Emerging strategies for nerve repair and regeneration in ischemic stroke:neural stem cell therapy

Ischemic stroke is a major cause of mortality and disability worldwide,with limited treatment options available in clinical practice.The emergence of stem cell therapy has provided new hope to the field of stroke treatment via the restoration of brain neuron function.Exogenous neural stem cells are beneficial not only in cell replacement but also through the bystander effect.Neural stem cells regulate multiple physiological responses,including nerve repair,endogenous regeneration,immune function,and blood-brain barrier permeability,through the secretion of bioactive substances,including extracellular vesicles/exosomes.However,due to the complex microenvironment of ischemic cerebrovascular events and the low survival rate of neural stem cells following transplantation,limitations in the treatment effect remain unresolved.In this paper,we provide a detailed summary of the potential mechanisms of neural stem cell therapy for the treatment of ischemic stroke,review current neural stem cell therapeutic strategies and clinical trial results,and summarize the latest advancements in neural stem cell engineering to improve the survival rate of neural stem cells.We hope that this review could help provide insight into the therapeutic potential of neural stem cells and guide future scientific endeavors on neural stem cells.

Anti-inflammatory sesquiterpene polyol esters from the stem and branch of Tripterygium wilfordii

The stem and branch extract of Tripterygium wilfordii(Celastraceae)afforded seven new dihydroagarofuran sesquiterpene polyesters[tripterysines A‒G(1‒7)]and eight known ones(8‒15).The chemical structures of these new compounds were established based on combinational analysis of HR-ESI-MS and NMR techniques.The absolute configurations of tripterysines A‒C(1‒3)and E‒G(5‒7)were determined by X-ray crystallographic analysis and circular dichroism spectra.All the compounds were screened for their inhibitory effect on inflammation through determining their inhibitory effect on nitric oxide production in LPS-induced RAW 264.7 cells and the secretion of inflammatory cytokines TNF-αand IL-6 in LPS-induced BV2 macrophages.Compound 9 exhibited significant inhibitory activity on NO production with an IC50 value of 8.77μmol·L−1.Moreover,compound 7 showed the strongest inhibitory effect with the secretion of IL-6 at 27.36%.

Exosomes:the next-generation therapeutic platform for ischemic stroke

Current therapeutic strategies for ischemic stroke fall short of the desired objective of neurological functional recovery.Therefore,there is an urgent need to develop new methods for the treatment of this condition.Exosomes are natural cell-derived vesicles that mediate signal transduction between cells under physiological and pathological conditions.They have low immunogenicity,good stability,high delivery efficiency,and the ability to cross the blood–brain barrier.These physiological properties of exosomes have the potential to lead to new breakthroughs in the treatment of ischemic stroke.The rapid development of nanotechnology has advanced the application of engineered exosomes,which can effectively improve targeting ability,enhance therapeutic efficacy,and minimize the dosages needed.Advances in technology have also driven clinical translational research on exosomes.In this review,we describe the therapeutic effects of exosomes and their positive roles in current treatment strategies for ischemic stroke,including their antiinflammation,anti-apoptosis,autophagy-regulation,angiogenesis,neurogenesis,and glial scar formation reduction effects.However,it is worth noting that,despite their significant therapeutic potential,there remains a dearth of standardized characterization methods and efficient isolation techniques capable of producing highly purified exosomes.Future optimization strategies should prioritize the exploration of suitable isolation techniques and the establishment of unified workflows to effectively harness exosomes for diagnostic or therapeutic applications in ischemic stroke.Ultimately,our review aims to summarize our understanding of exosome-based treatment prospects in ischemic stroke and foster innovative ideas for the development of exosome-based therapies.

A Corpus-Based Critical Discourse Analysis of Trump and Biden Administrations’China Policies

The theory of proximization is an effective discourse strategy to study the speaker’s ability to achieve his own legitimacy or reinforce the other’s illegitimacy,and its superiority can be maximized by means of quantitative and comparative analysis.In this study,we collected reports on Trump’s and Biden’s policies on China to build two small corpora,with a total of 11,030 words in the Trump corpus and 17,566 words in the Biden corpus.The critical discourse analysis is combined with proximization theory.With the help of BFSU Qualitative Coder 1.2,Antconc 3.5.7,and Log-Likelihood and Chi-Square Calculator 1.0,a critical cognitive score of the relevant discourse was conducted from the perspective of proximization theory.It has been found that:(1)Both Trump and Biden administrations resort to a large number of spatial proximization strategies to build ODCs converging to IDCs with China as the ODC,posing a threat to internal physical IDCs;(2)in the use of temporal proximization strategy,both administrations use primarily modal verbs and various entities to construct ODCs that extend indefinitely into the present and future,emphasizing the urgency and the threat of the effect and reinforcing the legitimacy of their actions;(3)in terms of axiological proximization strategy,the two administrations differ greatly from each other,indicating that there are still discursive biases.

西北太平洋是全球台风高发区域之一,我国每年受登录台风影响,造成沿海地区发生狂风、暴雨和风暴潮等灾害性天气,准确预测西北太平洋台风路径对我国防震减灾意义重大。基于循环神经网络的多元时间序列预测模型,以处理好的包含中国气象局(CMA)台风最佳路径数据集和欧洲中期天气预报中心(ERA5)数据集两种资料的多元时间序列数据为样本,训练3种循环神经网络并预测未来6、12、24 h的热带气旋(TC)中心位置。结果表明,预测失效为24 h时利用长短期记忆神经网络(LSTM)模型预测E值为173.15 km,误差要远小于门控循环神经网络(GRU)模型和循环神经网络(RNN)模型,认为利用LSTM网络对未来24 h TC中心位置的预测具有可行性和参考价值。

缺血性卒中是最常见的脑卒中类型，可导致严重的神经功能障碍，但目前缺乏有效的诊断方法和治疗手段。外泌体是一种天然的囊泡，可以通过递送蛋白质、脂质、核酸发挥细胞间通讯的作用。缺血性卒中发生后，外泌体分泌量增加且内容物发生变化，因此可以作为潜在的生物标志物，用以辅助缺血性卒中的诊断和治疗。本文旨在综述外泌体在缺血性卒中中潜在的诊断价值，并探讨外泌体在卒中后的修复作用以及作为药物载体的应用前景，最后我们简要介绍了基于外泌体的临床研究现状。

Agricultural Trading among BRICS Countries

This paper analyzes the characteristics of BRICS’s agricultural trading in four ways: export market competition, revealed comparative advantage, trade specialization index, and export similarity index. It shows that BRICS countries export different agricultural goods in the same market, so they do not have real competition in agricultural trading markets. BRICS countries have a comparative advantage in different products; especially Brazil with having total different RCA products. In BRICS countries, China and Brazil are the closest trading partners. Compared with other BRICS countries, China and India have a higher similarity in agricultural trading.

Tryptophan(Trp)carries a unique heteroaromatic indole side chain and plays a critical role in peptide or protein modification.Herein,we have reported a metal-free photoinduced N-H alkylation strategy using N-aryl glycines for specific modification of tryptophan-containing peptides.The robustness of our approach is demonstrated by its wide substrate scope,excellent isolated yields,as well as almost unobservable side effects.Using this highly efficiently metal-free condition,alkylated Trp-containing peptides can be smoothly assembled.This study provides a reliable and practical tool for the chemo-selective modification of various tryptophan containing oligopeptides.

Ferroptosis contributes to JEV-induced neuronal damage and neuroinflammation

Ferroptosis is a newly discovered prototype of programmed cell death (PCD) driven by iron-dependent phospholipid peroxidation accumulation, and it has been linked to numerous organ injuries and degenerative pathologies. Although studies have shown that a variety of cell death processes contribute to JEV-induced neuroinflammation and neuronal injury, there is currently limited research on the specific involvement of ferroptosis. In this study, we explored the neuronal ferroptosis induced by JEV infection in vitro and in vivo. Our results indicated that JEV infection induces neuronal ferroptosis through inhibiting the function of the antioxidant system mediated by glutathione (GSH)/glutathione peroxidase 4 (GPX4), as well as by promoting lipid peroxidation mediated by yes-associated protein 1 (YAP1)/long-chain acyl-CoA synthetase 4 (ACSL4). Further analyses revealed that JEV E and prM proteins function as agonists, inducing ferroptosis. Moreover, we found that treatment with a ferroptosis inhibitor in JEV-infected mice reduces the viral titers and inflammation in the mouse brains, ultimately improving the survival rate of infected mice. In conclusion, our study unveils a critical role of ferroptosis in the pathogenesis of JEV, providing new ideas for the prevention and treatment of viral encephalitis.
