LncRNA PCGEM1 facilitates cervical cancer progression via miR-642a-5p/KIF5B axis

Attribution of Biases of Interhemispheric Temperature Contrast in CMIP6 Models

One of the basic characteristics of Earth's modern climate is that the Northern Hemisphere(NH) is climatologically warmer than the Southern Hemisphere(sh.. Here, model performances of this basic state are examined using simulation results from 26 CMIP6 models. Results sh.w that the CMIP6 models underestimate the contrast in interhemispheric surface temperatures on average(0.8 K for CMIP6 mean versus 1.4 K for reanalysis data mean), and that there is a large intermodel spread, ranging from -0.7 K to 2.3 K. A box model energy budget analysis sh.ws that the contrast in interhemispheric sh.rtwave absorption at the top of the atmosphere, the contrast in interhemispheric greenhouse trapping, and the crossequatorial northward ocean heat transport, are all underestimated in the multimodel mean. By examining the intermodel spread, we find intermodel biases can be tracked back to biases in midlatitude sh.rtwave cloud forcing in AGCMs. Models with a weaker interhemispheric temperature contrast underestimate the sh.rtwave cloud reflection in the sh.but overestimate the sh.rtwave cloud reflection in the NH, which are respectively due to underestimation of the cloud fraction over the sh.extratropical ocean and overestimation of the cloud liquid water content over the NH extratropical *** that underestimate the interhemispheric temperature contrast exhibit larger double ITCZ biases, characterized by excessive precipitation in the sh.tropics. Although this intermodel spread does not account for the multimodel ensemble mean biases, it highlights that improving cloud simulation in AGCMs is essential for simulating the climate realistically in coupled models.

以多巴胺能神经细胞sh.SY5Y和PC12为实验模型,深入探讨了不同晶型(锐钛矿型、金红石型、混合型)和不同粒径(25,50,100nm)的纳米二氧化钛(TiO_(2)-NPs)暴露对多巴胺能神经细胞的毒性影响及机制.结果表明,金红石型TiO_(2)-NPs暴露具有更强的毒性作用,其次是混合型和锐钛矿型.此外,粒径较小的TiO_(2)-NPs对神经细胞的毒性更显著,特别是25nm粒径的TiO_(2)-NPs,其神经毒性作用与诱导活性氧自由基生成和细胞凋亡相关.蛋白免疫印迹法发现,TiO_(2)-NPs能上调Bax和细胞色素C的水平,下调Bcl-2的表达,同时激活Caspase-3,表明TiO_(2)-NPs通过影响线粒体凋亡途径相关蛋白的表达诱导多巴胺能神经细胞凋亡.

Donor safety and remnant liver volume in living donor liver transplantation

荧光显微镜是研究细胞结构和动态过程的最有力的工具之一,近年来发展的超分辨显微成像技术更是突破了传统衍射极限,能够在观测活体生物样本中达到纳米级的分辨率。但由于生物样品内部折射率变化的复杂性,进行超分辨观测时成像性能往往会因样品引起的像差而降低。自适应光学是一种矫正波前误差的技术,在超分辨显微成像技术中引入自适应光学技术可以显著提高超分辨系统的成像分辨率、成像深度、成像速度。为了进一步提高超分辨显微成像系统的成像性能,研究者不断创新自适应光学在超分辨技术的应用方法。介绍了自适应光学技术,阐述了其在显微成像中的应用;详细分析了近年来自适应光学在超分辨显微成像技术的应用,最终进行了总结,并对未来技术发展方向进行了展望。

Decitabine induces IRF7-mediated immune responses in p53-mutated triple-negative breast cancer:a clinical and translational study

p53 is mutated in half of cancer ***,no p53-targeting drugs have been ***,we reposition decitabine for triple-negative breast cancer(TNBC),a subtype with frequent p53 mutations and extremely poor *** a retrospective study on tissue microarrays with 132 TNBC cases,DNMT1 overexpression was associated with p53 mutations(P=0.037)and poor overall survival(OS)(P=0.010).In a prospective DEciTabinE and Carboplatin in TNBC(DETECT)trial(NCT03295552),decitabine with carboplatin produced an objective response rate(ORR)of 42%in 12 patients with stage IV *** the 9 trialed patients with available TP53 sequencing results,the 6 patients with p53 mutations had higher ORR(3/6 vs.0/3)and better OS(16.0 vs.4.0 months)than the patients with wild-type *** a mechanistic study,isogenic TNBC cell lines harboring DETECT-derived p53 mutations exhibited higher DNMT1 expression and decitabine sensitivity than the cell line with wild-type *** the DETECT trial,decitabine induced strong immune responses featuring the striking upregulation of the innate immune player IRF7 in the p53-mutated TNBC cell line(upregulation by 16-fold)and the most responsive patient with *** integrative studies reveal the potential of repurposing decitabine for the treatment of p53-mutated TNBC and suggest IRF7 as a potential biomarker for decitabine-based treatments.

基于近红外光谱特征的冷冻小龙虾鲜度快速检测方法

为建立快速检测冷冻小龙虾鲜度的近红外光谱模型,采集解冻的小龙虾虾尾、虾仁及虾糜的近红外光谱,分别利用一阶导数、多元散射校正、小波变换(wavelet transform,WT)和标准正态变换进行预处理,并利用偏最小二乘(partial least squares,PLS)与卷积神经网络(convolutional neural network,CNN)算法将预处理前后的光谱数据分别与总挥发性盐基氮(total volatile basic nitrogen,TVB-N)含量关联,构建定量预测模型并比较建模效果,选取较佳模型,探究模型预测准确度和适用性。结果显示,预处理方法明显影响了建立模型的精度,光谱经预处理建立的CNN模型与PLS模型相比,具备更好地预测小龙虾TVB-N含量的能力。其中,虾仁光谱经WT预处理建立的CNN模型对验证集的预测准确度最高,校正集与验证集的相关系数分别为0.97、0.96,校正集与验证集的均方根误差分别为1.26、0.93mg/100g。近红外光谱的准确度、精密度与灵敏度均在合理范围内,方法学验证结果良好。综合考虑实际应用中快速、准确、低损伤等需求,确定WT-CNN-虾仁模型为预测冷冻小龙虾中TVB-N含量的最优模型。这些结果表明,WT-CNN-虾仁模型在预测冷冻小龙虾TVB-N含量、快速评价新鲜度方面具有巨大潜力。
