Wave climate plays an important role in the air-sea interaction over marginal seas. Extreme wave height provides fundamental information for various ocean engineering practices, such as hazard mitigation, coastal structure design, and risk assessment. In this paper, we implement a third generation wave model and conduct a high-resolution wave hindcast over the East china Sea to reconstruct a 15-year wave field from 1988 to 2002 for derivation of monthly mean wave parameters and analysis of extreme wave conditions. The numerical results of the wave field are validated through comparison with satellite altimetry measurements, low-resolution reanalysis, and the ocean wave buoy record. The monthly averaged wave height and wave period show seasonal variation and refined spatial patterns of surface waves in the East china Sea. The climatological significant wave height and mean wave period decrease from the open ocean in the southeast toward the continental area in the northwest, with the pattern generally following the bathymetry. Extreme analysis on the significant wave height at the buoy station indicates the hindcast data underestimate the extreme values relative to the observations. The spatial pattern of extreme wave height shows single peak emerges at the southwest of Ryukyu Island although a wind forcing with multi-core structure at the extreme is applied.

In the establishment of differential equations,the determination of time-varying parameters is a difficult problem,especially for equations related to life ***,we propose a new framework named BioE-PINN based on a physical information neural network that successfully obtains the time-varying parameters of differential *** the proposed framework,the learnable factors and scale parameters are used to implement adaptive activation functions,and hard constraints and loss function weights are skillfully added to the neural network output to speed up the training convergence and improve the accuracy of physical information neural *** this paper,taking the electrophysiological differential equation as an example,the characteristic parameters of ion channel and pump kinetics are determined using *** results demonstrate that the numerical solution of the differential equation is calculated by the parameters predicted by BioE-PINN,the RootMean Square Error(RMSE)is between 0.01 and 0.3,and the Pearson coefficient is above 0.87,which verifies the effectiveness and accuracy of ***,realmeasuredmembrane potential data in animals and plants are employed to determine the parameters of the electrophysiological equations,with RMSE 0.02-0.2 and Pearson coefficient above *** conclusion,this framework can be applied not only for differential equation parameter determination of physiological processes but also the prediction of time-varying parameters of equations in other fields.

Schizophrenia is a common disorder with a high heritability, but its genetic architecture is still *** implemented whole-genome sequencing(WGS) analysis of 8 families with monozygotic(MZ) twin pairs discordant for schizophrenia to assess potential association of de novo mutations(DNMs) or inherited variants with susceptibility to schizophrenia. Eight non-synonymous DNMs(including one splicing site) were identified and shared by twins, which were either located in previously reported schizophrenia risk genes(p.V24689 I mutation in TTN, p.S2506 T mutation in GCN1L1, IVS3+1G > T in DOCK1) or had a benign to damaging effect according to in silico prediction analysis. By searching the inherited rare damaging or loss-of-function(LOF) variants and common susceptible alleles from three classes of schizophrenia candidate genes, we were able to distill genetic alterations in several schizophrenia risk genes, including GAD1, PLXNA2, RELN and FEZ1. Four inherited copy number variations(CNVs; including a large deletion at 16p13.11) implicated for schizophrenia were identified in four families, respectively. Most of families carried both missense DNMs and inherited risk variants, which might suggest that DNMs, inherited rare damaging variants and common risk alleles together conferred to schizophrenia susceptibility. Our results support that schizophrenia is caused by a combination of multiple genetic factors, with each DNM/variant showing a relatively small effect size.

Triple-negative breast cancer(TNBC)is a subtype of breast cancer with high mortality,and the efficacy of monotherapy for TNBC is still ***,we developed a novel combination therapy for TNBC based on a multifunctional nanohollow carbon *** intelligent material contains a superadsorbed silicon dioxide sphere,sufficient loading space,a nanoscale hole on its surface,a robust shell,and an outer bilayer,and it could load both programmed cell death protein 1/programmed cell death ligand 1(PD-1/PD-L1)small-molecule immune checkpoints and small-molecule photosensitizers with excellent loading contents,protect these small molecules during the systemic circulation,and achieve accumulation of them in tumor sites after systemic administration followed by the application of laser irradiation,thereby realizing dual attack of photodynamic therapy and immunotherapy on ***,we integrated the fasting-mimicking diet condition that can further enhance the cellular uptake efficiency of nanoparticles in tumor cells and amplify the immune responses,further enhancing the therapeutic ***,a novel combination therapy“PD-1/PD-L1 immune checkpoint blockade+photodynamic therapy+fasting-mimicking diet”was developed with the aid of our materials,which eventually achieved a marked therapeutic effect in 4T1-tumor-bearing *** concept can also be applied to the clinical treatment of human TNBC with guiding significance in the future.

全科医生作为居民健康“守门人”,在基层糖尿病等慢性疾病管理中发挥着重要作用。糖尿病管理可以延缓病情进展、减少糖尿病并发症和改善患者健康结局,这需要患者和医生之间的有效沟通并互相协作。全科医生具备良好的沟通能力有助于与糖尿病患者建立长期的照护关系、帮助患者建立有效的自我管理技能。本文总结了本研究团队在多种理论框架指导下的全科医生糖尿病沟通技能培训的设计和开发:基于系统综述发现的循证依据,通过定性研究进一步探索糖尿病患者与全科医生的沟通体验和想法,以及混合方法研究设计理念获取全科医学对于医患沟通培训内容的优先排序,旨在为高质量的基层糖尿病管理提供新思路,并为基于证据和医学教育框架设计全科医生相关培训项目提供借鉴。
