m^(6)A modification of plant virus enables host recognition by NMD factors in plants

N^(6)-methyladenosine(m^(6)A)is the most abundant eukaryotic mRNA modification and is involved in various biological processes.Increasing evidence has implicated that m^(6)Amodification is an important anti-viral defense mechanism in mammals and plants,but it is largely unknown how m^(6)Aregulates viral infection in plants.Here we report the dynamic changes and functional anatomy of m^(6)Ain Nicotiana benthamiana and Solanum lycopersicum during Pepino mosaic virus(PepMV)infection.m^(6)Amodification in the PepMV RNA genome is conserved in these two species.Overexpression of the m^(6)Awriters,mRNA adenosine methylase A(MTA),and HAKAI inhibit the PepMV RNA accumulation accompanied by increased viral m^(6)Amodifications,whereas deficiency of these writers decreases the viral RNA m^(6)Alevels but enhances virus infection.Further study reveals that the cytoplasmic YTH-domain family protein NbECT2A/2B/2C as m^(6)Areaders are involved in anti-viral immunity.Protein-protein interactions indicate that NbECT2A/2B/2C interact with nonsense-mediated mRNA decay(NMD)-related proteins,including NbUPF3 and NbSMG7,but not with NbUPF1.m^(6)Amodification-mediated restriction to PepMV infection is dependent on NMD-related factors.These findings provide new insights into the functionality of m^(6)Aanti-viral activity and reveal a distinct immune response that NMD factors recognize the m^(6)Areaders-viral m^(6)ARNA complex for viral RNA degradation to limit virus infection in plants.

MXenes are a family of two-dimensional(2D)layered transition metal carbides/nitrides that show promising potential for energy storage applications due to their high-specific surface areas,excellent electron conductivity,good hydrophilicity,and tunable terminations.Among various types of MXenes,Ti_(3)C_(2)T_(x) is the most widely studied for use in capacitive energy storage applications,especially in supercapacitors(SCs).However,the stacking and oxidation of MXene sheets inevitably lead to a significant loss of electrochemically active sites.To overcome such challenges,carbon materials are frequently incorporated into MXenes to enhance their electrochemical properties.This review introduces the common strategies used for synthesizing Ti_(3)C_(2)T_(x),followed by a comprehensive overview of recent developments in Ti_(3)C_(2)T_(x)/carbon composites as electrode materials for SCs.Ti_(3)C_(2)T_(x)/carbon composites are categorized based on the dimensions of carbons,including 0D carbon dots,1D carbon nanotubes and fibers,2D graphene,and 3D carbon materials(activated carbon,polymer-derived carbon,etc.).Finally,this review also provides a perspective on developing novel MXenes/carbon composites as electrodes for application in SCs.

Early Triassic Volcanic Rocks in the Western Yarlung Zangbo Suture Zone:Implications for the Opening of the Neo-Tethys Ocean

In this study,zircon U-Pb dating of volcanic rocks from the Zhongba ophiolite of the Yarlung Zangbo Suture Zone(YZSZ)in southern Xizang(Tibet)yielded an age of 247±3 Ma.According to whole rock geochemical and Sr-NdPb isotopic data,the Early Triassic samples could be divided into two groups:Group 1 with P-MORB affinity,showing initial^(87)Sr/^(86)Sr ratios of 0.70253–0.70602,ε_(Nd)(t)values of 4.2–5.3,(^(206)Pb/^(204)Pb)_(t)ratios of 16.353–18.222,(^(207)Pb/^(204)Pb)_(t)ratios of 15.454–15.564,and(^(208)Pb/^(204)Pb)_(t)ratios of 35.665–38.136;Group 2 with OIB affinity,showing initial^(87)Sr/^(86)Sr ratios of 0.70249–0.70513,ε_(Nd)(t)values of 4.4–4.9,(^(206)Pb/^(204)Pb)_(t)ratios of 17.140–18.328,(^(207)Pb/^(204)Pb)_(t)ratios of 15.491–15.575,and(^(208)Pb/^(204)Pb)_(t)ratios of 36.051–38.247.Group 2 rocks formed by partial melting of the mantle source enriched by a former plume,and assimilated continental crustal material during melt ascension.The formation of Group 1 rocks corresponds to the mixing of OIB melts,with the same components as Group 2 and N-MORBs.The Zhongba Early Triassic rocks belong to the continental margin type ophiolite and formed in the continental–oceanic transition zone during the initial opening of the Neo-Tethys in southern Xizang(Tibet).

Medium and long-term thermal coal contract embedded reparations from the perspective of an evolutionary game

Coal-fired electricity enterprises are caught in the dilemma of relative fixed prices and rising costs under the scenario of decarbonization.Meanwhile,soaring market-oriented coal pricing results in coal enterprises’increasing defaults on thermal coal medium-and long-term contracts(MLC).To investigate the implementation of MLC at the micro-level,this study formalized the contractual behaviors of coal and coal-fired electricity enterprises based on the asymmetric evolutionary game.We formalized the evolving behaviors of both parties using replicator dynamics equations and proved that there were two evolutionary stabilization strategies(ESSs):compliance and coal enterprises’unilateral default.A multi-agent-based simulation was applied to verify the evolving process of ESSs and determine the critical values of MLC design by sensitive analysis.From the simulation results,coal-fired electricity enterprises do not stop generation under the current carbon quota allocation mechanism,even if carbon emission trading increases electricity generation costs.Coal enterprises choose to“default”when the market price of coal is higher than the contracted price by 18%.However,if the original reparation is increased by 5%,the compliance rate of the coal enterprises improves.Dynamic reparations embedded in the MLC improved enforcement during the contracting period.Moreover,the proposed policy implications have practical significance for enhancing the coordinated operation of coal-electricity energy supply chains.

Industrial water splitting has long been suppressed by the sluggish kinetics of the oxygen evolution reaction(OER),which requires a catalyst to be efficient.Herein,we propose a molecular-level proton acceptor strategy to produce an efficient OER catalyst that can boost industrial-scale water splitting.Molecular-level phosphate(-PO_(4))group is introduced to modify the surface of PrBa_(0.5)Ca_(0.5)Co_(2)O_(5)+δ(PBCC).The achieved catalyst(PO_(4)-PBCC)exhibits significantly enhanced catalytic performance in alkaline media.Based on the X-ray absorption spectroscopy results and density functional theory(DFT)calculations,the PO_(4)on the surface,which is regarded as the Lewis base,is the key factor to overcome the kinetic limitation of the proton transfer process during the OER.The use of the catalyst in a membrane electrode assembly(MEA)is further evaluated for industrial-scale water splitting,and it only needs a low voltage of 1.66 V to achieve a large current density of 1 A cm^(-2).This work provides a new molecular-level strategy to develop highly efficient OER electrocatalysts for industrial applications.

Peculiarity of transcriptional and H3K27me3 dynamics during peach bud dormancy

Bud dormancy facilitates the survival of meristems under harsh environmental conditions.To elucidate how molecular responses to chilling accumulation controlling dormancy in peach buds,chromatin immunoprecipitation sequencing to identify the H3K27me3 modifications and RNA sequencing of two peach cultivars with pronounced differences in chilling requirement were carried out,the results showed that genes associated with abscisic acid and gibberellic acid signal pathways play key roles in dormancy regulation.The results demonstrated that peach flower bud differentiation occurred continuously in both cultivars during chilling accumulation,which was correlated with the transcript abundance of key genes involved in phytohormone metabolism and flower bud development under adverse conditions.The more increased strength in high chillingrequirement cultivar along with the chilling accumulation at the genome-wide level.The function of the dormancy-associated MADS-box gene PpDAM6 was identified,which is involved in leaf bud break in peach and flower development in transgenic Nicotiana tabacum(NC89).In addition,PpDAM6 was positively regulated by PpCBF,and the genes of putative dormancy-related and associated with metabolic pathways were proposed.Taken together,these results constituted a theoretical basis for elucidating the regulation of peach bud dormancy transition.

Regulation effects of water and nitrogen on yield,water,and nitrogen use efficiency of wolfberry

Wolfberry(Lycium barbarum L.)is important for health care and ecological protection.However,it faces problems of low productivity and resource utilization during planting.Exploring reasonable models for water and nitrogen management is important for solving these problems.Based on field trials in 2021 and 2022,this study analyzed the effects of controlling soil water and nitrogen application levels on wolfberry height,stem diameter,crown width,yield,and water(WUE)and nitrogen use efficiency(NUE).The upper and lower limits of soil water were controlled by the percentage of soil water content to field water capacity(θ_(f)),and four water levels,i.e.,adequate irrigation(W0,75%-85%θ_(f)),mild water deficit(W1,65%-75%θ_(f)),moderate water deficit(W2,55%-65%θ_(f)),and severe water deficit(W3,45%-55%θ_(f))were used,and three nitrogen application levels,i.e.,no nitrogen(N0,0 kg/hm^(2)),low nitrogen(N1,150 kg/hm^(2)),medium nitrogen(N2,300 kg/hm^(2)),and high nitrogen(N3,450 kg/hm^(2))were implied.The results showed that irrigation and nitrogen application significantly affected plant height,stem diameter,and crown width of wolfberry at different growth stages(P<0.01),and their maximum values were observed in W1N2,W0N2,and W1N3 treatments.Dry weight per plant and yield of wolfberry first increased and then decreased with increasing nitrogen application under the same water treatment.Dry weight per hundred grains and dry weight percentage increased with increasing nitrogen application under W0 treatment.However,under other water treatments,the values first increased and then decreased with increasing nitrogen application.Yield and its component of wolfberry first increased and then decreased as water deficit increased under the same nitrogen treatment.Irrigation water use efficiency(IWUE,8.46 kg/(hm^(2)·mm)),WUE(6.83 kg/(hm^(2)·mm)),partial factor productivity of nitrogen(PFPN,2.56 kg/kg),and NUE(14.29 kg/kg)reached their highest values in W2N2,W1N2,W1N2,and W1N1 treatments.Results of principal component analysis(PCA)showe

Crocus sativus (saffron) is a globally autumn-flowering plant, and its stigmas are the most expensive spice and valuable herb medicine. Crocus specialized metabolites, crocins, are biosynthesized in distant species, Gardenia (eudicot) and Crocus (monocot), and the evolution of crocin biosynthesis remains poorly understood. With the chromosome-level Crocus genome assembly, we revealed that two rounds of lineage-specific whole genome triplication occurred, contributing important roles in the production of carotenoids and apocarotenoids. According to the kingdom-wide identification, phylogenetic analysis, and functional assays of carotenoid cleavage dioxygenases (CCDs), we deduced that the duplication, site positive selection, and neofunctionalization of Crocus-specific CCD2 from CCD1 members are responsible for the crocin biosynthesis. In addition, site mutation of CsCCD2 revealed the key amino acids, including I143, L146, R161, E181, T259, and S292 related to the catalytic activity of zeaxanthin cleavage. Our study provides important insights into the origin and evolution of plant specialized metabolites, which are derived by duplication events of biosynthetic genes.

Background:At present,a large number of chronic obstructive pulmonary disease(COPD)patients are undiagnosed in China.Thus,this study aimed to develop a simple prediction model as a screening tool to identify patients at risk for COPD.Methods:The study was based on the data of 22,943 subjects aged 30 to 79 years and enrolled in the second resurvey of China Kadoorie Biobank during 2012 and 2013 in China.We stepwisely selected the predictors using logistic regression model.Then we tested the model validity through P-P graph,area under the receiver operating characteristic curve(AUROC),ten-fold cross validation and an external validation in a sample of 3492 individuals from the Enjoying Breathing Program in China.Results:The final prediction model involved 14 independent variables,including age,sex,location(urban/rural),region,educational background,smoking status,smoking amount(pack-years),years of exposure to air pollution by cooking fuel,family history of COPD,history of tuberculosis,body mass index,shortness of breath,sputum and wheeze.The model showed an area under curve(AUC)of 0.72(95%confidence interval[CI]:0.72-0.73)for detecting undiagnosed COPD patients,with the cutoff of predicted probability of COPD=0.22,presenting a sensitivity of 70.13%and a specificity of 62.25%.The AUROC value for screening undiagnosed patients with clinically significant COPD was 0.68(95%CI:0.66-0.69).Moreover,the ten-fold cross validation reported an AUC of 0.72(95%CI:0.71-0.73),and the external validation presented an AUC of 0.69(95%CI:0.68-0.71).Conclusion:This prediction model can serve as a first-stage screening tool for undiagnosed COPD patients in primary care settings.

凋落物输入能够影响土壤有机碳(SOC)输入与输出动态平衡。然而,针对气候变暖背景下凋落物输入对SOC矿化激发效应及温度敏感性的影响仍不清楚。本研究以^(13)C标记的杉木叶凋落物为对象,比较不同培养温度(15、25和35℃)条件下凋落物添加对不同来源CO_(2)释放速率和累积量、SOC矿化激发效应以及温度敏感性(Q_(10))的影响。结果发现:凋落物添加提高土壤总CO_(2)释放速率和累积量,但随着温度的升高,土壤总CO_(2)释放速率和累积量增加幅度呈下降趋势,且不同温度下土壤CO_(2)释放速率随培养时间的延长均呈降低趋势。与对照相比,凋落物添加促进土壤来源CO_(2)释放累积量,呈正激发效应。随着培养温度升高,添加凋落物处理的土壤来源和凋落物来源CO_(2)释放累积量均呈增加趋势,但SOC激发效应呈下降趋势。此外,凋落物添加降低土壤呼吸CO_(2)释放速率的温度敏感性,且凋落物来源CO_(2)释放速率的Q_(10)值低于土壤来源CO_(2)的Q_(10)值。本研究表明,不同培养温度下凋落物输入对土壤CO_(2)释放的影响存在差异,且凋落物输入能够降低土壤呼吸CO_(2)释放速率的温度敏感性。
