Fluorescent glycan nanoparticle-based FACS assays for the identification of genuine drug-resistant cancer cells with differentiation potential

Herein we develop a unique differentiated-uptake strategy capable of efficient and high-purity isolation of genuine drug-resistant(DR)cells from three types of drug-surviving cancer cells,which include paclitaxel-surviving human ovarian OVCAR-3 cancer cells and human lung carcinoma A549/Taxol cells,and doxorubicin-surviving human immortalized myelogenous leukemia K562/ADR *** using this strategy which relies on fluorescent glycan nanoparticle(FGNP)-based fluorescence-activated cell sorting(FACS)assays,two subpopulations with distinct fluorescences existing in drug-surviving OVCAR-3 cells were separated,and we found that the lower fluorescence(LF)subpopulation consisted of DR cells,while the higher fluorescence(HF)subpopulation was comprised of non-DR ***,the DR cells and their progenies were found distinct in their increased expression of drug-resistant *** intriguingly,by using the FGNP-based FACS assay to detect DR/non-DR phenotypes,we found that the DR phenotype had a potential to differentiate into the non-DR progeny,which demonstrates the differentiation feature of stem-like cancer *** research disclosed that the assay can quantitatively detect the degree of drug resistance in DR cells,as well as the reversal of drug resistance that are tackled by various therapeutic *** strategy thus paves the way to develop theranostic approaches associated with chemotherapy-resistance and cancer stemness.

Technical efficiency of cocoa farms at varying elevation levels in Davao City, Philippines: implications to sustainable upland farming systems

Elevation is one of many components that influence agriculture, and this in turn affects the level of both inputs and outputs of farmers. This article focuses on the productivity and technical efficiency of 100 cocoa farms using cross-sectional data from areas ranging from 190 to 1021 m above sea level which were classified as low, medium, and high elevation in Davao City, considered as the chocolate capital of the Philippines. Using stochastic frontier analysis, the results showed that the cost of inputs per ha and the number of cocoa trees per ha significantly increase yield. Farms at high elevations were less technically efficient, as this entails lower temperatures and increased rainfall, and cocoa farming in those areas and conditions can be more challenging, especially with changes in farming practices, terrain, and distance to markets. Other significant variables were age of cocoa farms, married farmers, and age of the farmers. Older farms may be more developed, farmers who are married benefit from their spouses being able to readily contribute as farm labor, and lastly, older farmers' inefficiency may likely stem from nonadaptation of newer farming practices. With an average technical efficiency of 0.61, 0.63, and 0.26 in low, medium, and high elevation areas, respectively, farmers therefore have an incentive to improve farm practices and consider topographical variations found in high elevation areas. Recommendations for the improvement of technical efficiency of cocoa farms are better connectivity to markets, enhancing farm practices, and continuation and improvement of government programs on cocoa with an added emphasis on research. For farmers in high elevation areas, mitigating solutions such as sustainable agriculture practices and ecolabelling are key to improving efficiency and minimizing the potential negative impact on upland farming systems. In late March 2020,SARS-CoV-2 arrived in Manaus,Brazil,and rapidly developed into a large-scale epidemic that collapsed the local health system and resulted in extreme death *** key studies reported that∼76%of residents of Manaus were infected(attack rate AR≃76%)by October 2020,suggesting protective herd immunity had been *** this,an unexpected second wave of COVID-19 struck again in November and proved to be larger than the first,creating a catastrophe for the unprepared *** has been suggested that this could be possible if the second wave was driven by ***,it is widely reported that reinfections were at a low rate(before the emergence of Omicron),and reinfections tend to be ***,we use novel methods to model the epidemic from mortality data without considering reinfection-caused deaths and evaluate the impact of interventions to explain why the second wave *** method fits a“flexible”reproductive numberR_(0)(t)that changes over the epidemic,and it is demonstrated that the method can successfully reconstruct R_(0)(t)from simulated *** Manaus,the method finds AR≃34%by October 2020 for the first wave,which is far less than required for herd immunity yet in-line with seroprevalence *** work is complemented by a two-strain *** genomic data,the model estimates transmissibility of the new P.1 virus lineage as 1.9 times higher than that of the ***,an age class model variant that considers the high mortality rates of older adults show very similar *** models thus provide a reasonable explanation for the two-wave dynamics in Manaus without the need to rely on large reinfection rates,which until now have only been found in negligible to moderate numbers in recent surveillance efforts.

The Dependence between Solar Flare Emergence and the Average Background Solar X-Ray Flux Emission

Solar flares, sudden bursts of intense electromagnetic radiation from the Sun, can significantly disrupt technological infrastructure, including communication and navigation satellites. To mitigate these risks, accurate forecasting of solar activity is crucial. This study investigates the potential of the Sun’s background X-ray flux as a tool for predicting solar flares. We analyzed data collected by solar telescopes and satellites between the years 2013 and 2023, focusing on the duration, frequency, and intensity of solar flares. We compared these characteristics with the background X-ray flux at the time of each flare event. Our analysis employed statistical methods to identify potential correlations between these solar phenomena. The key finding of this study reveals a significant positive correlation between solar flare activity and the Sun’s background X-ray flux. This suggests that these phenomena are interconnected within the framework of overall solar activity. We observed a clear trend: periods with increased occurrences of solar flares coincided with elevated background flux levels. This finding has the potential to improve solar activity forecasting. By monitoring background flux variations, we may be able to develop a more effective early warning system for potentially disruptive solar flares. This research contributes to a deeper understanding of the complex relationship between solar flares and the Sun’s overall radiative output. These findings indicate that lower-resolution X-ray sensors can be a valuable tool for identifying periods of increased solar activity by allowing us to monitor background flux variations. A more affordable approach to solar activity monitoring is advised.

Let’s Talk LGBTQIA+:Strategies and Activities for Teaching Diversity and Inclusion in the Foreign Language Classroom at the College Level

In this presentation,the author explores strategies and activities that create a welcoming and nurturing environment for LGBTQIA+foreign language students at the college level in the United ***,as it is commonly stated,there is a clear lack of visibility,acceptance/tolerance,and inclusion of LGBTQIA+voices(active and proactive)in foreign language teaching and learning(pedagogical material as well as in-class attitudes and delivery).The reasons for this deficiency are diverse since they cover not only outward discrimination against and/or indifference(passive-aggressive behavior)toward the LGBTQIA+community and its plight,but they also illustrate the sheer,physical inexistence of quality material that can be used to create,present,and foster a welcoming and all-encompassing classroom environment that addresses the diverse spectrum of the LGBTQIA+community while its members try to learn a foreign language and(positively)negotiate how to express their very identity in their new language of ***,the scant material that is available in the United States as well as in other Western countries where the LGBTQIA+community by and large is accepted-at least from the legal point of view-hardly represents and covers the subject matter in a satisfactory *** address this gap,the author will provide some examples in European Portuguese(EP)and Italian(IT),followed by a courtesy translation in English,to illustrate some positive activities and strategies for including LGBTQIA+-related subject matter in the foreign language classroom at the college *** is his desire that instructors of other foreign languages duplicate in their respective vernaculars the strategies and activities herein presented in order to create a welcoming and safe classroom environment that embraces the LGBTQIA+community as it goes through the enjoyable journey of learning a new language and culture.

Studying the membrane physiology of filamentous fungi is key to understanding their interactions with the environment and crucial for developing new therapeutic strategies for disease-causing ***,their plasma membrane has been inaccessible for a micron-sized patch-clamp pipette for pA current recordings due to the rigid chitinous cell ***,we report the first femtosecond IR laser nanosurgery of the cell wall of the filamentous fungi,which enabled patch-clamp measurements on protoplasts released from hyphae.A reproducible and highly precise(diffraction-limited,submicron resolution)method for obtaining viable released protoplasts was *** release from the nanosurgery-generated incisions in the cell wall was achieved from different regions of the *** plasma membrane of the obtained protoplasts formed tight and high-resistance(GΩ)contacts with the recording *** entire nanosurgical procedure followed by the patch-clamp technique could be completed in less than 1 *** to previous studies using heterologously expressed channels,this technique provides the opportunity to identify new ionic currents and to study the properties of the ion channels in the protoplasts of filamentous fungi in their native environment.
