Correlation between changes of central neurotransmitter expression and stress response in mice A restraint time-course analysis

BACKGROUND： Changes in central neurotransmitter expression play an important role in stress response and forms the basis for stress-induced psychological and behavior changes. OBJECTIVE： To observe the effects of different restraint stress intervals on brain monoamine neurotransmitter expression, and to investigate the correlation between stress response and neurotransmitter levels. DESIGN： Randomized controlled animal study. SETTING： Chinese Herb and Natural Medicine Institute, Pharmacological College of Jinan University. MATERIALS： Sixty 7-week-old male Kunming mice of clean grade, weighing 18-22 g, were provided by the Guangdong Medical Experimental Animal Center. The experiment was in accordance with animal ethics standards. METHODS： This study was performed at the Chinese Herb and Natural Medicine Institute, Pharmacological College of Jinan University from June 2006 to May 2007. A restraint device for mice was constructed according to published reports. Experimental mice were adaptively fed for 1 week and randomly divided into a control group （n = 10） and an experimental group （n = 50）. The experimental group was sub-divided into five restraint intervals： 4, 8, 12, 18, and 24 hours （n = 10 mice per time point）. Animals in the experimental group were not allowed to eat or drink during the restraint period. Mice in the control group did not undergo restraint, but had identical food and water restrictions. Cerebral cortex and hypothalamus were separated based on observational times and protein was extracted using perchloric acid. Central monoamine neurotransmitter levels were measured using high performance liquid chromatography with electrochemical detection. MAIN OUTCOME MEASURES： Levels of norepinephrine （NE）, dopamine hydrochloride （DA）, 3,4-dihydroxyphen-ylanetic acid （DOPAC）, homovanillic acid （HVA）, 5-hydroxytryptamine （5-HT）, and 5-hydroxyindoleac-etic acid （5-HIAA） in the cerebral cortex and hypothalamus of mice. Chronic superficial gastritis(CSG)is a common disease of the digestive system that possesses a serious *** tablet(JHT),a traditional Chinese medicine(TCM)prescription,exerts therapeutic effects against ***,the molecular basis of its therapeutic effect has not been ***,we employed ultra-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry(UPLC-Q/TOF-MS)based chemical profile identification to determine the chemical components in ***,we applied network pharmacology to illustrate its molecular mechanisms.A total of 96 chemical constituents were identified in JHT,31 of which were confirmed using reference *** on the bioinformatics analysis using the symptom-guided pharmacological networks of“chi,”“blood,”“pain,”and“inflammation,”and target screening through the interaction probabilities between compounds and targets,matrix metalloproteinase 2(MMP2),dopamine d2 receptor(DRD2),and Aldo-keto reductase family 1 member B1(AKR1B1)were identified as key targets in the therapeutic effect exhibited by JHT against ***,according to the inhibitory activities presented in the literature and binding mode analysis,the structural types of alkaloids,flavonoids,organic acids,including chlorogenic acid(10),caffeic acid(13),(-)-corydalmine(33),(-)-isocorypalmine(36),isochlorogenic acid C(38),isochlorogenic acid A(41),quercetin-3-O-a-L-rhamnoside(42),isochlorogenic acid B(47),quercetin(63),and kaempferol(70)tended to show remarkable activities against *** to the above findings,we systematically identified the chemical components of JHT and revealed its molecular mechanisms based on the symptoms associated with CSG.

Analysis of the CDR3 Length Repertoire and the Diversity of TCRα Chain in Human Peripheral Blood T Lymphocytes

Analysis of complementarity determining region 3 （CDR3） length of T lymphocyte receptors （TCRs） by immunoscope spectratyping technique has been used successfully to investigate the diversity of TCR in autoimmune diseases and infection diseases. In this study, we investigated the patterns of CDR3 length distribution for all 32 TCR AV gene families in human peripheral blood lymphocytes of four normal volunteers by the immunoscope spectratyping technique. It was found that PCR products exhibited an obscure band on 1.5% agarose gel electrophoresis. Each TCR AV family exhibited more than 8 bands on 6% sequencing gel electrophoresis. The CDR3 spectratyping of all TCR AV families showed a standard Gaussian distribution with different CDR3 length, and the expression frequency of CDR3 was similar among the gene families. Most of CDR3 in TCR AV family recombine in frame. However, some of the CDR3 showed out-of frame gene rearrangement. Additionally, we found that in some of TCR AV families there were 18 amino acid discrepancies between the longest CDR3 and shortest CDR3. These results may be helpful to further study the recombination mechanism of human TCR genes, the TCR CDR3 gene repertoire, and the repertoire drift in health people and disease state. Specific responses of monoamine neurotransmitters to various acute stressors

This study determined the composition of histamine,serotonin and dopamine using high performance liquid chromatography and electrochemical detection,and compared the changes in monoamine levels in plasma,the cortex and midbrain of mice exposed to acute stressors,such as blood-drawing stimulation or *** demonstrated that plasma histamine levels were markedly increased when mice were exposed to blood-drawing stimulation and restraint ***,serotonin levels decreased in plasma of mice treated with restraint stress,and dopamine levels in plasma had no significant response to the two acute *** three monoamines （histamine,serotonin and dopamine） increased at different degrees in restraint mice,but not in brain regions of blood-drawing stressed *** indicated that histaminergic,serotonergic or dopaminergic systems have their own specific response to different acute stressors.

Synergistic effect of trace elements and flavonoids from Epimedium koreanum Nakai on primary osteoblasts

Several trace elements,particularly,manganese(Mn)and zinc(Zn),are essential in bone metabolism as cofactors for specific *** has been reported that there exists the relationship between osteo-porosis and trace element-deficiency and the efficacy of Ca,Mn and Zn supplementation on spinal bone mineral density in postmenopausal *** Chinese medicines(TCM),such as Herba epimedii,were proved to be effective for prevention of osteoprosis in vivo;however,the efficacy of the main constituents and/or crude extract was not ideal in vitro,which suggested that they may work in another *** purpose of the present study was to examine whether the combination of icariin and total flavonoids(TF)from Herba epimedii with mineral elements,which were abundant in Herba epimedii,would have a more beneficial effect on the viability and differentiation of primary osteoblasts than either agent alone,and to analyze the dada for a possible synergistic,additive or antagonistic *** combinations of 10 μmol/L Zn,Ca and Mn with icariin and total flavonoids greatly improved the cell viability and meanwhile dramatically enhanced the alkaline phosphatase activity as compared to each agent *** the other hand,an increased cell growth inhibition was also observed by combining 0.1 μmol/L,1 μmol/L Zn with 10 μmol/L icariin,and 10 μmol/L Mn with 0.06 μg/mL total *** a decreased alkaline phosphatase activity was also found in several icariin-Zn/Mn and total flavonoids-Zn/Ca/Mn *** results suggested that mineral elements(Zn,Ca,Mn)greatly enhanced the efficacy of icariin and total flavonoids from Herba epimedii on the viability and differentiation of primary osteoblasts by certain combinations.

Codonopsis pilosula(CP),a well-known food medicine homology plant,is commonly used in many *** our preliminary study,a series of pyrrolidine alkaloids with high MS responses were detected as characteristic absorbed constituents in rat plasma after oral administration of CP ***,their structures were unclear due to the presence of various isomers and the lack of reference *** the present study,an MS-guided targeted isolation of pyrrolidine alkaloids of CP extract was performed by ultra-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry(UPLC/Q-TOF MS).For data analysis under fast data directed acquisition mode(Fast-DDA),an effective approach named characteristic fragmentation-assisted mass spectral networking was successfully applied to discover new pyrrolidine alkaloids with high MS response in CP *** a result,seven new pyrrolizidine alkaloids[codonopyrrolidiums C–I(3–9)],together with two known ones(1 and 2),were isolated and identified by NMR spectral *** them,codonopyrrolidium B(1),codonopyrrolidium D(4)and codonopyrrolidium E(5)were evaluated for lipid-lowering activity,and they could improve high fructose-induced lipid accumulation in HepG2 *** addition,the characteristic MS/MS fragmentation patterns of these pyrrolizidine alkaloids were investigated,and 17 pyrrolidine alkaloids were *** approach could accelerate novel natural products discovery and characterize a class of natural products with MS/MS fragmentation patterns from similar chemical *** research also provides a chemical basis for revealing in vivo effective substances in CP.

Four interesting sequoiatones stereoisomers(1-4) were isolated from a wetland soil-derived fungus Talaromyces flavus by chiral *** the basis of comprehensive NMR and mass analyses,their planar structures were elucidated as the same as that of sequoiatone *** them,1 and 3(or 2 and 4)were a pair of enantiomers,and 1 and 2(or 3 and 4) were a pair of stereoisomers with epimerization at C-12,which indicated that sequoiatione-type metabolites exist as enantiomers rather than as optically pure compounds in some *** the quantum chemical ECD calculations,the absolute configurations of C-8 in 1-4 were determined,which is the first report to establish the absolute configuration of C-8 in ***,the absolute configurations of C-12 in sequoiatones are still unsolved.

Chang-Kang-Fang(CKF)formula,a Traditional Chinese Medicine(TCM)prescription,has been widely used for the treatment of irritable bowel syndrome(IBS).However,its potential material basis and underlying mechanism remain ***,this study employed an integrated approach that combined ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q/TOF-MS)with network pharmacology to systematically characterize the phytochemical components and metabolites of CKF,as well as elucidating its underlying *** this comprehensive analysis,a total of 150 components were identified or tentatively characterized within the CKF ***,six N-acetyldopamine oligomers from Cicadae Periostracum and eight resin glycosides from Cuscutae Semen were characterized in this formula for the first ***,149 xenobiotics(58 prototypes and 91 metabolites)were detected in plasma,urine,feces,brain,and intestinal contents,and the in vivo metabolic pathways of resin glycosides were elaborated for the first ***,network pharmacology and molecular docking analyses revealed that alkaloids,flavonoids,chromones,monoterpenes,N-acetyldopamine dimers,p-hydroxycinnamic acid,and Cus-3/isomer might be responsible for the beneficial effects of CKF in treating IBS,and CASP8,MARK14,PIK3C,PIK3R1,TLR4,and TNF may be its potential *** discoveries offer a comprehensive understanding of the potential material basis and clarify the underlying mechanism of the CKF formula in treating IBS,facilitating the broader application of CKF in the field of medicine.
