Porcine enteric alphacoronavirus infection increases lipid droplet accumulation to facilitate the virus replication

Coronaviruses are widely transmissible between humans and animals, causing diseases of varying severity. Porcine enteric alphacoronavirus(PEAV) is a newly-discovered pathogenic porcine enteric coronavirus in recent years, which causes watery diarrhea in newborn piglets. The host inflammatory responses to PEAV and its metabolic regulation mechanisms remain unclear, and no antiviral studies have been reported. Therefore, we investigated the pathogenic mechanism and antiviral drugs of PEAV. The transcriptomic analysis of PEAV-infected host cells revealed that PEAV could upregulate lipid metabolism pathways. In lipid metabolism, steady-state energy processes, which can be mediated by lipid droplets(LDs), are the main functions of organelles. LDs are also important in viral infection and inflammation. In infected cells, PEAV increased LD accumulation, upregulated NF-κB signaling, promoted the production of the inflammatory cytokines IL-1β and IL-8, and induced cell death. Inhibiting LD accumulation with a DGAT-1 inhibitor significantly inhibited PEAV replication, downregulated the NF-κB signaling pathway, reduced the production of IL-1β and IL-8, and inhibited cell death. The NF-κB signaling pathway inhibitor BAY11-7082 significantly inhibited LD accumulation and PEAV replication. Metformin hydrochloride also exerted anti-PEAV effects and significantly inhibited LD accumulation, downregulated the NF-κB signaling pathway, reduced the production of IL-1β and IL-8, and inhibited cell death. LD accumulation in the lipid metabolism pathway therefore plays an important role in the replication and pathogenesis of PEAV, and metformin hydrochloride inhibits LD accumulation and the inflammatory response to exert anti-PEAV activity and reducing pathological injury. These findings contribute new targets for developing treatments for PEAV infections.

Background:Findings on the association of genetic factors and colorectal cancer(CRC)survival are limited and inconsistent,and revealing the mechanism underlying their prognostic roles is of great importance.This study aimed to explore the relationship between functional genetic variations and the prognosis of CRC and further reveal the possible mechanism.Methods:We first systematically performed expression quantitative trait locus(eQTL)analysis using The Cancer Genome Atlas(TCGA)dataset.Then,the Kaplan-Meier analysis was used to filter out the survival-related eQTL target genes of CRC patients in two public datasets(TCGA and GSE39582 dataset from the Gene Expression Omnibus database).The seven most potentially functional eQTL single nucleotide polymorphisms(SNPs)associated with six survival-related eQTL target genes were genotyped in 907 Chinese CRC patients with clinical prognosis data.The regulatory mechanism of the survival-related SNP was further confirmed by functional experiments.Results:The rs71630754 regulating the expression of endoplasmic reticulum aminopeptidase 1(ERAP1)was significantly associated with the prognosis of CRC(additive model,hazard ratio[HR]:1.43,95%confidence interval[CI]:1.08-1.88,P=0.012).The results of dual-luciferase reporter assay and electrophoretic mobility shift assay showed that the A allele of the rs71630754 could increase the binding of transcription factor 3(TCF3)and subsequently reduce the expression of ERAP1.The results of bioinformatic analysis showed that lower expression of ERAP1 could affect the tumor immune microenvironment and was significantly associated with severe survival outcomes.Conclusion:The rs71630754 could influence the prognosis of CRC patients by regulating the expression of the immune-related gene ERAP1.Trial Registration:No.NCT00454519(https://clinicaltrials.gov/)

Due to the benefits of the high sensitivity,real-time response,no labeling requirement,and good selectivity,fiber optic sensors based on surface plasmon resonance(SPR)have gained popularity in biochemical sensing in recent years.The current research on such sensors is hot in enhancing sensitivity,improving detection accuracy,and achieving the detection of biochemical molecules.The goal of this work is to present a thorough overview of recent developments in the optical fiber SPR biosensor research.Firstly,it explores the basic principles and sensing structures of optical fiber SPR biosensors,focusing on four aspects.Subsequently,this paper introduces three fiber optic surface plasmon biosensors:SPR,localized surface plasmon resonance(LSPR),and long-range surface plasmon resonance(LRSPR).Each concept is explained from the perspective of the basic principles of fiber optic SPR biosensors.Furthermore,a classification of fiber optic SPR biosensors in health monitoring,food safety,environmental monitoring,marine detection,and other applications is introduced and analyzed.Eventually,this paper summarizes the current research directions of SPR biosensors.Meanwhile,it provides a prospective outlook on how fiber optic SPR sensors will develop in the future.

Communication efficiency optimization of federated learning for computing and network convergence of 6G networks

Federated learning effectively addresses issues such as data privacy by collaborating across participating devices to train global models.However,factors such as network topology and computing power of devices can affect its training or communication process in complex network environments.Computing and network convergence(CNC)of sixth-generation(6G)networks,a new network architecture and paradigm with computing-measurable,perceptible,distributable,dispatchable,and manageable capabilities,can effectively support federated learning training and improve its communication efficiency.By guiding the participating devices'training in federated learning based on business requirements,resource load,network conditions,and computing power of devices,CNC can reach this goal.In this paper,to improve the communication eficiency of federated learning in complex networks,we study the communication eficiency optimization methods of federated learning for CNC of 6G networks that give decisions on the training process for different network conditions and computing power of participating devices.The simulations address two architectures that exist for devices in federated learning and arrange devices to participate in training based on arithmetic power while achieving optimization of communication efficiency in the process of transferring model parameters.The results show that the methods we proposed can cope well with complex network situations,effectively balance the delay distribution of participating devices for local training,improve the communication eficiency during the transfer of model parameters,and improve the resource utilization in the network.

Retrieval of Antarctic sea ice freeboard and thickness from HY-2B satellite altimeter data

Antarctic sea ice is an important part of the Earth’s atmospheric system,and satellite remote sensing is an important technology for observing Antarctic sea ice.Whether Chinese Haiyang-2B(HY-2B)satellite altimeter data could be used to estimate sea ice freeboard and provide alternative Antarctic sea ice thickness information with a high precision and long time series,as other radar altimetry satellites can,needs further investigation.This paper proposed an algorithm to discriminate leads and then retrieve sea ice freeboard and thickness from HY-2B radar altimeter data.We first collected the Moderate-resolution Imaging Spectroradiometer ice surface temperature(IST)product from the National Aeronautics and Space Administration to extract leads from the Antarctic waters and verified their accuracy through Sentinel-1 Synthetic Aperture Radar images.Second,a surface classification decision tree was generated for HY-2B satellite altimeter measurements of the Antarctic waters to extract leads and calculate local sea surface heights.We then estimated the Antarctic sea ice freeboard and thickness based on local sea surface heights and the static equilibrium equation.Finally,the retrieved HY-2B Antarctic sea ice thickness was compared with the CryoSat-2 sea ice thickness and the Antarctic Sea Ice Processes and Climate(ASPeCt)ship-based observed sea ice thickness.The results indicate that our classification decision tree constructed for HY-2B satellite altimeter measurements was reasonable,and the root mean square error of the obtained sea ice thickness compared to the ship measurements was 0.62 m.The proposed sea ice thickness algorithm for the HY-2B radar satellite fills a gap in this application domain for the HY-series satellites and can be a complement to existing Antarctic sea ice thickness products;this algorithm could provide long-time-series and large-scale sea ice thickness data that contribute to research on global climate change.

Large numbers of gas reservoirs have been discovered in overpressure basins.Fluid charging has a close relationship with paleo-pressure evolution,affecting the migration of gas reservoirs.To study fluid charging and the related pressure system,we analyzed burial histories and fluid inclusion(PVTx)simulations and conducted basin modeling of the Ledong Slope Zone in the Yinggehai Basin as an example.On the basis of fluid-inclusion assemblages(FIAs),homogenization temperature(Th),final melting temperature(Tm,ice)and Raman spectroscopy in fluid inclusions,there are three stages of fluid charging:during the first and second stage,methane-dominated fluid was charged at 2.2–1.7 Ma and 1.7–0.9 Ma,respectively.In the third stage,CO_(2)-rich hydrothermal fluid was charged since 0.9 Ma.It could be concluded from the well-logging data that the disequilibrium compaction in the Yinggehai Fm.,along with the fluid expansion and clay diagenesis in the Huangliu and Meishan formations,resulted in the overpressure in the Ledong slope zone.The evolution of paleo-pressure was affected by the sedimentation rate of the Yinggehai Fm.,as well as the hydrocarbon generation rate.Additionally,the Ledong Slope Zone is less affected by diapir activity than the nearby diapir area.Based on fluid inclusions,paleo-pressure,basin modeling and geological background,the gas migration history of the Ledong Slope Zone can be divided into four stages:in the first stage,excess pressure was formed around 5 Ma;from 2.2 to 1.7 Ma,there was a reduction in the charging of hydrocarbon fluid and steadily increasing excess pressure;during the 1.7–0.9 Ma period a large amount of hydrocarbon was generated,excess pressure increasing significantly and hydraulic fractures forming in mudstones,With gas reservoirs developing in structural highs;since 0.9 Ma,CO_(2)-rich hydrothermal fluid accumulated in reservoirs adjacent to faults and the pressure coefficient remained stable.The research results are helpful in the study of fluid migration and accumulation mechanisms i

酯类绝缘油在电力系统中的应用越来越广泛,其放电特性和产气规律与传统矿物油存在一定差异。为了研究酯类绝缘油变压器在放电故障发展过程中的放电特性和产气规律,该文以天然酯、改性酯以及合成酯为研究对象,搭建了油纸绝缘放电故障模拟试验平台,采用恒压法进行重复性试验,采集放电发展过程中的放电特征参量,并同时采集试验油样进行油中溶解气体分析。结果表明,酯类绝缘油放电脉冲幅值较高,脉冲重复率较低,其放电发展过程中所释放的能量较大;相同电应力作用下,酯类绝缘油易产生较多的气体产物,其中CO、C_(2)H_(6)和C_(2)H_(2)含量较多,尤其是天然酯FR3;根据放电统计特征参量和油中溶解气体产物可将整个放电过程划分为放电起始阶段、放电发展阶段以及预击穿阶段,其可为酯类绝缘油变压器故障诊断提供一定依据。

【目的】油气来源对比是油气勘探的基础,为扩大江陵凹陷油气资源勘探领域。【方法】基于前期饱和烃分析结果,利用色谱—质谱分析技术对江陵凹陷古近系新沟嘴组下段原油的芳烃馏分进行了分析研究,对原油来源进行了探讨。【结果】研究区原油可分为两大类:Ⅰ类原油取自万城断裂构造带和公安单斜带,其母源形成于一般还原环境并且水体具有一定盐度的半深湖相沉积环境,萘系列及三芳甾烷系列高碳数异构体比值体现其母质类型较好以及陆生高等植物输入较少的特点,该类原油整体处于成熟—高熟阶段,其中万城断裂构造带中的万13井和万12井和公安单斜带中的耀5井是高成熟原油,推测处于各自构造单元中最接近油藏充注点的位置;Ⅱ类原油取自荆州背斜带,成熟度低于Ⅰ类原油,母质类型以低等水生生物输入为主但相较于Ⅰ类原油接受了更多的陆生高等植物输入,形成于高盐度、强还原的深湖相沉积环境,且位于荆州背斜带中部及东南部的Ⅱ2类原油相较于荆州背斜带西北部的Ⅱ1类原油成熟度更低、陆生高等植物输入更多。【结论】万城断裂构造带的Ⅱ油组和Ⅲ油组为Ⅰ类原油的主要烃源岩,同时存在来自荆州背斜带Ⅰ油组的贡献,高的成熟度特征可能指示该类原油还存在除本文外的其他区域或层位的贡献;Ⅱ类原油主要来自荆州背斜带和万城断裂构造带的Ⅱ油组,同时存在荆州背斜带北部Ⅲ油组的贡献。

基于对武云亭地区烃源岩、储层、圈闭等油气藏形成基本条件的总结,系统分析油气特征及来源、压力分布与断层封闭、油气充注历史及油气运聚成藏过程,建立武云亭凝析气田成藏模式。研究表明,武云亭洼广泛分布的煤系烃源岩和宁波27主洼烃源岩生烃能力强、断层和砂岩尖灭耦合形成大型构造-岩性复合圈闭、三角洲沉积优质储集体发育、有利的断-砂配置、盖层质量好等因素共同构成了武云亭凝析气田形成的基础。武云亭油气田平湖组中下部成藏组合具有“武云亭洼煤系源岩供烃、源内持续弱超压充注”成藏特征;中上部成藏组合具有“武云亭洼和宁波27洼双源供烃、侧向持续强充注”成藏特征,早期双向来源原油与晚期大规模侧向来源高熟气混合形成凝析气藏。武云亭油气田总体上具有“早期油晚期气,多期成藏,连续充注”的特征,建立“多源持续强充注-断砂联控”成藏模式。
