Hsa-miR-214-3p inhibits breast cancer cell growth and improves the tumor immune microenvironment by downregulating B7H3

Background:Immune checkpoint inhibitors play an important role in the treatment of solid tumors,but the currently used immune checkpoint inhibitors targeting programmed cell death-1(PD-1),programmed cell death ligand-1(PD-l1),and cytotoxic T-lymphocyte antigen-4(CTlA-4)show limited clinical efficacy in many breast cancers.B7H3 has been widely reported as an immunosuppressive molecule,but its immunological function in breast cancer patients remains ***:We analyzed the expression of B7H3 in breast cancer samples using data from the Cancer Genome Atlas Program(TCGA)and the Gene Expression Omnibus(GEO)*** were selected using the TarBase,miRTarBase,and miRBase *** regulatory role of the microRNA hsa-miR-214-3p on B7H3 was investigated through dual-luciferase reporter assays,which identified the specific action sites of *** expression levels of B7H3 and hsa-miR-214-3p in human breast cancer tissues and adjacent normal tissues were quantified using Western blotting and quantitative PCR(qPCR).In vitro experiments were performed to observe the effects of modulating the expression of B7H3 or hsa-miR-214-3p on breast cancer cell proliferation and ***,the regulatory impact of hsa-miR-214-3p on B7H3 was ***-linked immunosorbent assays(ElISA)and flow cytometry were employed to assess the effects of co-cultured breast cancer cells and normal human peripheral blood mononuclear cells(PBMCs)on immune cells and associated ***:In breast cancer tissues,the expression level of B7H3 is inversely correlated with that of hsa-miR-214-3p,as well as with the regulatory effects on breast cancercell ***-miR-214-3p was found to inhibit breast cancer cell growth by downregulating ***,our research identified,for the first time,two binding sites for hsa-miR-214-3p on the 3’UTR of B7H3,both of which exert similar effects ***-culture experiments revealed that hsamiR-214-3p

Experimental Study for Effect of Multi-Site Spark Ignition on Dedicated Hybrid Engine Performance under High Dilution Condition

In this study,a triple spark ignition scheme was first designed on a three-cylinder 1.5-l dedicated hybrid engine（DHE）.On this basis,the effect of different ignition modes on engine combustion and emission characteristics was studied,especially under high dilution *** results tested at 2000 r/min and 0.8 MPa BMEP（brake mean effective pressure） show that with highly increased in-cylinder flow intensity,using only passive prechamber（PPC） has a lower lean limit than that with single central spark plug（CSP）,thereby leading to slightly higher minimum fuel consumption and nitrogen oxides（NOx） *** side spark plugs（SSP）based on PPC can result in improved capability of lean limit extension and engine performance than ***,the improvement level is lower than that with triple spark plugs（TSP）.As the excess air ratio λincreases,the advantage of PPC and PPC with SSP in improving the combustion phasing compared with CSP gradually ***,the increasing tendency of their ignition delay and combustion duration is more *** added SSP based on PPC can effectively shorten the ignition delay of leaner mixture,but the combustion duration can be only slightly *** a result,under extremely lean condition,the advantage of PPC and PPC with SSP in terms of combustion characteristics over CSP becomes much *** contrast,the TSP ignition can achieve much shorter ignition delay and combustion duration simultaneously under this *** to the highest available dilution level,the TSP ignition achieves the lowest raw ***,it can also reduce the raw carbon monoxide（CO） and hydrocarbons（HC） emissions compared to CSP due to a more thorough combustion of the end gas *** on the excellent performance of TSP,the highest engine brake thermal efficiency（BTE） was further *** results show that with normal RON 92 fuel,the engine finally achieved 43.69% and 45.02% BTE under stochiometric mode with exhaust

A hydrothermal reaction of zinc acetate with molybdenum trioxide, vanadium pentoxide, phosphorus pentoxide and 2,2?-bpy (2,2?-bipyridine) led to the title compound [Zn(2,2?- bpy)2(H2O)]2[HPMo12O40(VO)2] 1. Single-crystal X-ray diffraction revealed that 1 (C40H36- N8O44PV2Zn2Mo12) crystallizes in the triclinic system, space group P1 with a = 11.881(2), b = 12.468(2), c = 12.622(2) ?, α = 71.333(8), β = 74.485(4), γ = 86.896(5)o, V = 1705.8(4) ?3, Mr = 2747.64, Z = 1, Dc = 2.675 g/cm3, μ = 3.201 mm-1, F(000) = 1309, S = 1.070, the final R = 0.0697 and wR = 0.1905 for 5368 observed reflections with I > 2σ(I). Compound 1 is built on a mixed-metal bicapped [HPMo12V2O42] subunit covalently bonded to two [Zn(2,2?-bpy)2(H2O)]2+ clusters via terminal oxygen atoms of the capping V atoms. Each Zn atom is six-coordinated by four nitrogen atoms from two 2,2?-bpy ligands, one terminal oxygen atom from coordinated water mole- cule and another one from the capped {VO} unit.

Boosting cationic and anionic redox activity of li-rich layered oxide cathodes via li/Ni disordered regulation

lithium-rich layered oxides （llOs） are increasingly recognized as promising cathode materials for nextgeneration high-energy-density lithium-ion batteries （lIBs）.However,they suffer from voltage decay and low initial Coulombic efficiency （ICE） due to severe structural degradation caused by irreversible O ***,we introduce a three-in-one strategy of increasing Ni and Mn content,along with li/Ni disordering and TM–O covalency regulation to boost cationic and anionic redox activity simultaneously and thus enhance the electrochemical activity of *** target material,li1.2Ni0.168Mn0.558Co0.074O2（l1）,exhibits an improved ICE of 87.2%and specific capacity of 293.2 mA h g-1and minimal voltage decay of less than 0.53 m V cycle-1over 300 cycles at 1C,compared to li1.2Ni0.13Mn0.54Co0.13O2（ls）(274.4 mA h g-1for initial capacity,73.8%for ICE and voltage decay of 0.84 mV/cycle over 300 cycles at 1C).Theoretical calculations reveal that the density of states （DOS） area near the Fermi energy level for l1 is larger than that of ls,indicating higher anionic and cationic redox reactivity than ***,l1 exhibits increased O-vacancy formation energy due to higher li/Ni disordering of 4.76%（quantified by X-ray diffraction Rietveld refinement） and enhanced TM–O covalency,making lattice O release more difficult and thus improving electrochemical *** increased li/Ni disordering also leads to more Ni2+presence in the li layer,which acts as a pillar during li+de-embedding,improving structural *** research not only presents a viable approach to designing low-Co llOs with enhanced capacity and ICE but also contributes significantly to the fundamental understanding of structural regulation in high-performance lIB cathodes.

liquid-liquid Equilibrium for Extraction of Benzene from 1-Hexene Using Two Different Solvents

Two different solvents had been used to separate benzene from 1-hexene under atmosphere pressure by employing one of the following two solvents, viz.: dimethyl sulfoxide (DMSO) at 298.15 K and 318.15 K, and furfural at 298.15 K. A series of liquid-liquid equilibrium (llE) data had been obtained and the distribution coefficient together with the separation factor were calculated from them. Both the NRTl and the UNIQUAC models could fit in with the experimental data quite well.

Suppressing oxygen redox in layered oxide cathode of sodium-ion batteries with ribbon superstructure and solid-solution behavior

Sodium-ion batteries(SIBs)are proved as one of the most acceptable candidates for replacing lithium-ion batteries in some fields by virtue of a similar“rocking chair”mechanism and the abundance of *** voltage,rate performance,and energy density of these batteries are mainly determined by the ***,a li-Ni-Co co-substituted P2-Na_(0.67)[li_(0.1)(Mn_(0.7)Ni_(0.2)Co_(0.1))_(0.9)]O_(2)(NlMNC)with ribbon super-structure is prepared with the aim of multi-ion synergistic *** to the addition of Ni and Co,the Jahn-Teller distortion of Mn can be suppressed corresponding with the improved structural stability,and a little bit of oxygen redox activities is *** with the substitution of 10%li,the X-ray diffraction(XRD)peaks of NlMNC show the ribbon superstructure at about 21°and 22°.The smooth charge/discharge profiles of the NlMNC cathode exhibit the solid-solution *** addition,the platform at high voltage disappears corresponding with the existing oxygen redox activities being suppressed which may be related to the ribbon superstructure and the promotion of the Ni *** NlMNC cathode can deliver a reversible discharge capacity of 123.5 mA h g^(-1)at 10 mA g^(-1).Even if the current density increases to 500 mA g^(-1),a reversible discharge capacity of 112.8 mA h g^(-1)still can be *** distinguished cycling stability is related to the reversible migration of li+between the metal oxide layer and the interlayer and low volume change during *** is also needing to be mentioned that the capacity retention of NlMNC cathode is about 94.4%(based on the highest discharge capacity)after 100 *** work presents an effective route to develop high-performance cathodes for SIBs.

Biochemical characterization of chitin synthase activity and inhibition in the African malaria mosquito, Anopheles gambiae

Chitin synthase （CHS） is an important enzyme catalyzing the formation of chitin polymers in all chitin containing organisms and a potential target site for insect pest control. However, our understanding of biochemical properties of insect CHSs has been very limited. We here report enzymatic and inhibitory properties of CHS prepared from the African malaria mosquito, Anopheles gambiae. Our study, which represents the first time to use a nonradioactive method to assay CHS activity in an insect species, determined the optimal conditions for measuring the enzyme activity, including pH, temperature, and concentrations of the substrate uridine diphosphate N-acetyl-D-glucosamine （UDP- GlcNAc） and Mg＋＋. The optimal pH was about 6.5-7.0, and the highest activity was detected at temperatures between 37℃ and 44℃. Dithithreitol is required to prevent melanization of the enzyme extract. CHS activity was enhanced at low concentration of GIcNAc, but inhibited at high concentrations. Proteolytic activation of the activity is significant both in the 500×g supernatant and the 40 000×g pellet. Our study revealed only slight in vitro inhibition ofA. gambiae CHS activity by diflubenzuron and nikkomycin Z at the highest concentration （2.5μmol/l） examined. There was no in vitro inhibition by polyoxin D at any concentration examined. Furthermore, we did not observe any in vivo inhibition of CHS activity by any of these chemicals at any concentration examined. Our results suggest that the inhibition of chitin synthesis by these chemicals is not due to direct inhibition of CHS in A. gambiae.

Cooper Pairing Near the Boundary in the -U Hubbard Model

The Cooper pairing near the boundary of a superconductor is analyzed by using the linear-response theory The system is described by the square lattice(t,-U)Hubbard model occupying half *** half filling,the pairing also corresponds to the antiferromagnetic ordering in the repulsive Hubbard *** numerical result clearly shows that,at weak to intermediate coupling(U/t≤l),the boundary exerts strong influence on the order parameter;the weaker the coupling,the deeper the order parameter is suppressed near the *** present scheme is valid in the whole temperature regime of Cooper pairing.

Bending strength and fracture surface topography of natural fiber-reinforced shell for investment casting process

In order to improve the properties of silica sol shell for investment casting process, various contents of cattail fibers were added into the slurry to prepare a fiber-reinforced shell in the present study. The bending strength of fiber-reinforced shell was investigated and the fracture surfaces of shell specimens were observed using SEM. It is found that the bending strength increases with the increase of fiber content, and the bending strength of a green shell with 1.0 wt.% fiber addition increases by 44% compared to the fiber-free shell. The failure of specimens of the fiber-reinforced green shell results from fiber rupture and debonding between the interface of fibers and adhesive under the bending load. The micro-crack propagation in the matrix is inhibited by the micro-holes for ablation of f ibers in specimens of the f iber-reinforced shell during the stage of being fired. As a result, the bending strength of specimens of the fired shell had no significant drop. Particularly, the bending strength of specimens of the fired shell reinforced with 0.6wt.% fiber reached the maximum value of 4.6 MPa.
