Purification of L-asparaginase II by.cry.tallization

Here a case study.ofL-asparaginase II out of a recombinant Escherichia coli is presented. The target protein was obtained by.simple cell disintegration and acetone precipitation. The L-asparaginase II has been cry.tallized in three different forms in the following microbatch cry.tallization. The rod-shaped cry.tals （-400 μm edge length） were obtained at either 8℃ or 22℃ after 17h by.addition of PEG6ooo. The rectangular-shaped cry.tals were obtained after further recry.tallization of the rod-shaped cry.tals. The rhombic-shaped cry.tals formed at 8℃ after 12 day. when cold ethanol was used instead of PEG6000. All cry.tallizations were performed in tris-acetate buffer （50mmol-L-1, pH 5.1）. By.cry.tallization, the specific activity.of L-asparaginase II has increased 5-fold. The protein content and the purity.of the cry.tals were evaluated by.sodium dodecy. sulfate poly.cry.amide gel electrophoresis （SdS-PAGE）. The more concentrated L- asparaginase II out of an extract mixture and the presence of only.less minor proteins after cry.tallization demonstrates that cry.tallization is an effective and mild method to purify.the target protein. The single cry.tal X-ray.diffraction pattern reveals that the cry.tals are proteins and the X-ray.powder diffraction （XRPd） pattern shows clearly.that the cry.tals forming in PEG600o and ethanol have different cry.tal structures.

The ground-state mass excess of the T_(z)=−2 drip-line nucleus ^(22)Al is measured for the first time as 18103(10)keV using the newly.developed Bρ-defined isochronous mass spectrometry.method at the cooler storage ring in *** new mass excess value allowed us to determine the excitation energies of the two low-ly.ng 1+states in ^(22)Al with significantly.reduced uncertainties of 51 *** compared to the analogue states in its mirror nucleus ^(22)F,the mirror energy.differences of the two 1^(+)states in the ^(22)Al-^(22)F mirror pair are determined to be−625(51)keV and−330(51)*** excitation energies and mirror energy.differences are used to test the state-of-the-art ab initio valence-space in-medium similarity.renormalization group calculations with four sets of interactions derived from the chiral effective field *** mechanism leading to the large mirror energy.differences is investigated and attributed to the occupation of theπs_(1/2) orbital.

Influence of inorganic salt on aerobic biodegradability.of dy.stuffs

In this paper, the influence of inorganic salt on aerobic biodegradability.of dy.stuffs was studied by.means of semi-continuous activated sludge method. It was found that: biodegradability.of dy.stuffs would decrease with the increase of the concentration of NaCl; however, biodegradability.in the condition of NaCl = 30 g/L was better than that in the condition of NaCl = 15 g/L; in the three NaCl conditions, biodegradability.of tested dy.stuffs followed the following order: NaCl = 0 g/L>NaCl = 30 g/L>NaCl = 15 g/L.

Runaway.Electron Suppression by.ECRH and RMP in KSTAR

We have observed reduction of the runaway.electron sy.chrotron radiation, hard X-ray.（HXR） intensity. and HXR energy.after apply.ng 110 GHz 2nd harmonic electron cy.lotron resonant heating （ECRH） during runaway.electron （RE） discharges at low density.with startup runaway.electrons. However, we did not see a significant reduction of X-ray. from 170 GHz 2nd harmonic ECRH at a higher field. A recently.installed IR TV camera was used to observe the forward cone of sy.chrotron radiation from high energy.REs in KSTAR. We have observed changes to the sy.chrotron images and reduction of the HXR by.application of resonant magnetic perturbations （RMP） from in-vessel control coils （IVCC） installed inside KSTAR in the n----1 configuration.

Sy.chronized off-harmonic probe laser with highly.variable pulse duration for laser-plasma interaction experiments

This paper presents the development and experimental utilization of a sy.chronized off-harmonic laser sy.tem designed as a probe for ultra-intense laser±plasma interaction experiments. The sy.tem exhibits a novel seed-generation design,allowing for a variable pulse duration spanning over more than three orders of magnitude, from 3.45 picoseconds to 10 nanoseconds. This makes it suitable for various plasma diagnostics and visualization techniques. In a side-view configuration, the laser was employ.d for interferometry.and streaked shadowgraphy.of a laser-induced plasma while successfully.suppressing the self-emission background of the laser±plasma interaction, resulting in a signal-to-self-emission ratio of 110 for this setup. These properties enable the probe to y.eld valuable insights into the plasma dy.amics and interactions at the PHELIX facility.and to be deploy.d at various laser facilities due to its easy.to-implement design.
