Comparative Studies of Different Organs of Nyctanthes arbortristis in Modulation of Cytokines in Murine Model of Arthritis

Objective To study the modulation effect of pro- and anti-inflammatory cytokines following long term use of water soluble ethanol extracts from different organs of Nyctanthes arbortristis （NAT） in mouse model of arthritis. Methods Arthritis was induced in mice by two injections of Freund＇s complete adjuvant on days 0 and 12 in the sub-planter surface of the right hind paw. results Injection of adjuvant resulted in a maximum primary edema of the footpad with erythema, and edema and distortion of joints of the right hind paw after 24-48 hours. Second injection of FCA led to the formation of secondary swellings persisting more than four weeks that spread onto the other hind limb but to a lesser extent. Histological analysis of the ankle on day 47 showed marked evidence of cartilage destruction in association with pannus formation and moderate bone resorption. Proinfiammatory cytokine levels in the inflamed joint homogenate were elevated on days 2, 14, and 47. Oral administration of leaf and fruit extracts in arthritic mice reduced joint homogenate levels of tumor necrosis factor-α, interleukin-1β, and interleukin-6 on days 2, 14, and 47 in comparison to untreated arthritic mice. Intedeukin-10 level was elevated in the inflamed joint on days 2, 14, and 47 in comparisons to untreated arthritic mice. Confusion Evidence of lesser inflammation of the footpad and joint and associated histological observation support the therapeutic benefit of leaf and fruit extracts from Nyctanthes arbortristis. This study helps in understanding the mechanism of anti-inflammatory action of Nyctanthes arbortristis in the light of pro- and anti-inflammatory cytokine balance.

Intracranial Abscesses: An Institutional Study

Background: Brain abscess is defined as a focal intracranial infection that is initiated as an area of cerebritis and evolves into a collection of pus surrounded by a vascularized capsule. These are complications of head trauma, neurosurgical operations, meningitis, and otogenic, mastoid, and paranasal air sinus infections. Management involves both medical and surgical treatment. Surgical management includes either aspiration or excision of lesions larger than 2.5 cm in diameter, depending on brain location. However, literature on surgical treatment is replete with several procedures which, on their own, may not determine outcome. Aim: Aim is to study the epidemiology, management and outcome of various treatment modalities of brain abscess in our institute. Material and Methods: We conducted a retrospective study of demographic data as well as indications, treatment modalities, and outcomes of various surgical procedures for evacuation of intracranial abscesses in patients admitted to our Neuro-intensive care unit at r.N.T. Medical College, Udaipur, rajasthan, India from January 2013 to June 2019. results: We carried out 53 procedures in 43 (30 male and 13 female) patients with various intracranial abscesses. Most abscesses [16, i.e. 37%] occurred in the second decade and second most common in the first decade [15, i.e. 34%]. In infants 4 (9%) cases of intracranial abscesses were present. None of the infants had features of congenital heart disease. The predisposing factors were mostly otolaryngologic (15) or posttraumatic (8). Most commonly abscesses were located in frontal 13 (30%) followed by cerebellar 9 (21%). Burr hole evacuation was done in 74% of cases. 3 patients (7% of cases) died. Prognosis appears to worsen with ventriculitis, multiple abscesses especially in infants, and immunosuppression. Conclusion: In conclusion, brain abscess still continues to be a formidable challenge, with prognosis that dramatically improved over the last decades due to advances

Anti-Amnesic Activity of Vitex Negundo in Scopolamine Induced Amnesia in rats

In the present study we investigated the anti-amnesic activity of Vitex negundo in scopolamine induced amnesia in rats. Wistar rats (180-200 g) were trained on active avoidance task. Each animal received session of 15 trials with inter trial duration of 15 s for 5 days. Scopolamine (3 mg/kg, i.p) was administered at different time periods on the basis of stages of memory i.e acquisition, consolidation and retention in different groups (n = 6). Effect of Vitex negundo extract was evaluated and compared to a standard drug, Donepezil. Significant (p rease in the avoidance response on the 5th session has been observed as compared to 1st session in control group. Scopolamine treatment significantly (p reduced the avoidance response compared to control. Extract treated groups shown significant (p rease in number of avoidance responses as compared to scopolamine treated groups. Increased oxidative stress in brain after scopolamine treatment, as observed by increase in MDA &decrease in GSH &SOD, was lowered in the groups treated with extracts. AChE activity was also improved after V. negundo treatment. results of the study have shown that V. negundo treated groups decrease the phenomenon of amnesia by increasing learning of memory through antioxidant effect and decreasing AChE activity.

In the present work,zinc oxide(ZnO)and silver(Ag)doped ZnO nanostructures are synthesized using a hydrothermal *** quality of the products is attested using X-ray diffraction,which confirms the hexagonal wurtzite struc-ture of pure ZnO and Ag-doped ZnO *** further confirms the crystallite orientation along the c-axis,(101)*** field emission scanning electron microscope study reveals the change in shape of the synthesized ZnO particles from hexagonal nanoparticles to needle-shaped nanostructures for 3 wt%Ag-doped *** optical band gaps and lattice strain of nanostructures is increased significantly with the increase of doping concentration of Ag in ZnO *** anti-microbial activity of synthesized nanostructures has been evaluated against the gram-positive human pathogenic bacteria,Staphylococcus aureus via an agarose gel diffusion *** maximum value of zone of inhibition(22 mm)is achieved for 3 wt%Ag-doped ZnO nanostructure and it clearly demonstrates the remarkable antibacterial activity.
