Metabonomics and Molecular Biology-based Effects of Sugemule-3 in an Isoproterenol-induced Cardiovascular Disease Rat Model

Cardiovascular disease（CVD） is a common and serious disease in the elderly, which has characteristically high prevalence, disability, and mortality rates. However, the etiology of CVD is still not very clear. The traditional Mongolian medicine Sugemule-3（SM） is usually used for the treatment of CVD and exhibits a good curative effect. In this study, a sentm metabolite profile analysis was used to identify potential biomarkers associated with isoprote- renol（ISO）-induced CVD and investigate the mechanism of the action of SM. Ultra-perfornlance liquid chromato- graphy coupled with quadrupole time-of-flight mass spectrometry（UPLC-Q-TOF-MS） was used for the metabono- mics analysis. Principal component analysis（PCA） was used to process the acquired data to differentiate the results of the control, CVD, and SM treatment groups. Orthogonal partial least squares discriminant analysis（OPLS-DA） enabled the identification of 21 metabolites as potential biomarkers that were relevant to phospholipid and energy metabolism. The results indicate that SM played a protective role against ISO-induced CVD in rats by regulating phospholipid and energy metabolic pathways. Fttrther, we verified the apoptotic metabolic pathway using molecular biology methods, such as terminal deoxynucleotidyl transferase（TdT） deoxyuridine 5＇-triphosphate（dUTP） nick-end labeling（TUNEL） assay and Western blot analysis. Furthermore, this study identified early biomarkers of CVD and elucidated the underlying mechanism of the therapeutic actions of SM, which is worth further to be investigated for development as a clinical therapy.

Mesoscale simulation of discontinuous dynamic recrystallization using the cellular automaton method

A dynamic recrystallization （DRX） cellular automaton （CA） model that can mark the microstructure with DRX circle was developed. The effects of initial grain size on the stress-strain curve, mean grain size and DRX fraction were mainly investigated, and the simulated results were compared with those obtained from previous researches. The results show that the shape of the stress-strain curve is sensitive, while the stress and mean grain size at the steady state are insensitive to the initial grain size. The transition from a multiple-peak stress-strain curve to a single-peak one can be explained by variations in DRX circle fraction, and the initial grain size to make this transition is between 70 and 80 tim.

Performance of CH4MOD_(wetland) for the case study of different regions of natural Chinese wetland

Reliable national estimates of CH_4 emissions from natural wetlands depend on model validation based on site *** therefore evaluated the performance of the CH_4 MODwetlandmodel in simulating CH_4 emissions from 11 representative wetland sites in five regions of *** performance analysis showed that this method effectively simulates differences in the CH_4 fluxes between different sites and *** model efficiency for estimating the daily CH_4 fluxes in the northeastern China（NE）,Inner Mongolia and northwestern China（NW）,the North China plain and the Middle-Lower Yangtze Plain（E） and the Qinghai Tibetan Plateau（SW） was 0.51,0.20,0.52 and 0.65,*** efficiency for estimating the annual mean CH_4 fluxes in southern China（S） was *** negative deviation between the simulated and observed CH_4 emissions existed in all regions,especially in the NW region,which had a mean deviation（RMD） value of-36.7%.On the national scale,the root mean square error（RMSE）,the RMD,the model efficiency（EF）between the simulated and observed seasonal values were 28.7%,-7.8% and 0.93,*** CH_4 emissions showed the highest sensitivity to air temperature in the NE and SW regions,and to water table depth in the E *** on the sensitivity analysis,future climate warming and wetting are likely to increase the wetland CH_4 emissions at different levels in all regions of China.

OBJECTIVE:To investigate the antioxidant property and potential ameliorating effective ingredients for high altitude-induced fatigue from Gansu Maxianhao(Pedicularis Kansuensis Maxim.).METHODS:Macroporous adsorptive resin combined with polyamide chromatographic column was used to obtain water extract(P1),high polar part(P2),iridoid glycosides part(P3)and phenylethanoid glycosides part(P4)of Gansu Maxianhao(Pedicularis Kansuensis Maxim.).Antioxidant activity of each part was investigated employing a series of in vitro ***-eosin staining,analysis of blood biochemical parameters,along with molecular analyses examining oxidative stress makers,metabolite,metabolic enzyme and energy substance in liver,skeletal muscle and/or serum were further ***:The results showed phenylethanoid glycosides(Ph Gs)exhibited more effective with 1,1-diphenyl-2-picrylhydrazyl radical scavenging ability,2-2-Azinbis(3-ehtylbenzothiazolin-6-sulfnic Acid)Diammonium Salt+scavenging activity,hydroxyl radical scavenging activity,reducing ability and antioxidant activity to varying *** a hypobaric hypoxia attack in a hypobaric chamber(8000 m,7 consecutive days),Ph Gs was administered to BALB/C mice at doses of 50,200,400 mg/kg and antifatigue property was evaluated using a swimming test at an altitude of 4000 *** results showed that Ph Gs of Gansu Maxianhao(Pedicularis Kansuensis Maxim.)could significantly prolong the burden swimming time of mice,reduce the hypoxia-induced oxidative stress,remove the accumulated products of metabolism,improve the energy metabolism as well as improve preservation of endogenous glycogen ***:The ameliorating effect against altitude-induced fatigue of Ph Gs from Gansu Maxianhao(Pedicularis Kansuensis Maxim.)might come from the alleviation of oxidative stress,reduction of the adverse metabolic products,normalizing energy metabolism and increasing energy substances *** Gs is a potential antioxidant and novel rem

OBJECTIVE: To investigate the most effective weight ratio of Qiancao(Radix Rubiae Cordifoliae) and Haipiaoxiao(Endoconcha Sepiellae) used to produce decoction for the treatment of abnormal uterine bleeding(AUB) in rats with incomplete abortion, and to study the possible mechanism. METHODS: The models of AUB were established by incomplete drug abortion with mifepristone and misoprostol in pregnant rats. The therapeutic effects of decoctions made by eight different weight ratios of Qiancao(Radix Rubiae Cordifoliae) and Haipiaoxiao(Endoconcha Sepiellae) were observed. RESULTS: Statistical analysis revealed that the most effective weight ratio of Qiancao(Radix Rubiae Cordifoliae) and Haipiaoxiao(Endoconcha Sepiellae) to treat AUB in this study was 2∶1. CONCLUSION: The insights gained from this study would improve understanding of the mechanisms involved in the effect of Qiancao(Radix Rubiae Cordifoliae) and Haipiaoxiao(Endoconcha Sepiellae) on AUB.
