Diapause,an important strategy used by insects to avoid adverse environments,is regulated by various cell signaling pathways.The results of our previous studies demonstrated that the transforming growth factor-β(TGF-β)signaling pathway regulated pupal diapause in Helicoverpa armigera,which was accompanied by downregulation of proteins in TGF-βsignaling.However,to date the mechanism underlying this phenomenon remains unknown.Here,we cloned the E3 ubiquitin ligases gene Smurf.In vitro experiments showed that Smurf directly bound to TGF-βreceptor type I(TGFβRI)and Smad2.Overexpressing Smurf promoted ubiquitination of TGFβRI and Smad2,thereby downregulating their protein levels.Conversely,silencing of the Smurf gene suppressed ubiquitination of TGFβRI and Smad2 thereby increasing their protein levels.Results from in vivo co-immunoprecipitation assays revealed that the binding of Smurf to TGFβRI or Smad2 was stronger in diapause pupae than in nondiapause pupae.Injection of Smurf inhibitor A01 into diapause pupae markedly upregulated expression of TGFβRI and Smad2 proteins,leading to resumption of development in diapause pupae.Taken together,these findings suggested that ubiquitin ligase E3 Smurf participated in H.armigera diapause by regulating TGF-βsignaling,and thus could be playing a crucial role in insect diapause.

Effects of Chinese herbal monomers on oxidative phosphorylation and membrane potential in cerebral mitochondria isolated from hypoxia-exposed rats in vitro

Mitochondrial dysfunction is the key pathogenic mechanism of cerebral injury induced by high-altitude hypoxia. Some Chinese herbal monomers may exert anti-hypoxic effects through enhancing the efficiency of oxidative phosphorylation, in this study, effects of 10 kinds of Chinese herbal monomers on mitochondrial respiration and membrane potential of cerebral mitochondria isolated from hypoxia-exposed rats in vitro were investigated to screen anti-hypoxic drugs. Rats were exposed to a low-pressure environment of 405.35 mm Hg （54.04 kPa） for 3 days to establish high-altitude hypoxic models. Cerebral mitochondria were isolated and treated with different concentrations of Chinese herbal monomers （sinomenine, silymarin, glycyrrhizic acid, baicalin, quercetin, ginkgolide B, saffron, pipedne, ginsenoside Rgl and oxymatrine） for 5 minutes in vitro. Mitochondrial oxygen consumption and membrane potential were measured using a Clark oxygen electrode and the rhodamine 123 fluorescence analysis method, respectively. Hypoxic exposure significantly decreased the state 3 respiratory rate, respiratory control rate and mitochondrial membrane potential, and significantly increased the state 4 respiratory rate. Treatment with saffron ginsenoside Rgl and oxymatrine increased the respiratory control rate in cerebral mitochondria isolated from hypoxia-exposed rats in dose-dependent manners in vitro, while ginsenoside Rgl, piperine and oxymatrine significantly increased the mitochondrial membrane potential in cerebral mitochondria from hypoxia-exposed rats. The Chinese herbal monomers saffron, ginsenoside Rgl piperine and oxymatrine could thus improve cerebral mitochondrial disorders in oxidative phosphorylation induced by hypobaric hypoxia exposure in vitro.

Progress of MJO Prediction at CMA from Phase I to Phase II of the Sub-Seasonal to Seasonal Prediction Project

As one of the participants in the Subseasonal to Seasonal(S2S)Prediction Project,the China Meteorological Administration(CMA)has adopted several model versions to participate in the S2S Project.This study evaluates the models’capability to simulate and predict the Madden-Julian Oscillation(MJO).Three versions of the Beijing Climate Center Climate System Model(BCC-CSM)are used to conduct historical simulations and re-forecast experiments(referred to as EXP1,EXP1-M,and EXP2,respectively).In simulating MJO characteristics,the newly-developed high-resolution BCC-CSM outperforms its predecessors.In terms of MJO prediction,the useful prediction skill of the MJO index is enhanced from 15 days in EXP1 to 22 days in EXP1-M,and further to 24 days in EXP2.Within the first forecast week,the better initial condition in EXP2 largely contributes to the enhancement of MJO prediction skill.However,during forecast weeks 2–3,EXP2 shows little advantage compared with EXP1-M because the increased skill at MJO initial phases 6–7 is largely offset by the degraded skill at MJO initial phases 2–3.Particularly at initial phases 2–3,EXP1-M skillfully captures the wind field and Kelvin-wave response to MJO convection,leading to the highest prediction skill of the MJO.Our results reveal that,during the participation of the CMA models in the S2S Project,both the improved model initialization and updated model physics played positive roles in improving MJO prediction.Future efforts should focus on improving the model physics to better simulate MJO convection over the Maritime Continent and further improve MJO prediction at long lead times.

This paper studies the coordinated planning of transmission tasks in the heterogeneous space networks to enable efficient sharing of ground stations cross satellite systems.Specifically,we first formulate the coordinated planning problem into a mixed integer liner programming(MILP)problem based on time expanded graph.Then,the problem is transferred and reformulated into a consensus optimization framework which can be solved by satellite systems parallelly.With alternating direction method of multipliers(ADMM),a semi-distributed coordinated transmission task planning algorithm is proposed,in which each satellite system plans its own tasks based on local information and limited communication with the coordination center.Simulation results demonstrate that compared with the centralized and fully-distributed methods,the proposed semi-distributed coordinated method can strike a better balance among task complete rate,complexity,and the amount of information required to be exchanged.

The Mixed Inert Gas(MIG)produced by the novel Green On Board Inerting Gas Generation System(GOBIGGS)mainly consists of carbon dioxide,nitrogen and oxygen.Because of the large solubility of carbon dioxide in jet fuel compared with nitrogen,the no gas release or equilibrium model could not be employed any more.In this paper,first,a mathematical model of the ullage washing was set up to predict the variation of the oxygen concentration on ullage and in the fuel,and the gas evolution and dissolution rate were calculated by Fick's second law.Then,an experimental apparatus was constructed to verify the accuracy of the model.Finally,the numerical comparisons of ullage washing using Nitrogen Enriched Air(NEA)and MIG are presented under various flow rates and fuel loads,and the result reveals that the variation of the oxygen concentration on ullage is nearly identical whatever the inert gas is NEA or MIG.However,the variation of the oxygen concentration in the fuel is disparate,and the oxygen concentration decreases rapidly if the inert gas is MIG,especially when the fuel load is low or the flow rate of the inert gas is high.Besides,MIG could suppress the rising trend of the oxygen concentration on ullage when the aircraft ascends if the fuel tank is fully washed into an equilibrium state on ground.

Anaplastic lymphoma kinase(ALK)is the most common fusion gene involved in non-small cell lung cancer(NSCLC),and remarkable response has been achieved with the use of ALK tyrosine kinase inhibitors(ALK-TKIs).However,the clinical efficacy is highly variable.Pre-existing intratumoral heterogeneity(ITH)has been proven to contribute to the poor treatment response and the resistance to targeted therapies.In this work,we investigated whether the variant allele frequencies(VAFs)of ALK fusions can help assess ITH and predict targeted therapy efficacy.Through the application of next-generation sequencing(NGS),7.2%(326/4548)of patients were detected to be ALK positive.On the basis of the adjusted VAF(adjVAF,VAF normalization for tumor purity)of four different threshold values(adjVAF<50%,40%,30%,or 20%),the association of ALK subclonality with crizotinib efficacy was assessed.Nonetheless,no statistical association was observed between median progression-free survival(PFS)and ALK subclonality assessed by adjVAF,and a poor correlation of adjVAF with PFS was found among the 85 patients who received first-line crizotinib.Results suggest that the ALK VAF determined by hybrid capture-based NGS is probably unreliable for ITH assessment and targeted therapy efficacy prediction in NSCLC.

To enhance the thermoelectric effect of cement-based materials,conductive polyaniline(PANI)modified MnO_(2)powder was synthesized and used as a thermoelectric component in the cement composites.The nanostructured PANI was deposited on the surface of the nanorod-shapedα-MnO_(2)particle and the weight ratio of PANI to MnO_(2)was 22.3:77.7 in the composite.The synthesized PANI/MnO_(2)composite was nanostructured according to the SEM image.The test results of the thermoelectric properties proved that the PANI/MnO_(2)composite was effective as the Seebeck coefficient and electrical conductivity values of the cement composites with PANI/MnO_(2)inside were 3-4 orders of magnitude higher than those of pure cement paste and the thermal conductivity values of these cement samples were similar.The obtained maximum figure of merit(ZT)value(2.75×10^(-3))was much larger than that of conductive materials reinforced cement-based composites.The thermoelectric effect of cement composites is mainly enhanced by the increased Seebeck coefficient and electrical conductivity in this work.
