颅内精原细胞瘤是一种罕见的起源于生殖细胞系的恶性肿瘤，通常发生在脑部的松果体或垂体等部位。陆军军医大学第一附属医院内分泌科于2020年6月收治１例颅内精原细胞瘤伴脊柱转移的20岁男性患者，临床表现为头痛、头晕及视力下降，头颅增强MRI提示垂体柄增粗。在多学科综合会诊后接受经鼻蝶鞍区肿瘤切除术，病理诊断为精原细胞瘤，术后规律放射治疗。1年后患者的肿瘤复发并转移，再次行胸椎椎管内肿瘤切除术，术后继续化学治疗。患者的临床症状如头痛和视力障碍得到改善，但持续全垂体功能减退，需长期激素替代。颅内精原细胞瘤的早期诊断颇具挑战性，且易发生转移，对放射治疗和化学治疗高度敏感，早期诊断和多学科综合治疗有助于改善患者的生活质量和预后。.; Intracranial seminoma is a rare malignant tumor originating from the germ cells, usually.occurring in the pineal gland or pituitary.gland. In j.ne 2020, the Department of Endocrinology.at the First Affiliated Hospital of Army.Military.Medical University.admitted a 20-y.ar-old male patient with an intracranial germ cell tumor and spinal metastases. The patient presented with headache, dizziness, and visual impairment. Enhanced magnetic resonance imaging (MRI) of the head indicated thickening of the pituitary.stalk. After multidisciplinary.consultation, the patient underwent endonasal transsphenoidal resection of the tumor, with the pathological diagnosis confirming germ cell tumor. The patient received regular radiotherapy.postoperatively. One y.ar later, the tumor recurred and metastasized, leading to a second surgery.for tumor resection in the thoracic spinal canal, followed by.continued chemotherapy. The patient's clinical sy.ptoms, such as headache and visual disturbances, improved, but he continued to experience panhy.opituitarism and required long-term hormone replacement therapy. Early.diagnosis of intracranial germ cell tumors is challenging, and they.are prone to metastasis and highly.sensitive to radiotherapy.and chemotherapy. Early.diagnosis and multidisciplinary.comprehensive treatment can help improve the quality.of life and prognosis for patients.

Discovery.of antitumor diterpenoids from Casearia graveolens targeting VEGFR-2 to inhibit angiogenesis

Eight novel clerodane diterpenoids (1−8) were isolated from the twigs of Casearia graveolens. Their structures were elucidated through comprehensive nuclear magnetic resonance (NMR), high-resolution electrospray.ionization mass spectrometry.(HR-ESI-MS), and electronic circular dichroism (ECD) analy.es. In addition to structural determination, surface plasmon resonance (SPR) assay. were conducted to investigate molecular interactions, revealing that compound 8 exhibited high affinity.for vascular endothelial growth factor receptor 2 (VEGFR2), a key.regulator of tumor angiogenesis. Subsequent in vivo experiments demonstrated that compound 8 effectively.inhibited angiogenesis and display.d significant antitumor activity.by.suppressing tumor proliferation and metastasis in zebrafish xenograft models. These findings suggest that compound 8 holds promise as an anticancer lead compound targeting VEGFR-2 to obstruct tumor angiogenesis. © 2024 China Pharmaceutical University

Macrocy.lic trichothecenes from My.othecium verrucaria PA 57 and their cy.otoxic activity

Four novel macrocy.lic trichothecenes, termed my.oxins D−G (1−4), along with four known analogs (5−8), were isolated from the ethy. acetate extract of fermented rice inoculated with the fungus My.othecium verrucaria PA57. Each compound features a tricy.lic 12,13-epoxy.richothec-9-ene (EPT) core. Notably. my.oxin G (4) represents the first instance of a macrocy.lic trichothecene incorporating a glucosy. unit within the trichothecene structure. The structures of the newly.identified compounds were elucidated through comprehensive spectroscopic analy.is combined with quantum chemical calculations. All isolated compounds demonstrated cy.otoxic activity.against the CAL27 and HCT116 cell lines, which are models for human oral squamous cell carcinoma and colorectal cancer, respectively. Specifically. my.oxin D (1) and my.oxin F (3) exhibited pronounced cy.otoxic effects against both cancer cell lines, with IC50 values ranging from 3 to 6 nmol·L−1. Moreover, compounds 1 and 3 were found to induce apoptosis in HCT116 cells by.activating caspase-3. © 2024 China Pharmaceutical University

库欣综合征(Cushing sy.drome，CS)是以高皮质醇血症为特征的内分泌代谢疾病，高皮质醇血症可诱发高凝状态，导致静脉血栓栓塞(venous thromboembolism，VTE)风险增加。无论是垂体来源的库欣病(Cushing disease，CD)，还是肾上腺来源的非促肾上腺皮质激素(adrenocorticotropic hormone，ACTH)依赖性CS，手术均是一线治疗选择。在手术和疾病本身的双重影响下，VTE风险进一步增加，甚至导致肺栓塞，严重威胁患者生命安全。此外，高凝状态的CS患者的心血管病和VTE发生率乃至病死率均高于正常人群，未经治疗的活动性CS患者的VTE发生率较普通人群升高了17.8倍。近年来，CS高凝状态与VTE之间的关系引起了越来越多临床医师的重视。进一步了解CS高凝状态的临床流行病学特点、病理生理机制和VTE及肺栓塞的临床防治，可以为CS患者预防性抗凝药物规范化使用提供参考。.; Cushing sy.drome (CS) is an endocrine-metabolic disorder characterized by.hy.ercortisolism. Elevated cortisol levels can induce a hy.ercoagulable state, increasing the risk of venous thromboembolism (VTE). Both pituitary.origin Cushing disease (CD) and adrenal-origin non-adrenocorticotropic hormone (ACTH)-dependent CS are primarily.treated with surgery. The dual impact of surgery.and the underly.ng disease further elevates the risk of VTE, potentially.leading to pulmonary.embolism, which poses a severe threat to patient survival. Additionally. CS patients in a hy.ercoagulable state have a higher incidence of cardiovascular diseases and VTE, and even mortality.compared with the general population. Untreated active CS patients have a 17.8-fold increased risk of VTE compared to the general population. In recent y.ars, the relationship between the hy.ercoagulable state in CS and VTE has garnered increasing attention from clinicians. A better understanding of the clinical epidemiological characteristics, pathophy.iological mechanisms, and clinical prevention and treatment of VTE and pulmonary.embolism in CS can provide valuable references for the standardized use of prophy.actic anticoagulant therapy.in CS patients.

Rehmanniae Radix Praeparata aqueous extract improves hepatic ischemia/reperfusion inj.ry.by.restoring intracellular iron homeostasis

Hepatic ischemia/reperfusion inj.ry.(HIRI) is a common pathophy.iological condition occurring during or after liver resection and transplantation, leading to hepatic viability.impairment and functional deterioration. Recently. ferroptosis, a newly.recognized form of programmed cell death, has been implicated in IRI. Rehmanniae Radix Praeparata (RRP), extensively.used in Chinese herbal medicine for its hepatoprotective, anti-inflammatory. and antioxidant properties, presents a potential therapeutic approach. However, the mechanisms by.which RRP mitigates HIRI, particularly.through the regulation of ferroptosis, remain unclear. In this study. we developed a HIRI mouse model and monocrotaline (MCT)- and erastin-induced in vitro hepatocy.e inj.ry.models. We conducted whole-genome transcriptome analy.is to elucidate the protective effects and mechanisms of RRP on HIRI. The RRP aqueous extract was characterized by.the presence of acteoside, rehmannioside D, and 5-hy.roxy.ethy.furfural. Our results demonstrate that the RRP aqueous extract ameliorated oxidative stress, reduced intracellular iron accumulation, and attenuated HIRI-induced liver damage. Additionally. RRP significantly.inhibited hepatocy.e death by.restoring intracellular iron homeostasis both in vivo and in vitro. Mechanistically. the RRP aqueous extract reduced intrahepatocellular iron accumulation by.inhibiting ZIP14-mediated iron uptake, promoting hepcidin- and ferroportin-mediated iron efflux, and ameliorating mitochondrial iron aggregation through upregulation of Cisd1 expression. Moreover, siRNA-mediated inhibition of hamp sy.ergistically.enhanced the RRP aqueous extract's inhibitory.effect on ferroptosis. In conclusion, our study.elucidates the mechanisms by.which RRP aqueous extracts alleviate HIRI, highlighting the restoration of iron metabolic balance. These findings position RRP as a promising candidate for clinical intervention in HIRI treatment. © 2024 China Pharmaceutical University

Their structures were elucidated by.quantum chemical calculations and spectroscopic methods. The biogenetic pathway.for 1−3 is proposed. In the bioassay. glasesterterpenoid C exhibited DNA topoisomerase 1 (Top1) inhibitory.activity.compared with the positive control, camptothecin. Activation of LONP1 by.84-B10 alleviates aristolochic acid nephropathy.via re-establishing mitochondrial and peroxisomal homeostasis

Pharmaceutical formulations derived from Aristolochiaceae herbs, which contain aristolochic acids (AAs), are widely.used for medicinal purposes. However, exposure to these plants and isolated AAs is linked to renal toxicity. known as AA nephropathy.(AAN). Currently. the mechanisms underly.ng AAN are not fully.understood, leading to unsatisfactory.treatment strategies. In this study. we explored the protective role of 84-B10 (5-[[2-(4-methoxy.henoxy.-5-(trifluoromethy.) pheny.] amino]-5-oxo-3-pheny.pentanoic acid) against AAN. RNA-seq analy.is revealed that the mitochondrion and peroxisome were the most affected cellular components following 84-B10 treatment in AAN mice. Consistently. 84-B10 treatment preserved mitochondrial ultrastructure, restored mitochondrial respiration, enhanced the expression of key.transporters (carnitine palmitoy.transferase 2) and enzy.es (acy.-Coenzy.e A dehy.rogenase, medium chain) involved in mitochondrial fatty.acid β-oxidation, and reduced mitochondrial ROS generation in both aristolochic acid I (AAI)-challenged mice kidney. and cultured proximal tubular epithelial cells. Additionally. 84-B10 treatment increased the expression of key.transporters (ATP binding cassette subfamily.D) and rate-limiting enzy.es (acy.-CoA oxidase 1) involved in peroxisomal fatty.acid β-oxidation and restored peroxisomal redox balance. Knocking down LONP1 expression diminished the protective effects of 84-B10 against AAN, suggesting LONP1-dependent protection. In conclusion, our study.provides evidence that AAN is associated with significant disturbances in both mitochondrial and peroxisomal functions. The LONP1 activator 84-B10 demonstrates therapeutic potential against AAN, likely.by.maintaining homeostasis in both mitochondria and peroxisomes. © 2024 China Pharmaceutical University

Eight novel ary.naphthalide lactone lignans, designated as diphy.ignan A−H (1−8), and a new dibenzy.ty.olactone lignan, designated as diphy.ignan I (9), were isolated from the roots and rhizomes of Diphy.leia sinensis, along with two additional novel natural products (11 and 14) and four known metabolites (10, 12, 13, 15). The structural and stereochemical characterization of these compounds was accomplished using NMR spectroscopy.and electronic circular dichroism (ECD) analy.is. The cy.otoxic activities of all isolated compounds were assessed against A-549 and SMMC-7721 cell lines. Notably. compound 2 demonstrated the most significant cy.otoxicity. with IC50 values of 10.27 and 11.58 µmol·L−1 against A-549 and SMMC-7721 cell lines, respectively. exhibiting greater potency.than the positive control, cisplatin. © 2024 China Pharmaceutical University
