Constitutive Overexpression of Bcl-2, Survivin and ER Stress Chaperone GRP-78 Confers Intrinsic Radioresistance in Human Hepatocellular Carcinoma Cells: Insight into the Mechanistic Pathways Involved

We present evidence here that abundantly expressed b-catenin-triggered NF-kB-dependent upregulation of inducible nitric oxide synthase(iNOS) found in hepatoma Mahlavu cells (RT-resistant variant designated as RR-Mal), but not in Hep 3B cells (RT-sensitive variant designated as RS-3B) is a key element contribrting to the radioresisitance through the activation of two prominent radioprotective pathways. First, high iNOS expression found in RR-Mal, but not in RS-3B cells was found to perturb calcium homeostasis that triggered ER stress response leading to the overproduction of ER chaperone GRP-78 via robust generation of cleaved ATF-6a (50 kDa) subunits and their nuclear translocation. Meanwhile, both abundantly expressed NF-κB and COX-2 found in RR-Mal cells could also provoke an increased production of PGE2 resulting in robust production of Bcl-2. Interestingly, when RR-Mal cells were treated with PDTC (a NF-κB inhibitor) or celecoxib (a COX-2 inhibitor), a concentration-dependent downregulation of Bcl-2 could be demonstrated implying that Bcl-2 overexpression was indeed mediated through NF-κB/Cox-2/PGE2 pathway. Importantly, we also unveiled that siRNA-mediated silencing of survivin in RR-Mal cells could result in a concomitant downregulation of GRP-78 due to a severe inhibition of ATF-6a (50 kDa) expression. Taken together, our data demonstrate that constitutively overexpressed b-catenin/NF-κB/iNOS and NF-κB/COX-2/PGE2 pathways that overproducing GRP-78, survivin and Bcl-2 expressions are responsible for radioresistance acquisition in RR-Mal cells. Thus, both pathways could be served as potential targets for overcoming radioresistance.

Nickel sulfide exhibits excellent catalytic activity in the electrochemical 2,5-hydroxymethylfurfural oxidation reaction(HMFOR).However,due to the polydispersity of nanoparticles,it is difficult to establish a clear structure-activity relationship at the atomic *** this work,we have successfully synthesized atomically precise Ni_(6)(PET)_(12)and Ni_(4)(PET)_(8)clusters(PET:2-phenylethanethiol)for ***^(2+)and S_(2)-with atomic ratio of 1:2 was mainly existed in Ni_(6)(PET)_(12)and Ni_(4)(PET)_(8)to form Ni-S *** electrochemical test results have suggested both Ni_(6)(PET)_(12)and Ni_(4)(PET)_(8)displayed outstanding electrocatalytic ability for *** Ni_(6)(PET)_(12)exhibited better electrocatalytic ability than Ni_(4)(PET)_(8)with higher current density,lower overpotential and faster reaction *** superior electrochemical ability of Ni_(6)(PET)_(12)may be due to the enhanced adsorption towards HMF molecule with strong interaction towards hydroxyl group and furan ***,it found that the Ni^(2+)species in Ni_(6)(PET)_(12)could rapidly oxidized into Ni^(3+)species,which could spontaneously capture electron and proton from HMF for *** theoretical calculation demonstrated that the Ni_(6)(PET)_(12)process lower free energy barrier than Ni_(4)(PET)_(8)to display excellent electrocatalytic *** work is of great significance for designing efficient electrocatalysts for HMFOR.

Searching for single-particle resonances with the Green’s function method

Single-particle resonances in the continuum are crucial for studies of exotic *** this study,the Green’s function approach is employed to search for single-particle resonances based on the relativistic-mean-field ***^(120)Sn as an example,we identify singleparticle resonances and determine the energies and widths directly by probing the extrema of the Green’s *** contrast to the results found by exploring for the extremum of the density of states proposed in our recent study[***.C,44:084105(2020)],which has proven to be very successful,the same resonances as well as very close energies and widths are *** comparing the Green’s functions plotted in different coordinate space sizes,we also found that the results very slightly depend on the space *** findings demonstrate that the approach by exploring for the extremum of the Green’s function is also very reliable and effective for identifying resonant states,regardless of whether they are wide or narrow.

Fully enclosed hybrid electromagnetic-triboelectric nanogenerator to scavenge vibrational energy

We propose a fully enclosed hybrid nanogenerator consisting of five elec- tromagnetic generators （EMGs） and four triboelectric nanogenerators （TENGs）. Under a vibration frequency of 15.5 Hz, one TENG can deliver a high output voltage of approximately 24 V and a low output current of approximately 24 μA, whereas one EMG can deliver a low output voltage of approximately 0.8 V and a high output current of approximately 0.5 mA. By integrating five rectified EMGs in series and four rectified TENGs in parallel, the hybrid nanogenerator can be used to charge a home-made Li-ion battery from 1 to 1.9 V in 6.3 h. By using the hybrid nanogenerator to scavenge the vibrational energy produced by human hands, a temperature-humidity sensor can be sustainably powered by the nanogenerator, which is capable of charging the 200 ~F system power capacitor from 0 to 2 V in 15 s, and sustainably power the sensor in 29 s.

16.48% Efficient solution-processed CIGS solar cells with crystal growth and defects engineering enabled by Ag doping strategy

Solution-processed Cu（In,Ga）Se2（CIGS） solar cells suffer from serious carrier recombination and power conversion efficiency（PCE） loss because of the poor film properties and easy formation of ***, we propose Ag&Se co-selenization strategy to enhance the crystallization and passivate harmful defects of the CIGS films. The formation of Ag-Se phase during the selenization process enables the formation of large grains and suppresses the deep level defects. It is found that Ag doping can enlarge the depletion region width, lower the Urbach energy and prolong the carrier lifetime. As a result, a champion solution-processed CIGS solar cell presents a high efficiency of 16.48% with the highly improved opencircuit voltage（VOC） of 662 m V and fill factor（FF） of 75.8%. This work provides an efficient strategy to prepare high quality solution-processed CIGS films for high-performance CIGS solar cells.

Phase Separation, Wetting and Dewetting in PS/PVME Blend Thin Films:Dependence on Film Thickness and Composition Ratio

The effects of film thickness and composition ratio on the morphology evolution of polystyrene （PS）/poly（vinyl methyl ether）（PVME） blend thin films were investigated. Diverse morphology evolutions including droplet-matrix structure, hole emergence, bicontinuous structure formation, percolation-to-droplet transition could be observed under annealing in two-phase region, depending on film thickness and composition ratio. The mechanism for these morphology variations was related to the complex effects of phase separation, dewetting and preferential wetting. The comparison between the thickness of bottom PVME layer and the twice of gyration radius 2Rg（PVME） played a dominant role in morphology control. Only when the PS/PVME film had specific film thickness and compositional symmetry, phase separation and dewetting could happen in sequence.

A rational design of copper(Cu)nanocatalyst is critical to the efficient carbon dioxide(CO_(2))conversion toward C1/C2 chemical ***,the lack of actual information on the vicinal Cu interfaces hinders the intrinsic activitystructure correlation *** with atomic-scale in situ characterizations(AP-STM and synchrotron-based AP-XPS),Park and co-workers study and identify the adsorbate-driven morphological evolution of Cu nanoclusters on the Cu(997)surface,providing real-space evidence for the critical role of vicinal structures on Cu *** observed surface roughening trends caused by dissociated atomic oxygen and CO adsorbates subsequently affect the result of successive catalytic reactions to produce critical intermediates,such as carbonate and formate,during the CO_(2)reduction reaction.

With the development of information technology,rumors propagate faster and more widely than in the *** this paper,a stochastic rumor propagation model incorporating media coverage and driven by Lévy noise is *** global positivity of the solution process is proved,and further the basic reproductive number R_(0) is *** R_(0)1,the rumor will keep spreading and the system will oscillate randomly near the rumor equilibrium point of the deterministic *** results show that the oscillation amplitude is related to the disturbance of the *** addition,increasing media coverage can effectively reduce the final spread of ***,the above results are verified by numerical simulation.

Effects of Neuregulins on Invasion and Metastasis of Non-overexpression ErbB2 Breast Cancer Cells

Objective： To explore the effects of neuregulins on ErbB2 receptor signal transduction pathway activation, and invasion and metastasis of non-overexpression ErbB2 breast cancer cell MDA-MB-231. Methods： The expressions of neuregulin were detected by immunocytochemistry and Western blot. MDA-MB-231 cells were treated with ErbB2 kinase inhibitor AG825. Proliferations were measured with MTT assay. Invasion and metastasis of MDA-ME-231 cells were evaluated with transwell chamber. The enzyme activities of MMP-2 and MMP-9 were detected by gelatin zymography. The expressions of MMP-2 and HIF-1α were detected by Western blot. Results： MDA-MB-231 cells expressed a relatively higher level of neuregulin. In Western blot, the positive reaction band was found at 44KD which coincides with the molecular weight of NRG. When MDA-MB-231 cells were treated with AG825, the proliferation was inhibited in a time-dose-dependent manner （P〈0.01）, invasion and metastasis were also depressed （P〈0.05）. The enzyme activities of MMP-2 and MMP-9 were lower （P〈0.05）. The expression levels of MMP-2 and HIF-lct were decreased （P〈0.05）. Conclusion： Our study indicates that neuregulins are synthesized in MDA-MB-231 cells as transmembrane proteins, neuregulins could activate ErbB2 receptor signal transduction pathway by autocrine or paracrine secretion, and induce invasion and metastasis of MDA-MB-231 cells.
