Hyperplasia and migration of fibroblast-like synoviocytes(FLSs)are the key drivers in the pathogenesis of rheumatoid arthritis(RA)and joint *** Yes-associated protein(yaP),which is a powerful transcription co-activator for proliferative genes,was observed in the nucleus of inflammatory FLSs with unknown upstream *** Gene Expression Omnibus database analysis,it was found that Salvador homolog-1(SAV1),the pivotal negative regulator of the Hippo-yaP pathway,was slightly downregulated in RA ***,SAV1 protein expression is extremely ***,it was revealed that SAV1 is phosphorylated,ubiquitinated,and degraded by interacting with an important serine-threonine kinase,G protein-coupled receptor(GPCR)kinase 2(GRK2),which was predominately upregulated by GPCR activation induced by ligands such as prostaglandin E2(PGE2)in *** process further contributes to the decreased phosphorylation,nuclear translocation,and transcriptional potency of yaP,and leads to aberrant FLSs *** depletion of GRK2 or inhibition of GRK2 by paroxetine rescued SAV1 expression and restored yaP phosphorylation and finally inhibited RA FLSs proliferation and ***,paroxetine treatment effectively reduced the abnormal proliferation of FLSs in a rat model of collagen-induced arthritis which was accompanied by a significant improvement in clinical ***,these results elucidate the significance of GRK2 regulation of Hippo-yaP signaling in FLSs proliferation and migration and the potential application of GRK2 inhibition in the treatment of FLSs-driven joint destruction in RA.

First mirror(FM)cleaning,using radio frequency(RF)plasma,has been proposed to recover FM reflectivity in nuclear fusion reactors such as the International Thermonuclear Experimental Reactor(ITER).To investigate the influence of simultaneous cleaning of two mirrors on mirror cleaning efficiency and uniformity,experiments involving single-mirror cleaning and dual-mirror cleaning were conducted using RF capacitively coupled plasma in the *** the test and simultaneous cleaning of two mirrors,the FM and second mirror(SM),both measuring 110 mm×80 mm,were placed inside the first mirror unit(FMU).They were composed of 16 mirror samples,each with a dimension of 27.5 mm×20 *** mirror samples consist of a titanium-zirconium-molybdenum alloy substrate,a 500 nm Mo intermediate layer and a 30 nm Al_(2)O_(3) surface coating as a proxy for Be *** cleaning of a single first mirror(SFM)and the simultaneous cleaning of the FM and SM(DFM and DSM)lasted for 9 h using Ar plasma at a pressure of 1 *** total reflectivity of mirror samples on the DSM did not fully recover and varied with location,with a self-bias of−140 *** a self-bias of−300 V,the total reflectivity of mirror samples on the SFM and DFM was fully *** energy dispersive spectrometer results demonstrated that the Al_(2)O_(3) coating had been completely removed from these mirror ***,the mass loss of each mirror sample on the SFM and DFM before and after cleaning varied depending on its location,with higher mass loss observed for mirror samples located in the corners and lower loss for those in the *** with SM cleaning,the simultaneous cleaning of two mirrors reduced the difference between the highest and lowest mass ***,this mass loss for the mirror samples of the DFM facing the DSM was *** indicated that mirror samples cleaned face to face in the FMU simultaneously could influence each other,highlighting the need for special attention in

A Johann-type X-ray spectrometer was successfully developed at the hard X-ray branch (in-vacuum undulator with a 24-mm periodic length) of the energy material beamline (E-line) at the Shanghai Synchrotron Radiation Facility (SSRF). This spectrometer was utilized to implement X-ray emission spectroscopy (XES), high-energy resolution fluorescence-detected X-ray absorption spectroscopy (HERFD-XAS), and resonant inelastic X-ray scattering. Shale oil of the Qingshankou Formation of the Gulong Sag,the northern Songliao Basin,represents the first attempt at large-scale development of pure-shale-type shale oil in *** integrating the multiscale refined reservoir characterization with macro-micro-scale mechanical testing,it is clarified that the Gulong shale is characterized by high clay mineral content,high rock plasticity,highly-developed bedding,and prominent mechanical anisotropy.A three-dimensional(3D)fracture propagation model of hydraulic fracturing was built for the Gulong shale,which fully captures the hydraulic fracture distribution pattern affected by the high bedding density,in-situ stress,and fracturing treatment *** research showed that due to influences of bedding,hydraulic fracturing in the Gulong shale forms a complex fracture morphology featuring the main fracture with multiple perpendicular branches that have different lengths(like the outdoor directional TV antenna);however,the vertical propagation of fractures is inhibited,and the fracture height is commonly less than 10 *** limited stimulated reservoir volume(SRV)is the main problem facing the fracturing stimulation of the Gulong shale *** density has vital effects on fracture morphology,so case-specific fracturing designs shall be developed for shale intervals with different bedding development *** reservoirs with welldeveloped bedding,it is suggested to properly increase the perforation cluster spacing and raise the volume and proportions of viscous fluids of the pad,so as to effectively promote vertical fracture propagation and improve reservoir stimulation *** study integrates multi-scale fine reservoir characterization and macro-micro-scale mechanical testing,as well as the construction and numerical simulation of hydraulic fracturing models for high-density layered shale reservoirs,providing a new approach and methodological framework for the fracturing research of high-density layer

Plant high-affinity K^(+) transporters(HKTs)mediate Na^(+) and K^(+) uptake,maintain Na^(+)/K^(+) homeostasis,and therefore play crucial roles in plant salt *** this study,we present cryoelectron microscopy structures of HKTs from two classes,classI HKT1;1 from Arabidopsis thaliana(AtHKT1;1)and class II HKT2;1 from Triticum aestivum(TaHKT2;1),in both Na^(+) -and K^(+) -bound states at 2.6-to 3.0-A ***1;1and TaHKT2;1function *** HKT subunit consists of four tan-dem domain units(D1-D4)with a repeated K^(+) -channel-like M-P-M *** each subunit,D1-D4 assemble into an ion conduction pore with a pseudo-four-fold *** both TaHKT2;1and AtHKT1;1 have only one putative Na^(+) ion bound in the selectivity filter with a similar coordination pattern,the two HKTs display different K^(+) binding modes in the ***2;1 has three K^(+) ions bound in the selec-tivity filter,but AtHKT1;1 has only two K^(+) ions bound in the filter,which has a narrowed external entrance due to the presence of a Ser residue in the first filter *** structures,along with compu-tational,mutational,and electrophysiological analyses,enable us to pinpoint key residues that are critical for the ion selectivity of *** findings provide new insights into the ion selectivity and ion transport mechanisms of plant HKTs and improve our understanding about how HKTs mediate plant salt tolerance and enhance crop growth.
