Dear Editor,Primary mediastinal large B-cell lymphoma (PMBCL) is a rare subtype of aggressive B cell lymphoma, accounting for 2%to 4%of total non-Hodgkin’s lymphoma(NHL) cases (Savage, 2022). Genomic rearrangements (e.g., isochromosome9p) and copy-number alterations at9p24.1 loci, which contain the genes encoding programmed death-ligand 1/2(PDL1/2) and JAK2, lead to the overexpression of these genes in PMBCL,distinguishing PMBCL from diffuse large B-cell lymphoma (DLBCL)(Green et al.,2010). Recent thymic single-cell gene profiling has advanced our understanding of the presence and functions of B cells in the thymus (Afzali et al., 2024). However,the etiology of thymic B cell-derived PMBCL and its cellular microenvironment remain unknown. In this study, from a single-cell perspective, we compared the single-cell profiles of PMBCL with those of DLBCL and normal lymph nodes (NLNs),and discovered a unique phenotype of PMBCL-associated malignant plasma cells.

多囊卵巢综合征(PCOS)是育龄妇女常见的异质性内分泌疾病。PCOS患者心血管疾病、糖尿病、高尿酸血症、肥胖、血脂异常及其他代谢综合征的发生风险增加。胰岛素抵抗(IR)作为PCOS代谢紊乱中心环节,其改善对于PCOS治疗意义重大。近年发现钠-葡萄糖协同转运蛋白2(SGLT2)抑制剂是增加尿糖排泄的非胰岛素依赖性降血糖药,可改善PCOS患者IR与代谢,减轻患者体质量,保护多器官,降低PCOS心血管损伤、代谢综合征、血脂异常、肾病、肝脏代谢性疾病发生风险。该文就SGLT2抑制剂通过改善IR对PCOS患者代谢及心血管功能的作用及可能的机制进行综述。

目的:建立异体肾移植受者围术期静脉用卡泊芬净的群体药动学(population pharmacokinetics,PPK)模型,探讨并优化卡泊芬净用于治疗念珠菌属的剂量方案。方法:前瞻性收集常州市第一人民医院行同种异体肾移植术且术后静滴注射用醋酸卡泊芬净的51例受者共计134次血药浓度监测数据,考察的协变量包括年龄、性别、体质量(body weight,WT)、体质量指数,及谷丙转氨酶、谷草转氨酶、总胆红素、血浆白蛋白、血清肌酐、胱抑素C(serum cystatin C,Cys C)等临床指标,采用非线性混合效应模型程序(nonlinear mixed effect method,NONMEM)建立卡泊芬净的PPK模型,并采用自举法(Bootstrap)和正态化预测分布误差法(normal prediction distribution error,NPDE)进行模型验证。另采用Crystal Ball软件进行蒙特卡洛模拟,根据AUC_(0-24)/MIC计算不同剂量方案的达标概率(PTA),当PTA>90%时,判定为抗菌疗效良好。结果:基础模型采用一级消除的一室模型,WT和Cys C是影响卡泊芬净清除率(CL)的主要协变量,最终模型为CL(L·h^(–1))=0.411×(Cys C/3.17)~(–0.277)×(WT/57)~(0.519),分布容积(V)=10.7 L。Bootstrap法验证模型可靠稳定,且NPDE结果显示模型的预测能力较好。蒙特卡洛模拟显示,对于白色念珠菌(MIC_(90)=0.06 mg·L^(–1))和光滑念珠菌(MIC_(90)=0.125 mg·L^(–1)),各设定WT(40,60,80 kg)和设定Cys C(1,5,10 mg·L^(–1))下的不同剂量(50~150 mg·d^(–1))方案均可实现PTA>90%。对于近平滑念珠菌(MIC_(90)=1 mg·L^(–1)),所有剂量方案均不能达到药效学靶标。结论:在肾移植受者群体中初步建立了卡泊芬净的PPK模型,临床应用该模型结合患者的WT、肾功能状态,念珠菌种类和MIC值水平科学地制订个体化的给药方案,可达到改善临床结局、减少不良反应的目的。

Astragali radix(AR,the dried root of Astragalus)is a popular herbal remedy in both China and the United *** commercially available AR is commonly classified into premium graded(PG)and ungraded(UG)ones only according to the *** uncover novel sensitive and specific markers for AR grading,we took the integrated mass spectrometry-based untargeted and targeted metabolomics approaches to characterize chemical features of PG and UG samples in a discovery set(n=16 batches).A series of five differential compounds were screened out by univariate statistical analysis,including arginine,calycosin,ononin,formononetin,and astragalosideⅣ,most of which were observed to be accumulated in PG samples except for astragalosideⅣ.Then,we performed machine learning on the quantification data of five compounds and constructed a logistic regression prediction ***,the external validation in an independent validation set of AR(n=20 batches)verified that the five compounds,as well as the model,had strong capability to distinguish the two grades of AR,with the prediction accuracy>90%.Our findings present a panel of meaningful candidate markers that would significantly catalyze the innovation in AR grading.

This research systematically examined the degradation,antibacterial effects,and biocompatibility of micro-arc oxidation(MAO)coatings with nano CuO and ZnO on extruded Mg *** copper(Cu)and Zinc(Zn)possess antibacterial *** findings demonstrated that adding ZnO will appreciably reduce the degradation rate of MAO-coating alloy due to the self-sealing micro ***+MAO coating exhibited excellent antibacterial performance,with an antibacterial rate of over 90%within 6 h co-cultured with Staphylococcus ***,the antibacterial rate of ZnO+MAO coating reached 90%after 12 h *** test using MG63 cell indicated that the incorporation of CuO and ZnO did not notably affect the cell viability rate of the ***,after 14 days of culture,the CuO+MAO and ZnO+MAO coated samples exhibited higher alkaline phosphatase(ALP)activity than the MAO-coated and uncoated samples,suggesting favorable osteogenic properties.

Anderson acceleration(AA)is an extrapolation technique designed to speed up fixed-point *** optimization problems,we propose a novel algorithm by combining the AA with the energy adaptive gradient method(AEGD)[arXiv:2010.05109].The feasibility of our algorithm is ensured in light of the convergence theory for AEGD,though it is not a fixed-point *** provide rigorous convergence rates of AA for gradient descent(GD)by an acceleration factor of the gain at each implementation of *** experimental results show that the proposed AA-AEGD algorithm requires little tuning of hyperparameters and exhibits superior fast convergence.

Nitrate(NO_(3)^(-)),a nitrogen-containing pollutant,is prevalent in aqueous solutions,contributing to a range of environmental and health-related *** electrocatalytic reduction of NO_(3)^(-)holds promise as a sustainable approach to both eliminating NO_(3)^(-)and generating valuable ammonia(NH_(3)).Nevertheless,the reduction reaction of NO_(3)^(-)(NO_(3)^(-)RR),involving 8-electron transfer process,is intricate,necessitating highly efficient electrocatalysts to facilitate the conversion of NO_(3)^(-)to NH_(3).In this study,Fe-doped Co_(3)O_(4) nanowire strutted three-dimensional(3D)pinewood-derived carbon(Fe-Co_(3)O_(4)/PC)is proposed as a high-efficiency NO_(3)^(-)RR electrocatalyst for NH_(3) *** within 0.1 M NaOH containing NO_(3)^(-),Fe-Co_(3)O_(4)/PC demonstrates exceptional performance,obtain an impressively large NH_(3) yield of 0.55 mmol·h^(-1)·cm^(-2) and an exceptionally high Faradaic efficiency of 96.5%at-0.5 V,superior to its Co_(3)O_(4)/PC counterpart(0.2 mmol·h^(-1)·cm^(-2),73.3%).Furthermore,the study delves into the reaction mechanism of Fe-Co_(3)O_(4) for NO_(3)^(-)RR through theoretical calculations.

Reproduction and immune defense are costly functions,and they are expected to tradeoff with each other to drive *** brown planthopper(BPH),Nilaparvata lugens Stål(Hemiptera,Delphacidae),is a global superpest that mostly damages rice ***-like symbionts(YLS)exist in the abdominal fat body tissue and are tightly associated with the development,growth,and reproduction of *** previous research demonstrated that mating behavior promotes the release of YLS from the fat body into the hemolymph in the BPH,thereby triggering an immune ***,the fitness costs related to life-history traits of BPH(such as survival rate)have a strong dependence on the relative abundance of ***,the possible relationship between reproduction and the immune response in BPH has not been *** this study,an omics-based approach was used to analyze the transcriptome of fat body tissues in mated and unmated BPH at 72 h post-eclosion,from which two antimicrobial peptide genes,NlDefensin A(NlDfA)and NlDefensin B(NlDfB),were selected since they were highly expressed in mated ***,the full-length cDNA sequences of the NlDfA and NlDfB genes were cloned and *** results showed up-regulation of the NlDfA and NlDfB genes in mated BPH when compared to unmated ***-temporal expression analysis indicated that the NlDfA and NlDfB genes were expressed in all tissues and developmental stages,and they were most highly expressed in the fat body at 24 h ***,the symbionts in BPH were significantly inhibited by the in vitro expression of the NlDfA and NlDfB ***,RNA interference(RNAi)-mediated suppression of NlDfA and NlDfB dramatically increased the relative abundance of YLS in the fat body,while YLS in the hemolymph decreased *** BPHs also displayed some fitness disadvantages in survival,fecundity,hatchability,and possibly the vertical transmission of YLS from hemolymph to *** results

The development of highly efficient and durable oxygen evolution reaction(OER)catalysts for seawater electrolysis is of great importance for ***,an amorphous FeMoO_(4) nanorod array on Ni foam is reported as a highly active OER electrocatalyst in alkaline seawater,requiring only overpotentials of 303 and 332 mV to achieve 100 and 300 mA·cm^(-2),***,it shows strong long-term electrochemical durability for at least 50 h.
