3 inflammasome activation.CONCLUSION We conclude that aspirin has anti-inf

A Provable Positivity-Preserving Local Discontinuous Galerkin Method for the Viscous and Resistive MHD Equations

In this paper,we construct a high-order discontinuous Galerkin(DG)method which can preserve the positivity of the density and the pressure for the viscous and resistive magnetohydrodynamics(VRMHD).To control the divergence error in the magnetic field,both the local divergence-free basis and the Godunov source term would be employed for the multi-dimensional VRMHD.Rigorous theoretical analyses are presented for one-dimensional and multi-dimensional DG schemes,respectively,showing that the scheme can maintain the positivity-preserving(PP)property under some CFL conditions when combined with the strong-stability-preserving time discretization.Then,general frameworks are established to construct the PP limiter for arbitrary order of accuracy DG schemes.Numerical tests demonstrate the effectiveness of the proposed schemes.

Theoretical screening of cooperative N-bridged dual-atom sites for efficient electrocatalytic nitrogen reduction with remolding insight

The electrocatalytic nitrogen reduction reaction(e-NRR)is a promising alternative method for the Haber–Bosch process.However,it still faces many challenges in searching for high activity,stability,and selectivity catalysts and ascertaining the catalytic mechanism with complete insight.Here,a series of graphene-based N-bridged dual-atom catalysts(M1-N-M2/NC)are systematically investigated via first-principle calculation and a high-throughput screening strategy.The result unveils that N_(2) adsorption on M1-N-M2/NC in bridge-on adsorption mode can effectively break the scaling relationship on single-atom catalysts(SACs).Moreover,V-N-Ru/NC and V-N-Os/NC are systematically screened out as promising e-NRR catalysts,with extremely low limiting potentials of-0.20 and-0.18 V,respectively.Furthermore,the adsorption site competition between*N_(2) and*H,as well as the competitive twin reactions of hydrogen evolution reaction(HER)on intermediates(N_(n)H_(m))during the e-NRR process,is systematically evaluated to form a remodeling insight for the reactions in mechanism,and the e-NRR of new proposed dual-atom catalysts(DACs)is strategically optimized for its high-efficiency performance potential via our remolding insight in e-NRR mechanism.This work provides new ideas and insights for the design and mechanism of e-NRR catalysts and an effective strategy for rapidly screening highly efficient e-NRR catalysts.

Contrastive Consistency and Attentive Complementarity for Deep Multi-View Subspace Clustering

Deep multi-view subspace clustering (DMVSC) based on self-expression has attracted increasing attention dueto its outstanding performance and nonlinear application. However, most existing methods neglect that viewprivatemeaningless information or noise may interfere with the learning of self-expression, which may lead to thedegeneration of clustering performance. In this paper, we propose a novel framework of Contrastive Consistencyand Attentive Complementarity (CCAC) for DMVsSC. CCAC aligns all the self-expressions of multiple viewsand fuses them based on their discrimination, so that it can effectively explore consistent and complementaryinformation for achieving precise clustering. Specifically, the view-specific self-expression is learned by a selfexpressionlayer embedded into the auto-encoder network for each view. To guarantee consistency across views andreduce the effect of view-private information or noise, we align all the view-specific self-expressions by contrastivelearning. The aligned self-expressions are assigned adaptive weights by channel attention mechanism according totheir discrimination. Then they are fused by convolution kernel to obtain consensus self-expression withmaximumcomplementarity ofmultiple views. Extensive experimental results on four benchmark datasets and one large-scaledataset of the CCAC method outperformother state-of-the-artmethods, demonstrating its clustering effectiveness.

Rational surface charge engineering of haloalkane dehalogenase for boosting the enzymatic performance in organic solvent solutions

Biocatalysis in organic solvents(OSs)has numerous important applications,but native enzymes in OSs often exhibit limited catalytic performance.Herein,we proposed a computation-aided surface charge engineering strategy to improve the catalytic performance of haloalkane dehalogenase DhaA in OSs based on the energetic analysis of substrate binding to the DhaA surface.Several variants with enhanced OS resistance were obtained by replacing negative charged residues on the surface with positive charged residue(Arg).Particularly,a four-substitution variant E16R/E93R/E121R/E257R exhibited the best catalytic performance(five-fold improvement in OS resistance and seven-fold half-life increase in 40%(vol)dimethylsulfoxide).As a result,the overall catalytic performance of the variant could be at least 26 times higher than the wild-type DhaA.Fluorescence spectroscopy and molecular dynamics simulation studies revealed that the residue substitution mainly enhanced OS resistance from four aspects:(a)improved the overall structural stability,(b)increased the hydrophobicity of the local microenvironment around the catalytic triad,(c)enriched the hydrophobic substrate around the enzyme molecule,and(d)lowered the contact frequency between OS molecules and the catalytic triad.Our findings validate that computationaided surface charge engineering is an effective and ingenious rational strategy for tailoring enzyme performance in OSs.

Covalent organic frameworks(COFs),a rapidly developing category of crystalline conjugated organic polymers,possess highly ordered structures,large specific surface areas,stable chemical properties,and tunable pore microenvironments.Since the first report of boroxine/boronate ester-linked COFs in 2005,COFs have rapidly gained popularity,showing important application prospects in various fields,such as sensing,catalysis,separation,and energy storage.Among them,COFs-based electrochemical(EC)sensors with upgraded analytical performance are arousing extensive interest.In this review,therefore,we summarize the basic properties and the general synthesis methods of COFs used in the field of electroanalytical chemistry,with special emphasis on their usages in the fabrication of chemical sensors,ions sensors,immunosensors,and aptasensors.Notably,the emerged COFs in the electrochemiluminescence(ECL)realm are thoroughly covered along with their preliminary applications.Additionally,final conclusions on state-of-the-art COFs are provided in terms of EC and ECL sensors,as well as challenges and prospects for extending and improving the research and applications of COFs in electroanalytical chemistry.

Cobalt doping amount determines dominant reactive species in peroxymonosulfate activation via porous carbon catalysts co-doped by cobalt and nitrogen

Switching the reaction routes in peroxymonosulfate(PMS)-based advanced oxidation processes have attracted much attention but remain challenging.Herein,a series of Co-N/C catalysts with different compositions and structures were prepared by using bimetallic zeolitic imidazolate frameworks based on ZIF-8 and ZIF-67(x Zn/Co-ZIFs).Results show that Co doping amount could mediate the transformation of the activation pathway of PMS over CoN/C.When Co doping amount was less than 10%,the constructed x Co-N/C/PMS system(x≤10%)was singlet oxygen-dominated reaction;however further increasing Co doping amount would lead to the generation and coexistence of sulfate radicals and high-valent cobalt,besides singlet oxygen.Furthermore,the nitrogen-coordinated Co(Co-NX)sites could serve as main catalytically active sites to generate singlet oxygen.While excess Co doping amount caused the formation of Co nanoparticles from which leached Co ions were responsible for the generation of sulfate radicals and high-valent cobalt.Compared to undoped N/C,Co doping could significantly enhance the catalytic performance.The 0.5%Co-N/C could achieve the optimum degradation(0.488 min^(-1))and mineralization abilities(78.4%)of sulfamethoxazole among the investigated Co-N/C catalysts,which was superior to most of previously reported catalysts.In addition,the application prospects of the two systems in different environmental scenarios(pH,inorganic anions and natural organic matter)were assessed and showed different degradation behaviors.This study provides a strategy to regulate the reactive species in PMS-based advanced oxidation process.
