Forster resonance energy transfer （FRET） techniques have been widely used in biological studies in vitro andin vivo and are powerful tools for elucidating protein interactions in many regulatory cascades. FRET occurs between oscillating dipoles of two fluorophores with overlapping emission and excitation wavelengths and is dependent on the spectroscopic and geometric properties of the donor-acceptor pair. Various efforts have been made to develop quantitative FRET methods to accurately determine the interaction affinity and kinetics parameters. SUMOylation is an important post-translational protein modification with key roles in multiple biological processes. Conjugating SUMO to substrates requires an enzymatic cascade. Sentrin/SUMO-specific proteases （SENP） act as endopeptidases to process the pre-SUMO or an isopeptidase to deconjugate SUMO from its substrate. Here we also summarize recent developments of theoretical and experimental procedures for determining the protein interaction dissociation constant, Kd, and protease kinetics parameters, kcat and Kin, in the SUMOylation pathway. The general principles of these quantitative FRET-based measurements can be applied to other protein interactions and proteases.

In vehicle Ad-hoc netwok （VANET）, traffic load is often unevenly distributed among access points （APs）. Such load imbalance hampers the network from fully utilizing the network capacity. To alleviate such imbalance, the paper introduces a novel pricing game model. The research scene is at the intersection when the traffic light is green. As vehicles are highly mobile and the network typology changes dynamically, the paper divides the green light time into equal slots and calculates APs＇ prices with the presented pricing game in each time slot. The whole process is a repeated game model. The final equilibrium solution set is APs＇ pricing strategy, and the paper claim that this equilibrium solution set can affect vehicles＇ selection and ensure APs＇ load-balancing. Simulation results based on a realistic vehicular traffic model demonstrate the effectiveness of the game method.

We report the discovery of a peculiar X-ray transient,EP240408a,by Einstein Probe （EP） and follow-up studies made with EP,Swift,NICER,GROND,ATCA and other ground-based multiwavelength *** new transient was first detected with Wide-field X-ray Telescope （WXT） on board EP on April 8th,2024,manifested in an intense yet brief X-ray flare lasting for *** flare reached a peak flux of 3.9×10-9erg cm-2s-1in 0.5-4 ke V,～300 times brighter than the underlying X-ray emission detected throughout the *** and more precise follow-up observations by EP/FXT,Swift and NICER confirmed the finding of this new *** X-ray spectrum is non-thermal in 0.5-10 ke V,with a power-law photon index varying *** X-ray light curve shows a plateau lasting for ～4 d,followed by a steep decay till becoming undetectable ～10 d after the initial *** on its temporal property and constraints from previous EP observations,an unusual timescale in the range of 7-23 d is found for EP240408a,which is intermediate between the commonly found fast and long-term *** counterparts have been found in optical and near-infrared,with the earliest observation at 17 h after the initial X-ray detection,suggestive of intrinsically weak emission in these *** demonstrate that the remarkable properties of EP240408a are inconsistent with any of the transient types known so far,by comparison with,in particular,jetted tidal disruption events,gamma-ray bursts,X-ray binaries and fast blue optical *** nature of EP240408a thus remains an *** suggest that EP240408a may represent a new type of transients with intermediate timescales of the order of～10 *** detection and follow-ups of more of such objects are essential for revealing their origin.

The cuticle, an essential structure for insects, is produced from cuticular proteins and chitin via a series of biochemical reactions. Tweedle genes are important members of the cuticular protein family and have four conserved motifs binding to chitin. Tweedle family genes have been found to play a profound effect on cuticle development. Here, we report that the cuticular protein gene LmTwdll ofLocusta migratoria belongs to the Tweedle family. In situ hybridization showed that LmTwdll is localized to epidermal cells of the cuticle. The expression patterns of LmTwdll showed low expression in the cuticle during the early and middle stages of the fifth-instar nymphs; in contrast, its expression rapidly increased in the late stages of fifth-instar nymphs. We performed RNA interference to examine the function of LmTwdll in locusts. Silencing of LmTwdll resulted in high mortality during the molting process before the next stage. Also, the epicuticle of nymphs failed to molt, tended to be thinner and the arrangement of chitin in the procuticle appeared to be disordered compare to the control group. These results demonstrate that LmTwdl! plays a critical role in molting, which contributes to a better understanding of the distinct functions of the Tweedle family in locusts.

Outcomes of Chinese Patients with End-stage Pulmonary Disease while Awaiting Lung Transplantation： A Single-center Study

Background： The factors affecting the outcome of patients referred for lung transplantation （LTx） still have not been investigated extensively. The aim of this study was to characterize the patient outcomes and identify the prognostic factors for death while awaiting the LTx. Methods： From January 2003 to November 2013, the clinical data of 103 patients with end-stage lung disease that had been referred for LTx to Department of Thoracic Surgery, Shanghai Pulmonary Hospital were analyzed retrospectively. The relationship between predictors and survival was evaluated using the Kaplan-Meier method and the Cox proportional hazards model. Results： Twenty-five patients （24.3%） died while awaiting the LTx. Fifty patients （48.5%） underwent LTx, and 28 patients （27.2%） were still on the waitlist. Compared to the candidates with chronic obstructive puhnonary disease （COPD）, patients with idiopathic pulmonary fibrosis （IPF） had a higher mortality while awaiting the LTx （40.0% vs. 12.3%, P= 0.003）. Patients requiring mechanical ventilation （MV） had a higher mortality while waiting than others （50.0% vs. 20.2%, P = 0.038）. Two variables, using MV and IPF but not COPD as primary disease, emerged as significant independent risk factors for death on the waitlist （hazard ratio [HR] 56.048, 95% confidence interval [CI]： 3.935-798.263, P= 0.003 and MR = 14.859, 95% CI： 2.695 81.932, P= 0.002, respectively）. Conclusion： The type of end-stage lung disease, pulmonary hypertension, and MV may be distinctive prognostic factors for death while awaiting the LTx.
