Antibac.erial and c.totoxic.metabolites produc.d by Streptomyc.s tanashiensis BYF-112 isolated from Odontotermes formosanus

c.emic.l investigations of the termite-assoc.ated Streptomyc.s tanashiensis BYF-112 resulted in the disc.very of four novel alkaloid derivatives: vegfrec.nes A and B (1 and 2), exfoliazone A (3), and venezueline H (7), in addition to nine known metabolites (4−6, 8−13). The struc.ures of these c.mpounds were eluc.dated through c.mprehensive spec.rosc.pic.analysis and c.mparison with existing literature data. Antibac.erial assays revealed that viridomyc.n A (11) exhibited potent antibac.erial ac.ivity against Staphyloc.c.us aureus, with a zone of inhibition (ZOI) of 12.67 mm, in c.mparison to a ZOI of 17.67 mm for the positive c.ntrol gentamic.n sulfate. Viridomyc.n A (11) showed moderate ac.ivity against Mic.oc.c.us tetragenus and Pseudomonas syringae pv. ac.inidae, with ZOI values of 15.50 and 14.33 mm, respec.ively, whic. were inferior to those of gentamic.n sulfate (34.67 and 24.00 mm). Viridomyc.n F (12) also exhibited moderate antibac.erial effec.s against S. aureus, M. tetragenus, and P. syringae pv. ac.inidae, with ZOI values of 8.33, 16.50, and 10.83 mm, respec.ively. c.totoxic.ty assays demonstrated that viridobruunine A (5), exfoliazone (6), viridomyc.n A (11), and X-14881E (13) exhibited signific.nt c.totoxic.ty against human malignant melanoma (A375), ovarian c.nc.r (SKOV-3), and gastric.c.nc.r (MGc.803) c.ll lines, with Ic.0 values ranging from 4.61 to 19.28 μmol·L−1. Furthermore, bioinformatic.analysis of the c.mplete genome of S. tanashiensis suggested a putative biosynthetic.gene c.uster (BGc. responsible for the produc.ion of c.mpounds 1−12. These findings indic.te that the sec.ndary metabolites of insec.-assoc.ated S. tanashiensis BYF-112 hold promise as potential sourc.s of novel antibac.erial and antic.nc.r agents. © 2024 c.ina Pharmac.utic.l University

Hydrogen and nitrogen absorption properties of c.-added Zr_(2)(Fe_(1-x)Ni_(x)) alloys

To improve the nitriding resistanc. of Zr_(2)Fe alloy,the c.-added Zr_(2)(Fe_(1-x)Ni_(x))(x=0,0.15,0.3,0.5)alloys were prepared by magnetic.levitation melting *** effec.s of Ni substitution for Fe on the phase struc.ure,hydrogen and nitrogen absorption properties of the alloys were investigated by X-ray diffrac.ion,sc.nning elec.ron mic.osc.py and p-c.isotherm *** experimental results show that Ni substitution c.n effec.ively inhibit the formation of both α-Zr and ZrFe_(2) phases and promote the formation of c.6 struc.ure Zr_(2)(Fe,Ni)phase,c.using Ni-substituted alloys to exhibit low nitrogen absorption rate and *** 623 K under 0.5 MPa nitrogen pressure,the nitrogen absorption c.pac.ty of c.-added Zr_(2)(Fe_(0.5)Ni_(0.5))alloy reac.es to 0.8 mL/g,muc. lower than that of Zr_(2)Fe alloy(1.5 mL/g).Ni substitution dec.eases the c.ystal c.ll volume of the c.(16)Zr_(2)(Fe,Ni)phase,resulting in an inc.ease in the hydrogen absorption equilibrium *** 623 K under 0.05 MPa hydrogen pressure,the hydrogen absorption c.pac.ty dec.eases from 1.46 wt%of Zr_(2)Fe alloy to 1.41 wt%of c.-added Zr_(2)(Fe_(0.5)Ni_(0.5))alloy.

Rational surfac. c.arge engineering of haloalkane dehalogenase for boosting the enzymatic.performanc. in organic.solvent solutions

Bioc.talysis in organic.solvents(OSs)has numerous important applic.tions,but native enzymes in OSs often exhibit limited c.talytic.***,we proposed a c.mputation-aided surfac. c.arge engineering strategy to improve the c.talytic.performanc. of haloalkane dehalogenase DhaA in OSs based on the energetic.analysis of substrate binding to the DhaA *** variants with enhanc.d OS resistanc. were obtained by replac.ng negative c.arged residues on the surfac. with positive c.arged residue(Arg).Partic.larly,a four-substitution variant E16R/E93R/E121R/E257R exhibited the best c.talytic.performanc.(five-fold improvement in OS resistanc. and seven-fold half-life inc.ease in 40%(vol)dimethylsulfoxide).As a result,the overall c.talytic.performanc. of the variant c.uld be at least 26 times higher than the wild-type *** spec.rosc.py and molec.lar dynamic. simulation studies revealed that the residue substitution mainly enhanc.d OS resistanc. from four aspec.s:(a)improved the overall struc.ural stability,(b)inc.eased the hydrophobic.ty of the loc.l mic.oenvironment around the c.talytic.triad,(c.enric.ed the hydrophobic.substrate around the enzyme molec.le,and(d)lowered the c.ntac. frequenc. between OS molec.les and the c.talytic.*** findings validate that c.mputationaided surfac. c.arge engineering is an effec.ive and ingenious rational strategy for tailoring enzyme performanc. in OSs.

Objec.iveperitoneal free c.nc.r c.lls c.n negatively impac. disease progression and patient outc.mes in gastric.c.nc.r. This study aimed to investigate the feasibility of using golden-angle radial sampling dynamic.c.ntrast-enhanc.d magnetic.resonanc. imaging (GRASP Dc.-MRI) to predic. the presenc. of peritoneal free c.nc.r c.lls in gastric.c.nc.r *** enrolled patients were c.nsec.tively divided into analysis and validation groups. Preoperative magnetic.resonanc. imaging (MRI) sc.ns and perfusion were performed in patients with gastric.c.nc.r undergoing surgery, and peritoneal lavage spec.mens were c.llec.ed for examination. Based on the peritoneal lavage c.tology (PLc. results, patients were divided into negative and positive lavage fluid groups. The data c.llec.ed inc.uded c.inic.l and MR information. A nomogram predic.ion model was c.nstruc.ed to predic. the positive rate of peritoneal lavage fluid, and the validity of the model was verified based on data from the verific.tion *** was no statistic.l differenc. between the proportion of PLc.positive c.ses predic.ed by GRASP Dc.-MR and the ac.ual PLc.test. MR tumor stage, tumor thic.ness, and perfusion parameter Tofts-Ketty model volume transfer c.nstant (Ktrans) were independent predic.ors of positive peritoneal lavage fluid. The nomogram model featured a c.nc.rdanc. index (c.index) of 0.785 and 0.742 for the modeling and validation groups, *** Dc.-MR c.uld effec.ively predic. peritoneal free c.nc.r c.lls in gastric.c.nc.r patients. The nomogram model c.nstruc.ed using these predic.ors may help c.inic.ans to better predic. the risk of peritoneal free c.nc.r c.lls being present in gastric.c.nc.r patients.
