Prediction and quantification of effective gas source rocks in a lacustrine basin: Western Depression in the liaohe Subbasin, China

Due to limited data on the geochemical properties of natural gas, estimations are needed for the effective gas source rock in evaluating gas potential. However, the pronounced heterogeneity of mudstones in lacustrine successions complicates the prediction of the presence and geochemical characteristics of gas source rocks. In this paper, the liaohe Subbasin of Northeast China is used as an example to construct a practical methodology for locating effective gas source rocks in typical lacustrine basins. Three types of gas source rocks, microbial, oil-type, and coal-type, were distinguished according to the different genetic types of their natural gas. A practical three-dimensional geological model was developed, refined, and applied to determine the spatial distribution of the mudstones in the Western Depression of the liaohe Subbasin and to describe the geochemical characteristics(the abundance, type, and maturation levels of the organic matter). Application of the model in the subbasin indicates that the sedimentary facies have led to heterogeneity in the mudstones, particularly with respect to organic matter types. The effective gas source rock model constructed for the Western Depression shows that the upper sequence(SQ2) of the Fourth member(Mbr 4) of the Eocene Shahejie Formation(Fm) and the lower and middle sequences(SQ3 and SQ4) of the Third member(Mbr 3) form the principal gas-generating interval. The total volume of effective gas source rocks is estimated to be 586 km3. The effective microbial, oil-type, and coal-type gas source rocks are primarily found in the shallow western slope, the central sags, and the eastern slope of the Western Depression, respectively. This study provides a practical approach for more accurately identifying the occurrence and geochemical characteristics of effective natural gas source rocks, enabling a precise quantitative estimation of natural gas reserves.

Response of ecosystem carbon storage to land use change from 1985 to 2050 in the Ningxia Section of Yellow River Basin,China

Regional sustainable development necessitates a holistic understanding of spatiotemporal variations in ecosystem carbon storage(ECS),particularly in ecologically sensitive areas with arid and semi-arid *** this study,we calculated the ECS in the Ningxia Section of Yellow River Basin,China from 1985 to 2020 using the Integrated Valuation of Ecosystem Services and Tradeoffs(InVEST)model based on land use *** further predicted the spatial distribution of ECS in 2050 under four land use scenarios:natural development scenario(NDS),ecological protection scenario(EPS),cultivated land protection scenario(CPS),and urban development scenario(UDS)using the patch-generating land use simulation(PLUS)model,and quantified the influences of natural and human factors on the spatial differentiation of ECS using the geographical detector(Geodetector).Results showed that the total ECS of the study area initially increased from 1985 until reaching a peak at 402.36×10^(6) t in 2010,followed by a decreasing trend to *** spatial distribution of ECS was characterized by high values in the eastern and southern parts of the study area,and low values in the western and northern *** 1985 and 2020,land use changes occurred mainly through the expansion of cultivated land,woodland,and construction land at the expense of unused *** total ECS in 2050 under different land use scenarios(ranked as EPS>CPS>NDS>UDS)would be lower than that in *** light was the largest contributor to the spatial differentiation of ECS,with soil type and annual mean temperature being the major natural driving *** of this study could provide guidance on the ecological construction and high-quality development in arid and semi-arid areas.

On-site rapid detection of multiple pesticide residues in tea leaves by lateral flow immunoassay

The application of pesticides (mostly insecticides and fungicides) during the tea-planting process will undoubtedly increase the dietary risk associated with drinking tea. Thus, it is necessary to ascertain whether pesticide residues in tea products exceed the maximum residue limits. However, the complex matrices present in tea samples comprise a major challenge in the analytical detection of pesticide residues. In this study, nine types of lateral flow immunochromatographic strips (LFICSs) were developed to detect the pesticides of interest (fenpropathrin, chlorpyrifos, imidacloprid, thiamethoxam, acetamiprid, carbendazim, chlorothalonil, pyraclostrobin, and iprodione). To reduce the interference of tea substrates on the assay sensitivity, the pretreatment conditions for tea samples, including the extraction solvent, extraction time, and purification agent, were optimized for the simultaneous detection of these pesticides. The entire testing procedure (including pretreatment and detection) could be completed within 30 min. The detected results of authentic tea samples were confirmed by ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), which suggest that the LFICS coupled with sample rapid pretreatment can be used for on-site rapid screening of the target pesticide in tea products prior to their market release.

Short-term Preoperative Octreotide for Thyrotropin-secreting Pituitary Adenoma

Background： Thyrotropin-secreting pituitary adenomas （TSHomas） are a rare cause of hyperthyroidism. Somatostatin （SST） analogs work by interacting with somatostatin receptors （SSTRs）. This study aimed to evaluate short-term preoperative octreotide （OCT） use in TSHoma patients and to investigate SSTR2 and SSTR5 expression and observe structural changes in tumor tissue. Methods： We reviewed records and samples from eight TSHoma patients treated between July 2012 and July 2015. We tested immunohistochemically for SSTR2/5 expression and examined TSHoma cells for morphological changes. Signed rank sum test was used to compare the efficacy of short-term preoperative OCT treatment. Results： OCT treatment （median time： 7.9 days, range： 3-16 days; median total dose： 1.8 mg, range： 0.94.2 mg） led to significant decrease in all patients＇ thyroid hormone levels （FT3 [nmol/L]： 8.33 [7.02, 12.29] to 4.67 [3.52, 5.37] [P = 0.008]; FT4 [pmol/L]： 25.36 [21.34, 28.99] to 16.66 [14.88, 21.49] [P = 0.016]; and TSH [gU/ml]： 5.80 [4.37, 6.78] to 0.57 [0.19, 1.24] [P = 0.008]）. All the eight tumor specimens expressed high SSTR2 protein levels; 5/8 expressed high SSTRS, but 3/8 that expressed low SSTR5 presented a significantly higher TS H suppression rate （P = 0.036）. Electron microscopy showed subcellular level impairments, including clumped nuclear chromatin and reduced cytoplasmic volume. Golgi complexes were observed in the OCT-treated TSHoma specimens. Conclusions： OCT can control hormone levels and damage the ultrastructure of tumor cells and organelles. Short-term response to OCT may be related to SSTR5 expression. Preoperative SST analog treatment for TSHoma could be considered as a combination therapy.

Analysis of the influencing factors and clinical related characteristics of pulmonary tuberculosis in patients with type 2 diabetes mellitus

BACKGROUND In China,the prevalence of type 2 diabetes mellitus(T2DM)among diabetic patients is estimated to be between 90%-95%.Additionally,China is among the 22 countries burdened by a high number of tuberculosis cases,with approximately 4.5 million individuals affected by active ***,T2DM poses a significant risk factor for the development of tuberculosis,as evidenced by the increased incidence of T2DM coexisting with pulmonary tuberculosis(T2DMPTB),which has risen from 19.3%to 24.1%.It is evident that these two diseases are intricately interconnected and mutually reinforcing in *** To elucidate the clinical features of individuals diagnosed with both T2DM and tuberculosis(T2DM-PTB),as well as to investigate the potential risk factors associated with active tuberculosis in patients with *** T2DM-PTB patients who visited our hospital between January 2020 and January 2023 were selected as the observation group,Simple DM patients presenting to our hospital in the same period were the control group,Controls and case groups were matched 1:2 according to the principle of the same sex,age difference(±3)years and disease duration difference(±5)years,patients were investigated for general demographic characteristics,diabetes-related characteristics,body immune status,lifestyle and behavioral habits,univariate and multivariate analysis of the data using conditional logistic regression,calculate the odds ratio(OR)values and 95%CI of OR *** A total of 315 study subjects were included in this study,including 105 subjects in the observation group and 210 subjects in the control *** of the results of both anthropometric and biochemical measures showed that the constitution index,systolic blood pressure,diastolic blood pressure and lymphocyte count were significantly lower in the case group,while fasting blood glucose and high-density lipoprotein cholesterol levels were significantly higher than those in the control ***

Petroleum ether extract of Derris eriocarpa How.(PEEDEH) investigate antidiabetic nephropathy effect and mechanism through TLR4/NF-κB/caspase-3 pathways

OBJECTIVE To investigate anti-diabetic nephropathy effect and mechanism through Toll-like receptor 4（TLR4）/NF-κB/Caspase-3 pathway of petroleum ether extract of Derris eriocarpa How.（PEEDEH）.METHODS Type 2 diabetes was induced in male Kunming（KM） mice through a combination of high-sugar and highfat diet for 2 months, after that, which was injected with streptozotocin（STZ, 80 mg·kg-1） via tail vein. After 3 d,mice with a fasting blood glucose（FBG）≥ 11.1 mmol·L;were considered diabetic. The mice were randomly assigned to normal control group, model group, metformin positive group(320 mg · kg;) and once that received low-, middle-, high-dose(200, 400, 800 mg · kg;) of PEEDEH for 21 days. RESULTS The intraperitoneal administration of PEEDEH reduced the levels of FBG,blood urine nitrogen（BUN）, serum creatinine（Scr）, kidney index, interleukin-6（IL-6） and tumor necrosis factor-α（TNF-α）. Pathological changes of kidney observed by HE staining revealed that the pathology was improved. The expression of TLR-4, NF-κB, caspase-3 and COX-2 detected using immunohistochemical analysis indicated that their expressions were reduced after PEEDEH administration. PEEDEH also significantly reduced m RNA and protein expression levels of TLR-4/NF-κB/caspase-3 pathway in kidney compared to the diabetic group. CONCLUSION PEEDEH ameliorated DN through TLR-4/NF-κB/Caspase-3 signaling pathways.

A High-Performance Electrochemical Analysis Platform for Ultrasensitive Detection of HTLV-1 DNA: Integrating Cascade Signal Amplifi cation with λ-Exonuclease-Assisted Target Recycling and Enzyme Catalysis

Human T-cell lymphophilic virus type 1 (HTLV-1), the known retrovirus causing cancer in humans, is closely associated with adult T-cell leukemia/lymphoma and HTLV-1 associated myelopathy/tropical spastic paraparesis. Due to its ability to evade the host's defense mechanisms, early tracking of HTLV-1 becomes crucial. In this study, we integrateλ-Exonuclease(λ-Exo)-assisted target recycling with a terminal deoxynucleotidyl transferase (TdT)-mediated template-free DNA extension process to develop an electrochemical analysis platform for the specifi c and sensitive detection of HTLV-1 DNA. During theλ-Exo-assisted target recycling, HTLV-1 DNA is recognized by hairpin DNA (Hp-DNA), forming double-stranded DNA(dsDNA) through DNA hybridization. The dsDNA, featuring blunt 5'terminal phosphorylation, is cleaved byλ-Exo, generating abundant short output sequence (sDNA). HTLV-1 DNA is released, initiating a cyclic hybridization-cleavage ***, thiol-labelled capture DNA (CP-DNA) assembled on gold electrode surface captures a substantial amount of the generated sDNA, forming CP-DNA-sDNA nanostructures. When TdT and dNTPs are present on the electrode surface, the3'-OH terminal of sDNA extends to generate long single-stranded DNA (ssDNA) structure. Methylene blue (MB) is selected as the electrochemical signal molecule. MB not only binds with ssDNA but also interacts specifi cally with dsDNA, resulting in a signifi cantly enhanced electrochemical signal on modifi ed electrode surface. The detection limit of HTLV-1 DNA is as low as 19 amol/L (S/N=3) when the two signal amplifi cation strategies are combined. The analysis platform exhibits excellent analytical performance and holds promise as a novel tool for the early tracing and diagnosis of HTLV-1 DNA.
