Objective: Long non-coding RNAs(lnc RNAs) are involved in numerous biological processes in lung cancer cells. In our previous studies, we identified a lnc RNA, ENST00000439577, which is highly expressed in lung carcinomas, and termed it lung cancer progression-associated transcript 1(LCPAT1). To characterize the role of LCPAT1 in lung cancer, we conducted the current ***: Expression of LCPAT1 and autophagy-associated markers in tumor tissues and lung cancer cell lines was determined by real-time quantitative polymerase chain reaction(q PCR). Hematoxylin and eosin(HE) staining, q PCR, Western blot, and immunohistochemistry were performed to evaluate xenografted tumor tissues. Autophagy induced by rapamycin was detected by Western blot and immunofluorescence in lung cancer cell ***: Expression of LCPAT1 and microtubule-associated protein 1 light chain 3 beta(LC3B) was positively correlated in lung cancer. Knockdown of LCPAT1 inhibited tumor growth and suppressed cell autophagy in vivo. Moreover, LCPAT1 knockdown in lung cancer cell lines resulted in decreased autophagy-associated gene expression and alleviated the cell autophagy induced by ***: We speculate that LCPAT1 plays a crucial role in regulating autophagy in lung cancer.

Pancreatic cancer is a fatal malignancy with an increasing incidence in Shanghai, China. A genomewide association study(GWAS) and other work have shown that ABO alleles are associated with pancreatic cancer risk. We conducted a population-based case-control study involving 256 patients with pathologically confirmed pancreatic ductal adenocarcinoma(PDAC) and 548 healthy controls in Shanghai, China, to assess the relationships between GWAS-identified ABO alleles and risk of PDAC. Carriers of the C allele of rs505922 had an increased cancer risk [adjusted odds ratio(OR) = 1.42, 95% confidence interval(CI): 1.02-1.98] compared to TT carriers. The T alleles of rs495828 and rs657152 were also significantly associated with an elevated cancer risk(adjusted OR = 1.58, 95% CI: 1.17-2.14; adjusted OR = 1.51, 95% CI: 1.09-2.10). The rs630014 variant was not associated with risk. We did not find any significant gene-environment interaction with cancer risk using a multifactor dimensionality reduction(MDR) method. Haplotype analysis also showed that the haplotype CTTC was associated with an increased risk of PDAC(adjusted OR = 1.46, 95% CI: 1.12-1.91) compared with haplotype TGGT. GWAS-identified ABO variants are thus also associated with risk of PDAC in the Chinese population.

Rapid growth in biomedical research coupled with dramatic advancement in biotechnology has signi fi cantly improved our understanding of the molecular basis involving cancer development and progression. This improvement has led to the discovery of new molecular markers for cancer diagnosis and prognosis as well as new molecular targets for cancer treatment and intervention. Continuous emergence of some new developing area in molecular profi ling, new therapeutic agents, tissue microenvironment and systems biology have made signifi cant progress in clinical oncology. Clinical research and investi-gation that focus on these new developments have begun to show exciting results that indicate future promises in improving patient management and survival.

Promoter methylation of Wnt/β-Catenin signal inhibitor TMEM88 is associated with unfavorable prognosis of nonsmall cell lung cancer

Objective:Recent research has indicated that altered promoter methylation of oncogenes and tumor suppressor genes is an important mechanism in lung cancer development and *** this study,we investigated the association between promoter methylation of TMEM88,a possible inhibitor of the Wnt/β-Catenin signaling,and the survival of patients with nonsmall cell lung cancer(NSCLC).Methods:Twelve pairs of tumor and adjacent non-tumor samples were used for microarray analyses of DNA methylation and gene *** validation,more than two hundred additional samples were analyzed for methylation using bisulfite pyrosequencing and for gene expression using q *** the cell function were tested by wound healing,transwell,CCK8 and cell cycle ***:Our analysis of patient specimens showed that TMEM88 methylation was higher in NSCLC tumors(82.2%±10.3,P56 months;P=0.021).In addition,we found that demethylation treatment could inhibit tumor cell proliferation,migration,and invasion,which was supportive of an association between methylation and ***:Based on these consistent observations,we concluded that TMEM88 may play an important role in NSCLC progression and that promoter methylation of TMEM88 may serve as a biomarker for NSCLC prognosis and treatment.

＜正＞Background:The cellular functions of DNA repair and programmed cell death are believed to play an important role in controlling the fate of tumor cells in response to chemotherapy or *** has been speculated that single nucleotide polymorphisms （SNPs） in related genes may alter their expression or activity,affect the functions of DNA repair and/or apoptosis,and in turn influence the effects of cancer treatment and *** assess this possibility,we genotyped four single nucleotide polymorphisms （SNPs） in ATM （A60G）,ERCCl （Asn118Asn）,APEl （Asn148Glu）,XRCCl（Arg194Trp）,XRCCl （Arg399Gln） and iASPP （A67T）,and examined their associations with treatment response and overall survival among patients with advanced non-small cell lung cancer （NSCLC）. Methods:Included in the study were 230 patients diagnosed with inoperable advanced *** these patients,76 received platinum-based chemotherapy,125 received chemotherapy plus radiation,and 29 received radiotherapy *** SNPs were genotyped using the TaqMan methods. Associations between clinical variables,treatment response and genotype were analyzed using the Chi-square *** unconditional logistic regression model was used to analyze the association between genotype and treatment response while adjusting for *** analysis was performed for overall survival using Kaplan-Meier survival curves and Cox proportional hazards regression. Results:No significant correlation between genotype and overall survival was found,but ERCCl （Asn118Asn） and iASPP （A67T） genotype was significantly associated with treatment response separately among the patients who received chemotherapy only and the patients who were treated with both chemotherapy and *** with either one or two T alleles （T/T+C/T） at Asn118Asn of ERCCl were more likely not to respond to platinum-based chemotherapy compared to those without the T allele （OR=4.10,95% CI:1.31-12.85）.Patients carrying an A allele （A/T+A/A） at A67T o

＜正＞Insulin - like growth factors （IGFs） are essential peptide hormones in regulation of cell proliferation, differentiation and apoptosis. As these cellular activities are critically related to tumorigenesis, IGFs are believed to play important roles in cancer development and progression. IGFs consist of two polypeptides, IGF - Ⅰ and IGF - Ⅱ , which bind to a specific cell membrane receptor with tyrosine kinase activity. Although the two peptides share extensive similarities in molecular structure and biologic action, these hormones are regulated differently and act at specific time. IGF - Ⅱ regulates cell proliferation and apoptosis during embryonic development and fetal growth, while IGF -Ⅰ exerts its actions in the postnatal life. Both IGF-Ⅰ and IGF - Ⅱ are subject to endocrine and paracrine regulation, and are expressed in ovarian tissue regulating its functions. Ovarian caner is one of the most fatal diseases for women and its etiology remains largely unknown. To evaluate IGF’s role in ovarian cancer, we investigated the effect and regulation of IGFs in ovarian cancer. The questions addressed in this study include a) which IGF, IGF-Ⅰ or IGF - Ⅱ , plays a more important role in ovarian cancer progression, b) whether IGF-Ⅰ genotype affects its phenotype in ovarian tissue, c) which IGF- Ⅱ promoters are more important for its expression in ovarian cancer, d) whether the promoter activity is affected by promoter methylation, and e) which laboratory methods are more reliable in assessing promoter methylation. We collected fresh frozen tumor tissue samples from 215 patients with primary epithelial ovarian cancer for the study. DNA, total RNA and cytosol proteins were extracted from the tissue samples and were analyzed for IGF-Ⅰ CA repeat polymorphism, IGF- Ⅱ promoter methylation, promoter- specific RNA expressions of IGF-Ⅰ and IGF - Ⅱ , as well as cytosol levels of IGF-Ⅰ, IGF- Ⅱ , and IGFBP - 3 proteins. 本文叙述了用于控制电子交换系统的一台低成本微程序处理机的内部工作。为了减轻维修难度和进一步提高组装的效率,机器的结构进行了规格化。已经从数量有限的校验电路和适量的实时消耗得到了高度的并行自校验。所有数据操作设备都是组合的并采用奇偶预测技术来检出所产生的数据的误差。由于处理机的自校验特性,因此可实行具有故障保护的独立操作。整个机器包括取下条指令在内,都在微程序控制之下。由于指令系统易于修改,因此微程序设计的灵活性可使软件和硬件并行改进。已经设计出一种充分利用存贮器的简单命令结构。外加微指令已被用于并行校验。由于存贮器具有预读能力,因此主存贮器访问时间对微程序周期时间的影响得以消除。一台实验室样机已于1971年2月作成并开始运行,运算数据表明已满足了设计指标。
