The Dianchi golden-line barbel,Sinocyclocheilus grahami(Regan,1904),is one of the“Four Famous Fishes”of Yunnan Province,China.Given its economic value,this species has been artificially bred successfully since 2007,with a nationally selected breed(“S.grahami,Bayou No.1”)certified in 2018.For the future utilization of this species,its growth rate,disease resistance,and wild adaptability need to be improved,which could be achieved with the help of molecular markerassisted selection(MAS).In the current study,we constructed the first chromosome-level genome of S.grahami,assembled 48 pseudo-chromosomes,and obtained a genome size of 1.49 Gb.We also performed QTL-seq analysis of S.grahami using the highest and lowest bulks(i.e.,largest and smallest size)in both a sibling and random population.We screened two quantitative trait loci(QTLs)(Chr3,14.9-39.1 Mb and Chr17,4.1-27.4 Mb)as major growth-related locations.Several candidate genes(e.g.,map2k5,stat1,phf21a,sox6,and smad6)were also identified,with functions related to growth,such as cell differentiation,neuronal development,skeletal muscle development,chondrogenesis,and immunity.These results built a solid foundation for in-depth MAS studies on the growth traits of S.grahami.

Rational surface charge engineering of haloalkane dehalogenase for boosting the enzymatic performance in organic solvent solutions

Biocatalysis in organic solvents(OSs)has numerous important applications,but native enzymes in OSs often exhibit limited catalytic performance.Herein,we proposed a computation-aided surface charge engineering strategy to improve the catalytic performance of haloalkane dehalogenase DhaA in OSs based on the energetic analysis of substrate binding to the DhaA surface.Several variants with enhanced OS resistance were obtained by replacing negative charged residues on the surface with positive charged residue(Arg).Particularly,a four-substitution variant E16R/E93R/E121R/E257R exhibited the best catalytic performance(five-fold improvement in OS resistance and seven-fold half-life increase in 40%(vol)dimethylsulfoxide).As a result,the overall catalytic performance of the variant could be at least 26 times higher than the wild-type DhaA.Fluorescence spectroscopy and molecular dynamics simulation studies revealed that the residue substitution mainly enhanced OS resistance from four aspects:(a)improved the overall structural stability,(b)increased the hydrophobicity of the local microenvironment around the catalytic triad,(c)enriched the hydrophobic substrate around the enzyme molecule,and(d)lowered the contact frequency between OS molecules and the catalytic triad.Our findings validate that computationaided surface charge engineering is an effective and ingenious rational strategy for tailoring enzyme performance in OSs.

Clinical relevance of distolingual roots and periodontal status in mandibular first molars: a cross-sectional study employing CBCT analysis

Objectives:Distolingual root of the permanent mandibular first molar(PMFM-DLR)has been frequently reported,which may complicate the treatment of periodontitis.This study aimed to assess the morphological features of PMFM-DLR and investigate the correlation between the morphological features of PMFM-DLR and periodontal status in patients with Eastern Chinese ethnic background.Materials and methods:A total of 836 cone beam computed tomography(CBCT)images with 1497 mandibular first molars were analyzed to observe the prevalence of PMFM-DLR at the patients and tooth levels in Eastern China.Among them,complete periodontal charts were available for 69 Chinese patients with 103 teeth.Correlation and regression analyses were used to evaluate the correlation between the morphological features of DLR,bone loss,and periodontal clinical parameters,including clinical attachment loss(CAL),probing pocket depth(PPD),gingival recession(GR),and furcation involvement(FI).Results:The patient-level prevalence and tooth-level prevalence of DLR in mandibular first molars were 29.4%and 26.3%,respectively.Multiple linear regression analysis suggested that bone loss at the lingual site and CAL were negatively affected by the angle of separation between distolingual and mesial roots in the transverse section,while they were significantly influenced by age and the angle of separation between distobuccal and mesial roots in the coronal section.Conclusions:The prevalence of PMFM-DLR in Eastern China was relatively high in our cohort.The morphological features of DLR were correlated with the periodontal status of mandibular first molars.This study provides critical information on the morphological features of DLR for improved diagnosis and treatment options of mandibular molars with DLR.

UPLC-Q-TOF/MS-Based Serum Metabolomics Reveals Potential Anti-tumor Mechanism of Banxia Xiexin Decoction in Colorectal Cancer Mice

Objective: To clarify the potential mechanism of Banxia Xiexin Decoction(BXD) on colorectal cancer(CRC) from the perspective of metabolomics. Methods: Forty male C57BL/6 mice were randomly divided into normal control(NC), azoxymethane/dextran sulfate sodium(AOM/DSS) model, low-dose BXD(L-BXD), high-dose BXD(H-BXD) and mesalamine(MS) groups according to a random number table, 8 mice in each group. Colorectal cancer model was induced by AOM/DSS. BXD was administered daily at doses of 3.915(L-BXD) and 15.66 g/kg(H-BXD) by gavage for consecutive 21 days, and 100 mg/kg MS was used as positive control. Following the entire modeling cycle, colon length of mice was measured and quantity of colorectal tumors were counted. The spleen and thymus index were determined by calculating the spleen/thymus weight to body weight. Inflammatory cytokine and changes of serum metabolites were analyzed by enzyme-linked immunosorbent assay kits and ultra performance liquid chromatography-quadrupole/time-of-flight mass spectrometry(UPLC-Q/TOF-MS), respectively. Results: Notably, BXD supplementation protected against weight loss, mitigated tumor formation, and diminished histologic damage in mice treated with AOM/DSS(P<0.05 or P<0.01). Moreover, BXD suppressed expression of serum inflammatory enzymes, and improved the spleen and thymus index(P<0.05). Compared with the normal group, 102 kinds of differential metabolites were screened in the AOM/DSS group, including 48 potential biomarkers, involving 18 main metabolic pathways. Totally 18 potential biomarkers related to CRC were identified, and the anti-CRC mechanism of BXD was closely related to D-glutamine and D-glutamate metabolism, phenylalanine, tyrosine and tryptophan biosynthesis, arginine biosynthesis, nitrogen metabolism and so on. Conclusion: BXD exerts partial protective effects on AOM/DSS-induced CRC by reducing inflammation, protecting organism immunity ability, and regulating amino acid metabolism.

Social hierarchy is a general self-organizational scheme in animal societies in which each member achieves a certain status of domination or subordination when getting access to resources,with profound consequences for survival,health,reproductive success,and multiple behaviors[1,2].The prefrontal cortex(PFC)was recognized as a central regulator of neuronal circuits determining the formation of social hierarchy;brain areas upstream of the PFC convey information about social status,and downstream brain regions execute dominance behavior[3].Social deficits have been suggested to be due to an elevation of excitatory/inhibitory(E/l)balance within the mouse dorsomedial(dm)PFC[4].A compensatory increase of inhibitory cell excitability partially rescued social deficits caused by such a disturbance of E/l balance.

Centrifuge modeling of unreinforced and multi-row stabilizing piles reinforced landslides subjected to reservoir water level fluctuation

With the construction of the Three Gorges Reservoir dam,frequent reservoir landslide events have been recorded.In recent years,multi-row stabilizing piles(MRSPs)have been used to stabilize massive reservoir landslides in China.In this study,two centrifuge model tests were carried out to study the unreinforced and MRSP-reinforced slopes subjected to reservoir water level(RWL)operation,using the Taping landslide as a prototype.The results indicate that the RWL rising can provide lateral support within the submerged zone and then produce the inward seepage force,eventually strengthening the slope stability.However,a rapid RWL drawdown may induce outward seepage forces and a sudden debuttressing effect,consequently reducing the effective soil normal stress and triggering partial pre-failure within the RWL fluctuation zone.Furthermore,partial deformation and subsequent soil structure damage generate excess pore water pressures,ultimately leading to the overall failure of the reservoir landslide.This study also reveals that a rapid increase in the downslope driving force due to RWL drawdown significantly intensifies the lateral earth pressures exerted on the MRSPs.Conversely,the MRSPs possess a considerable reinforcement effect on the reservoir landslide,transforming the overall failure into a partial deformation and failure situated above and in front of the MRSPs.The mechanical transfer behavior observed in the MRSPs demonstrates a progressive alteration in relation to RWL fluctuations.As the RWL rises,the mechanical states among MRSPs exhibit a growing imbalance.The shear force transfer factor(i.***.the ratio of shear forces on pile of the n th row to that of the first row)increases significantly with the RWL drawdown.This indicates that the mechanical states among MRSPs tend toward an enhanced equilibrium.The insights gained from this study contribute to a more comprehensive understanding of the failure mechanisms of reservoir landslides and the mechanical behavior of MRSPs in reservoir banks.

Numerical Simulation of Droplet Generation in Coaxial Microchannels

In this study,numerical simulations of the pinching-off phenomena displayed by the dispersed phase in a continuous phase have been conducted using COMSOL Multiphysics(level-set method).Four flow patterns,namely“drop flow”,“jet flow”,“squeeze flow”,and“co-flow”,have been obtained for different flow velocity ratios,channel diameter ratios,density ratios,viscosity ratios,and surface tension.The flow pattern map of two-phase flow in coaxial microchannels has been obtained accordingly,and the associated droplet generation process has been critically discussed considering the related frequency,diameter,and pinch-off length.In particular,it is shown that the larger the flow velocity ratio,the smaller the diameter of generated droplets and the shorter the pinch-off length.The pinch-off length of a droplet is influenced by the channel diameter ratio and density ratio.The changes in viscosity ratio have a negligible influence on the droplet generation pinching frequency.With an increase in surface tension,the frequency of generation and pinch-off length of droplets decrease,but for small surface tension the generation diameter of droplet increases.

Tuning the membrane rejection behavior by surface wettability engineering for an effective water-in-oil emulsion separation

Membrane-based separation is a promising technology to eliminate water impurities from the oil phase.However,it remains a great challenge to separate water from highly emulsified viscous oil owing to the high stability of the water droplets in oil.Herein we report a surface wettability engineering on an alumina ceramic membrane to achieve an efficient separation of a water-in-oil(W/O)emulsion.Silanes with different carbon chain lengths and fluorinated status were introduced to endow the alumina membrane with different surface wettabilities.While all the modified membranes exhibited excellent separation of the W/O without Span 80(surfactant),the one with amphiphobic wettability and lowest surface energy failed to separate the Span 80 stabilized W/O.The presence of Span 80 reduced the interfacial tension of water droplets,making them easier to deform and penetrate the modified membrane with the lowest surface energy.It reveals that engineering proper surface wettability is the key to separating the oil and water phases.Besides,the modified membranes maintained decent separation performance and stability under long-term run separation of the emulsified W/O.

Multiphase Interfacial Regulation Based on Hierarchical Porous Molybdenum Selenide to Build Anticorrosive and Multiband Tailorable Absorbers

Electromagnetic wave(EMW)absorbing materials have an irreplaceable position in the field of military stealth as well as in the field of electromagnetic pollution control.And in order to cope with the complex electromagnetic environment,the design of multifunctional and multiband high efficiency EMW absorbers remains a tremendous challenge.In this work,we designed a three-dimensional porous structure via the salt melt synthesis strategy to optimize the impedance matching of the absorber.Also,through interfacial engineering,a molybdenum carbide transition layer was introduced between the molybdenum selenide nanoparticles and the three-dimensional porous carbon matrix to improve the absorption behavior of the absorber.The analysis indicates that the number and components of the heterogeneous interfaces have a significant impact on the EMW absorption performance of the absorber due to mechanisms such as interfacial polarization and conduction loss introduced by interfacial engineering.Wherein,the prepared MoSe_(2)/MoC/PNC composites showed excellent EMW absorption performance in C,X,and Ku bands,especially exhibiting a reflection loss of−59.09 dB and an effective absorption bandwidth of 6.96 GHz at 1.9 mm.The coordination between structure and components endows the absorber with strong absorption,broad bandwidth,thin thickness,and multi-frequency absorption characteristics.Remarkably,it can effectively reinforce the marine anticorrosion property of the epoxy resin coating on Q235 steel substrate.This study contributes to a deeper understanding of the relationship between interfacial engineering and the performance of EMW absorbers,and provides a reference for the design of multifunctional,multiband EMW absorption materials.

Classifying Chinese Medicine Constitution Using Multimodal Deep-Learning Model

Objective:To develop a multimodal deep-learning model for classifying Chinese medicine constitution,i.e.,the balanced and unbalanced constitutions,based on inspection of tongue and face images,pulse waves from palpation,and health information from a total of 540 subjects.Methods:This study data consisted of tongue and face images,pulse waves obtained by palpation,and health information,including personal information,life habits,medical history,and current symptoms,from 540 subjects(202 males and 338 females).Convolutional neural networks,recurrent neural networks,and fully connected neural networks were used to extract deep features from the data.Feature fusion and decision fusion models were constructed for the multimodal data.Results:The optimal models for tongue and face images,pulse waves and health information were ResNet18,Gate Recurrent Unit,and entity embedding,respectively.Feature fusion was superior to decision fusion.The multimodal analysis revealed that multimodal data compensated for the loss of information from a single mode,resulting in improved classification performance.Conclusions:Multimodal data fusion can supplement single model information and improve classification performance.Our research underscores the effectiveness of multimodal deep learning technology to identify body constitution for modernizing and improving the intelligent application of Chinese medicine.
