OBJECTIVE Cue-induced drug craving progressively increase after prolong abstinence in animal and human beings.A behavioral phenomenon termed incubation of drug craving is considered as a key reason for drug relapse,but the transcriptional mechanisms that contribute to this incubation are unknown.It has been demonstrated that circular RNAs(circR NAs),act as miR NA sponges,play important roles in the regulation of gene expression and the pathogenesis of disease.The present study aims to explore the transcriptional profiles associated with incubation of morphine craving in nucleus accumbens(NAc),an important brain area previously implicated in drug seeking.METHODS The animal model of the incubation of drug craving was induced by CPP paradigms with six morphine(5 mg·kg-1) injections and 14 d drug abstinence in mice.The brain tissues of NAc were collected after the behavioral tests for circRNA-sequencing by Illumina Hiseq X sequencer.We identified differentially expressed genes(DEGs) by qRT-PCR and used bioinformatics methods for further function analysis.RESULTS The progressive increase of CPP scores during the 14 d drug abstinence indicated the establishment of animal model.The data of circRNA-sequencing reported that 16 circRNAs were significantly altered after 28 d drug abstinence in NAc of morphine treated mice(FC≥2 and P<0.05).Among those circRNAs,9 were significantly up-regulated,while 7 were down-regulated.Furthermore,we subsequently tested circRNAs expression using quantitative real-time PCR,and the consistent data were obtained.The results of KEGG pathway analysis indicated that these genes were enriched for several biological processes,including RNA transport,protein ubiquitination and histone methylation,etc.CONCLUSION These findings provide a unique resource of gene expression data for future studies examining transcriptional mechanisms in NAc that mediate opioids seeking after prolonged withdrawal.

Comprehensive Summary,The Pd-catalyzed dipolar cycloaddition represents a significant synthetic strategy for the construction of useful heterocyclic compounds.This study developed the dipolar[4+2]and[6+2]cycloaddition reactions of benzo[d]isothiazole 1,1-dioxides(BDs)leading to the synthesis of BD-fused 1,3-oxazinane and 1,3-oxazocane derivatives,respectively.In particular,the synthesis of BD-fused 1,3-oxazinanes demonstrated regio-and enantioselective characteristics,resulting in products with good yields,enantioselectivity and regioselectivity(if applicable).Furthermore,the[6+2]cycloaddition reaction developed in this work represented the first strategy for the synthesis of medium-sized ring compounds based on BDs.

Light-induced recognition,assemblies,and materials are intensive areas of research due to their high spatiotemporal resolution.Herein,we demonstrated photoswitchable molecular recognition via dithienylethene-triggeredreversible structural regulation of dynamic covalent hydrazone bonds.By combining dithienylethenes and cyclic hemiacetals,the photochemical open-ring and closed-ring forms enabled turningoff and on the creation of awide range of hydrazones when desired.Light-induced bidirectional switching between hydrazones and their cyclization structures promoted by a neighboring carboxyl group was further achieved.By taking advantage of reversible structural changes totoggleon andoff the binding pocket,photoswitchable recognitionofmetal ionswas realized.Finally,the construction of an acylhydrazone polymer offered a facile way for light-mediated selective extraction/release.The strategies and results reported here should find applications in many contexts,such asdynamicassemblies,molecular switches,and smart materials.

An imperfect propagation environment or optical system would introduce wavefront aberrations to vortex beams.The phase aberrations and orbital angular momentum in a vortex beam are proved to be mutually restrictive in parameter measurement.Aberrations make traditional topological charge(TC)probing methods ineffective while the phase singularity makes phase retrieval difficult due to the aliasing between the wrapped phase jump and the vortex phase jump.An interactive probing method is proposed to make measurements of the aberrated phase and orbital angular momentum in a vortex beam assist rather than hinder each other.The phase unwrapping is liberated from the phase singularity by an annular shearing interference technique while the TC value is determined by a Moirétechnique immune to aberrations.Simulation and experimental results proving the method effective are presented.It is of great significance to judge the characteristics of vortex beams passing through nonideal environments and optical systems.

BACKGROUND Intracranial atherosclerosis,a leading cause of stroke,involves arterial plaque formation.This study explores the link between plaque remodelling patterns and diabetes using high-resolution vessel wall imaging(HR-VWI).AIM To investigate the factors of intracranial atherosclerotic remodelling patterns and the relationship between intracranial atherosclerotic remodelling and diabetes mellitus using HR-VWI.METHODS Ninety-four patients diagnosed with middle cerebral artery or basilar artery INTRODUCTION Intracranial atherosclerotic disease is one of the main causes of ischaemic stroke in the world,accounting for approx-imately 10%of transient ischaemic attacks and 30%-50%of ischaemic strokes[1].It is the most common factor among Asian people[2].The adaptive changes in the structure and function of blood vessels that can adapt to changes in the internal and external environment are called vascular remodelling,which is a common and important pathological mechanism in atherosclerotic diseases,and the remodelling mode of atherosclerotic plaques is closely related to the occurrence of stroke.Positive remodelling(PR)is an outwards compensatory remodelling where the arterial wall grows outwards in an attempt to maintain a constant lumen diameter.For a long time,it was believed that the degree of stenosis can accurately reflect the risk of ischaemic stroke[3-5].Previous studies have revealed that lesions without significant luminal stenosis can also lead to acute events[6,7],as summarized in a recent meta-analysis study in which approximately 50%of acute/subacute ischaemic events were due to this type of lesion[6].Research[8,9]has pointed out that the PR of plaques is more dangerous and more likely to cause acute ischaemic stroke.Previous studies[10-13]have found that there are specific vascular remodelling phenomena in the coronary and carotid arteries of diabetic patients.However,due to the deep location and small lumen of intracranial arteries and limitations of imaging techniques,the relationship between intra

The interaction between cluster of differentiation 47(CD47)and signal regulatory proteinα(SiRPa)protects healthy cells from macrophage attack,which is crucial for maintain-ing immune homeostasis.Overexpression of CD47 occurs widely across various tumor cell types and transmits the"don't eat me"signal to macrophages to avoid phagocytosis through binding to SIRPa.Blockade of the CD47-SIRPa axis is therefore a promising approach for cancer treat-ment.Lymphoma is the most common hematological malignancy and is an area of unmet clin-ical need.This review mainly described the current strategies targeting the CD47-SIRPa axis,including antibodies,SiRPaFc fusion proteins,small molecule inhibitors,and peptides both in preclinical studies and clinical trials with Hodgkin lymphoma and non-Hodgkin lymphoma.

Derivation of Expanded Isospectral-Nonisospectral Integrable Hierarchies via the Column-vector Loop Algebra

A scheme for generating nonisospectral integrable hierarchies is introduced.Based on the method,we deduce a nonisospectral hierarchy of soliton equations by considering a linear spectral problem.It follows that the corresponding expanded isospectral and nonisospectral integrable hierarchies are deduced based on a 6 dimensional complex linear space ■.By reducing these integrable hierarchies,we obtain the expanded isospectral and nonisospectral derivative nonlinear Schr?dinger equation.By using the trace identity,the biHamiltonian structure of these two hierarchies are also obtained.Moreover,some symmetries and conserved quantities of the resulting hierarchy are discussed.

BACKGROUND A decreased autophagic capacity of bone marrow mesenchymal stromal cells(BMSCs)has been suggested to be an important cause of decreased osteogenic differentiation.A pharmacological increase in autophagy of BMSCs is a potential therapeutic option to increase osteoblast viability and ameliorate osteoporosis.AIM To explore the effects of sinomenine(SIN)on the osteogenic differentiation of BMSCs and the underlying mechanisms.METHODS For in vitro experiments,BMSCs were extracted from sham-treated mice and ovariectomized mice,and the levels of autophagy markers and osteogenic differentiation were examined after treatment with the appropriate concen-trations of SIN and the autophagy inhibitor 3-methyladenine.In vivo,the therapeutic effect of SIN was verified by establishing an ovariectomy-induced mouse model and by morphological and histological assays of the mouse femur.RESULTS SIN reduced the levels of AKT and mammalian target of the rapamycin(mTOR)phosphorylation in the phosphatidylinositol 3-kinase(PI3K)/AKT/mTOR signaling pathway,inhibited mTOR activity,and increased autophagy ability of BMSCs,thereby promoting the osteogenic differentiation of BMSCs and effectively alleviating bone loss in ovariectomized mice in vivo.CONCLUSION The Chinese medicine SIN has potential for the treatment of various types of osteoporosis,bone homeostasis disorders,and autophagy-related diseases.

Multiscale observation in wide-spatial radar surveillance based on coherent FDA design

Wide-spatial radar surveillance missions are challenging tasks, requiring an increased power budget and agility in transmission aimed at extracting information from multiple targets in different environments. These requirements necessitate high transmitting degrees of freedom(DOF) to achieve the objective of multiscale observation for specific tasks in specialized regions of interest. Herein, we exploit the multiscale observation ability in wide-spatial radar surveillance based on frequency diverse array(FDA)radar. The proposed method facilitates spatial anisotropic control of multiple radar resources, including the transmitting waveforms, beampattern, and bandwidth. By utilizing a coherent FDA radar, we offer principles for the selection of baseband waveforms, in addition to the quantitative design of the beampattern gain and optimal bandwidth from the perspective of detection. The feasibility of the proposed method is validated through numerical experiments, thus indicating the potential in wide-spatial radar surveillance. Moreover,this work can be regarded as a preliminary attempt to gauge the efficacy of the computational array, a novel academic concept.
