Biomolecular analysis at the single-cell level is increasingly important in the study of cellular heterogeneity and its consequences,particularly in organismic development and complex diseases such as ***-cell molecular analyses have led to the identification of new cell types1 and the discovery of novel targets for diagnosis and *** these analyses are performed predominantly on dissociated single cells,emerging techniques seek understanding of cellular state,cellular function and cell–cell interactions within the native tissue environment by combining optical microscopy and single-cell molecular *** techniques include in situ multiplexed imaging of fluorescently labeled proteins and nucleotides,as well as low-throughput ex vivo methods in which specific cells are isolated for downstream molecular ***,these methods are limited in either the number and type of molecular species they can identify or the number of cells that can be ***-throughput methods are needed for comprehensive profiling of many cells(>1000)to detect rare cell types,discriminate relevant biomarkers from intrinsic population noise,and reduce the time and cost of *** established,highthroughput single-cell analyses are not directly applicable because they require tissue dissociation,leading to a loss of spatial *** current methods exist that can seamlessly connect spatial mapping to single-cell *** this Perspective,we review current methods for spatially resolved single-cell analysis and discuss the prospect of novel multiplexed imaging probes,called laser particles,which allow individual cells to be tagged in tissue and analyzed subsequently using high-throughput,comprehensive single-cell techniques.

Gold nanoparticles （AuNPs） stabilized with sodium alginate （SA）, glycol chitosan （GC）, polyvinyl alcohol （PVA）, and polyethyleneimine （PEI） were synthesized in aqueous solutions at room temperature. Different sizes and size distributions of AuNPs were obtained according to the polymeric stabilizers. The mean particle diameters of AuNPs synthesized with GC and PVA as stabilizers were about 5.1 and 5.3 nm, respectively, while those of AuNPs stabilized with SA and PEI were 8.4 and 10.8 nm, respectively. When SA was used, relatively large particles aggregated to form large clusters. In the case of GC, a number of small particles built up large clusters. When PVA was used, the size and the number of nanoperticles were estimated small with a low weight content of Au. For this reason, the PVA-stabilized AuNPs formed clusters with a small hydrodynamic size. The AuNPs synthesized in the aqueous solution of PEI without a reducing agent, sodium borohydride, were uniform in size and formed the smallest nanoclusters （67.4 nm）. In this case, the highest Au content was obtained. Sufficiently grown nanoparticles with a high Au content and a limited hydrodynamic size are considered to be suitable for a variety of applications.

Treatment-induced apoptosis of cancer cells is one goal of cancer ***,more heat is generated by mitochondria during apoptosis,especially the uncoupled apoptotic state,^(1,2) compared to the resting *** this case study,we explore these thermal effects by longitudinally measuring temperature variations in a breast lesion of a pathological complete responder during neoadjuvant chemotherapy(NAC).Diffuse Optical Spectroscopic Imaging(DOSI)was employed to derive absolute deep tissue temperature using subtle spectral features of the water peak at 975 nm.^(3)A significant temperature increase was observed in time windows during the anthracycline and cyclophosphamide(AC)regimen but not in the paclitaxel and bevacizumab *** concentration changes generally did not follow temperature,suggesting the measured temperature increases were likely due to mitochondrial uncoupling rather than a direct vascular effect.A simultaneous increase of tissue oxygen saturation with temperature was observed,suggesting that oxidative stress also contributes to *** preliminary,this study indicates longitudinal DOSI tissue temperature monitoring provides information that can improve our understanding of the mechanisms of tissue response during NAC.

Lithofacies modeling by multipoint statistics and economic evaluation by NPV volume for the early Cretaceous Wabiskaw Member in Athabasca oilsands area, Canada

The static modeling and dynamic simulation are essential and critical processes in petroleum exploration and development. In this study, lithofacies models for Wabiskaw Member in Athabasca, Canada are generated by multipoint statistics(MPS) and then compared with the models built by sequential indicator simulation(SIS). Three training images(Tls) are selected from modern depositional environments;the Orinoco River Delta estuary, Cobequid bay-Salmon River estuary, and Danube River delta environment. In order to validate lithofacies models, average and variance of similarity in lithofacies are calculated through random and zonal blind-well *** random six-blind-well test, similarity average of MPS models is higher than that of SIS model. The Salmon MPS model closely resembles facies pattern of Wabiskaw Member in subsurface. Zonal blind-well tests show that successful lithofacies modeling for transitional depositional setting requires additional or proper zonation information on horizontal variation, vertical proportion, and secondary *** Wabiskaw Member is frontier oilsands lease, it is impossible to evaluate the economics from production data or dynamic simulation. In this study, a dynamic steam assisted gravity drainage(SAGD)performance indicator(SPIDER) on the basis of reservoir characteristics is calculated to build 3 D reservoir model for the evaluation of the SAGD feasibility in Wabiskaw Member. SPIDER depends on reservoir properties, economic limit of steam-oil ratio, and bitumen price. Reservoir properties like porosity,permeability, and water saturation are measured from 13 cores and calculated from 201 well-logs. Three dimensional volumes of reservoir properties are constructed mostly based on relationships among properties. Finally, net present value(NPV) volume can be built by equation relating NPV and SPIDER. Deformation behavior of 1 Al containing Mg alloy has been investigated in the present *** annealing,the Mg-1 Al alloy shows a typical basal *** compared to the pure Mg having a similar texture and grain size,the Mg-1 Al alloy shows much higher strength and larger *** trace analyses of the tensile strained specimens show that non-basal slips such as pyramidal I and II slips can be easily activated at an early stage of deformation in the Mg-1 Al alloy and the grains in the Mg-1 Al alloy are seen to accommodate a larger degree of deformation than those in the pure Mg at a given *** increasing tensile strain,however,there is a strain localization along the initially formed slip lines of non-basal slips,forming surface steps without activating multiple slip lines.

Intercomparison of Precipitation Simulated by Regional Climate Models over East Asia in 1997 and 1998

Regional climate simulations in Asia from May 1997 to August 1998 were performed using the Seoul National University regional climate model （SNURCM） and Iowa State University regional climate model （***5/LSM）, which were developed by coupling the NCAR/Land Surface Model （LSM） and the Mesoscale Model （MM5）. However, for physical processes of precipitation, the SNURCM used the Grell scheme for the convective parameterization scheme （CPS） and the simple ice scheme for the explicit moisture scheme （EMS）, while the ***5/LSM used the Betts-Miller scheme for CPS and the mixed phase scheme for EMS.The simulated precipitation patterns and amounts over East Asia for the extreme climatic summer in 1997 （relative drought conditions） and 1998 （relative flood conditions） were especially focused upon. The ***5/LSM simulated more precipitation than was observed in 1997 due to more moisture and cloud water in the lower levels, despite weak upward motion. In the SNURCM, strong upward motion resulted in more precipitation than that was observed in 1998, with more moisture and cloud water in the middle levels. In the ***5/LSM, weak upward motion, unchanged moisture in the lower troposphere, and the decrease in latent heat flux at the surface increased convective precipitation only by 3% for the 1998 summer event. In the SNURCM, strong upward motion, the increase in moisture in the lower troposphere, and the increase in latent heat flux at the surface increased convective precipitation by 48% for the summer of 1998. The main differences between both simulations were moisture availability and horizontal momentum transport in the lower troposphere, which were also strongly influenced by large-scale forcing.

We introduce a fast solver for the phase field crystal(PFC)and functionalized Cahn-Hilliard(FCH)equations with periodic boundary conditions on a rectangular domain that features the preconditioned Nesterov’s accelerated gradient descent(PAGD)*** discretize these problems with a Fourier collocation method in space,and employ various second-order schemes in *** observe a significant speedup with this solver when compared to the preconditioned gradient descent(PGD)*** the PAGD solver,fully implicit,second-order-in-time schemes are not only feasible to solve the PFC and FCH equations,but also do so more efficiently than some semi-implicit schemes in some cases where accuracy issues are taken into *** computations of four different schemes for the PFC and FCH equations are conducted and the results indicate that,for the FCH experiments,the fully implicit schemes(midpoint rule and BDF2 equipped with the PAGD as a nonlinear time marching solver)perform better than their IMEX versions in terms of computational cost needed to achieve a certain *** the PFC,the results are not as conclusive as in the FCH experiments,which,we believe,is due to the fact that the nonlinearity in the PFC is milder nature compared to the FCH *** also discuss some practical matters in applying the *** introduce an averaged Newton preconditioner and a sweeping-friction strategy as heuristic ways to choose good preconditioner *** sweeping-friction strategy exhibits almost as good a performance as the case of the best manually tuned parameters.
