Regulations of m6A and other RNA modifications and their roles in cancer

RNA modification is an essential component of the epitranscriptome, regulating RNA metabolism and cellular functions. Several types of RNA modifications have been identified to date; they include N-6-methyladenosine (m(6)A), N-1-methyladenosine (m(1)A), 5-methylcytosine (m(5)C), N-7-methylguanosine (m(7)G), N-6,2 '-O-dimethyladenosine (m(6)A(m)), N-4-acetylcytidine (ac(4)C), etc. RNA modifications, mediated by regulators including writers, erasers, and readers, are associated with carcinogenesis, tumor microenvironment, metabolic reprogramming, immunosuppression, immunotherapy, chemotherapy, etc. A novel perspective indicates that regulatory subunits and post-translational modifications (PTMs) are involved in the regulation of writer, eraser, and reader functions in mediating RNA modifications, tumorigenesis, and anticancer therapy. In this review, we summarize the advances made in the knowledge of different RNA modifications (especially m(6)A) and focus on RNA modification regulators with functions modulated by a series of factors in cancer, including regulatory subunits (proteins, noncoding RNA or peptides encoded by long noncoding RNA) and PTMs (acetylation, SUMOylation, lactylation, phosphorylation, etc.). We also delineate the relationship between RNA modification regulator functions and carcinogenesis or cancer progression. Additionally, inhibitors that target RNA modification regulators for anticancer therapy and their synergistic effect combined with immunotherapy or chemotherapy are discussed.

Assessment of subtalar joint neutral position： a cadaveric study

Background Subtalar joint （STJ） neutral position is the position typically used by clinicians to obtain a cast representation of a patient＇s foot before fabrication of biomechanical functional orthosis. But no method for measuring STJ neutral position has been proven accurate and reproducible by different testers. This study was conducted to investigate the STJ neutral position in normal feet in cadavers. Methods Twelve fresh-frozen specimens of amputated lower legs were used. Pressure-sensitive films were inserted into the anterior and posterior articulation of STJ. The contact areas for various foot positions and under axial loads of 600 N were determined based on the gray level of the digitized film. The STJ neutral positions were determined as the ankle-foot position where the maximum contact area was achieved, because the neutral position of a joint was defined as the position where the concave and convex surfaces were completely congruous. Results In ankle-foot neutral position, the contact area of STJ was （2.79±0.24） cm^2. In the range of motion of adduction-abduction （ADD-ABD）, the maximum contact area was （3.00±0.26） cm^2 when the foot was positioned 10° of ABD （F=-221.361, P 〈0.05）. In the range of motion of dorsiflexion-plantarflexion （DF-PF）, the maximum contact area was （3.61±0.25） cm^2 when the foot was positioned 20° of DF （F=-121.067, P 〈0.05）. In the range of motion of inversion-eversion （INV-EV）, the maximum contact area was （3.14±0.26） cm^2 when the foot was positioned 10° of EV （F=-256.252, P〈0.05）. Conclusions Joints, such as STJ, therefore, are not necessarily in neutral position when the ankle-foot is placed in the traditional concept of neutral position. The results demonstrate that the most approximate STJ neutral position was in the foot Dosition of 10° of abduction, 20° of dorsiflexion and 10° of eversion.

COMPARISON OF PERSIANN AND TMPA DAILY PRECIPITATION ESTIMATES OVER HUNAN PROVINCE OF CHINA

This study presented a detailed comparison of daily precipitation estimates from Precipitation Estimation from Remote Sensing Information using Artificial Neural Network(PERSIANN) and Tropical Rainfall Measuring Mission(TRMM) Multi-satellite Precipitation Analysis(TMPA) over Hunan province of China from 1998 to 2014. The ground gauge observations are taken as the reference. It is found that overall TMPA clearly outperforms PERSIANN, indicating by better statistical metrics(including correlation coefficient, root mean square error and relative bias). For the geospatial pattern, although both products are able to capture the major precipitation features(e.g., precipitation geospatial homogeneity) in Hunan, yet PERSIANN largely underestimates the precipitation intensity throughout all seasons. In contrast, there is no clear bias tendency from TMPA estimates. Precipitation intensity analysis showed that both the occurrence and amount histograms from TMPA are closer to the gauge observations from spring to ***, in the winter season PERSIANN is closer to gauge observation, which is likely due to the ground contamination from the passive microwave sensors used by TMPA.

A Ray-Testing Approach to Automatically Identify Surface Visibility in Parting Line Generation

It is critical to identify core/cavity and undercut surfaces of molds in parting line generation. A new Ray-Testing approach is presented to detect these surfaces by identifying the visibility of surfaces. A simple, direct and efficient algorithm to identify surface visibility is developed. Case studies show that the approach is practical and valuable in automated parting line generation.

The whole theoretical framework of CAXASolid is introduced. Users can customize their system through application integration, data integration, integration by DLL or integration by COM. Focusing on the customization techniques with COM components, the development method and process are illustrated with some case studies.

In this paper,we study spatial cross-sectional data models in the form of matrix exponential spatial specification(MESS),where MESS appears in both dependent and error *** empirical likelihood(EL)ratio statistics are established for the parameters of the MESS *** is shown that the limiting distributions of EL ratio statistics follow chi-square distributions,which are used to construct the confidence regions of model *** experiments are conducted to compare the performances of confidence regions based on EL method and normal approximation method.

Background The exudative form of age-related macular degeneration(AMD)is characterized by abnormal blood vessel growth,which is stimulated by vascular endothelial growth factor(VEGF)released from retinal pigment epithelium(RPE).The angiogenic behaviors of vascular endothelial cells in vitro depend on forkhead box protein P1(Foxp1),a transcription repressor widely expressed in human and murine tissues during *** this study,we aimed to determine whether loss of Foxp1 affects laser-induced choroidal neovascularization(CNV)in *** Eye-selective deletion of Foxp1 was obtained by crossing Foxp1flox/flox with Six3-Cre *** photocoagulation was delivered to six-to eight-week-old mice to induce *** expression of Foxp1 and Cre was determined by immunofluorescence in cryostat sections of the *** fluorescein angiography(FFA),optical coherence tomography(OCT),and B4 isolectin staining were applied to analyze the leakage,bulge height,and area of CNV lesions,***-choroid tissues were isolated for the determination of VEGF and pigment epithelium derived factor(PEDF)by Western *** Foxp1 was expressed in retinal ganglion cells,RPE,and the choroidal endothelial *** photocoagulation increased the number of Foxp1+-endothelial cells and induced ***3-Cre reduced Foxp1 expression in RPE but not the endothelium,leading to a lower level of VEGF in the ***1 knockout inhibited pathological angiogenesis and vascular leakage of the laser-induced CNV *** Foxp1 regulates the expression of VEGF in the RPE,and inhibition of Foxp1 could potentially be a novel strategy for the prevention and therapy of neovascularization related to AMD.

A deoxidation thermodynamic model for 304 stainless steel considering multiple-components coupled reactions

A thermodynamic model for predicting the equilibrium oxygens of 304 stainless steel was developed based on the theory of slag-steel equilibrium,the law of mass conservation,and the ion and molecule coexistence *** the developed model,the Fe-Cr-Mn-Si-Al-S-O-melts reaction system and CaO-MgO-CaF_(2)-FeO-MnO-Al_(2)O_(3)-SiO_(2)-Cr2O_(3)slags were *** oxygen contents calculated by the model are in good agreement with experimental results and reference *** equilibrium oxygen contents in 304 stainless steel mainly decrease with increasing binary basicity(w(CaO)/w(SiO_(2)),where w(i)is the mass percentage of component i)and decreasing *** binary basicity at 2.0 while maintaining temperatures lower than 1823 K will keep the oxygen contents in the 304 stainless steel lower than 15×10^(-6).The equilibrium oxygen contents may also be decreased with increasing content of MgO in slags,which is more significant at lower binary ***,a small amount of FeO,MnO,and Al_(2)O_(3)(about 0-2.5 wt.%)in slags has little effect on equilibrium oxygen ***,it is found that the[C]-[O]reaction may occur during refining process but will not significantly affect the equilibrium oxygen contents.

Objective The tricho-rhino-phalangeal syndrome 1(TRPS1) gene, which was initially found to be associated with tricho-rhino-phalangeal syndrome, is critical for the development and differentiation of bone, hair follicles and kidney. However, its role in cancer progression is largely unknown. Method The functions of TRPS1 were analyzed by cellular biological assays in vitro and in vivo. Result We here demonstrated that down-regulation of TRPS1 correlated with distant metastasis, tumor recurrence and survival rate in cancer patients. TRPS1 was frequently down-regulated in high-metastatic breast, colon and nasopharyngeal cancers, and silencing of TRPS1 stimulated epithelial-mesenchymal transition(EMT), migration and invasion in vitro and metastasis in vivo, while TRPS1 over-expression exhibited the opposite effects. Using quantitative proteomics, the negative EMT regulator FOXA1 was shown to be down-regulated by TRPS1 knockdown. Ectopic expression of FOXA1 blocked the enhancement of EMT, migration and invasion induced by TRPS1 silencing. Mechanistically, TRPS1, acting as a transcription activator, directly induced FOXA1 transcription by binding to the FOXA1 promoter. We further showed that down-regulation of TRPS1 was induced by mi R-373 binding to the 3‘UTR of TRPS1. TRPS1, but not TRPS1-3‘UTR, over-expression blocked the enhancement of migration and invasion induced by mi R-373. Conclusion Loss of TRPS1 by mi R-373, acting as a transcriptional activator, promotes EMT and metastasis by repressing FOXA1 transcription, expanding upon its previously reported role as a transcription repressor.
