Punicalagin alleviates hypercholesterolemia in mice through modulating farnesoid X receptor signaling and modulating gut microbiota

Punicalagin has been demonstrated to exhibit anti-oxidant and anti-inflammatory properties， but whether and how it could impact hypercholesterolemia remains not fully explored. The aim of this study was to investigate the influence of punicalagin on hypercholesterolemia in mice and its related mechanisms. After 6 weeks’ intervention， punicalagin significantly reduced serum total cholesterol and low density lipoprotein cholesterol (LDL-C) levels in mice fed a high-fat high-cholesterol (HFHC) diet. Meanwhile， punicalagin supplementation lowered hepatic cholesterol level， which corresponded to the down-regulation of cholesterol synthesis genes (Fdps， Cyp51) and up-regulated bile acid synthesis genes (Cyp7a1， Cyp27a1). In addition， bile acid reabsorption was retarded in punicalagin-fed mice through down-regulating ileal apical sodium-dependent BA transporter (ASBT). Furthermore， intestinal farnesoid X receptor (FXR)-fibroblast growth factor 15 (Fgf15) pathway was inhibited while hepatic FXR-small heterodimeric partner (SHP) pathway was activated in punicalagin group. Microbiota analysis and targeted metabolomics showed that punicalagin decreased the abundance of bile-salt hydrolase (BSH)-producing bacteria (Clostridiaceae and Bifidobacteriaceae) and the ratio of primary BAs to secondary BAs. In conclusion， the cholesterol-lowering effect of punicalagin partly through down-regulating cholesterol synthesis and increasing cholesterol catabolism， which could be achieved by regulating gut microbiota， altering bile acid composition and modulating FXR signaling pathway. These findings indicate the potential application of punicalagin-related products as an alternative strategy for hypercholesterolemia prevention and mitigation.

Alzheimer ’s disease(AD) is a leading cause of dementia in the ***-activated protein kinase phosphatase 1(MKP-1) plays a neuroprotective role in ***,the molecular mechanisms underlying the effects of MKP-1 on AD have not been extensively ***(miRNAs) regulate gene expression at the post-transcriptional level,thereby repressing mRNA ***,we reported that the microRNA-429-3p(miR-429-3p) was significantly increased in the brain of APP23/PS45 AD model mice and N2AAPPAD model *** further found that miR-429-3p could downregulate MKP-1 expression by directly binding to its 3’-untranslated region(3’ UTR).Inhibition of miR-429-3p by its antagomir(A-miR-429) restored the expression of MKP-1 to a control level and consequently reduced the amyloidogenic processing of APP and Aβ *** importantly,intranasal administration of A-miR-429 successfully ameliorated the deficits of hippocampal CA1 long-term potentiation and spatial learning and memory in AD model mice by suppressing extracellular signal-regulated kinase(ERK1/2)-mediated GluAl hyperphosphorylation at Ser831 site,thereby increasing the surface expression of GluAl-containing α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors(AMPARs).Together,these results demonstrate that inhibiting miR-429-3p to upregulate MKP-1 effectively improves cognitive and synaptic functions in AD model mice,suggesting that miR-429/MKP-1 pathway may be a novel therapeutic target for AD treatment.

aiM:To compare high or low concentration of hyaluronic acid eye drops(HY)for dry eye syndromes(DES).METHODS:Randomized controlled trials(RCTs)comparing various concentrations of HY were searched in PubMed,Embase,Web of Science,Cochrane,SinoMed,CNKI,Wanfang Database,CQVIP,and Chinese journals databases between inception and July *** standardized mean differences(SMD)or weighted mean difference(WMD)with 95%confidence intervals(CI)from RCTs evaluating Schirmer’s I test(SIT),corneal fluorescein staining score(CFS),tear breakup time(TBUT),DES score(DESS),and Ocular Surface Disease Index(OSDI)were *** analysis,Egger’s test and Meta-regression analysis were performed for all ***:We conducted a Meta-analysis of 10 RCTs that met the inclusion criteria,involving 1796 ***-concentrations group significantly improved the outcome of CFS according to random effects modelling(SMD,-3.37;95%CI,-5.25 to-1.48;P=0.0005).The rest of the results were not statistically significant,including indicators such as SIT,TBUT,DESS and ***:For dry eyes with positive corneal staining,a high concentration of HY is recommended,whereas in other cases,a high concentration of HY does not offer a more pronounced advantage over a low concentration of HY in the treatment of dry eyes.

Ultrashort and powerful laser interactions with a target generate intense wideband electromagnetic pulses(EMPs).In this study,we report EMPs generated by the interactions between petawatt(30 fs,1.4×10^(20) W/cm^(2))femtosecond(fs)lasers with metal flat,plastic flat,and plastic nanowire-array(NWA)*** analyses are conducted on the EMPs in terms of their spatial distribution,time and frequency domains,radiation energy,and *** results indicate that EMPs from metal targets exhibit larger amplitudes at varying angles than those generated by other types of targets and are enhanced significantly for NWA *** a plastic target holder and increasing the laser focal spot can significantly decrease the radiation energy of the ***,the composite shielding materials indicate an effective shielding effect against *** simulation results show that the NWA targets exert a collimating effect on thermal electrons,which directly affects the distribution of *** study provides guidance for regulating EMPs by controlling the laser focal spot,target parameters,and target rod material and is beneficial for electromagnetic-shielding design.

Semitransparent organic photovoltaics(STOPVs)have gained wide attention owing to their promising applications in building-integrated photovoltaics,agrivoltaics,and floating *** semiconductors with high charge carrier mobility usually have planar and conjugated structures,thereby showing strong absorption in visible *** this work,a new concept of incorporating transparent inorganic semiconductors is proposed for high-performance ***(I)thiocyanate(CuSCN)is a visible-transparent inorganic semiconductor with an ionization potential of 5.45 eV and high hole *** transparency of CuSCN benefits high average visible transmittance(AVT)of *** energy levels of CuSCN as donor match those of near-infrared small molecule acceptor BTP-eC9,and the formed heterojunction exhibits an ability of exciton *** mobility of CuSCN contributes to a more favorable charge transport channel and suppresses charge *** control STOPVs based on PM6/BTP-eC9 exhibit an AVT of 19.0%with a power conversion efficiency(PCE)of 12.7%.Partial replacement of PM6 with CuSCN leads to a 63%increase in transmittance,resulting in a higher AVT of 30.9%and a comparable PCE of 10.8%.

Current Status of Key Competencies among Medical Students: A Latent Profile Analysis

The purpose of this study was to understand the overall level of key competencies of medical students and explore the potential profile of key competencies, promoting quality education, and improving the quality talent cultivation in medical colleges. A stratified random sampling method selected 734 medical students from four medical colleges in Chongqing Province of China. A general information questionnaire and a key competencies survey questionnaire were used to conduct the survey. The overall score and scores of each dimension of key competencies were analyzed. Latent profile analysis was conducted to classify the key competencies of medical students and compare the distribution differences of demographic variables among different categories. The results showed that 26% of medical students have never heard of the concept of key competencies, and 59% of them are not familiar with the content related to key competencies. The score of key competencies is 3.66 ± 0.60, with the highest score in the dimension of responsibility and the lowest score in the dimension of humanistic accomplishment. The latent profile analysis classified them into three categories: “low key competencies group (14.71%)”, “medium key competencies group (36.79%)”, and “high key competencies group (48.50%)”. The R3STEP regression analysis results showed statistically significant differences in educational level and whether they served as student cadres among different key competencies categories of medical students. This paper discusses three different potential key competencies categories among medical students, and the overall level of key competencies is relatively good. However, medical students lack a comprehensive and systematic understanding of key competencies. Humanistic accomplishment, healthy living, and practical innovation are the three dimensions with lower scores and should be given more attention.
