Effect of recombinant human interleukin-11 on expressions of interleukin-11 receptorα-chain and glycoprotein 130 in intestinal epithelium cell line-6 after neutron irradiation

AIM： To explore the effect of recombinant human interleukin-11 （rhIL-11） on the expressions of interleukin-11 receptor α-chain （IL-11Rα） and an additional signal transducer glycoprotein 130 （gp130） in intestinal epithelium cell line-6 （IEC-6） after neutron irradiation. METHODS： Cultured IEC-6 cells were exposed to 4.0Gy neutron and treated with 100 ng/mL rhIL-11 12 h prior to or immediately after irradiation. The apoptosis and necrosis rates and expressions of IL-11Rα and gp130 were observed by flow cytometry, immunohistochemistry, Western blot and image analysis. RESULTS： The apoptosis rate of IEC-6 cells was increased by irradiation at 6 h （P 〈 0.01）, IL-11 stimulation resulted in a decreased apoptosis rate in irradiated IEC-6 cells （P 〈 0.05）. In normal control IEC-6 cells, intense immunoreactivity of IL-11Rα was located within the cell membrane and cytoplasm. The level of IL-11Rα expression significantly decreased at 6 h after irradiation （P 〈 0.01） and restored at 24 h after irradiation. In IEC-6 cells treated with both radiation and rhIL-11, the level of IL- 11Rα expression was higher than that of irradiated cells （P 〈 0.05）. When it came to gp130 protein, it was located in the cytoplasm of IEC-6 cells. After irradiation, we found a progressive decrease in the expression of gp130 protein （P 〈 0.05） in 48 hours post-radiation, while in rhIL-11-stimulated cells, it came back to normal level at 24 h after irradiation and decreased at 48 h, but was still higher than that of only irradiated cells （P 〈 0.05）. CONCLUSION： rhIL-11 can protect IEC-6 cells from neutron irradiation. The protective effect of rhIL-11 might be connected with its ability to up-regulate the expressions of specific ligand-binding subunit IL-11Rα and signal-transducing subunit gp130.

BACKGROUND In recent years,intraoperative radiotherapy(IORT)has been increasingly used for the treatment of rectal ***,the efficacy and safety of IORT for the treatment of rectal cancer are still *** To evaluate the value of IORT for patients with rectal *** We searched PubMed,Embase,Cochrane Library,Web of Science databases,and conference abstracts and included randomized controlled trials and observational studies on IORT vs non-IORT for rectal *** variables were evaluated by odds ratio(OR)and 95%confidence interval(CI),hazard ratio(HR)and 95%CI was used as a summary statistic of survival *** analyses were performed using Stata V.15.0 and Review Manager 5.3 *** In this study,3 randomized controlled studies and 12 observational studies were included with a total of 1460 patients,who are mainly residents of Europe,the United States,and *** results did not show significant differences in 5-year overall survival(HR=0.80,95%CI=0.60-1.06;P=0.126);5-year disease-free survival(HR=0.94,95%CI=0.73-1.22;P=0.650);abscess(OR=1.10,95%CI=0.67-1.80;P=0.713),fistulae(OR=0.79,95%CI=0.33-1.89;P=0.600);wound complication(OR=1.21,95%CI=0.62-2.36;P=0.575);anastomotic leakage(OR=1.09,95%CI=0.59-2.02;P=0.775);and neurogenic bladder dysfunction(OR=0.69,95%CI=0.31-1.55;P=0.369).However,the meta-analysis of 5-year local control was significantly different(OR=3.07,95%CI=1.66-5.66;P=0.000).CONCLUSION The advantage of IORT is mainly reflected in 5-year local control,but it is not statistically significant for 5-year overall survival,5-year disease-free survival,and complications.

Dear Editor,Multiple experiments have established TNF-α-induced protein 3 (A20/TNFAIP3) as a critical regulator associated with rheumatoid arthritis(ra)[1,2].The lack of TNF-α-induced protein 3 (A20) promotes the NOD-like receptor protein 3 (NLRP3) inflammasome and induces spontaneous arthritis,while increase of A20 reduces the secretion of IL-1β and favors immunological tolerance[3,4]. Hence, we investigate the feasibility of recombinant adeno-associated virus 6 (raAV6)-mediated A20 gene therapy in a collageninduced arthritis (CIA) model.

针对移动边缘计算(Mobile Edge Computing,MEC)网络中的多服务器联合任务卸载和资源分配问题,提出一种MEC多服务器启发式联合任务卸载和资源分配策略。将原始优化问题表述为一个混合整数非线性规划,并分解成具有固定任务卸载决策的资源分配(Resource Allocation,ra)优化问题和任务卸载(Task Offloading,TO)最优值函数问题。将ra问题进一步解耦为上行链路功率分配和计算资源分配两个独立子问题,并分别采用拟凸和凸优化技术加以解决。提出一种启发式算法解决TO问题,可以在多项式时间内实现目标问题的次优解。仿真结果表明,与其他优化策略相比,该策略能够很好地实现最优解,显著提高系统的平均卸载效率。

Fetal Hemodynamics and Fetal Growth Indices by Ultrasound in Late Pregnancy and Birth Weight in Gestational Diabetes Mellitus

Background： The offspring of women with gestational diabetes mellitus （GDM） are prone to macrosomia. However, birth weight is difficult to be correctly estimated by ultrasound because of fetal asymmetric growth characteristics. This study aimed to investigate the correlations between fetal hemodynamics, fetal growth indices in late pregnancy, and birth weight in GDM. Methods： A total of 147 women with GDM and 124 normal controls （NC） were enrolled in this study. Fetal hemodynamic indices, including the systolic/diastolic ratio （S/D）, resistance index （RI）, pulsatility index （PI） of umbilical artery （UA）, middle cerebral artery （MCA）, and renal artery （ra）, were collected. Fetal growth indices, including biparietal diameter （BPD）, head circumference （HC）, abdominal circumference （AC）, and femur length, were also measured by ultrasound. Birth weight, newborn gender, and maternal clinical data were collected. Results： The independent samples t-test showed that BPD, HC, and AC were larger in GDM than in NC （P 〈 0.05）. Fetal hemodynamic indices of the UA and MCA were lower （P 〈 0.05）, but those of the ra were higher （P 〈 0.001 ） in GDM than in NC. Birth weight was higher in GDM than in NC （P 〈 0.001）. Pearson＇s correlation analysis showed that hemodynamic indices of the UA were negatively correlated with birth weight, BPD, HC, and AC in both groups （P 〈 0.05）. MCA （S/D, PI, and RI） was negatively correlated with birth weight, HC, and AC in GDM （i- = -0.164, -0.206, -0.200, -0.226, -0.189, -0.179, -0.196, -0.177, and - 0.172, respectively, P 〈 0.05）, but there were no correlations in NC （P 〉 0.05）. ra （S/D, PI, and RI） was positively correlated with birth weight in GDM （r = 0.168, 0.207, and 0.184, respectively, P 〈 0.05）, but there were no correlations in NC （P 〉 0.05）. Conclusion： Fetal hemodynamic indices in late pregnancy might be helpful for estimating newborn birth weight in women with GDM.
