Mechanism of Wendan decoction in preventing obesity by regulating multiple signal pathway networks based on gene promoter methylation

Objective:To investigate the potential mechanism of Wendan decoction in obesity by screening target genes with promoter region methylation changes and constructing a multiple signaling pathways network based on promoter ***:The methylation degree of Itgad,Col8a1,Adra2b,Jund,Rab2a,Wnt8b,Fzd9,B4galt7,Pik3cd,Creb1,Stard8,and Mmp1 in the abdominal adipose tissue of obese rats was determined using the Agena MassARRAY *** blot was performed to assess protein expression *** genes were identified based on the methylation degree in the promoter region and protein *** analysis of signaling pathways was conducted to identify relevant target genes and obtain a multiple signaling pathway network associated with *** and terminal effector molecules in the pathway networks were selected as research targets for reverse transcription-polymerase chain reaction(RT-PCR)***:Four genes(Adra2b,Creb1,Itgad,and Pik3cd)showed a degree of promoter methylation consistent with their respective protein expression *** them,Adra2b,Creb1,and Pik3cd expression increased,while that of Itgad *** analysis revealed that Creb1 and Pik3cd were involved in 6 signaling pathways related to obesity:tumor necrosis factor(TNF)signaling pathway,growth hormone synthesis/secretion and action,adenosine 5'-monophosphate-activated protein kinase(AMPK)signaling pathway,relaxin signaling pathway,cyclic nucleotide(cAMP)signaling pathway,and phosphatidylinositol 3-kinase/protein kinase B(PI3K/Akt)signaling ***,a multiple signaling pathways network was constructed based on promoter *** molecules including protein kinase B(AKT),mechanistic target of rapamycin complex 1(mTORC1),and unc-51 like autophagy activating kinase 1(ULK1),as well as terminal effector molecules interleukin-1β(IL-1β),interleukin-6(IL-6),and chemokine(C-X-C motif)ligand 2(CXCL2)were selected as research *** decoct

In 2022,Yellow Sea green tide caused by Ulva prolifera reached a historic minimum and the coastal areas of Shandong Peninsula were less ***,the largest amount of seaweed biomass has been washed ashore on Lianyungang and Rizhao coasts since *** studied the development pattern of Yellow Sea green tide in 2022,and analyzed the key environmental factors on the growth and drifting,then discussed the possible reasons that resulted in the massive stranding of green tide biomass in Lianyungang and *** show under the combined influence of the east to southeast winds and currents with shoreward anomalies,green tide drifted to the coastal waters between Shandong and Jiangsu provinces and the distribution areas located westward compared with previous years(2008–2021).Floating *** rafts from the coastal waters of Binhai and Sheyang drifted continuously into the coastal waters of Lianyungang and Rizhao,providing important supplements for Yellow Sea green *** green tide in 2022 distributed close to the coastal waters,the abundant nutrients might support their continuous high growth *** addition,the amount of rainfall around Shandong Peninsula from late June to early July were significantly higher than in previous years,which might promote the development of green tide to some extent.

The Zhongjiang gas field is a typical large gas field in terrigenous strata of the Western Sichuan *** remains debatable which member of the Upper Triassic Xujiahe Formation served as the source rocks and how significant the member contributed to the gas accumulations in the Zhongjiang gas *** this study,we analyzed the essential characteristics of the Lower Jurassic source rocks and the geochemical features of light hydrocarbons in natural gas from the 2nd(T_(3)χ^(2))and 4th members(T_(3)χ^(4))of the Upper Triassic Xujiahe Formation(T_(3)χ),as well as the Middle Jurassic Shaximiao(J_(2)s)and Qianfoya(J_(2)q)*** on this,we explored the sources of the natural gas in the Zhongjiang gas field and determined the natural gas migration patterns and their effects on the properties of light hydrocarbons in the natural *** results indicate that the Lower Jurassic lacustrine source rocks of the Zhongjiang gas field contain humic organic matter,with vitrinite reflectance(R_(0))values ranging from 0.86%to 0.98%.Samples meeting the criterion for effective source rocks[total organic carbon(TOC)content≥0.75%]exhibited an average TOC content of merely 1.02%,suggesting significantly lower hydrocarbon generation potential than source rocks in the underlying T3x,which show higher thermal maturity and TOC *** natural gas samples from T_(3)χ^(2),T_(3)χ^(4),J_(2)s,and J_(2)q reservoirs,their C_(5-7)iso-alkane content was significantly higher than their n-alkane content,and their methylcyclohexane(MCH)index ranged from 59.0%to 77.3%,indicating the predominance of methylcyclohexane in C_(7)light *** indicated by the origin identification and gas-source correlation based on the geochemical features of light hydrocarbons,the natural gas in the Zhongjiang gas field is typical coal-derived *** gas from the primary pay zone of the Shaximiao Formation,with significantly high K_(1),(P_(2)+N_(2))/C_(7),and P_(3)/C_(7)values,predominantly ori

Continuous-flow synthesis of polysubstituted γ-butyrolactones via enzymatic cascade catalysis

Polysubstituted chiral γ-butyrolactones are the core structural units of many natural products and high value-added flavors and fragrances used in the food and cosmetic industry. Current enzymatic cascade synthesis of these molecules faces the problems of low enzyme activity and phase separation in batch reaction, resulting in low productivity. Herein, we report a new continuous-flow process to synthesize the optically pure Nicotiana tabacum lactone(3S,4S)-4a and whisky lactone(3R,4S)-4b from α,β-unsaturatedγ-ketoesters. A new ene reductase(ER) from Swingsia samuiensi(Ss ER) and a carbonyl reductase(Ss CR)were engineered by directed evolution to improve their activity and thermostability. The continuous-flow preparative reactions were performed in two 3D microfluidic reactors, generating(3S,4S)-4a(99% ee and87% de) and(3R,4S)-4b(99% ee and 98% de) with space-time yields 3 and 7.4 times higher than those of the batch reactions. The significant enhancement in the productivity of enzyme cascade catalysis brought by cutting-edge continuous microfluidic technology will benefit the general multi-enzyme catalytic systems in the future.

Nitrogen-doped carbon loaded single-atom catalysts(SACs)are promising candidates for electrocatalytic conversion of CO_(2)into high-valuable chemicals,and the modification of catalysts by heteroatom-doping strategy is an effective approach to enhance the CO_(2)reduction ***,the large difference exists in atomic radius between nitrogen atoms and the doped heteroatoms may lead to the poor stability of active *** this study,we have synthesized a Ni single atom catalyst with S doping at the secondshell on the ultrathin carbon nanosheets support(Ni-N_(4)-SC)by solid-phase *** S atom in the second-shell contributes to the higher efficiency of CO_(2)conversion at lower potentials while the Ni-N_(4)-SC can be more *** experimental results and theoretical calculations indicate that the S atom in second-shell breaks the uniform charge distribution and reduces the free energy of hydrogenation,which can increase the adsorption of CO_(2),accelerate charge transfer,and reduce the reaction energy *** work reveals the close relationship between the second-shell and the electrocatalytic activity of single atom sites,which also provides a new perspective to design efficient single atom catalysts.

In nuclear collisions at RHIC energies, an excess of Ω hyperons over ■ is observed, indicating that Ω has a net baryon number despite s and s quarks being produced in pairs. The baryon number in Ω may have been transported from the incident nuclei and/or produced in the baryon-pair production of Ω with other types of anti-hyperons such as Ξ. To investigate these two scenarios, we propose to measure the correlations between Ω and K and between Ω and anti-hyperons. We use two versions, the default and string-melting, of a multiphase transport(AMPT) model to illustrate the method for measuring the correlation and to demonstrate the general shape of the correlation. We present the Ω-hadron correlations from simulated Au+Au collisions at ■ =7.7 and 14.6 Ge V and discuss the dependence on the collision energy and on the hadronization scheme in these two AMPT versions. These correlations can be used to explore the mechanism of baryon number transport and the effects of baryon number and strangeness conservation on nuclear collisions.

Mechanical properties of semi-solid casting are dependent on multiple processing parameters,and improper processing parameters will not only reduce mean data but also increase *** present study investigated the impact of parameters in slurry preparation and heat treatment on the yield strength and ductility of T6 heat-treated A356 Al-Si alloy using rapid slurry forming(RSF)semi-solid *** focus was primarily on the robustness of mechanical properties based on Taguchi design *** analyzing signal-to-noise ratio and minimum value calculated from-3S,the optimum slurry preparation parameters and heat treatment parameters were determined to be no quench,enthalpy exchange material(EEM)temperature of 140℃,EEM-to-melt ratio of 6mass%,stirring time of 18 s,solution heat treated at 520℃ for 2 h,and ageing heat treated at 190℃ for 6 *** a small batch validation,the-3S yield strength and-3S elongation reach 256.1 MPa and 5.03% respectively,showing a satisfactory *** hardness and microstructure of heat-treated samples with the best and worst properties were characterized to gain insight into the underlying mechanisms affecting the mean value and variations of mechanical properties.
