Comparative proteome analysis of Helicobacter pylori clinical strains by two-dimensional gel electrophoresis

Objective： To investigate the pathogenic properties of Helicobacterpylori by comparing the proteome map of H. pylori clinical strains. Methods： Two wild-type H. pylori strains, YN8 （isolated from biopsy tissue of a gastric cancer patient） and YN14 （isolated from biopsy tissue of a gastritis and duodenal ulcer patient）, were used. Proteomic analysis, using a pH range of 3-10 and 5-8, was performed. The individual proteins were identified by quadrupole time-of-flight （Q-TOF） mass spectrometer and protein database search. Results： Variation in spot patterns directed towards differential protein expression levels was observed between the strains. The gel revealed prominent proteins with several protein ＂families＂. The comparison of protein expressions of the two strains reveals a high variability. Differentially present or absent spots were observed. Nine differentially expressed protein spots identified by Q-TOF included adenosine triphosphate （ATP）-binding protein, disulfide oxidoreductase B （DsbB）-Iike protein, N utilization substance A （NusA）, ATP-dependent protease binding subunit/heat shock protein, hydantoin utilization protein A, seryl-tRNA synthetase, molybdenum ABC transporter ModD, and hypothetical proteins. Conclusions： This study suggests that H. pylori strains express/repress protein variation, not only in terms of the virulence proteins, but also in terms of physiological proteins, when they infect a human host. The difference of protein expression levels between H. pylori strains isolated from gastric cancer and gastritis may be the initiator of inflammation, and result in the different clinical presentation. In this preliminary study, we report seven differential proteins between strains, with molecule weights from approximately 10 kDa to approximately 40 kDa. Further studies are needed to investigate those proteins and their function associated with H. pylori colonization and adaptation to host environment stress.

This paper proposes a highly sensitive,compact,and low-cost optical fiber salinity sensor based on the Mach-Zehnder *** sensor is constructed using a single mode fiber(SMF)-no-core fiber-double-C fiber(DCF)-NCF-SMF structure,with the DCF prepared by etching the dual side-hole fiber with HF *** DCF’s large-size exposed microfluidic channels solve the previous microstructured optical fiber’s challenging liquid filling and replacement *** simulations and experiments demonstrate that the sensor is suitable for high-sensitivity salinity *** sensor exhibits a high salinity sensitivity of-2.26 nm/‰ in the salinity range of 10‰-50‰,as demonstrated by the experimental ***,the sensor exhibits some fascinating characteristics,including high repeatability,hysteresis,reversibility,and stability.

Maternal Murine Cytomegalovirus Infection during Pregnancy Up-regulates the Gene Expression of Toll-like Receptor 2 and 4 in Placenta

Increasing evidence has revealed that maternal cytomegalovirus （CMV） infection may be associated with neurodevelopmental disorders in offspring. Potential relevance between the placental inflammation and CMV-related autism has been reported by clinical observation. Meanwhile, abnormal expression of Toll-like receptor 2 （TLR2） and TLR4 in placenta of patients with chorioamnionitis was observed in multiple studies. IL-6 and IL- 10 are two important maternal inflammatory mediators involved in neurodevelopmental disorders. To investigate whether murine CMV （MCMV） infection causes alterations in placental IL-6/10 and TLR2/4 levels, we analyzed the dynamic changes in gene expression of TLR2/4 and IL-6/10 in placentas following acute MCMV infection. Mouse model of acute MCMV infection during pregnancy was created, and pre-pregnant MCMV infected, lipopolysaccharide （LPS）-treated and uninfected mice were used as controls. At E13.5, E 14.5 and E 18.5, placentas and fetal brains were harvested and mRNA expression levels of placental TLR2/4 and IL-6/10 were analyzed. The results showed that after acute MCMV infection, the expression levels of placental TLR2/4 and IL-6 were elevated at E13.5, accompanied by obvious placental inflammation and reduction of placenta and fetal brain weights. However, LPS 50 ktg/kg could decrease the IL-6 expression at E13.5 and E14.5. This suggests that acute MCMV infection during pregnancy could up-regulate the gene expression of TLR2/4 in placental trophoblasts and activate them to produce more pro- inflammatory cytokine IL-6. High dose of LPS stimulation （50 gg/kg） during pregnancy can lead to down-regulation of IL-6 levels in the late stage. Imbalance of IL-6 expression in placenta might be associated with the neurodevelopmental disorders in progeny.

The simultaneous high-capacity and fast removal of radioactive barium ion(Ba^(2+))from nuclear wastewater is necessary but still faces large *** this study,a novel adsorbent containing sulfonic group,MOF-808-SO_(3)H,was prepared based on the oxidation of-SH *** confirm the successful introduction of-SO_(3)H,good porosity,and stability of the *** adsorbent exhibits excellent capture ability for Ba^(2+)due to strong hard acid-base and electrostatic interactions,with the adsorption capacity of 152.0 mg g^(-1)and fast equilibrium adsorption time,superior to other reported *** pH was found to be almost neutral and temperature cannot affect the adsorption ***,the irreversible adsorption was demonstrated and thereby the secondary pollution may be ***,our work provides an efficient adsorbent for capturing Ba^(2+)irreversibly,and proposes a feasible strategy for constructing adsorbents with simultaneous high adsorption capacity and fast kinetics.

Short-lived AIM2 Inflammasome Activation Relates to Chronic MCMV Infection in BALB/c Mice

Absent in melanoma 2(AIM2)inflammasome is a crucial link bridging the innate host defense and the subsequent adaptive immunity when activated by exogenous double stranded DNA(dsDNA).Through establishing models of disseminated murine cytomegalovirus(MCMV)infection in BALB/c and C57BL/6 mice,we evaluated dynamic expression of AIM2 inflammasome components and its relationship with pathological damage and viral replication,trying tofigure out whether AIM2 inflammasome is related to the chronic mechanism of ***/c and C57BL/6 mice were sacrificed on day 0,1,3,7,14 and 28 post *** levels of AIM2,pro-caspase-1,caspase-1 p20,pro-IL1β and mature IL1β in primary peritoneal macrophages(PMs)and spleens were detected by Western *** of IL18 in the serum were detected by *** examinations of livers were performed,and mRNA levels of MCMV glycoprotein B(gB)in salivary glands also *** showed that expression levels of AIM2 in PMs and spleens of C57BL/6 mice increased on day 3,even continued to day 28;caspase-1 p20 and mature IL1β increased on day 7,14 and 28;the persistently high expression of IL1β in the serum started on day 1,showing a double peak *** for BALB/c mice,expression of AIM2 in PMs increased on day 1 and day 7,while contents of AIM2 in spleens increased on day 1 and day 3;caspase-1 p20 and mature ILip merely increased 7 days fter ***,expression levels of AIM2,caspase-1 p20,mature IL1β and IL18 were limited;the duration of AIM2 inflammasome activation in BALB/c mice was much shorter than that in C57BL/6 *** severer pathological damage and more viral replications in BALB/c mice further proved the deficient antiviral immunity to *** conclusion,the activation of AIM2 inflammasome in BALB/c mice was short-lived,which is quite possibly related to the chronicity of MCMV infection.

In Vitro Inhibition of Alphaviruses by Lycorine

Chikungunya virus(CHIKV) is a mosquito-borne alphavirus. As an emerging virus, CHIKV imposes a threat to public health. Currently, there are no vaccines or antivirals available for the prevention of CHIKV infection. Lycorine, an alkaloid from Amaryllidaceae plants, has antiviral activity against a number of viruses such as coronavirus, flavivirus and enterovirus. In this study, we found that lycorine could inhibit CHIKV in cell culture at a concentration of 10 lmol/L without apparent cytotoxicity. In addition, it exhibited broad-spectrum anti-alphavirus activity, including Sindbis virus(SINV),Semliki Forest virus(SFV), and Venezuelan equine encephalomyelitis virus(VEEV). The time of addition studies indicated that lycorine functions at an early post-entry stage of CHIKV life cycle. The results based on two different CHIKV replicons provided further evidence that lycorine exerts its antiviral activity mainly by inhibiting CHIKV ***, our study extends the antiviral spectrum of lycorine.

Effect of BRM S1 expression on proliferation, migration and adhesion of mouse forestomach carcinoma

Objective: To discuss the ef ect of BRMS1 on the proliferation, migration and adhesion of mouse forestomach carcinoma(MFC). Methods: The constructed p CMV-myc-BRMS1 recombinant plasmid and blank plasmid were transfected into mouse forestomach carcinoma. MTT method was employed to measure the activity of gastric cancer cell; the scratch assay and Transwell assay to measure the migration and invasion of gastric cancer cell; the adhesion assay to measure the adhesion of gastric cancer cell; while the Western blot assay to measure the expression of The NF-毷B signal pathway, downstream matrix metalloproteinase(MMP-2), MMP-9 and osteopontin and E-cadherin in the gastric cancer cell. Besides, the transplanted animal model of gastric cancer in mice was constructed to measure the size of tumor xenograft. Results: Results of MTT assay showed that, compared with the empty vector control group, the activity of gastric cancer cell was not af ected in the BRMS1 transfection group. The improved expression of BRMS1 could inhibit the adhesion, migration and invasion of gastric cancer cell(P<0.01). Besides, compared with the empty vector control group, the phosphorylation of NF-毷B p65 and I毷Bα was reduced in the BRMS1 transfection group, with the decreased expression of MMP 2, MMP 9 and osteopontin and the increased expression of E-cadherin(P<0.01). Results of animal experiment also showed that the expression of BRMS1 did not af ect the transplanted tumor. Conclusions: The expression of BRMS1 can signii cantly inhibit the adhesion, migration, invasion and metastasis of MCF gastric cancer cell, which is related to The NF-毷B signal pathway.

The lung is the prophylaxis target against SARS-CoV-2 infection,and neutralizing antibodies are a leading class of biological products against various infectious viral *** this study,we develop a safe and cost-effective platform to express neutralizing antibody in the lung with replicating mRNA basing on alphavirus replicon particle(VRP)delivery system,to prevent SARS-CoV-2 ***,a modified VEEV replicon with two subgenomic(sg)promoters was engineered to translate the light and heavy chains of antibody simultaneously,for expression and assembly of neutralizing anti-SARS-CoV-2 antibody ***,the feasibility and protective efficacy of replicating mRNA against SARS-CoV-2 infection were demonstrated through both in vitro and in vivo *** lung target delivery with the help of VRP system resulted in efficiently block SARS-CoV-2 infection with reducing viral titer and less tissue damage in the lung of ***,our data suggests that expressing neutralizing antibodies in the lungs with the help of self-replicating mRNA could potentially be a promising prophylaxis approach against SARS-CoV-2 infection.

Dear Editor,Omsk hemorrhagic fever virus(OHFV)belongs to the tickborne virus group within Flavivirus,in Flaviviridae family(Holbrook et al.2005)which consists of several important human pathogens including tick-borne encephalitis virus(TBEV),Powassan virus(POWV),Louping ill virus(LIV),Langat virus(LGTV)and Kyasnaur Forest disease virus(KFDV)(Yoshii and Holbrook 2009).OHFV is an enveloped virus containing a single-stranded positive-sense RNA genome of approximately 11,000 *** genome has a single open reading frame,which encodes three structural proteins[capsid(C),pre-membrane(prM)or membrane(M),and envelop(E)]and seven non-structural proteins(NS1,NS2a,NS2b,NS3,NS4a,NS4b and NS5)between the 5'untranslated region(UTR)and 3'UTR(Chambers et al.1990).

Dear Editor,Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),has spread rapidly and developed into a global pandemic since its outbreak in December ***,there is no antiviral treatment available for human *** compounds,such as remdesivir and chloroquine,have been reported to inhibit SARS-CoV-2 replication effectively in vitro,but for most of them,the in vivo efficacies against SARS-CoV-2 are still under clinical studies,and for chloroquine,a drug with prominent in vitro antiviral activity,it has been found no beneficial effect for COVID-19 patients in the recent largest *** is thus urgent to speed up large-scale screening to discover drug candidates to treat COVID-19.
