TRIM21-dependent ultrasmall chiral gold nanoparticles for preventing microglia senescence against Alzheimer's disease

Tripartite motif 21(TRIM21)is an E3 ubiquitin ligase that shows great promise for protein degradation through ***,ultrasmall chiral gold nanoparticles(D-or L-NPs)modified by D-or L-glutathione ligands were *** conjugated with an NLR family pyrin domain-containing protein 3(NLRP3)antibody(D-NP-a NLRP3 or L-NP-a NLRP3),DNP-a NLRP3 showed effective delivery efficiency of the antibody into microglia and prevented Aβ-mediated microglia senescence,and p16^(ink4a),a marker of senescence,in the microglia was reduced by 90.3%±7.8% while L-NP-a NLRP3 decreased by 48.01%±3.1%.Mechanistic investigations revealed that the D-NP-a NLRP3((1.5±0.3)×10^(7)M^(-1))exhibited sixteen-fold larger binding affinity to transmembrane glycoprotein SLC3A2 than L-type((9.5±2.7)×10^(5)M^(-1)),which led to a high efficiency of antibody delivery and TRIM21-dependent NLRP3 ***,the blood-brain barrier(BBB)-crossing ability of chiral NP-a NLRP3 as well as NLRP3 degradation was demonstrated in *** APP/PS1 Alzheimer's disease(AD)model mice experiments exhibited a reduction of 89.7%±6.8% for the NLRP3 protein and 84.2±7.5% for p16^(ink4a)following the intravenous administration of D-NP-a NLRP3 once a week for 60 ***,the levels of the AD markers Aβ and phosphorylatedTau in the brains were reduced by 86.2%±8.2% and 81.6%±9.1%;these were 2.1-fold and 1.9-fold higher than those treated with L-NP-a NLRP3,*** studies provide a method to rescue AD-like pathologies and prevent senescence.

The [α/Fe] ratios in stars are good tracers to probe the formation history of stellar populations and the chemical evolution of the Galaxy. The spectroscopic survey of LAMOST provides a good opportunity to determine [α/Fe] of millions of stars in the Galaxy. We present a method of measuring the [α/Fe]ratios from LAMOST spectra using the template-matching technique of the LSP3 pipeline. We use three test samples of stars selected from the ELODIE and MILES libraries, as well as the LEGUE survey to validate our method. Based on the test results, we conclude that our method is valid for measuring [α/Fe]from low-resolution spectra acquired by the LAMOST survey. Within the range of the stellar parameters Teff= [5000, 7500] K, log g = [1.0, 5.0] dex and [Fe/H]= [onsistent with values derived from high-resolution spectra,-1.5, ＋0.5] dex, our [α/Fe] measurements are c and the accuracy of our [α/Fe] measurements from LAMOST spectra is better than 0.1 dex with spectral signal-to-noise higher than 20.

Inflammation scores predict survival for hepatitis B virus-related hepatocellular carcinoma patients after transarterial chemoembolization

AIM:To compare the prognostic ability of inflammation scores for patients with hepatitis B virus(HBV)-related hepatocellular carcinoma(HCC)undergoing transarterial chemoembolization(TACE).METHODS:Data of 224 consecutive patients who underwent TACE for unresectable HBV-related HCC from September 2009 to November 2011 were retrieved from a prospective *** association of inflammation scores with clinicopathologic variables and overall survival(OS)were analyzed,and receiver operating characteristic curves were generated,and the area under the curve(AUC)was calculated to evaluate the discriminatory ability of each inflammation score and staging system,including tumor-node-metastasis,Barcelona Clinic Liver Cancer,and Cancer of the Liver Italian Program(CLIP)***:The median follow-up period was 390 d,the one-,two-,and three-year OS were 38.4%,18.3%,and 11.1%,respectively,and the median OS was *** Glasgow Prognostic Score(GPS),modifed GPS,neutrophil-lymphocyte ratio,and Prognostic Index were associated with *** GPS consistently had a higher AUC value at 6 mo(0.702),12 mo(0.676),and24 mo(0.687)in comparison with other inflammation *** consistently had a higher AUC value at6 mo(0.656),12 mo(0.711),and 24 mo(0.721)in comparison with tumor-node-metastasis and Barcelona Clinic Liver Cancer staging *** analysis revealed that alanine aminotransferase,GPS,and CLIP were independent prognostic factors for *** combination of GPS and CLIP(AUC=0.777)was superior to CLIP or GPS alone in prognostic ability for ***:The prognostic ability of GPS is superior to other inflammation scores for HCC patients undergoing *** GPS and CLIP improved the prognostic power for OS.

The Ordos Basin is a large cratonic basin with stable deposition in Northwest *** its mostly stable subsidence background and the urgent need for saline water disposal and carbon dioxide storage in the coal mining and coal chemical industries,the Ordos Basin has been chosen as a pilot demonstration site for carbon dioxide and saline water storage in ***,few studies have been made to evaluate the sedimentary and sequence stratigraphy characteristics of this region,as well as their influence on carbon dioxide and saline water storage *** address this research gap,we conducted a sedimentary study of the Lower Triassic Liujiagou Formation in the northeastern Ordos Basin utilizing the stratigraphy theory,laboratory test analysis,and pilot project demonstration,to evaluate the area's viability for the deep geological storage of gas and liquid *** studied the tectonic setting,petrological features,and sedimentary characteristics of the favorable strata and predicted favorable areas based on sequence stratigraphy *** lithology predominantly consists of feldspathic graywacke,with a fine grain size and mostly fine-to-medium-grained *** distribution of thick-grained sandstone and fine-grained sediment was identified by dividing the sequence,and a favorable reservoir-cap assemblage configuration was ***'s concluded that the Lower Triassic Liujiagou Formation exhibits suitable characteristics for the deep geological storage of carbon dioxide and saline *** study demonstrates the importance of basic theory in guiding practical applications and provides a reference for the scientific selection of favorable areas for deep basin storage.

Due to its difﬁculty in early diagnosis and lack of sensitivity to chemotherapy and radiotherapy,renal cell carcinoma(RCC)remains to be a frequent cause of cancer-related ***,we probed into new targets for its early diagnosis and treatment for ***(miRNA)data of M2-EVs and RCC were searched on the Gene Expression Omnibus database,followed by the prediction of the potential downstream *** of target genes was measured via RT-qPCR and Western blot,respectively.M2 macrophage was obtained viaﬂow cytometry with M2-EVs *** binding ability of miR-342-3p to NEDD4L and to CEP55 ubiquitination was studied with their roles in the physical abilities of RCC cells *** tumor-bearing mouse models and lung metastasis models were prepared to observe in vivo role of target genes.M2-EVs induced RCC growth and ***-342-3p showed high expression in both M2-EVs and RCC cells.M2-EVs carrying miR-342-3p promoted RCC cell abilities to proliferate,invade and *** RCC cells,M2-EV-derived miR-342-3p could speciﬁcally bind to NEDD4L and consequently elevate CEP55 protein expression via suppressing NEDD4L,thereby exerting tumor-promoting ***55 could be degraded by ubiquitination under the function of NEDD4L,and miR-342-3p delivered by M2-EVs facilitated the RCC occurrence and development by activating the PI3K/AKT/mTOR signaling *** conclusion,M2-EVs promote RCC growth and metastasis by delivering miR-342-3p to suppress NEDD4L and subsequently inhibit CEP55 ubiquitination and degradation via activation of the PI3K/AKT/mTOR signaling pathway,strongly driving the proliferative,migratory and invasive of RCC cells.

Association between Genetic Variants and Characteristic Symptoms of Type 2 Diabetes:A Matched Case-Control Study

Objective： To examine the association of genetic variants with characteristic symptoms of type 2 diabetes mellitus （T2DM）. Methods： A matched case-control study was performed to investigate the association between common variants in four genes （CDKAL1, GLIS3, GRK5, and TCFTL2） and symptoms of T2DM. Symptoms were examined with questionnaire for 710 subjects. Genomic DNA was extracted from peripheral blood mononuclear cell by salting-out procedure. Genotyping was carried out by direct sequencing of the unpurified polymerase chain reaction products. Result： Most of the T2DM patients pressented characteristic symptoms, such as feeling weak in limbs （P=0.0057）, hand tremor （P=0.0208）, bradymasesis （P=0.0234）, and polyuria （P=0.0051）. Some of the T2DM patients shared characteristic symptoms, such as desire for cold drinks （P=0.0304）, polyphagia （P=0.0051）, and furred tongue （P=0.028）. The impaired glucose regulation （IGR） cases took only one characteristic symptom of frequent micturition （P=0.0422）. GLIS3 rs7034200 and GRK5 rs10886471 were significantly associated with increased T2DM risk （GLIS3 rs7034200 under dominant model： P=0.0307; GRK5 rs 10886471 under recessive model： P=0.0092）. However, only the rs10886471 polymorphism in GRK5 showed a significant effect on both differentiated symptoms and T2DM risk. The C-allele was involved in both dampness-heat encumbering Pi （Spleen） syndrome （P=0.047） and qi-yin deficiency syndrome （P=0.002） via increased GRK5 expression. Conclnsions： Both T2DM and IGR exhibited its corresponding characteristic symptoms. The variants of GRK5 were involved with both qi-yin deficiency syndrome and dampness-heat encumbering Pi syndrome.

Two pairs of Pt(Ⅱ) enantiomers((RR)/(SS)-Py Pt,((RR)/(SS)-Py: N,N’-(1,2-diphenylethane-1,2-diyl)dipicolinamide;(RR)-P/M-QPt,((RR)/(SS)-Q: N,N’-((1R,2R)-1,2-diphenylethane-1,2-diyl)bis(quinoline-2-carboxamide)) were synthesized, respectively, with good circularly polarized luminescence(CPL) and tunable dissymmetry factors(g) by molecular self-induction with(RR)/(SS)-1,2-diphenylethane-1,2-diamine as carbon chiral sources. In the(RR)-P-QPt and(SS)-M-QPt, specific P-and M-configurations were effectively induced from intrinsic chiral carbon centres(R or S), ingeniously avoiding the racemic mixture formation and chiral separation. Furthermore, the chirality originating from both chiral carbon centres and helicene-like structure improves the g factor significantly, which provides a new molecular design strategy for chiral Pt(II) enantiomers with good CPL properties.

Wood-based hydrogel with a unique anisotropic structure is an attractive soft material,but the presence of rigid crystalline cellulose in natural wood makes the hydrogel less *** this study,an all-wood hydrogel was constructed by cross-linking cellulose fibers,polyvinyl alcohol(PVA)chains,and lignin molecules through the Hofmeister *** all-wood hydrogel shows a high tensile strength of 36.5 MPa and a strain up to~438%in the longitudinal direction,which is much higher than its tensile strength(~2.6 MPa)and strain(~198%)in the radial direction,*** high mechanical strength of all-wood hydrogels is mainly attributed to the strong hydrogen bonding,physical entanglement,and van der Waals forces between lignin molecules,cellulose nanofibers,and PVA *** to its excellent flexibility,good conductivity,and sensitivity,the all-wood hydrogel can accurately distinguish diverse macroscale or subtle human movements,including finger flexion,pulse,and swallowing *** particular,when“An Qi”was called four times within 15 s,two variations of the pronunciation could be *** recyclable,biodegradable,and adjustable mechanical properties,the all-wood hydrogel is a multifunctional soft material with promising applications,such as human motion monitoring,tissue engineering,and robotics materials.
