Succinylation modification:a potential therapeutic target in stroke

Stroke is a leading cause of mortality and disability *** cell death triggered by the compromised supply of blood oxygen and glucose is one of the major pathophysiology of strokeinduced brain *** mitochondrial energy metabolism is observed minutes after stroke and is closely associated with the progression of ***,a new type of posttranslational modification,known as lysine succinylation,has been recognized to play a significant role in mitochondrial energy metabolism after ***,the role of succinylation modification in cell metabolism after stroke and its regulation are not well *** aimed to review the effects of succinylation on energy metabolism,reactive oxygen species generation,and neuroinflammation,as well as Sirtuin 5 mediated desuccinylation after *** also highlight the potential of targeting succinylation/desuccinylation as a promising strategy for the treatment of *** succinylation level is dynamically regulated by the nonenzymatic or enzymatic transfer of a succinyl group to a protein on lysine residues and the removal of succinyl catalyzed by *** evidence has suggested that succinylation can regulate the metabolic pathway through modulating the activity or stability of metabolic ***,especially Sirtuin 5,are characterized for their desuccinylation activity and have been recognized as a critical regulator of metabolism through desuccinylating numerous metabolic *** between succinylation and desuccinylation has been implicated in the pathophysiology of *** agents that enhance the activity of Sirtuin 5 have been employed to promote desuccinylation and improve mitochondrial metabolism,and neuroprotective effects of these agents have been observed in experimental stroke ***,their therapeutic efficacy in stroke patients should be validated.

Development and validation of a novel non-invasive test for diagnosing nonalcoholic fatty liver disease in Chinese children

With the exploding prevalence of obesity,many children are at risk of developing nonalcoholic fatty liver *** anthropometric and laboratory parameters,our study aimed to develop a model to quantitatively evaluate liver fat content(LFC)in children with *** A well-characterized cohort of 181 children between 5 and 16 years of age were recruited to the study in the Endocrinology Department as the derivation *** external validation cohort comprised 77 *** assessment of liver fat content was performed using proton magnetic resonance *** and laboratory metrics were measured in all subjects.B-ultrasound examination was carried out in the external validation *** Kruskal-Wallis test,Spearman bivariate correlation analyses,univariable linear regressions and multivariable linear regression were used to build the optimal predictive *** The model was based on indicators including alanine aminotransferase,homeostasis model assessment of insulin resistance,triglycerides,waist circumference and Tanner *** adjusted R^(2)of the model was 0.589,which presented high sensitivity and specificity both in internal[sensitivity of 0.824,specificity of 0.900,area under curve(AUC)of 0.900 with a 95%confidence interval:0.783–1.000]and external validation(sensitivity of 0.918 and specificity of 0.821,AUC of 0.901 with a 95%confidence interval:0.818–0.984).Conclusions Our model based on five clinical indicators was simple,non-invasive,and inexpensive;it had high sensitivity and specificity in predicting LFC in ***,it may be useful for identifying children with obesity who are at risk for developing nonalcoholic fatty liver disease.

Effect of streamlined nose length on aerodynamic performance of high-speed train with a speed of 400 km/h

The streamlined nose length(SNL)plays a crucial role in determining the aerodynamic performance of high-speed *** appropriate SNL can not only effectively reduce the magnitude of aerodynamic drag and lift forces,but also improve the performance of the high-speed train in tunnel passing and crosswind *** this study,a numerical simulation of the aerodynamic performance of high-speed trains at a speed of 400 km/h,with varying SNLs,is conducted using the k-ωshear stress transport(SST)turbulence *** different SNLs include 6.0,7.0,8.0,9.0,9.8,12.0,15.0,and 18.0 *** order to validate the accuracy of the numerical simulation,its results are compared with wind tunnel test data obtained from the *** simulation is carried out using compressible and incompressible gases to determine the effect of gas compressibility on *** impact of SNL on the aerodynamic performance of the trains is analyzed in terms of aerodynamic forces,velocity,and pressure *** comparison to the original train,the train with a 6.0 m SNL experienced a 10.8%increase in overall aerodynamic ***,the lift forces on the head and tail cars increased by 35.7%and 75.5%,*** the other hand,the train with an 18.0 m SNL exhibited a 16.5%decrease in aerodynamic ***,the lift forces on the head and tail cars decreased by 21.9%and 49.7%,*** aerodynamic drag force of the entire train varies linearly with the SNL,while the aerodynamic lift of the tail car follows a quadratic function in relation to the SNL.

Polymers have become an essential part of modern life and the global economy on account of their costeffectiveness and versatile ***,most postconsumer polymer wastes are unrecycled,leading to environmental pollution and resource ***,as an efficient chemical recycling approach,holds great promise in establishing a circular polymer *** this review,we attempt to highlight recent and significant advancements in depolymerization *** key research topics are discussed:(1)depolymerization of commodity polymers to produce reusable monomers and high-value chemicals;(2)depolymerization of intrinsically depolymerizable *** is anticipated that this review will reflect the present status and future trends of this rapidly evolving realm of depolymerization.

Enhanced bright green luminescence from GdOF:Ho^(3+) up-conversion phosphor via Yb^(3+) doping

In this paper,a series of GdOF-based green phosphors doped with Ho^(3+) and Yb^(3+) ions was prepared by a simple co-precipitation *** crystal structure s and optical prope rties were investigated in *** intensity of the emission of GdOF:Yb^(3+),Ho^(3+) phosphors is enhanced by the effective energy transfer from Yb^(3+) to Ho^(3+),and the optimal luminescence is achieved with the Ho^(3+)doping amount of 0.1 mol%as well as Yb^(3+) doping amount of 5 mol%.Tunable UC emission of samples is achieved by adjusting the doping concentration of Ho^(3+) or Yb^(3+).The purity of the green UC emission is further improved by enhancing the excitation pumping *** addition,logarithmic functions of green,red and nearinfrared radiation intensities were calculated versus logarithmic function of pump power for further study.A possible UC luminescence mechanism between Yb^(3+) and Ho^(3+) is discussed based on powerdependent UC luminescence spectrum and decay *** results suggest that GdOF:Yb^(3+),Ho^(3+)phosphors with bright and high-purity green UC emission have potential applications in the fields of bioimaging,display and illumination.

The stance and factors of international organizations towards countries from a Chinese perspective

The degree and scope of constraints imposed by International Organizations(IOs)on States are increasing,and identifying the factors affecting the IOs’stances on States is helpful to enhance the state's discourse power and influence in the international ***,by coding the records of Regular Press Conferences of the Speaking Office of the Chinese Ministry of Foreign Affairs during the period of 20182022,we obtained a dataset of IOs’stances on China-related ***,we constructed political relation,economic relation,and humanistic relation indicators to complement the influence factors,adopted the Bayesian logit model,and applied the Monte Carlo Markov chain algorithm and Gibbs sampling to analyze the probability of IOs’positive stances towards *** result shows that IOs’category,length of establishment,functional position,and relationship with China are all related to their tendency of making a statement about *** terms of the heterogeneity of event types,forum-type IOs are significantly inclined to give positive assessment compared to service-type IOs on events focusing on China's own *** analysis reveals that the model for analyzing and predicting the attitudes of IOs is more effective when the international situation is in a stable period.

Integrated multi-omics analysis reveals liver metabolic reprogramming by fish iridovirus and antiviral function of alpha-linolenic acid

Iridovirus poses a substantial threat to global aquaculture due to its high mortality rate;however,the molecular mechanisms underpinning its pathogenesis are not well ***,a multi-omics approach was applied to groupers infected with Singapore grouper iridovirus(SGIV),focusing on the roles of key *** showed that SGIV induced obvious histopathological damage and changes in metabolic enzymes within the ***,SGIV significantly reduced the contents of lipid droplets,triglycerides,cholesterol,and *** analysis indicated that the altered metabolites were enriched in 19 pathways,with a notable down-regulation of lipid metabolites such as glycerophosphates and alpha-linolenic acid(ALA),consistent with disturbed lipid homeostasis in the *** of transcriptomic and metabolomic data revealed that the top enriched pathways were related to cell growth and death and nucleotide,carbohydrate,amino acid,and lipid metabolism,supporting the conclusion that SGIV infection induced liver metabolic *** integrative transcriptomic and proteomic analysis indicated that SGIV infection activated crucial molecular events in a phagosome-immune depression-metabolism dysregulation-necrosis signaling *** note,integrative multi-omics analysis demonstrated the consumption of ALA and linoleic acid(LA)metabolites,and the accumulation of L-glutamic acid(GA),accompanied by alterations in immune,inflammation,and cell death-related *** experimental data showed that ALA,but not GA,suppressed SGIV replication by activating antioxidant and anti-inflammatory responses in the ***,these findings provide a comprehensive resource for understanding host response dynamics during fish iridovirus infection and highlight the antiviral potential of ALA in the prevention and treatment of iridoviral diseases.

Microphase Separation of Semiflexible Ring Diblock Copolymers

Aiming at the difficult problem of solving the conformation statistics of complex polymers, this study presents a novel and concise conformation statistics theoretical approach based on Monte Carlo and Neural Network method. This method offers a new research idea for investigating the conformation statistics of complex polymers, characterized by its simplicity and practicality. It can be applied to more complex topological structure, more higher degree of freedom polymer systems with higher dimensions, theory research on dynamic self-consistent field theory and polymer field theory, as well as the analysis of scattering experimental data. The conformation statistics of complex polymers determine the structure and response properties of the system. Using the new method proposed in this study, taking the semiflexible ring diblock copolymer as an example, Monte Carlo simulation is used to sample this ring conformation to construct the dataset of polymer. The structure factor describing conformation statistics are expressed as continuous functions of structure parameters by neural network supervised learning. This is the innovation of this work. As an application, the structure factors represented by neural networks were introduced into the random phase approximation theory to study the microphase separation of semiflexible ring diblock copolymers. The influence of the ring's topological properties on the phase transition behavior was pointed out.
