Four centrosome-related genes to predict the prognosis and drug sensitivity of patients with colon cancer

BACKGROUND As the primary microtubule organizing center in animal cells,centrosome abnormalities are involved in human colon cancer.AIM To explore the role of centrosome-related genes(CRGs)in colon cancer.METHODS CRGs were collected from public databases.Consensus clustering analysis was performed to separate the Cancer Genome Atlas cohort.Univariate Cox and least absolute shrinkage selection operator regression analyses were performed to identify candidate prognostic CRGs and construct a centrosome-related signature(CRS)to score colon cancer patients.A nomogram was developed to evaluate the CRS risk in colon cancer patients.An integrated bioinformatics analysis was conducted to explore the correlation between the CRS and tumor immune microenvironment and response to immunotherapy,chemotherapy,and targeted therapy.Single-cell transcriptome analysis was conducted to examine the immune cell landscape of core prognostic genes.RESULTS A total of 726 CRGs were collected from public databases.A CRS was constructed,which consisted of the following four genes:TSC1,AXIN2,COPS7A,and MTUS1.Colon cancer patients with a high-risk signature had poor survival.Patients with a high-risk signature exhibited decreased levels of plasma cells and activated memory CD4+T cells.Regarding treatment response,patients with a high-risk signature were resistant to immunotherapy,chemotherapy,and targeted therapy.COPS7A expression was relatively high in endothelial cells and fibroblasts.MTUS1 expression was high in endothelial cells,fibroblasts,and malignant cells.CONCLUSION We constructed a centrosome-related prognostic signature that can accurately predict the prognosis of colon cancer patients,contributing to the development of individualized treatment for colon cancer.

Sustainable production of high-energy-density jet fuel via cycloaddition reactions

Developing an energy supply-chain based on renewable biomass holds great potential to build a low carbon society. High-energy-density(HED) jet fuel, featuring unique fused/strained cycloalkanes, is of great significance for volume-limited military aircrafts, as their high density and combustion heat can extend flight duration and increase the payload. Therefore, the exploration of biomass-based routes towards HED fuel has drawn much attention over the past decade. Cycloaddition reaction features rapid construction of various carbocycles in an atom-and step-economical fashion. This elegant strategy has been widely applied in the manufacture of sustainable HED fuel. Here we carefully summarize the progress achieved in this fascinating area and the review is categorized by the cycloaddition patterns including [4+2], [2+2],[4+4], and [2+1] cycloadditions. Besides, the energy densities of the as-prepared biofuels and petroleumbased fuels(conventional Jet-A and advanced JP-10) are also compared. This review will provide important insights into rational design of new HED fuel with different ring-types/sizes and inspire the chemists to turn those literature studies into practical applications in military field.

Permanent lining castable with low bulk density and thermal conductivity:bauxite castable for tundish obtained by adding pearlescent sand

The low thermal conductivity and light mass of castables for tundish permanent linings are crucial for minimizing the heat loss of molten steel.In consideration of the low bulk density and thermal conductivity of pearlescent sand,the thermal insulation performance of castables was attempted to be improved by adding pearlescent sand.Pearlescent sand was modified to prevent the strength of its porous structure from deteriorating.The modification mechanism of pearlescent sand and the effect of pearlescent sand on the performance of bauxite castables were studied.The results suggested that the addition of the modified pearlescent sand significantly raised the apparent porosity and decreased the bulk density of bauxite castable.At 1000℃,the bulk density of more than 60%of the modified pearlescent sand-bauxite castable was only 2.03 g/cm^(3).The mechanical properties and thermal shock resistance of the modified pearlescent sand-bauxite castable were inferior to those of conventional bauxite castable but were adequate to meet the use conditions of casta-bles for tundish permanent linings.At high temperatures of 200-800℃,the thermal conductivity of more than 60%of the modified pearlescent sand-bauxite castable was smaller than that of conventional bauxite castable.The addition of the modified pearlescent sand can greatly reduce the thermal conductivity and bulk density of bauxite castable.

Clinical relevance of distolingual roots and periodontal status in mandibular first molars: a cross-sectional study employing CBCT analysis

Objectives:Distolingual root of the permanent mandibular first molar(PMFM-DLR)has been frequently reported,which may complicate the treatment of periodontitis.This study aimed to assess the morphological features of PMFM-DLR and investigate the correlation between the morphological features of PMFM-DLR and periodontal status in patients with Eastern Chinese ethnic background.Materials and methods:A total of 836 cone beam computed tomography(CBCT)images with 1497 mandibular first molars were analyzed to observe the prevalence of PMFM-DLR at the patients and tooth levels in Eastern China.Among them,complete periodontal charts were available for 69 Chinese patients with 103 teeth.Correlation and regression analyses were used to evaluate the correlation between the morphological features of DLR,bone loss,and periodontal clinical parameters,including clinical attachment loss(CAL),probing pocket depth(PPD),gingival recession(GR),and furcation involvement(FI).Results:The patient-level prevalence and tooth-level prevalence of DLR in mandibular first molars were 29.4%and 26.3%,respectively.Multiple linear regression analysis suggested that bone loss at the lingual site and CAL were negatively affected by the angle of separation between distolingual and mesial roots in the transverse section,while they were significantly influenced by age and the angle of separation between distobuccal and mesial roots in the coronal section.Conclusions:The prevalence of PMFM-DLR in Eastern China was relatively high in our cohort.The morphological features of DLR were correlated with the periodontal status of mandibular first molars.This study provides critical information on the morphological features of DLR for improved diagnosis and treatment options of mandibular molars with DLR.

Unraveling the mechanism of potassium metal capacitor for highly efficient charge storage

Although hybrid metal ion capacitors(MICs) are highly desired to achieve both high power density of supercapacitors and high energy density of rechargeable batteries, the mismatch problem of electrochemical kinetics of negative and positive electrodes in MICs hampers the realization of this goal. Here, a new hybrid capacitor concept-potassium metal capacitor(PMC) is proposed for the first time, where potassium metal and commercial activated carbon(AC) without any modification are applied as negative and positive electrodes, respectively, and the electrolyte is the same as that of non-aqueous potassium ion batteries. The simplest PMC prototype exhibits a good combination of high energy density(184.9 Wh kg^(-1)) and power density(12.4 kW kg^(-1)), which benefits from the synergistic effect of potassium metal and AC electrode. The former experiences fast potassium plating/striping during charging and discharging, and the later possesses complex multiple charge behaviors driven by low potential of potassium metal. Specifically, below open-circuit voltage, transportation of solvated cations in AC pores plays an important role;beyond this voltage, synergy actions of cations and anions, including adsorption/desorption of solvated cations and anions, and ions exchange between them, dominate the capacitance contribution. This work enriches the types of MICs, and deepens the understanding of the energy storage mechanism of non-aqueous hybrid metal capacitors.

BACKGROUND It is possible that this condition will lead to urosepsis and progressive deterioration of renal function in the absence of surgical intervention.Several recent clinical studies have shown that multi-tract percutaneous nephrolithotomy(MPCNL)has a similar stone free rate(SFR)as standard percutaneous nephrolithotomy(S-PCNL).As a result,M-PCNL was also recommended as a treatment option for staghorn calculi.AIM To examine the perioperative and long-term results of ultrasonography-guided single-and M-PCNL.METHODS This was a retrospective cohort study.Between March 2021 and January 2022,the urology department of our hospital selected patients for the treatment of staghorn calculi using percutaneous nephrolithotomy.The primary outcomes were com plication rate and SFR,and the characteristics of patients,operative parameters,laboratory measurements were also collected.RESULTS In total,345 patients were enrolled in the study(186 in the S-PCNL group and 159 in the M-PCNL group).The SFR in the M-PCNL group was significantly higher than that in the S-PCNL group(P=0.033).Moreover,the incidence rates of hydrothorax(P=0.03)and postoperative infection(P=0.012)were higher in the M-PCNL group than in the S-PCNL group.Logistic regression analysis demonstrated that post-operative white blood cell count(OR=2.57,95%CI:1.90-3.47,P<0.001)and stone size(OR=1.59,95%CI:1.27-2.00,P<0.001)were associated with a higher overall complication rate in the S-PCNL group.Body mass index(OR=1.22,95%CI:1.06-1.40,P=0.004)and stone size(OR=1.70,95%CI:1.35-2.15,P<0.001)were associated with increased overall complications in the M-PCNL group.CONCLUSION Multiple access tracts can facilitate higher SFR while slightly increasing the incidence of acceptable complications.

BACKGROUND Acute liver failure(ALF)is a common cause of postoperative death in patients with hepatocellular carcinoma(HCC)and is a serious threat to patient safety.The neutrophil-to-lymphocyte ratio(NLR)is a common inflammatory indicator that is associated with the prognosis of various diseases,and the albumin-bilirubin score(ALBI)is used to evaluate liver function in liver cancer patients.Therefore,this study aimed to construct a predictive model for postoperative ALF in HCC tumor integrity resection(R0)based on the NLR and ALBI,providing a basis for clinicians to choose appropriate treatment plans.AIM To construct an ALF prediction model after R0 surgery for HCC based on NLR and ALBI.METHODS In total,194 patients with HCC who visited The First People’s Hospital of Lianyungang to receive R0 between May 2018 and May 2023 were enrolled and divided into the ALF and non-ALF groups.We compared differences in the NLR and ALBI between the two groups.The risk factors of ALF after R0 surgery for HCC were screened in the univariate analysis.Independent risk factors were analyzed by multifactorial logistic regression.We then constructed a prediction model of ALF after R0 surgery for HCC.A receiver operating characteristic curve,calibration curve,and decision curve analysis(DCA)were used to evaluate the value of the prediction model.RESULTS Among 194 patients with HCC who met the standard inclusion criteria,46 cases of ALF occurred after R0(23.71%).There were significant differences in the NLR and ALBI between the two groups(P<0.05).The univariate analysis showed that alpha-fetoprotein(AFP)and blood loss volume(BLV)were significantly higher in the ALF group compared with the non-ALF group(P<0.05).The multifactorial analysis showed that NLR,ALBI,AFP,and BLV were independent risk factors for ALF after R0 surgery in HCC.The predictive efficacy of NLR,ALBI,AFP,and BLV in predicting the occurrence of ALT after R0 surgery for HCC was average[area under the curve(AUC)NLR=0.767,AUCALBI=0.755,AUCAFP=0.599,AUCBLV=0.718].The predictio

Porous Ti_(3)C_(2)T_(x) MXene nanosheets sandwiched between polyimide fiber mats for electromagnetic interference shielding

With the rapid development of wireless communication technology and electronic devices,the issue of electromagnetic interference(EMI)is becoming increasingly severe.Developing a new and flexible electromagnetic interference shielding material has become a challenging task.Here,a sandwich-structured EMI shielding composite film was prepared using electrospinning and vacuum filtration methods.In this process,a porous MXene was synthesized through a reaction with cobalt acetate and served as the intermediate layer in the composite film to shield electromagnetic waves.The electrospun polyimide(PI)fibers were used as the top and bottom layers of the composite film,which can protect the porous MXene from oxidation.This lightweight and flexible composite film integrates electromagnetic interference shielding and thermal insulation capabilities,showing excellent comprehensive performance.The composite film achieves an EMI shielding effectiveness of 48.8 dB in X-band(8.2–12.4 GHz),and absolute shielding effectiveness of the composite film reached a satisfying 4142.43(dB·cm^(2))/g.Owing to the design of a multi-layer porous structure,the density of the composite film is 0.65 g/cm3.Furthermore,the thermal conductivity of the film is 0.042 W/(m·K)due to the clamping of electrospun PI fibers,showing excellent thermal insulation performance.Additionally,the composite film exhibits excellent high and low-temperature resistance.In summary,this work provides a feasible strategy for preparing a lightweight polymer-based EMI shielding film.

A Real-time Lithological Identification Method based on SMOTE-Tomek and ICSA Optimization

In petroleum engineering,real-time lithology identification is very important for reservoir evaluation,drilling decisions and petroleum geological exploration.A lithology identification method while drilling based on machine learning and mud logging data is studied in this paper.This method can effectively utilize downhole parameters collected in real-time during drilling,to identify lithology in real-time and provide a reference for optimization of drilling parameters.Given the imbalance of lithology samples,the synthetic minority over-sampling technique(SMOTE)and Tomek link were used to balance the sample number of five lithologies.Meanwhile,this paper introduces Tent map,random opposition-based learning and dynamic perceived probability to the original crow search algorithm(CSA),and establishes an improved crow search algorithm(ICSA).In this paper,ICSA is used to optimize the hyperparameter combination of random forest(RF),extremely random trees(ET),extreme gradient boosting(XGB),and light gradient boosting machine(LGBM)models.In addition,this study combines the recognition advantages of the four models.The accuracy of lithology identification by the weighted average probability model reaches 0.877.The study of this paper realizes high-precision real-time lithology identification method,which can provide lithology reference for the drilling process.

BACKGROUND Thrombocytopenia 2,an autosomal dominant inherited disease characterized by moderate thrombocytopenia,predisposition to myeloid malignancies and normal platelet size and function,can be caused by 5’-untranslated region(UTR)point mutations in ankyrin repeat domain containing 26(ANKRD26).Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1)have been identified as negative regulators of ANKRD26.However,the positive regulators of ANKRD26 are still unknown.AIM To prove the positive regulatory effect of GATA binding protein 2(GATA2)on ANKRD26 transcription.METHODS Human induced pluripotent stem cells derived from bone marrow(hiPSC-BM)INTRODUCTION Ankyrin repeat domain containing protein 26(ANKRD26)acts as a regulator of adipogenesis and is involved in the regulation of feeding behavior[1-3].The ANKRD26 gene is located on chromosome 10 and shares regions of homology with the primate-specific gene family POTE.According to the Human Protein Atlas database,the ANKRD26 protein is localized to the Golgi apparatus and vesicles,and its expression can be detected in nearly all human tissues[4].Moreover,UniProt annotation revealed that ANKRD26 is localized in the centrosome and contains coiled-coil domains formed by spectrin helices and ankyrin repeats[5,6].The most common disease related to ANKRD26 is thrombocytopenia 2(THC2),which is a rare autosomal dominant inherited disease characterized by lifelong mild-to-moderate thrombocytopenia and mild bleeding[7-9].Caused by the variants in the 5’-untranslated region(UTR)of ANKRD26,THC2 is defined by a decrease in the number of platelets in circulating blood and results in increased bleeding and decreased clotting ability[8,10].Due to the point mutations that occur in the 5’-UTR of ANKRD26,its negative transcription factors(TFs),Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1),lose their repression effect[11].The persistent expression of ANKRD26 increases the activity of the mitogen activated protein kinase and e
