Effects of Irradiation with Light of Different Photon Densities on the Growth of young Green Barley Plants

In recent years, young green barley has been increasingly used as a functional food ingredient. Studies have shown that cultivated young green barley has different levels of amino acids, vitamin c, and polyphenol content depending on the harvest time. Furthermore, some studies have investigated the effect of the growing conditions on cultivated barley in particular the use of light-emitting diodes (LED). young green barley was exposed to three light sources of different wavelengths (sun light [ASL], light-emitting diode[LED] - Red 9 + Blue 1 [LED-R9:B1], LED - Red 4 + Green 1+ Blue 1 [LED-R4: G1:B1]). Under light with photon densities of 200 μmolmL-2·s-1, the dry weight of young green barley was not affected, while the differences were observed in the morphology of the underground portion of the plant (roots) depending on the light source. The roots of the plant grown in ASL weighed twice as much as those that were grown under LED irradiation. Furthermore, LED irradiation caused an increase in the amino acid content in plants;the amino acid content of plants grown under LED-R9:B1 was twice as much as that grown under ASL irradiation. Four kinds of cyanogenicglucosides (cGs) were isolated, identified, and their levels were measured. The plant grown under LED-RGB irradiation, including LED-G, produced approximately 20% more cGs as compared to the plants grown under the other two light sources. Thus, it was inferred that young green barley exhibited a stress response under LED-G light and accumulated cGs in the stems and leaves with prepare for any damage that may occur on the leaf surface. The nitrogen (N) content in the root was the lowest, while the cG content was the highest in the plant grown under LED-RGB irradiation. It is inferred that the N content decreased because N was eliminated from the roots to allow for accumulation of cGs response to stress. BAcKGROUND: The present study aimed to compare outcome of primary and secondary Illizarov's fi xator application as a treatment method for type III open tibial fractures in terms of nonunion and wound ***: This prospective study was done in a tertiary.c.re center. Forty-eight type III tibial fractures were treated with Illizarov's apparatus between 2008 and 2011. The patients were divided into two groups depending on the treatment protocol, timing of wound closure and Illizarov's application, primary(n=28) and secondary(n=20).RESULTS: In the primary group, healing was achieved in all 28 patients. The median time to recovery was 24 weeks, and the median number of operations was 3. There were 6 patients with a bone defect. In the secondary group, complete recovery was achieved in 18 out of 20 patients. The median time to recovery was 30 weeks, and the median number of operations 5. There were 9 patients with a bone defect. The median time to recovery and the number of operations were signifi cantly smaller in patients undergoing primary operation. Union was 100% in the primary group and more than 95% in the secondary group. chronic osteomyelitis persisted in one patient and below amputation was done in one patient in the secondary ***: Primary wound closure and Illizarov's fixation required a smaller number of operations and shorter time to recovery than secondary wound closure and Illizarov's fi xation, mostly due to a signifi cantly less number of patients with a bone defect in the primary group.

Genomic analysis of a novel strain of Bacillus nealsonii, isolated from Surti buffalo rumen

Aim: Whole genome sequencing and functional annotation of Bacillus nealsonii strain AAU1, an amylolytic anaerobic spore forming isolate from ruminal contents of buffalo. Methods and Results: Morphologically, the strain was observed as slender Grampositive rods, occurring in pairs. Optimal growth was observed at 40°c (range: 30°c to 45°c) and pH 6.5 (range: 5.5 to 7.5) when cultivated in Hungate’s medium supplemented with starch. The microorganism showed extracellular constitutive amylolytic activity, proving to be capable of utilizing glucose, maltose, mannose, trehalose, dextrin and starch under anaerobic conditions. Sequence analysis revealed a Gc content of 35.1 mol%. comparison of housekeeping gene sequences for RNA polymerase subunit B (rpoB) and gyrase A (gyrA) identified sequence similarity within the Bacillus genus, confirmed by 16S rRNA gene sequence similarity which identified Bacillus nealsonii DSM 15077 as the closest publically available relative. chemotaxonomic analysis provided conflicting results with straight-chain saturated c16: 0/c16:0 aldehyde, c16:0 DMA, c14:0 and monounsaturated 16:1w7c and 16:1w9c the major fatty acids in contrast to those reported for B. nealsonii DSM15077. Further characterization using AN-Bilog and physiological parameters provided genotypic and phenotypic support for taxonomic classification of isolate AAU1 with published Bacillus species including B. licheniformis, B. subtilis, B. circulans and B. nealsonii. conclusion: Based on the data presented, isolate is likely to represent a new strain/subspecies, for which the identifier B. nealsonii AAU1 is proposed. Significance and Impact of Study: The strict anaerobic conditions prevailing in the bovine rumen from where AAU1 was isolated may have resulted in genetic polymorphism influencing its metabolic characteristics.

Several obstacles to the production,expansion and genetic modification of immunotherapeutic T cells in vitro have restricted the widespread use of T-cell *** the context of HScT,delayed naïve T-cell recovery.c.ntributes to poor outcomes.A novel approach to overcome the major limitations of both T-cell immunotherapy and HScT would be to transplant human T-lymphoid progenitors(HTLPs),allowing reconstitution of a fully functional naïve T-cell pool in the patient ***,it is challenging to produce HTLPs in the high numbers required to meet clinical ***,we found that adding tumor necrosis factor alpha(TNFα)to a DL-4-based culture system led to the generation of a large number of nonmodified or genetically modified HTLPs possessing highly efficient in vitro and in vivo T-cell potential from either cB HSPcs or mPB HSPcs through accelerated T-cell differentiation and enhanced HTLP cell cycling and *** study provides a clinically suitable cell culture platform to generate high numbers of clinically potent nonmodified or genetically modified HTLPs for accelerating immune recovery after HScT and for T-cell-based immunotherapy(including cAR T-cell therapy).

Plecanatide-mediated activation of guanylate cyclase-c suppresses inflammation-induced colorectal carcinogenesis in Apc+/Min-Fccc mice

AIM To evaluate the effect of orally administered plecanatide on colorectal dysplasia in Apc^(+/Min-Fccc) mice with dextran sodium sulfate(DSS)-induced inflammation. METHODS Inflammation driven colorectal carcinogenesis was induced in Apc^(+/Min-Fccc) mice by administering DSS in their drinking water. Mice were fed a diet supplemented with plecanatide(0-20 ppm) and its effect on the multiplicity of histopathologically.c.nfirmed polypoid,flat and indeterminate dysplasia was evaluated. Plecanatide-mediated activation of guanylate cyclase-c(Gc-c) signaling was assessed in colon tissues by measuring cyclic guanosine monophosphate(cG MP) by ELISA, protein kinase G-II and vasodilator stimulated phosphoprotein by immunoblotting. Ki-67, c-myc and cyclin D1 were used as markers of proliferation. cellular levels and localization of b-catenin in colon tissues were assessed by immunoblotting and immunohistochemistry, respectively. Uroguanylin(UG) and Gc-c transcript levels were measured by quantitative reverse transcription polymerase chain reaction(RT-PcR). A mouse cytokine array panel was used to detect cytokines in the supernatant of colon explant cultures. RESULTS Oral treatment of Apc^(+/Min-Fccc) mice with plecanatide produced a statistically significant reduction in the formation of inflammation-driven polypoid, flat and indeterminate dysplasias. This anti-carcinogenic activity of plecanatide was accompanied by activation of cG MP/Gc-c signaling mediated inhibition of Wnt/b-catenin signaling and reduced proliferation. Plecanatide also decreased secretion of pro-inflammatory.c.tokines(IL-6, IL-1 TNF), chemokines(MIP-1, IP-10) and growth factors(GcSF and GMcSF) from colon explants derived from mice with acute DSS-induced inflammation. The effect of plecanatidemediated inhibition of inflammation/dysplasia on endogenous expression of UG and Gc-c transcripts was measured in intestinal tissues. Road traffic accidents in hilly regions of northern India: What has to be done?

BAcKGROUND: Road traffi c accidents(RTA) are responsible for 1.2 million deaths worldwide each year. RTA will become the 3rd largest contributor to the global burden of diseases after ischemic heart diseases(IHD) and depression. We conducted a retrospective study on RTA in a tertiary.c.nter in the hilly district of Uttarakhand in ***: The number of RTA, pattern of RTA, the number of patients killed and injured, the pattern of injury.c.using death and disability, the severity of accidents, and the type of disability were noted from December 2009 to November 2011. The accident severity was calculated as the number of patients killed per 100 accidents. The methods for reducing the incidence of RTA were observed, and the role of policy makers was ***: The majority of deaths and disabilities in Uttarakhand were due to road traffic accidents in the hilly districts of the states. The most common cause of RTA was driving fault followed by defective ***: Proper designing of roads and minimizing the fault of drivers are essential to prevent road traffi c accidents in hilly regions.
