Background:DNA methylation and gene expression are known to play important roles in the etiology of human diseases such as prostate cancer(PCa).However,it has not yet been possible to incorporate information of DNA methylation and gene expression into polygenic risk scores(PRSs).Here,we aimed to develop and validate an improved PRS for PCa risk by incorporating genetically predicted gene expression and DNA methylation,and other genomic information using an integrative ***:Using data from the PRACTICAL consortium,we derived multiple sets of genetic scores,including those based on available single-nucleotide polymorphisms through widely used methods of pruning and thresholding,LDpred,LDpred-funt,AnnoPred,and EBPRS,as well as PRS constructed using the genetically predicted gene expression and DNA methylation through a revised pruning and thresholding *** the tuning step,using the UK Biobank data(1458 prevalent cases and 1467 controls),we selected PRSs with the best *** an independent set of data from the UK Biobank,we developed an integrative PRS combining information from individual ***,in the testing step,we tested the performance of the integrative PRS in another independent set of UK Biobank data of incident cases and ***:Our constructed PRS had improved performance(C statistics:76.1%)over PRSs constructed by individual benchmark methods(from 69.6%to 74.7%).Furthermore,our new PRS had much higher risk assessment power than family *** overall net reclassification improvement was 69.0%by adding PRS to the baseline model compared with 12.5%by adding family ***:We developed and validated a new PRS which may improve the utility in predicting the risk of developing *** innovative method can also be applied to other human diseases to improve risk prediction across multiple outcomes.

An Alternative Proof of the Largest Number of Maximal Independent Sets in Connected Graphs Having at Most Two Cycles

G. C. ying, y. y. Meng, B. E. Sagan, and V. R. Vatter [1] found the maximum number of maximal independent sets in connected graphs which contain at most two cycles. In this paper, we give an alternative proof to determine the largest number of maximal independent sets among all connected graphs of order n ≥ 12, which contain at most two cycles. We also characterize the extremal graph achieving this maximum value.

Undoubtfully there is a strong interrelationship between sediment transport and river bed dynamics. Instable river beds are the result and the reason for erosion, transport and for accumulation of bed sediments. Since the investigations of Simons & Richardson (1962) the interpretation on flow regime, bed roughness and resistance to flow is well investigated for Sandy bed material. In contrast there is a certain neglect of knowledge in the case of coarser material. The big difference between these river beds are caused by the distinctive erodibility of the coarse materials and their restricted availability. Nevertheless the regime concept is a good tool not only for the classification of many phenomena but will help to understand the dynamics of both river beds and transports of mountain torrents.

Cropping frequency and N fertilizer effects on soil water distribution from spring to fall in the semiarid Canadian prairies

In the semiarid Canadian prairies, water is the most limiting and nitrogen (N) the second most limiting factor influencing spring wheat (Triticum aestivum L.) production. The efficiency of water-and nitrogen use needs to be assessed in order to maintain this production system. The effects of cropping frequency and N fertilization on trends in soil water distribution and water use were quantified for an 18-yr (1967-1984) field experiment conducted on a medium textured Orthic Brown Chernozem (Aridic Haploboroll) in southwestern Saskatchewan, Canada. Soil water contents were measured eight times each year and plant samples were taken at five phenological growth stages. The treatments studied were continuous wheat (Cont W), summer fallow - wheat, F-(W) and summer fallow - wheat - wheat, F-W-(W) each receiving recommended rates of N and phosphorus (P) fertilizer, and (F)-W-W and (Cont W) each receiving only P fertilizer, with the examined rotation phase shown in parentheses. Soil water conserved under fallow during the summer months averaged 25 mm in the root zone, and was related to the initial water content of the soil, the amount of precipitation received, its distribution over time, and potential evapotranspiration. Under a wheat crop grown on fallow, soil water contents between spring and the five-leaf stage remained relatively constant at about 250 mm, but those under a stubble crop, with 40 mm lower spring soil water reserves, increased slightly until about the three-leaf stage. During the period of expansive crop growth (from the five-leaf to the soft dough stage) soil water was rapidly lost from all cropped phases at rates of 1.87 ***–1 for F-(W) (N+P), 1.23 ***–1 for Cont W (N+P) and 1.17 ***–1 for Cont W (+P).
