Diverse nucleotide substitutions in rice base editing mediated by novel TadA variants

CRISPR-mediated base editors have been widely used to correct defective alleles and create novel alleles by artificial evolution for the rapid genetic improvement of *** editing capabilities of base editors strictly rely on the performance of various nucleotide modification *** with the welldeveloped adenine base editors(ABEs),cytosine base editors(CBEs)and dual base editors suffer from unstable editing efficiency and patterns at different genomic loci in rice,significantly limiting their ***,we comprehensively examined the base editing activities of multiple evolved TadA8e variants in *** found that both TadA-CDd and TadA-E27R/N46l achieved more robust C-to-T editing than previously reported hyperactive hAID*D,and TadA-CDd outperformed TadA-E27R/N46l.A C-to-G base editor(CGBE)engineered with TadA-CDd and OsUNG performed highly efficient C-to-G editing in rice compared with that of *** addition,a dual base editor constructed with a single protein,TadDE,enabled simultaneous,highly efficient C-to-T and A-to-G editing in ***,our results demonstrate that TadA8e derivatives improve both CBEs and dual base editors in rice,providing a powerful way to induce diverse nucleotide substitutions for plant genome editing.

Efficient guiding and focusing of intense laser pulse using periodic thin slits

Slits have been widely used in laser-plasma interactions as plasma optical components for generating high-harmonic light and controlling laser-driven particle ***,we propose and demonstrate that periodic thin slits can be regarded as a new breed of optical elements for efficient focusing and guiding of intense laser *** fundamental physics of intense laser interaction with thin slits is studied,and it is revealed that relativistic effects can lead to enhanced laser focusing far beyond the pure diffractive focusing *** addition,the interaction of an intense laser pulse with periodic thin slits makes it feasible to achieve multifold enhancement in both laser intensity and energy transfer efficiency compared with conventional *** results provide a novel method for manipulating ultra-intense laser pulses and should be of interest for many laser-based applications.

GSW3.1,a novel gene controlling grain size and weight in rice

Grain size and weight are closely related traits determining yield in rice(Oryza sativa l.).Since indica and japonica rice varieties differ significantly in multiple traits,a high-generation recombinant inbred line(RIl)population derived from the crossing lH9(indica)and RPy(japonica)was used to map grainrelated traits in six *** of the quantitative trait loci(QTl)for thousand-grain weight showed that combinations of multiple QTl significantly increased the phenotypic effect.A novel gene named GSW3.1 controlling grain size and weight was discovered using the major QTl for the colocalization of grain width and thousand-grain weight on chromosome *** editing revealed that GSW3.1(lOC_Os03g16850)was pleiotropic,positively regulating grain size and weight while affecting several other agronomic *** analysis indicated that some traits,including grain width and weight,were highly correlated with indica-japonica differentiation.

Effect of initial ammonium concentration on a one-stage partial nitrification/anammox biofilm system:Nitrogen removal performance and the microbial community

One-stage partial nitrification coupled with anammox(PN/A)technology effectively reduces the energy consumption of a biological nitrogen removal *** nitrite-oxidizing bacteria(NOB)is essential for this technology to maintain efficient nitrogen removal *** ammonium concentration(IAC)affects the degree of inhibited *** this study,the effect of the IAC on a PN/A biofilm was investigated in a moving bed biofilm *** results showed that nitrogen removal efficiency decreased from 82.49%±1.90%to 64.57%±3.96%after the IAC was reduced from 60 to 20 mg N/l,while the nitrate production ratio increased from 13.87%±0.90%to 26.50%±3.76%.NOB activity increased to1,133.86 mg N/m^(2)/day after the IAC decreased,approximately 4-fold,indicating that the IAC plays an important inhibitory role in *** rate-limiting step in the mature biofilm of the PN/A system is the nitritation process and is not shifted by the *** analysis of the microbial community structure in the biofilm indicates that the IAC was the dominant factor in changes in community *** and *** were the main anammox bacteria,and Nitrosomonas and Nitrospira were the main AOB and NOB,*** did not affect the difference in growth between *** and ***,modulating the IAC promoted the PN/A process with efficient nitrogen removal performance at medium to low ammonium concentrations.

Turbo-shaft engine adaptive neural network control based on nonlinear state space equation

Intelligent Adaptive Control(AC) has remarkable advantages in the control system design of aero-engine which has strong nonlinearity and uncertainty. Inspired by the Nonlinear Autoregressive Moving Average(NARMA)-l2 adaptive control, a novel Nonlinear State Space Equation(NSSE) based Adaptive neural network Control(NSSE-AC) method is proposed for the turbo-shaft engine control system design. The proposed NSSE model is derived from a special neural network with an extra layer, and the rotor speed of the gas turbine is taken as the main state variable which makes the NSSE model be able to capture the system dynamic better than the NARMA-l2 model. A hybrid Recursive least-Square and levenberg-Marquardt(RlS-lM) algorithm is advanced to perform the online learning of the neural network, which further enhances both the accuracy of the NSSE model and the performance of the adaptive controller. The feedback correction is also utilized in the NSSE-AC system to eliminate the steady-state tracking error. Simulation results show that, compared with the NARMA-l2 model, the NSSE model of the turboshaft engine is more accurate. The maximum modeling error is decreased from 5.92% to 0.97%when the lM algorithm is introduced to optimize the neural network parameters. The NSSE-AC method can not only achieve a better main control loop performance than the traditional controller but also limit all the constraint parameters efficiently with quick and accurate switching responses even if component degradation exists. Thus, the effectiveness of the NSSE-AC method is validated.

Application of surfactant for improving leaching process of weathered crust elution-deposited rare earth ores

Weathered crust elution-deposited rare earth ores(WREOs)in China are the main source of medium and heavy rare earths in the *** order to improve the seepage and the mass transfer of traditional ammonium salt leaching process of WREOs,surfactants were added in the ammonium sulfate leaching solution and the ammonium chloride leaching *** leaching kinetics,the mass transfer process,and the adhesion work reduction factor calculated from the interfacial properties were studied to reveal the strengthening mechanism of *** the presence of the tested five surfactants,cetyl trimethyl ammonium bromide(CTAB),dodecyl trimethyl ammonium bromide(DTAB),sodium dodecyl sulfate(SDS),sodium oleate and oleic acid,the permeability of WREOs is improved,the rare earth(RE)leaching efficiency increase and the impurity aluminum(Al)leaching efficiency decrease,indicating its promotional effect on the leaching process of ***,CTAB shows a better leaching enhancement,and the optimal addition dosage is 0.4 g/l for the two ammonium salt leaching agent *** kinetics analysis shows the internal diffusion controls model of RE and Al leaching process,and the leaching kinetics equations of RE and Al related to CTAB concentration were obtained for the two ammonium salt leaching *** to the chromatographic plate theory,the mass transfer efficiency of RE increases with the CTAB concentration increasing until 0.4 g/l,which confirms that the optimal CTAB addition is 0.4 g/*** adhesion work and adhesion work reduction factor indicate more favorable permeation as well as the leaching process.

Certain plant species within the Apiales order accumulate triterpenoid saponins that feature a distinctive glucose-glucose-rhamnose(G-G-R)sugar chain attached at the C-28 position of the pentacyclic triterpene *** recently,the genomic basis underlying the biosynthesis and evolution of this sugar chain has remained *** this study,we identified two novel glycoside glycosyltransferases(GGTs)that can sequentially install the sugar chain’s second D-glucose and third l-rhamnose during the biosynthesis of asiaticoside and madecassoside,two representative G-G-R sugar chain-containing triterpenoid saponins produced by Centella *** assays revealed the remarkable substrate promiscuity of the two GGTs and the key residues crucial for sugar-donor selectivity of the glucosyltransferase and rhamnosyl*** further identified syntenic tandem gene duplicates of the two GGTs in the Apiaceae and Araliaceae families,suggesting a well-conserved genomic basis underlying sugar chain assembly that likely has evolved in the early ancestors of the Apiales ***,expression patterns of the two GGTs in pierced leaves of *** were found to be correlated with the production of asiaticoside and madecassoside,implying their involvement in host defense against herbivores and *** work sheds light on the biosynthesis and evolution of complex saponin sugars,paving the way for future engineering of diverse bioactive triterpenoids with unique glycoforms.

A refined representation method of the particle size distribution curve based on grading entropy

The particle size distribution(PSD)curve is an important expression of the basic properties of the *** characteristic parameters such as median particle size D_(50),effective particle size D_(10),and some combinations of characteristic parameters can only represent some points of the PSD curve,failing to represent all information of the PSD *** this paper,a fraction characteristic parameter F that could reflect the fraction size variation of the PSD curve was introduced based on grading *** a new presentation method of the A-B-F three parameters was proposed for a refined representation of the PSD *** was found that the newly constructed model not only better represents the differences in the width of PSD curves but also has a higher sensitivity than the A-B two-parameter model proposed by lő*** PSD curves with similar distributions,the new presentation method has a higher degree of discrimination than that of the D_(50)-C_(u)-C_(c) three-parameter or the four-parameter combination G_(c) proposed by ***,the application of this new method in describing the spatial nonuniformity distribution of depositions and predicting the soil hydraulic conductivity was *** research results can provide a reference for the refined representation of PSD curves.

Oat arabinoxylan promoted lactobacillus gasseri to alleviate metabolic disorders in high-fat diet-induced obese mice

Arabinoxylan (AX) has been found to improve an imbalanced gut microbiota. lactobacillus gasseri is a beneficial endogenous bacterium that has been shown to have several health benefits in the human gut， particularly its lipid-lowering activity. However， it is not known whether AX can promote the action of l. gasseri. The results of in vitro experiments showed that AX promoted biofilm formation in l. gasseri， its acid and bile salt resistance could be enhanced， and enabled better colonization of l. gasseri in the intestinal tract of mice. In vivo experiments showed that the AX+ l. gasseri group could effectively ameliorate weight gain and fat accumulation in high-fat diet-induced obese mice， and the l. gasseri group or AX+l. gasseri alleviated liver injury in mice. 16S rRNA sequencing showed that l. gasseri can colonize the mice intestine and AX+l. gasseri can ameliorate gut microbiota dysbiosis in obese mice by increasing lactobacillus spp. and Coriobacteriaceae＿UCG-002 and decreasing Peptococcaceae. In addition， metabolomics results indicated that the l. gasseri group and the AX+l. gasseri group could alleviate metabolic disorders by decreasing the levels of l-phenylalanine， l-tyrosine， kynurenine acid， and arachidonic acid in obese mice. The effect of AX+l. gasseri group was better than that of the l. gasseri group， suggesting that AX promotes the lipid-lowering activity of l. gasseri， and the mechanism may be due to the activation of retrograde endocannabinoid signaling. AX can be used as a functional food ingredient to potentially alleviate obesity and metabolic syndrome.
