Regeneration of immunocompetent B lymphopoiesis from pluripotent stem cells guided by transcription factors

Regeneration of functional B lymphopoiesis from pluripotent stem cells(PSCs)is challenging,and reliable methods have not been developed.Here,we unveiled the guiding role of three essential factors,Lhx2,Hoxa9,and Runx1,the simultaneous expression of which preferentially drives B lineage fate commitment and in vivo B lymphopoiesis using PSCs as a cell source.In the presence of Lhx2,Hoxa9,and Runx1 expression,PSC-derived induced hematopoietic progenitors(iHPCs)immediately gave rise to pro/pre-B cells in recipient bone marrow,which were able to further differentiate into entire B cell lineages,including innate B-1a,B-1b,and marginal zone B cells,as well as adaptive follicular B cells.In particular,the regenerative B cells produced adaptive humoral immune responses,sustained antigen-specific antibody production,and formed immune memory in response to antigen challenges.The regenerative B cells showed natural B cell development patterns of immunoglobulin chain switching and hypermutation via cross-talk with host T follicular helper cells,which eventually formed T cell-dependent humoral responses.This study exhibits de novo evidence that B lymphopoiesis can be regenerated from PSCs via an HSC-independent approach,which provides insights into treating B cell-related deficiencies using PSCs as an unlimited cell resource.

Effects of Different Seedling Raising Methods on Seedling Growth of Two Kinds of Fritillaria

[Objectives] To reveal the effects of film mulching and open ground seedling raising on the differences between Fritillaria przewalskii Maxim. and Fritillaria pallidiflora Schrenkf., find the optimal seedling raising method, and provide scientific basis for standard seedling raising technique and protecting wild resources of Fritillaria. [Methods] This experiment counted the seedling emergence number and calculated the seedling emergence indicator and the yield. [Results] The results showed that under the condition of film mulching and open ground seedling raising, the changes of Fritillaria seedlings showed an "S" curve, conforming to the Logistic equation, and the rapid growth period appeared 5-13 d after seedling emergence. Compared with the open ground seedling raising, the seedlings of F. przewalskii Maxim. and F. pallidiflora Schrenkf. raised through film mulching emerged 2-4 d earlier, and the yield increased by 56.8% and 21.71%, respectively, and the individual quality was excellent. [Conclusions] The film mulching of F. przewalskii Maxim. and F. pallidiflora Schrenkf. has great significance to protect the wild resources of Fritillaria in Eryin Mountain Area. It is recommended to adopt the film mulching method for Fritillaria seedling raising, to reduce the yield decline due to lodging, and adopt sun-shading net covering after sowing.

Determination of Aromatic Components of Rosa davurica Pall. by Headspace Solid Phase Microextraction Combined with GC-MS

[Objectives] To determine the aromatic components of Rosa davurica Pall. [Methods] 42 kinds of aromatic components were identified from the flowers of R. davurica by headspace solid phase microextraction( HS-SPME) combined with gas chromatography-mass spectrometry( GC-MS). The main compounds were alcohols( 54. 88%) and aldehydes( 19. 55%). [Results] The top five components with the highest relative content were phenylethyl alcohol( 12. 69%),geraniol( 9. 85%),citronellol( 8. 80%),nerol( 7. 84%) and 2-n-pentylfuran( 7. 45%). [Conclusions] Headspace solid phase microextraction( HS-SPME) combined with gas chromatography-mass spectrometry( GC-MS) can provide basis for further development and utilization of R. davurica.

在甘肃省永登县开展芍药直接引种(T1)和引种3年后再分株栽培(T2)对比试验,揭示两种栽培方式下芍药生长的差异性,探讨国外芍药品种在国内的适宜性。通过分析原产地和引种地气候因子,测定物候期、农艺性状、个体质量及含量,在大田管理条件下,甘肃永登气候适宜引进国外芍药,T1和T2栽培方式下国外芍药品种均能生长开花,花色纯正,花型饱满,茎秆直立粗壮,叶片浓郁。与T2栽培方式比较,T1栽培方式下芍药生长表现更优,成活率提高4.26%(P<0.05),成花率提高8.34%(P<0.01),单根鲜重、根粗、根长、侧根数和芽数分别增加了0.31 kg(P<0.05)、3.27 mm(P<0.05)、7.00 cm(P<0.05)和5个(P<0.01),根部主要成分芍药苷含量提高48.68%(P<0.01)。以上表明,直接引种更有利于芍药在甘肃永登地区的生长,具有更高的市场前景和经济价值。

Objective:To investigate the therapeutic effect of Biling Weitong Granules combined with oryz-aspergillus enzyme and pancreatin tablets on patients with reflux esophagitis with functional dyspepsia.Methods:Sixty patients diagnosed with reflux esophagitis with functional dyspepsia who were admitted to the Affiliated Hospital of Hebei University between June 2020 and June 2023 were selected and divided into two groups:the control group and the observation group,each consisting of 30 cases.The control group received oryz-aspergillus enzyme and pancreatin tablets only,while the observation group received Biling Weitong Granules in addition to the tablets.The clinical efficacy,Chinese medicine syndrome points,esophageal kinetic indexes,gastrointestinal hormone levels,and therapeutic safety of both groups were evaluated.Results:The total efficiency of the observation group reached 93.33%,significantly higher than the 73.33%of the control group(P0.05).Conclusion:The combination of Biling Weitong Granules with oryz-aspergillus enzyme and pancreatin tablets demonstrates significant efficacy in the treatment of reflux esophagitis with functional dyspepsia,with a better safety profile.This finding warrants

丙烷脱氢制丙烯技术由于选择性高,价格相对便宜,是丙烯生产的一种非常有前景的方法。在本研究中,设计出一种Pd-C_(2)N二维单原子催化剂。通过密度泛函理论研究Pd-C_(2)N上丙烷脱氢的反应,所有模拟计算使用B3LYP泛函,对Pd原子使用LAN2DZ赝势,对于C,H,N非金属原子使用6-31g^(**)基组。分析催化剂静电势,态密度及反应物在催化剂上的吸附作用及反应机理。计算研究表明,Pd-C_(2)N催化丙烷脱氢反应路径为先脱甲基上的氢原子,再脱亚甲基上的氢原子从而生成丙烯,能垒为25.09 kcal·mol^(-1)。丙烯在催化剂上将进一步进行脱氢反应,能垒为42.81 kcal·mol^(-1)。Pd-C_(2)N催化剂在丙烷脱氢反应中表现出优异的催化性能,为设计潜在的新型丙烷脱氢潜在的催化剂提供了理论依据。

Automatic modeling algorithm of stochastic error for inertial sensors

This paper proposes an automatic algorithm to determine the properties of stochastic processes and their parameters for inertial error. The proposed approach is based on a recently developed method called the generalized method of wavelet moments (GMWM), whose estimator was proven to be consistent and asymptotically normally distributed. This algorithm is suitable mainly (but not only) for the combination of several stochastic processes, where the model identification and parameter estimation are quite difficult for the traditional methods, such as the Allan variance and the power spectral density analysis. This algorithm further explores the complete stochastic error models and the candidate model ranking criterion to realize automatic model identification and determination. The best model is selected by making the trade-off between the model accuracy and the model complexity. The validation of this approach is verified by practical examples of model selection for MEMS-IMUs (micro-electro-mechanical system inertial measurement units) in varying dynamic conditions.
