Tissue optical clearing enhances efficacy of vascular targeted photodynamic therapy of mouse dorsal skin

Vascular-targeted photodynamic therapy(V-PDT)is an effective treatment for port wine stains(PWS).However,repeated treatment is usually needed to achieve optimal treatment outcomes,possibly due to the limited treatment light penetration depth in the PWS lesion.The optical clearing technique can increase light penetration in depth by reducing light scattering.This study aimed to investigate the V-PDT in combination with an optical clearing agent(OCA)for the therapeutic enhancement of V-PDT in the rodent skinfold window chamber model.Vascular responses were closely monitored with laser speckle contrast imaging(LSCI),optical coherence tomography angiography,and stereo microscope before,during,and after the treatment.We further quantitatively demonstrated the effects of V-PDT in combination with OCA on the blood flow and blood vessel size of skin microvasculature.The combination of OCA and V-PDT resulted in significant vascular damage,including vasoconstriction and the reduction of blood flow.Our results indicate the promising potential of OCA for enhancing V-PDT for treating vascular-related diseases,including PWS.

Hollow Ni Mo-based nitride heterojunction with super-hydrophilic/aerophobic surface for efficient urea-assisted hydrogen production

Hydrogen evolution reaction（HER） and urea oxidation reaction（UOR） are key reactions of the watercycling associated catalytic process/device.The design of catalysts with a super-hydrophilic/aerophobic structure and optimized electron distribution holds great promise.Here,we have designed a threedimensional（3D） hollow Ni/NiMoN hierarchical structure with arrayed-sheet surface based on a onepot hydrothermal route for efficient urea-assisted HER based on a simple hydrothermal process.The Ni/NiMoN catalyst exhibits super-hydrophilic/aerophobic properties with a small droplet contact angle of 6.07° and an underwater bubble contact angle of 155.7°,thus facilitating an escape of bubbles from the electrodes.Density functional theory calculations and X-ray photoelectron spectroscopy results indicate the optimized electronic structure at the interface of Ni and NiMoN,which can promote the adsorption/desorption of reactants and intermediates.The virtues combining with a large specific surface area endow Ni/NiMoN with efficient catalytic activity of low potentials of 25 mV for HER and 1.33 V for UOR at10 mA cm-2.The coupled HER and UOR system demonstrates a low cell voltage of 1.42 V at 10 mA cm-2,which is approximately 209 mV lower than water electrolysis.

Lobe-dominatedγ-Ray Emission of Compact Symmetric Objects

Theγ-ray emitting compact symmetric objects(CSOs)PKS 1718-649,NGC 3894,and TXS 0128+554 are lobedominated in the radio emission.In order to investigate theirγ-ray radiation properties,we analyze the~14yr Fermi/LAT observation data of the three CSOs.They all show the low luminosity(1041-1043 erg s-1)and no significant variability in theγ-ray band.Theirγ-ray average spectra can be well fitted by a power-law function.These properties ofγ-rays are clearly different from theγ-ray emitting CSOs CTD 135 and PKS 1413+135,for which theγ-rays are produced by a restarted aligned jet.In the L_(γ)-Γ_(γ)plane,the three CSOs are also located at the region occupied by radio galaxies(RGs)while CTD 135 and PKS 1413+135 display a similar feature to blazars.Together with a similar radio emission property toγ-ray emitting RGs Cen A and Fornax A,we speculate that theγ-rays of the three CSOs stem from their extended mini-lobes.The broadband spectral energy distributions of the three CSOs can be well explained by the two-zone leptonic model,where theirγ-rays are produced by the inverse Compton process of the relativistic electrons in extended regions.By extrapolating the observed Fermi/LAT spectra to the very high energy band,we find that TXS 0128+554 among the three CSOs may be detected by the Cherenkov Telescope Array in the future.

Continual evolution of the severe acute respiratory syndrome coronavirus(SARS-CoV-2)virus has allowed for its gradual evasion of neutralizing antibodies(nAbs)produced in response to natural infection or vaccination.The rapid nature of these changes has incited a need for the development of superior broad nAbs(bnAbs)and/or the rational design of an antibody cocktail that can protect against the mutated virus strain.Here,we report two angiotensin-converting enzyme 2 competing nAbs—8H12 and 3E2—with synergistic neutralization but evaded by some Omicron subvariants.Cryo-electron microscopy reveals the two nAbs synergistic neutralizing virus through a rigorous pairing permitted by rearrangement of the 472-489 loop in the receptor-binding domain to avoid steric clashing.Bispecific antibodies based on these two nAbs tremendously extend the neutralizing breadth and restore neutralization against recent variants including currently dominant XBB.1.5.Together,these findings expand our understanding of the potential strategies for the neutralization of SARS-CoV-2 variants toward the design of broad-acting antibody therapeutics and vaccines.

Anti-PD1 antibody and not anti-LAG-3 antibody improves the antitumor effect of photodynamic therapy for treating metastatic breast cancer

Photodynamic therapy(PDT)has limited effects in treating metastatic breast cancer.Immune checkpoints can deplete the function of immune cells;however,the expression of immune checkpoints after PDT is unclear.This study investigates whether the limited e±cacy of PDT is due to upregulated immune checkpoints and tries to combine the PDT and immune checkpoint inhibitor to observe the e±cacy.A metastatic breast cancer model was treated by PDT mediated by hematoporphyrin derivatives(HpD-PDT).The anti-tumor effect of HpD-PDT was observed,as well as CD4þT,CD8þT and calreticulin(CRT)by immunohistochemistry and immunofluorescence.Immune checkpoints on T cells were analyzed byflow cytometry after HpD-PDT.When combining PDT with immune checkpoint inhibitors,the antitumor effect and immune effect were assessed.For HpD-PDT at 100 mW/cm2 and 40,60 and 80 J/cm2,primary tumors were suppressed and CD4þT,CD8þT and CRT were elevated;however,distant tumors couldn't be inhibited and survival could not be prolonged.Immune checkpoints on T cells,especially PD1 and LAG-3 after HpD-PDT,were upregulated,which may explain the reason for the limited HpD-PDT effect.After PDT combined with anti-PD1 antibody,but not with anti-LAG-3 antibody,both the primary and distant tumors were signi-cantly inhibited and the survival time was prolonged,additionally,CD4þT,CD8þT,IFN-þCD4þT and TNF-þCD4þT cells were signi-cantly increased compared with HpD-PDT.HpD-PDT could not combat metastatic breast cancer.PD1 and LAG-3 were upregulated after HpD-PDT.Anti-PD1 antibody,but not anti-LAG-3 antibody,could augment the antitumor effect of HpD-PDT for treating metastatic breast cancer.

Small-sized Ni-Co/Mo_(2)C/Co_(6)Mo_(6)C_(2)@C for efficient alkaline and acidic hydrogen evolution reaction by an anchoring calcination strategy

A novel,cheap and highly efficient Ni-Co/Mo_(2)C/Co_(6)Mo_(6)C_(2)@C nanocomposite has been successfully constructed through simple one-step carbonization method in a nitrogen atmosphere.Polyethyleneimine in the precursor can effectively anchor molybdenum-based Keggin-type polyoxometallate and NiCo-layered double hydroxide through electrostatic and coordination interactions,which avoids the aggregation of catalyst particles during the pyrolysis process.After optimization,the obtained Ni-Co/Mo_(2)C/Co_(6)Mo_(6)C_(2)@C possesses small size(3-8 nm),large specific surface area and hierarchical pore structure.More importantly,Ni-Co/Mo_(2)C/Co_(6)Mo_(6)C_(2)@C presents remarkable hydrogen evolution reaction activity with low overpotentials in 0.5 mol·L^(-1)H_(2)SO_(4)(102.3 mV)and 1 mol·L^(-1)KOH(95 mV)to afford the current density of 10 mA·cm^(-2),as well as small Tafel slopes of 82.49 and 99.92 mV·dec^(-1),respectively.Simultaneously,this catalyst also shows outstanding stability for 12 h without a significant change in current density.The excellent catalytic performance of Ni-Co/Mo_(2)C/Co_(6)Mo_(6)C_(2)@C can put down to the synergistic effect between multiple components and the small size of the catalyst.This work provides unique insights into the preparation of efficient transition metal-based catalysts for HER.

A study on carbon emission calculation in operation stage of residential buildings based on micro electricity usage behavior:Three case studies in China

Along with the improvement of social productivity and living standard,residential buildings generate a growing portion of carbon emissions,especially during the operation stage.However,energy use behaviors are usually ignored in carbon emission calculation.This study focuses on calculating carbon emissions during the operation stage for residential buildings based on the characteristics of energy use behaviors in different regions.Firstly,we investigated energy use behaviors in dwellings across three cities in China:Xi'an,Shanghai and Fuzhou.Then,we established calibrated carbon emission models and optimization models with different green building measures for residential buildings.The results of this research reveal a significant disparity between the energy usage habits of residents in different climate regions.The carbon emissions of residential electricity bills in Xi'an,Shanghai and Fuzhou are 13.6 kgCO_(2)/(m^(2)·a)(excluding central heating),29.3 kgCO_(2)/(m^(2)·a)and 17.2 kgCO_(2)/(m^(2)·a),respectively.Equipment carbon emissions account for 32.2%-64.1% of the total.In comparison to the model based on internal standard setting,the accuracy of the models using actual internal has improved by 25.9%-37.4%.The three-star green building methods have the highest carbon reduction rate among different star buildings,the emission reduction rates are around 30%.This study's findings are useful for carbon emission calculation and green building design of residential buildings in the future.

Asymmetric copolyimide membranes fabricated by nonsolvent-induced phase separation for He/CH_(4) and He/N_(2) separation

Membrane gas separation is considered an energy-saving technique to extract He from natural gas due to no phase change and room temperature operation.However,the membrane performance was strongly limited by the trade-off between permeance and selectivity.Herein,novel 4,4′-(hexafluoroisopropylidene)diphthalic anhydride(6FDA)-2,2′-bis(3-amino-4-hydroxyphenyl)hexafluoropropane(APAF)-5-amino-2-(4-aminobenzene)benzimidazole(BIA)asymmetric membranes with a thickness of 300 nm were successfully prepared by the non-solvent induced phase separation method.The membrane performance was modulated by regulating dope solution compositions(e.g.,tetrahydrofuran and polymer concentration).The ideal He/CH_(4) selectivity was 124 and the optimized He permeance reached 87 GPU,beyond the current upper bound.He/CH_(4) selectivity was 75 and He permeance was 73 GPU for the binary mixture feed containing 0.2 mol%He.The membrane showed good resistance to CO_(2) and C_(2)H_(6),which are the typical impurities in natural gas.The 6FDA-APAF-BIA membranes have good stability(>160 h),which can provide great potential in He extraction from natural gas.

Amlodipine inhibits the proliferation and migration of esophageal carcinoma cells through the induction of endoplasmic reticulum stress

BACKGROUND L-type calcium channels are the only protein channels sensitive to calcium channel blockers,and are expressed in various cancer types.The Cancer Genome Atlas database shows that the mRNA levels of multiple L-type calcium channel subunits in esophageal squamous cell carcinoma tumor tissue are significantly higher than those in normal esophageal epithelial tissue.Therefore,we hypothesized that amlodipine,a long-acting dihydropyridine L-type calcium channel blocker,may inhibit the occurrence and development of esophageal cancer(EC).AIM To investigate the inhibitory effects of amlodipine on EC through endoplasmic reticulum(ER)stress.METHODS Cav1.3 protein expression levels in 50 pairs of EC tissues and corresponding paracancerous tissues were examined.Subsequently,the inhibitory effects of amlodipine on proliferation and migration of EC cells in vitro were detected using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide and Transwell assays.In vivo experiments were performed using murine xenograft model.To elucidate the underlying mechanisms,in vitro cell studies were performed to confirm that ER stress plays a role in inhibition proliferation and migration of EC cells treated with amlodipine.RESULTS The expression level of Cav1.3 in esophageal carcinoma was 1.6 times higher than that in paracancerous tissues.Amlodipine treatment decreased the viability of esophageal carcinoma cells in a dose-and time-dependent manner.In vivo animal experiments also clearly indicated that amlodipine inhibited the growth of EC tumors in mice.Additionally,amlodipine reduces the migration of tumor cells by inhibiting epithelial-mesenchymal transition(EMT).Mechanistic studies have demonstrated that amlodipine induces ER stress-mediated apoptosis and suppresses EMT.Moreover,amlodipine-induced autophagy was characterized by an increase in autophagy lysosomes and the accumulation of light chain 3B protein.The combination of amlodipine with the ER stress inhibitor 4-phenylbutyric acid further confirmed the role of

As positioning sensors,edge computation power,and communication technologies continue to develop,a moving agent can now sense its surroundings and communicate with other agents.By receiving spatial information from both its environment and other agents,an agent can use various methods and sensor types to localize itself.With its high flexibility and robustness,collaborative positioning has become a widely used method in both military and civilian applications.This paper introduces the basic fundamental concepts and applications of collaborative positioning,and reviews recent progress in the field based on camera,LiDAR(Light Detection and Ranging),wireless sensor,and their integration.The paper compares the current methods with respect to their sensor type,summarizes their main paradigms,and analyzes their evaluation experiments.Finally,the paper discusses the main challenges and open issues that require further research.
