Background:Atopic dermatitis(AD)affects approximately 10%of adults ***310 is a humanized monoclonal antibody targeting interleukin-4 receptor alpha that blocks interleukin-4 and interleukin-13 *** trial aimed to evaluate the efficacy and safety of CM310 in Chinese adults with moderate-to-severe ***:This multicenter,randomized,double-blind,placebo-controlled,phase 2b trial was conducted in 21 medical institutions in china from February to November *** 120 eligible patients were enrolled and randomized(1:1:1)to receive subcutaneous injections of 300 mg CM310,150 mg CM310,or placebo every 2 weeks for 16 weeks,followed by an 8-week follow-up *** primary endpoint was the proportion of patients achieving≥75%improvement in the Eczema Area and Severity Index(EASI-75)score from baseline at week *** and pharmacodynamics were also ***:At week 16,the proportion of EASI-75 responders from baseline was significantly higher in the CM310 groups(70%[28/40]for high-dose and 65%[26/40]for low-dose)than that in the placebo group(20%[8/40]).The differences in EASI-75 response rate were 50%(high ***,95%CI 31%-69%)and 45%(low ***,95%CI 26%-64%),with both P values10 at both doses also significantly improved the EASI score,Investigator’s Global Assessment score,daily peak pruritus Numerical Rating Scale,AD-affected body surface area,and Dermatology Life Quality Index compared with ***310 treatment reduced levels of thymus and activation-regulated chemokine,total immunoglobulin E,lactate dehydrogenase,and blood *** incidence of treatment-emergent adverse events(TEAEs)was similar among all three groups,with the most common TEAEs reported being upper respiratory tract infection,atopic dermatitis,hyperlipidemia,and *** severe adverse events were deemed to be attributed to ***:CM310 at 150 mg and 300 mg every 2 weeks demonstrated significant efficacy and was well-tol

In this paper,we provide a new theoretical framework of pyramid Markov processes to solve some open and fundamental problems of blockchain selfish mining under a rigorous mathematical *** first describe a more general model of blockchain selfish mining with both a two-block leading competitive criterion and a new economic incentive *** we establish a pyramid Markov process and show that it is irreducible and positive recurrent,and its stationary probability vector is matrix-geometric with an explicitly representable rate ***,we use the stationary probability vector to study the influence of orphan blocks on the waste of computing ***,we set up a pyramid Markov reward process to investigate the long-run average mining profits of the honest and dishonest mining pools,*** a by-product,we build one-dimensional Markov reward processes and provide some new interesting interpretation on the Markov chain and the revenue analysis reported in the seminal work by Eyal and Sirer(2.14).Note that the pyramid Markov(reward)processes can open up a new avenue in the study of blockchain selfish *** we hope that the methodology and results developed in this paper shed light on the blockchain selfish mining such that a series of promising research can be developed potentially.

采用溶胶-凝胶-常压干燥的方法,以耐热杂化酚醛树脂(PF)为基体,复合碳纤维编织物(CF)制备树脂基防隔热一体化热防护复合材料(PF/CF-HT01)。利用热分析(TG)、电子万能试验机研究材料热稳定性和高温力学性能,利用氧乙炔装置研究材料耐烧蚀性能,利用扫描电子显微镜(SEM)、X线衍射仪(XRD)研究材料微观结构演变过程。结果表明:空气中树脂基体的初始分解温度为387.3℃,最大分解温度为644.7℃,800℃时残炭率为13.8%;复合材料初始分解温度为405.3℃,800℃时残炭率为42.8%;复合材料常温压缩强度最大为542.6 MPa,经1 000℃原位热处理30和60 s后的最大压缩强度分别为166.2.149.9 MPa。复合材料具有良好的防隔热一体化性能,其线烧蚀率可达0.039 mm/s,单次热考核结束时背温低于100℃、继续热传导后最高背温低于200℃。高温作用下材料快速陶瓷化形成致密的SiO2和BN瓷化层,赋予材料突出的耐烧蚀抗冲刷性能,而底层仍然保留着多孔结构使得材料保持较好的隔热性能。

Brown adipose tissue (BAT) plays an essential role in non-shivering thermogenesis. The phosphatidylinositol transfer protein, cytoplasmic 1 (PITPNC1) is identified as a lipid transporter that reciprocally transfers phospholipids between intracellular membrane structures. However, the physiological significance of PITPNC1 and its regulatory mechanism remain unclear. Here, we demonstrate that PITPNC1 is a key player in thermogenesis of BAT. While Pitpnc1^(−/−) mice do not differ with wildtype mice in body weight and insulin sensitivity on either chow or high-fat diet, they develop hypothermia when subjected to acute cold exposure at 4℃. The Pitpnc1^(−/−) brown adipocytes exhibit defective β-oxidation and abnormal thermogenesis-related metabolism pathways in mitochondria. The deficiency of lipid mobilization in Pitpnc1^(−/−) brown adipocytes might be the result of excessive accumulation of phosphatidylcholine and a reduction of phosphatidic acid. Our findings have uncovered significant roles of PITPNC1 in mitochondrial phospholipid homeostasis and BAT thermogenesis.

To determine whether RNA interference （RNAi） could block cytotoxic T-lymphocyte antigen 4 （CTLA4） in human lymphocytes in vitro and promote IFN-T and IL-2 secretions, three small interfering RNAs （siRNAs） were selected based on target specificity sequences of human CTLA4 and transfected into human lymphocytes of chronic HBV patients. As a result, the expression of human CTLA4 mRNA was efficiently suppressed by all the three siRNAs. Compared with negative control （siRNA-co）, siRNA-1 inhibited the expression of CTLA4 most efficiently and was used in the further study. The expressions of IFN-γ and IL-2 were upregulated and the level of IL-4 was almost unchanged in lymphocytes transfected with siRNA-1 compared with the blank control. These results indicated that siRNA-1 led to IFN-γ and IL-2 secretions, which is a main response of Th1/Th2. In a conclusion, RNAi significantly suppressed the expression of human CTLA4 mRNA in human lymphocytes in vitro, and could induce Th1/Th2 response. It could be a new therapeutic strategy for chronic HBV infection.

Dehydro-Diels-Alder(DDA)reaction is a textbook reaction for preparing six-membered rings in solution but is scarcely seen in solid-state *** this work,using multiple characterization techniques,we demonstrate that the bowl-shaped clusters C_(18)Te_(3)Br_(4)(Bu-O)_(6) might experience a DDA reaction at room temperature and high pressure between 5.5 and 7.4 *** 17.0 GPa,it is found that the bonding conversion from the intramolecular sp^(2) to the intermolecular spa occurred,in the form of pressure-induced *** recovered samples from 20.0 and 36.1 GPa showed incomplete reversibility,while the decompression-induced graphitization of glassy carbon was observed during decompression from 46.5 *** electrochemical impedance spectroscopy results indicated that the transport properties changed from grain boundary dominant to grain dominant due to the DDA reaction and the grain boundary effect disappeared as the intermolecular sp3 bonding building-up and carrier transmission channel formation above 17.0 *** results in this study open a new route to construct the crystalline carbon materials with different transport properties.

目的探讨采用颈椎后路单侧显露椎管扩大椎板成形术治疗颈椎后路单开门椎管扩大椎板成形术后骨化进展的可行性和有效性。方法2021年6月—2023年3月,采用颈椎后路单侧显露椎管扩大椎板成形术治疗颈椎后路单开门椎管扩大椎板成形术后骨化进展患者12例。记录翻修手术的手术时间、术中出血量、术后引流量及手术并发症情况。于术前、术后12个月及末次随访时采用疼痛视觉模拟量表(VAS)评分评估颈部疼痛程度;采用颈椎功能障碍指数(NDI)评价颈椎功能;采用日本骨科学会(JOA)评分评估神经功能状况。测量术前、术后12个月及末次随访时C_(2~7)Cobb角、颈椎活动度(ROM)、T1倾斜角、C_(2~7)矢状面平衡(SVA)及椎管横截面积。结果所有手术顺利完成,所有患者均未发生明显神经损伤、深部感染等严重并发症。手术时间为(105.38±19.06)min,术中出血量为(128.44±33.62)mL,术后引流量为(142.51±25.38)mL。所有患者随访12~33(22.64±6.72)个月。翻修术后12个月、末次随访VAS评分、NDI及JOA评分较术前明显改善,差异均有统计学意义(P12个月、末次随访时C_(2~7)Cobb角、ROM、T1倾斜角及C_(2~7)SVA较术前无明显变化,差异无统计学意义(P>0.05)。翻修术后12个月、末次随访时椎管横截面积较术前明显增加,差异均有统计学意义(P<0.05)。随访中无钛板断裂等严重内置物并发症发生。结论采用颈椎后路单侧显露椎管扩大椎板成形术治疗颈椎后路单开门椎管扩大椎板成形术后的骨化进展安全、有效。翻修手术明显改善了患者的神经功能、保留了颈椎ROM。
