Parallelized fluorescence imaging has been a long-standing pursuit that can address the unmet need for a comprehensive three-dimensional(3D)visualization of dynamical biological processes with minimal ***,the available approaches are limited to incomplete parallelization in only two dimensions or sparse sampling in three *** hereby develop a novel fluorescence imaging approach,called coded light-sheet array microscopy(CLAM),which allows complete parallelized 3D imaging without mechanical *** the concept of an“infinity mirror”,CLAM generates a light-sheet array with controllable sheet density and degree of ***,CLAM circumvents the common complications of multiple coherent light-sheet generation in terms of dedicated wavefront engineering and mechanical dithering/***,the encoding of multiplexed optical sections in CLAM allows the synchronous capture of all sectioned images within the imaged *** demonstrate the utility of CLAM in different imaging scenarios,including a light-scattering medium,an optically cleared tissue,and microparticles in fluidic *** can maximize the signal-to-noise ratio and the spatial duty cycle,and also provides a further reduction in photobleaching compared to the major scanning-based 3D imaging *** flexible implementation of CLAM regarding both hardware and software ensures compatibility with any light-sheet imaging modality and could thus be instrumental in a multitude of areas in biological research.

A tV-Based Iterative Regularization Method for the Solutions of thermal Convection Problems

Linear/nonlinear and Stokes based-stabilizations for the filter equations for damping out primitive variable(PV)solutions corrupted by uniformly distributed randomnoises are numerically studied through the natural convection(NC)aswell as the mixed convection(MC)*** most recognizable filter-scheme is based on a combination of the negative Laplace equation multiplied with the selection of the spatial scale and a linear function in order to preserve the uniqueness of the filtered solution.A more complicated filter-scheme,based on a Stokes problem which couples a filtered velocity and a filtered(artificial)pressure(or Lagrange multiplier)in order to enforce the incompressibility constraint,is also *** and Stokes basedfilters via nested iterative(NI)filters and the consistent splitting scheme(CSS)are proposed for the NC/MC *** by the total-variation(tV)model of image diffusion,well preserved feature flow patterns from the corrupted NC/MC environment are obtained by tV-Stokes based-filters together with the *** experimental results show that our proposed algorithms are effective and efficient in eliminating the unwanted spurious oscillations and preserving the accuracy of thermal convective fluid flows.

High-speed high-resolution laser diode-based photoacoustic microscopy for in vivo microvasculature imaging

Laser diodes(LDs)have been considered as cost-effective and compact excitation sources to overcome the requirement of costly and bulky pulsed laser sources that are commonly used in photoacoustic microscopy(PAM).However,the spatial resolution and/or imaging speed of previously reported LD-based PAM systems have not been optimized *** this paper,we developed a high-speed and high-resolution LD-based PAM system using a continuous wave LD,operating at a pulsed mode,with a repetition rate of 30 kHz,as an excitation source.A hybrid scanning mechanism that synchronizes a one-dimensional galvanometer mirror and a two-dimensional motorized stage is applied to achieve a fast imaging capability without signal averaging due to the high signal-to-noise *** optimizing the optical system,a high lateral resolution of 4.8μm has been *** vivo microvasculature imaging of a mouse ear has been demonstrated to show the high performance of our LD-based PAM system.

Converging multi-modality datasets to build efficient drug repositioning pipelines against Alzheimer’s disease and related dementias

Alzheimer’s disease and related dementias(AD/ADRD)affects more than 50 million people worldwide but there is no clear therapeutic option affordable for the general patient ***,drug repositioning studies featuring collaborations between academic institutes,medical centers,and hospitals are generating novel therapeutics candidates against these devastating diseases and filling in an important area for healthcare that is poorly represented by pharmaceutical *** drug repositioning studies converge expertise from bioinformatics,chemical informatics,medical informatics,artificial intelligence,high throughput and high-content screening and systems *** also take advantage of multi-scale,multi-modality datasets,ranging from transcriptomic and proteomic data,electronical medical records,and medical imaging to social media information of patient behaviors and emotions and epidemiology profiles of disease populations,in order to gain comprehensive understanding of disease mechanisms and drug *** proposed a recursive drug repositioning paradigm involving the iteration of three processing steps of modeling,prediction,and validation to identify known drugs and bioactive compounds for AD/*** recursive paradigm has the potential of quickly obtaining a panel of robust novel drug candidates for AD/ADRD and gaining in-depth understanding of disease mechanisms from those repositioned drug candidates,subsequently improving the success rate of predicting novel hits.

Afteruse development of former landfll sites in Hong Kong

Landfll is one of the major methods in disposing of municipal solid waste(MSW) in Hong Kong. there are now 13 closed landfll sites in Hong Kong, and a restoration program has been launched since 1999 to transform the closed landfll sites back into recreational area. the MSW underneath these closed landfll sites will biodegrade slowly and gradually, releasing toxic gases and leachate. As there have only been a few afteruse development ever completed in Hong Kong, this paper describes the consideration in planning and design of two former landfll sites in Hong Kong–Ngau Chi Wan(NCW) and Jordan Valley(JV) landflls, in particular a discussion on the MSW settlement. Various models in predicting the longterm MSW settlement have been available. this paper reviews the applicability of these models to predict MSW settlement. However, as the predicted settlements can be signifcantly in error, they need sitespecifc calibration and validation. this paper therefore presents the settlement monitoring data in these former landfll sites over the last 30 years, and suggests modifying the Bjarngard and Edgers’ method for estimating this long-term settlement as well as the effect of additional surcharge due to afteruse development. Based on the regression of these data, compression indices Cce and C at different stages of the settlement in the modifed method have been calibrated and validated.

the disruptive effects of the COVID-19 pandemic has rapidly shifted how individuals navigate in *** are concerned that travel behavior will shift toward a car-driven and homeworking future,shifting demand away from public transport *** concerns place the recovery of public transport in a possible crisis.A resilience perspective may aid the discussion around recovery-particularly one that deviates from pre-pandemic *** paper presents an empirical study of London’s public transport demand and introduces a perspective of spatial resilience to the existing body of research on post-pandemic public transport *** study defines spatial resilience as the rate of recovery in public transport demand within census boundaries over a period after lockdown restrictions were *** relationship between spatial resilience and urban socioeconomic factors was investigated by a global spatial regression model and a localized perspective through Geographically Weighted Regression(GWR)*** this case study of London,the analysis focuses on the period after the first COVID-19 lockdown restrictions were lifted(June 2020)and before the new restrictions in mid-September *** analysis shows that outer London generally recovered faster than inner *** of income,car ownership and density of public transport infrastructure were found to have the greatest influence on spatial patterns in ***,influential relationships vary locally,inviting future research to examine the drivers of this spatial ***,this research recommends transport policymakers capture the influences of homeworking,ensure funding for a minimum level of service,and advocate for a polycentric recovery post-pandemic.

Optical-resolution photoacoustic microscopy(OR-PAM)has demonstrated high-spatial-resolution imaging of optical absorption in biological *** date,most OR-PAM systems rely on mechanical scanning with confocally aligned optical excitation and ultrasonic detection,limiting the wide-field imaging speed of these *** several multifocal OR-PA(MFOR-PA)systems have attempted to address this limitation,they are hindered by the complex design in a constrained physical ***,we present a two-dimensional(2D)MFOR-PAM system that utilizes a 2D microlens array and an acoustic ergodic *** a single-element ultrasonic transducer,this system can detect PA signals generated from 400 optical foci in parallel and then raster scan the optical foci patterns to form an MFOR-PAM *** system improves the imaging resolution of an acoustic ergodic relay system from 220 to 13μm and enables 400-folds shorter scanning time than that of a conventional OR-PAM system at the same resolution and laser repetition *** demonstrated the imaging ability of the system with both in vitro and in vivo experiments.

the use of sensors for monitoring livestock has opened up new possibilities for the management of livestock in extensive grazing *** work presented in this paper aimed to develop a model for predicting the metabolisable energy intake(MEI)of sheep by using temperature,pitch angle,roll angle,distance,speed,and grazing time data obtained directly from wearable sensors on the sheep.A Deep Belief Network(DBN)algorithm was used to predict MEI,which to our knowledge,has not been attempted *** results demonstrated that the DBN method could predict the MEI for sheep using sensor data *** mean square error(MSE)values of 4.46 and 20.65 have been achieved using the DBN model for training and testing datasets,*** also evaluated the influential sensor data variables,i.e.,distance and pitch angle,for predicting the *** study demonstrates that the application of machine learning techniques directly to on-animal sensor data presents a substantial opportunity to interpret biological interactions in grazing systems directly from sensor *** expect that further development and refinement of this technology will catalyse a step-change in extensive livestock management,as wearable sensors become widely used by livestock producers.
